| 2 | Probiotic effects on intestinal fermentation patterns in patients with irritable bowel syndrome显示文摘AIM: To determine whether Lactobacillus casei strain Shirota (Yakult ) can alter small intestinal bacterial overgrowth (SIBO), as tested by the lactulose breath test, and whether this is associated with changes in symptoms in irritable bowel syndrome (IBS). METHODS: 18 patients with IBS (Rome Ⅱ criteria), who showed an early rise in breath hydrogen with lactulose (ERBHAL), consumed 65 mL of Yakult daily for 6 wk. Lactulose breath test was repeated at the end of the treatment period. Symptoms were recorded daily using a 10 cm visual analogue scale. RESULTS: 14 patients completed the study, 9 (64%) had reversal of ERBHAL, with the median time of f irst rise in breath hydrogen increasing from 45 to 75 min (P = 0.03). There was no signifi cant improvement in the symptom score with probiotic therapy, except for wind (P=0.04). Patients commencing with at least moderate symptoms and who no longer had ERBHAL at the end of treatment, showed improvement in the overall symptoms scores [median fi nal score 5.3 (IQR3.9-5.9), 55% reduction; n=6] to a greater extent than those who had had persisting ERBHAL [final score 6.9 (5.0-7.0), 12% reduction; n = 5; P = 0.18]. CONCLUSION: Yakult is effective in altering fermentation patterns in the small bowel, consistent with reducing SIBO. The loss of ERBHAL was associated with reduced symptoms. | 3 | Satellite remote sensing of grasslands:from observation to management显示文摘Aims Grasslands are the world’s most extensive terrestrial ecosystem,and are a major feed source for livestock.Meeting increasing demand for meat and other dairy products in a sustainable manner is a big challenge.At a field scale,Global Positioning System and ground-based sensor technologies provide promising tools for grassland and herd management with high precision.With the growth in availability of spaceborne remote sensing data,it is therefore important to revisit the relevant methods and applications that can exploit this imagery.In this article,we have reviewed the(i)current status of grassland monitoring/observation methods and applications based on satellite remote sensing data,(ii)the technological and methodological developments to retrieve different grassland biophysical parameters and management characteristics(i.e.degradation,grazing intensity)and(iii)identified the key remaining challenges and some new upcoming trends for future development.Important Findings The retrieval of grassland biophysical parameters have evolved in recent years from classical regression analysis to more complex,efficient and robust modeling approaches,driven by satellite data,and are likely to continue to be the most robust method for deriving grassland information,however these require more high quality calibration and validation data.We found that the hypertemporal satellite data are widely used for time series generation,and particularly to overcome cloud contamination issues,but the current low spatial resolution of these instruments precludes their use for field-scale application in many countries.This trend may change with the current rise in launch of satellite constellations,such as RapidEye,Sentinel-2 and even the microsatellites such as those operated by Skybox Imaging.Microwave imagery has not been widely used for grassland applications,and a better understanding of the backscatter behaviour from different phenological stages is needed for more reliable products in cloudy regions.The development of hyperspectral satellite instrumentation and analytical methods will help for more detailed discrimination of habitat types,and the development of tools for greater end-user operation. | Iftikhar Ali Fiona Cawkwell Edward Dwyer Brian Barrett Stuart Green | 2016 | Journal of Plant Ecology2016,9,6: | 15 |

| 5 | Specific shRNA targeting of FAK influenced collagen metabolism in rat hepatic stellate cells显示文摘AIM:To investigate the effects and mechanism of disruption of focal adhesion kinase(FAK) expression on collagen metabolism in rat hepatic stellate cells(HSC).METHODS:The plasmids expressing FAK short hairpin RNA(shRNA) were transfected into HSC-T6 cells,and the level of FAK expression was determined by both real-time quantitative polymerase chain reaction(QPCR) and Western blotting analysis.The production of type collagen and type collagen in FAK-disrupted cells was analyzed by real-time Q-PCR.The level of collagen metabolism proteins,including matrix metalloproteinases-13(MMP-13) and tissue inhibitors of metalloproteinases-1(TIMP-1) was also determined by both real-time Q-PCR and Western blotting analysis.RESULTS:The transfection of FAK shRNA plasmids into HSC resulted in disrupted FAK expression.Compared with the HK group,the levels of type collagen