1Mutagenic and cytotoxic potential of Endosulfan and Lambda-cyhalothrin—In vitro study describing individual and combined effects of pesticides显示文摘Excessive use of pesticides poses increased risks to non target species including humans. In the developing countries, lack of proper awareness about the toxic potential of pesticides makes the farmer more vulnerable to pesticide linked toxicities, which could lead to diverse pathological conditions. The toxic potential of a pesticide could be determined by their ability to induce genetic mutations and cytotoxicity. Hence, determination of genetic mutation and cytotoxicity of each pesticide is unavoidable to legislate health and safety appraisal about pesticides. The objective of current investigation was to determine the genotoxic and cytotoxic potential of Endosulfan(EN) and Lambda-cyhalothrin(LC); individually and in combination. 3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium Bromide(MTT) assay was utilized to determine cytotoxicity, while two mutant histidine dependent Salmonella strains(TA98, TA100) were used to determine the mutagenicity of EN and LC.Moreover, mutagenicity assay was conducted with and without S9 to evaluate the effects of metabolic activation on mutagenicity. Even though a dose dependent increase in the number of revertant colonies was detected with EN against both bacterial strains, a highly significant(p < 0.05) increase in the mutagenicity was detected in TA98 with S9. In comparison, data obtained from LC revealed less mutagenic potential than EN. Surprisingly,the non-mutagenic individual-concentrations of EN and LC showed dose dependent mutagenicity when combined. Combination of EN and LC synergistically induced mutagenicity both in TA98 and TA100. MTT assay spotlighted comparable dose dependent cytotoxicity effects of both pesticides. 2Group psychological intervention for maternal depression: A nested qualitative study from Karachi, Pakistan显示文摘AIM To understand the experience of maternal depression, the factors implicated in accessing health, and the acceptability of the psychosocial intervention. METHODS The participants were recruited from the paediatrics outpatient department of Civil Hospital Karachi, Pakistan. The study started in December 2009 and completed in December 2010. Women with maternal depression, aged 18-44 years with children aged 0-30 mo who had received nutritional supplements, and participated in the intervention programme [called Learning through Play(LTP) plus] were included in the study. Qualitative interviews were conducted with 8 participants before the intervention and 7 participants after the intervention. A semi structured topic guide was used to conduct the interviews.RESULTS Framework analysis procedures were used to analyse the qualitative data. Four themes emerged:(1) the women's contextual environment: Interpersonal conflicts, lack of social support and financial issues being the major barriers in assessing healthcare;(2) women's isolation and powerlessness within the environment: Sense of loneliness was identified as a restricting factor to access healthcare;(3) the impact of the intervention(LTP-Plus): Women felt 'listened to' and seemed empowered; and(4) empowered transformed women within the same contextual environment: The facilitator provided a 'gardening role' in nurturing the women resulting in a positive transformation within the same environment. 3Factors associated with cervical cancer knowledge and practice among Bahraini women显示文摘The study was conducted to assess the women’s knowledge and attitude towards cervical cancer and cervical cancer screening,to support the health education programs in this field.A total sample of 350 women(n = 350 ),70 women from each health center was chosen based on a multistage sampling technique according to a number of 21 health centers that serve the population.One health center from each government was selected based on the population it serves,thus the health center with the highest population number was chosen.The main tool of data collection was a face to face interview questionnaire.When assessing the knowledge,attitude, and practice(KAP) of our sample,we observed that approximately equal percentages(70%) of women have heard of both Pap smear and cervical cancer.Only 47.4%had ever had Pap smear and of those 35.1% had it during a period of 5 years or less.The main source of knowledge for those who have done the procedure is the family physician(30.8%).When asked about the reason behind having a Pap smear,the majority (62.3%) had it during postnatal visits.Nevertheless,those that didnt have it,almost half(46.6%) disapproved to do so because they are healthy even after being told that Pap smear is a painless significant procedure. 