| 4 | Schaftoside inhibits 3CL^(pro)and PL^(pro)of SARS-CoV-2 virus and regulates immune response and inflammation of host cells for the treatment of COVID-19显示文摘It is an urgent demand worldwide to control the coronavirus disease 2019(COVID-19)pandemic caused by the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)virus.The 3-chymotrypsin-like protease(3CL^(pro))and papain-like protease(PL^(pro))are key targets to discover SARS-CoV-2 inhibitors.After screening 12 Chinese herbal medicines and 125 compounds from licorice,we found that a popular natural product schaftoside inhibited 3CL^(pro)and PL^(pro)with IC_(50)values of 1.73±0.22 and 3.91±0.19μmol/L,respectively,and inhibited SARS CoV-2 virus in Vero E6 cells with EC_(50)of 11.83±3. 23μmol/L. Hydrogen-deuterium exchange mass spectrometry analysis, quantum mechanics/molecular mechanics calculations, together with site-directed mutagenesis indicated the antiviral activities of schaftoside were related with non-covalent interactions with H41, G143 and R188 of3CLpro, and K157, E167 and A246 of PLpro. Moreover, proteomics analysis and cytokine assay revealed that schaftoside also regulated immune response and inflammation of the host cells. The antiinflammatory activities of schaftoside were confirmed on lipopolysaccharide-induced acute lung injury mice. | 5 | Induced Pib Expression and Resistance to Magnaporthe grisea are Compromised by Cytosine Demethylation at Critical Promoter Regions in Rice显示文摘Pib is a well-characterized rice blast-resistance gene belonging to the nucleotide binding site (NBS) and leucine-rich repeat (LRR) superfamily.Expression of Pib was low under non-challenged conditions,but strongly induced by the blast-causing fungal pathogen Magnaporthe grisea,thereby conferring resistance to the pathogen.It is generally established that cytosine methylation of the promoter-region often plays a repressive role in modulating expression of the gene in question.We report here that two critical regions of the Pib promoter were heavily CG cytosine-methylated in both cultivars studied.Surprisingly,induced expression of Pib by M.grisea infection did not entail its promoter demethylation,and partial demethylation by 5-azacytidine-treatment actually reduced Pib expression relative to wildtype plants.Accordingly,the blast disease-resistance was compromised in the 5-azaC-treated plants relative to wild-type.In contrast,the disease susceptibility was not affected by the 5-azaC treatment in another two rice cultivars that did not contain the Pib gene,ruling out effects of other R genes and non-specific genotoxic effects by the drug-treatment as a cause for the compromised Pib-conditioned blast-resistance.Taken together,our results suggest that promoter DNA methylation plays a novel enhancing role in conditioning high-level of induced expression of the Pib gene in times of M.grisea infection,and its conferred resistance to the pathogen. | Yuan Li Qiong Xia Hongping Kou Dan Wang Xiuyun Lin Ying Wu Chunming Xu Shaochen Xing Bao Liu | 2011 | Journal of Integrative Plant Biology2011,53,10: | 0 |

| 7 | Distribution, evolution and structural properties of Wushenqi paleo-uplift in Ordos Basin, NW China显示文摘This paper depicts the distribution of the Wushenqi paleo-uplift in the Ordos Basin by using the latest drilling and seismic data, and analyzes the tectonic evolution of the paleo-uplift with the support of Bischke curve and balanced section. The compressional Wushenqi paleo-uplift which developed in the Early Caledonian orogeny(Huaiyuan orogeny) is approximately a ellipse extending in S-N direction. Its long axis is about 194 km and short axis is about 55-94 km in nearly W-E direction. The denudation thickness and area of the Cambrian in the core are 170-196 m and 11 298 km^(2), respectively. It was mainly formed during the Huaiyuan orogeny according to the chronostratigraphic framework. It was in the embryonic stage in the Middle-Late Cambrian, denuded after developed obviously at the end of Late Cambrian. The paleo-uplift of the 3rd member of the Ordovician Majiagou Formation was reactivated and reduced in area. In the sedimentary period of the Ma 4 Member-pre-Carboniferous, the paleo-uplift experienced non-uniform uplift and denudation. It entered the stable period of burial and preservation in the Carboniferous and later period. The Wushenqi paleo-uplift was formed on the weak area of the basement and tectonic belts, into an compressional structure with irregular morphology, under the control of the non-coaxial compression in the south and north and the stress transmitted by the uplift in the basin.
