| 1 | Role of PERK/eIF2α/CHOP Endoplasmic Reticulum Stress Pathway in Oxidized Low-density Lipoprotein Mediated Induction of Endothelial Apoptosis显示文摘Objective PERK/eI F2α/CHOP is a major signaling pathway mediating endoplasmic reticulum(ER) stress related with atherosclerosis.Oxidized LDL(ox-LDL) also induces endothelial apoptosis and plays a vital role in the initiation and progression of atherosclerosis.The present study was conducted to explore the regulatory effect of ox-LDL on PERK/e IF2α/CHOP signaling pathway in vascular endothelial cells.Methods The effects of ox-LDL on PERK and p-e IF2α protein expression of primary human umbilical vein endothelial cells(HUVECs) were investigated by Western blot analysis.PERK gene silencing and selective eI F2α phosphatase inhibitor,salubrinal were used to inhibit the process of ox-LDL induced endothelial cell apoptosis,caspase-3 activity,and CHOP mR NA level.Results Ox-LDL treatment significantly increased the expression of PERK,PERK-mediated inactivation of e IF2α phosphorylation,and the expression of CHOP,as well as the caspase-3 activity and apoptosis.The effects of ox-LDL were markedly decreased by knocking down PERK with stable transduction of lentiviral sh RNA or by selective eI F2α phosphatase inhibitor,salubrinal.Conclusion This study provides the first evidence that ox-LDL induces apoptosis in vascular endothelial cells mediated largely via the PERK/eI F2α/CHOP ER-stress pathway.It adds new insights into the molecular mechanisms underlying the pathogenesis and progression of atherosclerosis. | TAO Yong Kang YU Pu Lin BAI Yong Ping YAN Sheng Tao ZHAO Shui Ping ZHANG Guo Qiang | 2016 | Biomedical and Environmental Sciences2016,29,12: | 21 |

| 4 | Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. | 8 | Establishment and validation of a computer-assisted colonic polyp localization system based on deep learning显示文摘BACKGROUND Artificial intelligence in colonoscopy is an emerging field,and its application may help colonoscopists improve inspection quality and reduce the rate of missed polyps and adenomas.Several deep learning-based computer-assisted detection(CADe)techniques were established from small single-center datasets,and unrepresentative learning materials might confine their application and generalization in wide practice.Although CADes have been reported to identify polyps in colonoscopic images and videos in real time,their diagnostic performance deserves to be further validated in clinical practice.AIM To train and test a CADe based on multicenter high-quality images of polyps and preliminarily validate it in clinical colonoscopies.METHODS With high-quality screening and labeling from 55 qualified colonoscopists,a dataset consisting of over 71000 images from 20 centers was used to train and test a deep learning-based CADe.In addition,the real-time diagnostic performance of CADe was tested frame by frame in 47 unaltered full-ranged videos that contained 86 histologically confirmed polyps.Finally,we conducted a selfcontrolled observational study to validate the diagnostic performance of CADe in real-world colonoscopy with the main outcome measure of polyps per colonoscopy in Changhai Hospital.RESULTS The CADe was able to identify polyps in the test dataset with 95.0%sensitivity and 99.1%specificity.For colonoscopy videos,all 86 polyps were detected with 92.2%sensitivity and 93.6%specificity in frame-by-frame analysis.In the prospective validation,the sensitivity of CAD in identifying polyps was 98.4%(185/188).Folds,reflections of light and fecal fluid were the main causes of false positives in both the test dataset and clinical colonoscopies.Colonoscopists can detect more polyps(0.90 vs 0.82,P<0.001)and adenomas(0.32 vs 0.30,P=0.045)with the aid of CADe,particularly polyps<5 mm and flat polyps(0.65 vs 0.57,P<0.001;0.74 vs 0.67,P=0.001,respectively).However,high efficacy is not realized in colonoscopies with inadequate bowel preparation and withdrawal time(P=0.32;P=0.16,respectively).CONCLUSION CADe is feasible in the clinical setting and might help endoscopists detect more polyps and adenomas,and further confirmation is warranted. | Sheng-Bing Zhao Wei Yang Shu-Ling Wang Peng Pan Run-Dong Wang Xin Chang Zhong-Qian Sun Xing-Hui Fu Hong Shang Jian-Rong Wu Li-Zhu Chen Jia Chang Pu Song Ying-Lei Miao Shui-Xiang He Lin Miao Hui-Qing Jiang Wen Wang Xia Yang Yuan-Hang Dong Han Lin Yan Chen Jie Gao Qian-Qian Meng Zhen-Dong Jin Zhao-Shen Li Yu Bai | 2021 | World Journal of Gastroenterology2021,27,31: | 5 |