5Environmental Impacts of the Existing Solid Waste Management System of Northern Lahore显示文摘With the substantial increase in solid waste due to industrialization and urbanization,the environmental damage has also aggravated,making the management of solid waste an important issue throughout the world.Global warming,species extinction,imbalance in nutrient cycle and random disposal of hazardous waste are some environmental problems threatening sustainable development.The solid waste from the study area mainly consists of organic waste(66%),recyclables(25%) and miscellaneous waste(9%).About 10% of the organic waste is composted by public facilities funded by the government;whereas the entire miscellaneous waste is dumped at dumping sites without going through any treatment.About 41% of the recyclables are sold to junk shops by households,and 28% are sorted out by scavengers at dumping sites.An EASEWASTE model is used to evaluate the impacts of existing solid waste management system on environment.The major gases which contribute to life cycle impact assessment are carbon dioxide(CO;),methane(CH4),nitrous oxide(N;O),CFCs(CFC-11,CFC-12,CFC-113,CFC-114 and CFC-115),HCFCs(HCFC-22,HCFC-123,HCFC-124 and HCFC-141 b),HFCs(HFC-125,HFC-134 a and HFC-152 a),halons,tetra chloromethane(CCl;),1,1,1-trichloroethane(CCl;CH;) and carbon monoxide(CO),and global warming potential is calculated by the EASEWASTE model at different timelines,i.e.20,100 and 500 years,respectively.Human toxicity via water and air is also evaluated and it found that the existing system is polluting the environment in many ways.Syeda Amber FATIMA Mohammad Nawaz CHAUDHRY Syeda Adila BATOOL 2019Chinese Journal of Urban and Environmental Studies2019,7,3:0
6Genomic Epidemiology of SARS-CoV-2 in Pakistan显示文摘COVID-19 has swept globally and Pakistan is no exception.To investigate the initial introductions and transmissions of the SARS-CoV-2 in Pakistan,we performed the largest genomic epidemiology study of COVID-19 in Pakistan and generated 150 complete SARS-CoV-2 genome sequences from samples collected from March 16 to June 1,2020.We identified a total of 347 mutated positions,31 of which were over-represented in Pakistan.Meanwhile,we found over 1000 intra-host single-nucleotide variants(iSNVs).Several of them occurred concurrently,indicating possible interactions among them or coevolution.Some of the high-frequency iSNVs in Pakistan were not observed in the global population,suggesting strong purifying selections.The genomic epidemiology revealed five distinctive spreading clusters.The largest cluster consisted of 74 viruses which were derived from different geographic locations of Pakistan and formed a deep hierarchical structure,indicating an extensive and persistent nation-wide transmission of the virus that was probably attributed to a signature mutation(G8371T in ORF1ab)of this cluster.Furthermore,28 putative international introductions were identified,several of which are consistent with the epidemiological investigations.In all,this study has inferred the possible pathways of introductions and transmissions of SARS-CoV-2 in Pakistan,which could aid ongoing and future viral surveillance and COVID-19 control.Shuhui Song Cuiping Li Lu Kang Dongmei Tian Nazish Badar Wentai Ma Shilei Zhao Xuan Jiang Chun Wang Yongqiao Sun Wenjie Li Meng Lei Shuangli Li Qiuhui Qi Aamer Ikram Muhammad Salman Massab Umair Huma Shireen Fatima Batool Bing Zhang Hua Chen Yun-Gui Yang Amir Ali Abbasi Mingkun Li Yongbiao Xue Yiming Bao 2021Genomics, Proteomics & Bioinformatics2021,19,5:0