and type collagen mRNA transcripts in FAK shRNA plas-mid group were signif icantly decreased(0.69 ± 0.03 vs 1.96 ± 0.15,P = 0.000;0.59 ± 0.07 vs 1.62 ± 0.12,P = 0.020).The production of TIMP-1 in this cell type was also signif icantly reduced at both mRNA and protein levels(0.49 ± 0.02 vs 1.72 ± 0.10,P = 0.005;0.76 ± 0.08 vs 2.31 ± 0.24,P = 0.000).However,the expression of MMP-13 mRNA could be significantly up-regulated by the transfection of FAK shRNA plasmids into HSC(1.74 ± 0.20 vs 1.09 ± 0.09,P = 0.000).CONCLUSION:These data support the hypothesis that shRNA-mediated disruption of FAK expression could attenuate extracellular matrix(ECM) synthesis and promote ECM degradation,making FAK a potential target for novel anti-f ibrosis therapies. | Zhi-Na Dun Xiao-Lan Zhang Jun-Yan An Li-Bo Zheng Robert Barrett Shu-Rui Xie | 2010 | World Journal of Gastroenterology2010,16,32: | 8 |
| 6 | Adenosquamous carcinoma of the pancreas: Molecular characterization of 23 patients along with a literature review显示文摘Adenosquamous carcinoma of the pancreas(ASCP)is a rare entity. Like adenocarcinoma of the pancreas,overall survival is poor. Characteristics of ASCP include central tumor necrosis, along with osteoclasts and hypercalcemia. Various theories exist as to why this histological subtype exists, as normal pancreas tissue has no benign squamous epithelium. Due to the rarity of this disease, limited molecular analysis has been performed, and those reports indicate unique molecular features of ASCP. In this paper, we characterize 23 patients diagnosed with ASCP through molecular profiling using immunohistochemistry staining, fluorescent in situ hybridization, chromogenic in situ hybridization, and gene sequencing, Additionally, we provide a comprehensive literature review of what is known to date of ASCP.Molecular characterization revealed overexpression in MRP1(80%), MGMT(79%), TOP2A(75), RRM1(42%),TOPO1(42%), PTEN(45%), CMET(40%), and C-KIT(10%) among others. One hundred percent of samples tested were positive for KRAS mutations. This analysis shows heretofore unsuspected leads to be considered for treatments of this rare type of exocrine pancreas cancer. Molecular profiling may be appropriate to provide maximum information regarding the patient's tumor. | 9 | Malaria: Global progress 2000-2015 and future challenges显示文摘Background:2015 was the target year for malaria goals set by the World Health Assembly and other international institutions to reduce malaria incidence and mortality.A review of progress indicates that malaria programme financing and coverage have been transformed since the beginning of the millennium,and have contributed to substantial reductions in the burden of disease.Findings:Investments in malaria programmes increased by more than 2.5 times between 2005 and 2014 from US$960 million to US$2.5 billion,allowing an expansion in malaria prevention,diagnostic testing and treatment programmes.In 2015 more than half of the population of sub-Saharan Africa slept under insecticide-treated mosquito nets,compared to just 2%in 2000.Increased availability of rapid diagnostic tests and antimalarial medicines has allowed many more people to access timely and appropriate treatment.Malaria incidence rates have decreased by 37%globally and mortality rates by 60%since 2000.It is estimated that 70%of the reductions in numbers of cases in sub-Saharan Africa can be attributed to malaria interventions.Conclusions:Reductions in malaria incidence and mortality rates have been made in every WHO region and almost every country.However,decreases in malaria case incidence and mortality rates were slowest in countries that had the largest numbers of malaria cases and deaths in 2000;reductions in incidence need to be greatly accelerated in these countries to achieve future malaria targets.Progress is made challenging because malaria is concentrated in countries and areas with the least resourced health systems and the least ability to pay for system improvements.Malaria interventions are nevertheless highly cost-effective and have not only led to significant reductions in the incidence of the disease but are estimated to have saved about US$900 million in malaria case management costs to public providers in sub-Saharan Africa between 2000 and 2014.Investments in malaria programmes can not only reduce malaria morbidity and mortality,thereby contributing to the health targets of the Sustainable Development Goals,but they can also transform the well-being and livelihood of some of the poorest communities across the globe. | Richard E.Cibulskis Pedro Alonso John Aponte Maru Aregawi Amy Barrette Laurent Bergeron Cristin A.Fergus Tessa Knox Michael Lynch Edith Patouillard Silvia Schwarte Saira Stewart Ryan Williams | 2016 | Infectious Diseases of Poverty2016,5,1: | 4 |