| 9 | New insights on the enhanced non-hydroxyl radical contribution under copper promoted TiO_(2)/GO for the photodegradation of tetracycline hydrochloride显示文摘TiO_(2)/graphene oxide(GO)as photocatalyst in the photo-degradation of multitudinous pollutants has been extensively studied.But its low photocatalytic efficiency is attributed to the high band gap energy which lead to low light utilization.Cu-TiO_(2)/GO was synthesized via the impregnation methods to enhance the catalytic performance.The Cu-TiO_(2)/GO reaction rate constant for photo-degradation of pollutants(tetracycline hydrochloride,TC)was about 1.4 times that of TiO_(2)/GO.In 90 min,the removal ratio of Cu-TiO_(2)/GO for TC was 98%,and the maximum degradation ratio occurred at p H 5.After five cycles,the removal ratio of Cu-Ti O_(2)/GO still exceeded 98%.UV-visible adsorption spectra of Cu-Ti O_(2)/GO showed that its band gap was narrower than TiO_(2)/GO.Electron paramagnetic resonance(EPR)spectra test illustrated the generation rate of·O_(2)^(-)and·OH was higher in Cu-TiO_(2)/GO system than TiO_(2)/GO and TiO_(2) system.The contribution sequence of oxidative species was·O_(2)^(-)>holes(h+)>·OH in both TiO_(2)/GO and Cu-Ti O_(2)/GO system.Interestingly,the contribution of·OH in Cu-TiO_(2)/GO was less than that in TiO_(2)/GO during the photo-degradation process.This phenomenon was attributed to the better adsorption performance of Cu-Ti O_(2)/GO which could reduce the accessibility of TC to·OH in liquid.The enhanced non-hydroxyl radical contribution could be attributed to that the more other active species or sites on(nearby)the surface of Cu-TiO_(2)/GO generated after doping Cu.These results provide a new perspective for the tradition metal-doped conventional catalysts to enhance the removal of organic pollutants in the environment. | Sijia Qian Shengyan Pu Ying Zhang Peng Wang Yingchen Bai Bo Lai | 2021 | Journal of Environmental Sciences2021,33,2: | 4 |

Methods A cross-sectional study was conducted in 472 elderly in the Longtan community of Dongcheng district, Beijing in 2009. Data regarding the incidence of fall and recurrent falls in the previous year, as well as associated factors were collected from the elderly through face-to-face interviews. Results The incidence of falls and recurrent falls was 17.8% and 6.1%, respectively, and it increased with age(χ2for trend=21.06, 19.20, P=0.001, 0.002). Binary logistic stepwise regression analysis showed that age(OR=2.20), living alone(OR=4.67) and gait disturbance(OR=1.27) were risk factors, while housing with elevators(OR=0.35), appropriate width/height of stair steps(OR=0.78), sufficient lighting for stairway(OR=0.45) and regular exercise(OR=0.12) could lower the risk for single fall; factors such as low monthly family income(OR=1.39), poor vision(OR=1.83), low physical ability(OR=4.47), abnormal static balance(OR=2.48), and fear of falls(OR=2.23) were risk factors, while appropriate width/height of stair steps(OR=0.49) and easiness of access to daily supplies(OR=0.41) were protective factors for recurrent falls. | 20 | Metal Exposure and Risk of Diabetes and Prediabetes among Chinese Occupational Workers显示文摘Objective To study the association between metal exposure and risk of diabetes and prediabetes among Chinese workers exposed to metals.Methods We used data obtained from the baseline survey of the Jinchang Cohort Study of workers in Jinchang Industry,the largest nickel production company in China.A total of 42,122 workers ≥20 years of age were included in the study.A standardized,structured questionnaire was used to collect epidemiological information.Physical examinations and laboratory tests were conducted to evaluate the health status of the participants and to measure various biomarkers including blood sugar,lipids,and urinary metal concentrations.Logistic regression was used to study the association between occupational groups categorized according to the measured metal levels(office workers,low-level;mining/production workers,mid-level;and smelting/refining workers,high-level)and risk of diabetes and prediabetes.Results The overall prevalence of diabetes and prediabetes was 7.5% and 16.8%,respectively.The adjusted odds ratios for diabetes among mining/production workers and smelting/refining workers compared to office workers were 1.5(95% CI:1.3,1.7)and 3.8(95% CI:3.4,4.3),respectively.No association was observed between these occupational groups and prediabetes in this study.Conclusion Occupations associated with higher levels of metal exposure were associated with an increased risk of diabetes in this cohort.More studies are needed to confirm this observed association. | YANG Ai Min CHENG Ning PU Hong Quan LIU Si Min LI Juan Sheng BASSIG Bryan A.