7Identification,Isolation and Characterization of GaCyPI Gene in Gossypium arboreum under Cotton Leaf Curl Virus Disease Stress显示文摘Pakistan is facing the threat of Cotton Leaf Curl Virus(CLCuV)which is transmitted through whitefly to cotton crop.Molecular mechanism of leaf epicuticular wax protects the plants from different pathogens including insect attack and disease transmission.Objective of current study is the isolation and characterization of a wax related gene GaCyPI from Gossypium arboreum under CLCuV infection.A fragment of 475 bp was isolated from the total RNA and 3’and 5’RACE-PCR products were arranged by overlapping the extended sequences at both the ends.Deduced protein sequence of GaCyPI showed homology with Cyclophilin cis-trans isomerase gene of Gossypium ramondii and Gossypium barbadanse.Multiple sequence alignment also revealed homology among the coding sequences of same gene.GaCyPI protein comprised of 173 amino acids and ORF finder revealed the 69 bases upstream at 5’while 350 bp at 3’UTR.InterProScan revealed that it belongs to Cyclophilin-type peptidyl prolyl cis/trans isomerase(PPIase)family.Active sites are visible at specific amino acid positions and 3D structure was stable in Ramachandran plot.Prosa server showed protein residues have average 3D-1D score>=0.2 and Z-Score was−6.74.Phylogenetic analysis revealed that G.raimondii is the closest species that shares the same sequence.Hence,GaCyPI has strong role in plants’epicuticular wax and its genetic transformation may protect the cotton from whitefly which transmits CLCuV.Zunaira Sher Muhammad Umair Majid Sameera Hassan Fatima Batool Beenish Aftab Bushra Rashid 2021Phyton-International Journal of Experimental Botany2021,90,6:0
8Umbilical cord-derived mesenchymal stem cells preconditioned with isorhamnetin:potential therapy for burn wounds显示文摘BACKGROUND Impaired wound healing can be associated with different pathological states.Burn wounds are the most common and detrimental injuries and remain a major health issue worldwide.Mesenchymal stem cells(MSCs)possess the ability to regenerate tissues by secreting factors involved in promoting cell migration,proliferation and differentiation,while suppressing immune reactions.Preconditioning of MSCs with small molecules having cytoprotective properties can enhance the potential of these cells for their use in cell-based therapeutics.AIM To enhance the therapeutic potential of MSCs by preconditioning them with isorhamnetin for second degree burn wounds in rats.METHODS Human umbilical cord MSCs(hU-MSCs)were isolated and characterized by surface markers,CD105,vimentin and CD90.For preconditioning,hU-MSCs were treated with isorhamnetin after selection of the optimized concentration(5μmol/L)by cytotoxicity analysis.The migration potential of these MSCs was analyzed by the in vitro scratch assay.The healing potential of normal,and preconditioned hU-MSCs was compared by transplanting these MSCs in a rat model of a second degree burn wound.Normal,and preconditioned MSCs(IH+MSCs)were transplanted after 72 h of burn injury and observed for 2 wk.Histological and gene expression analyses were performed on day 7 and 14 after cell transplantation to determine complete wound healing.RESULTS The scratch assay analysis showed a significant reduction in the scratch area in the case of IH+MSCs compared to the normal untreated MSCs at 24 h,while complete closure of the scratch area was observed at 48 h.Histological analysis showed reduced inflammation,completely remodeled epidermis and dermis without scar formation and regeneration of hair follicles in the group that received IH+MSCs.Gene expression analysis was time dependent and more pronounced in the case of IH+MSCs.Interleukin(IL)-1β,IL-6 and Bcl-2 associated X genes showed significant downregulation,while transforming growth factorβ,vascular endothelial growth factor,Bcl-2 and matrix metallopeptidase 9 showed significant upregulation compared to the burn wound,showing increased angiogenesis and reduced inflammation and apoptosis.CONCLUSION Preconditioning of hU-MSCs with isorhamnetin decreases wound progression by reducing inflammation,and improving tissue architecture and wound healing.The study outcome is expected to lead to an improved cell-based therapeutic approach for burn wounds.Shazmeen Aslam Irfan Khan Fatima Jameel Midhat Batool Zaidi Asmat Salim 2020World Journal of Stem Cells2020,12,12:0
