| 1 | Constitutive STAT3 phosphorylation and IL-6/IL-10 co-expression are associated with impaired T-cell function in tuberculosis patients显示文摘T-cells critically contribute to protection against Mycobacterium tuberculosis infection,and impaired T-cell responses can lead to disease progression.Pro-inflammatory and immunosuppressive cytokines affect T-cells,and fine-tuned regulation of cytokine signaling via the Jak/STAT signaling pathways is crucial for appropriate T-cell function.Constitutive STAT3 phosphorylation as a consequence of aberrant cytokine signaling has been described to occur in pathognomonic T-cell responses in inflammatory and autoimmune diseases.We characterized blood samples from tuberculosis patients(n=28)and healthy contacts(n=28)from Ghana for M.tuberculosis-specific T-cell responses,constitutive cytokine production,and SOCS3 and pSTAT3 expression.Lentiviral modulation of primary CD4+T-cells was performed to determine the effects of SOCS3 on T-cell functions.T-cells from tuberculosis patients expressed higher levels of IL-10 and IL-6 and lower levels of T helper type(TH)17 cytokines after M.tuberculosis-specific stimulation compared to healthy contacts.In addition,tuberculosis patients had higher IL-10 and IL-6 levels in the supernatants of non-stimulated immune cells and plasma samples compared to healthy contacts.Notably,aberrant cytokine expression was accompanied by high constitutive pSTAT3 levels and SOCS3 expression in T-cells.Multivariate analysis identified an IL-6/IL-10 co-expression-based principal component in tuberculosis patients that correlated with high pSTAT3 levels.SOCS3 contributed to a regulatory component,and tuberculosis patients with high SOCS3 expression showed decreased TH1 cytokine expression and impaired IL-2-induced STAT5 phosphorylation.SOCS3 over-expression in primary CD4+T-cells confirmed the SOCS3 inhibitory function on IL-2-induced STAT5 phosphorylation.We conclude that constitutive pSTAT3 and high SOCS3 expression are influential factors that indicate impaired T-cell functions in tuberculosis patients. | Kirstin Harling Ernest Adankwah Alptekin Güler Anthony Afum-Adjei Awuah Louis Adu-Amoah Ertan Mayatepek Ellis Owusu-Dabo Norman Nausch Marc Jacobsen | 2019 | Cellular & Molecular Immunology2019,16,3: | 15 |
| 2 | A review of the effects of forest fire on soil properties显示文摘Forest fires are key ecosystem modifiers affecting the biological,chemical,and physical attributes of forest soils.The extent of soil disturbance by fire is largely dependent on fire intensity,duration and recurrence,fuel load,and soil characteristics.The impact on soil properties is intricate,yielding different results based on these factors.This paper reviews research investigating the effects of wildfire and prescribed fire on the biological and physico-chemical attributes of forest soils and provides a summary of current knowledge associated with the benefits and disadvantages of such fires.Low-intensity fires with ash deposition on soil surfaces cause changes in soil chemistry,including increase in available nutrients and pH.High intensity fires are noted for the complete combustion of organic matter and result in severe negative impacts on forest soils.High intensity fires result in nutrient volatilization,the break down in soil aggregate stability,an increase soil bulk density,an increase in the hydrophobicity of soil particles leading to decreased water infiltration with increased erosion and destroy soil biota.High soil heating(> 120℃) from high-intensity forest fires is detrimental to the soil ecosystem,especially its physical and biological properties.In this regard,the use of prescribed burning as a management tool to reduce the fuel load is highly recommended due to its low intensity and limited soil heating.Furthermore,the use of prescribed fires to manage fuel loads is critically needed in the light of current global warming as it will help prevent increased wildfire incidences.This review provides information on the impact of forest fires on soil properties,a key feature in the maintenance of healthy ecosystems.In addition,the review should prompt comprehensive soil and forest management regimes to limit soil disturbance and restore fire-disturbed soil ecosystems. | Alex Amerh Agbeshie Simon Abugre Thomas Atta-Darkwa Richard Awuah | 2022 | Journal of Forestry Research2022,33,5: | 5 |
| 3 | Hypercalciuria switches Ca^(2+) signaling in proximal tubular cells,induces oxidative damage to promote calcium nephrolithiasis显示文摘Proximal tubule(PT)transports most of the renal Ca^(2+),which was usually described as paracellular(passive).We found a regulated Ca^(2+)entry pathway in PT cells via the apical transient receptor potential canonical 3(TRPC3)channel,which initiates transcellular Ca^(2+)transport.Although TRPC3 knockout(−/−)mice were mildly hypercalciuric and displayed luminal calcium phosphate(CaP)crystals at Loop of Henle(LOH),no CaP+calcium oxalate(CaOx)mixed urine crystals were spotted,which are mostly found in calcium nephrolithiasis(CaNL).Thus,we used oral calcium gluconate(CaG;2%)to raise the PT luminal[Ca^(2+)]o further in TRPC3−/−mice for developing such mixed stones to understand the mechanistic role of PT-Ca^(2+)signaling in CaNL.Expectedly,CaG-treated mice urine samples presented with numerous mixed crystals with remains of PT cells,which were pronounced in TRPC3−/−mice,indicating PT cell damage.Notably,PT cells from CaG-treated groups switched their mode of Ca^(2+)entry from receptor-operated to store-operated pathway with a sustained rise in intracellular[Ca^(2+)]([Ca^(2+)]i),indicating the stagnation in PT Ca^(2+)transport.Moreover,those PT cells from CaG-treated groups demonstrated an upregulation of calcification,inflammation,fibrotic,oxidative stress,and apoptotic genes;effects of which were more robust in TRPC3 ablated condition.Furthermore,kidneys from CaG-treated groups exhibited fibrosis,tubular injury and calcifications with significant reactive oxygen species generation in the urine,thus,indicating in vivo CaNL.Taken together,excess PT luminal Ca^(2+)due to escalation of hypercalciuria in TRPC3 ablated mice induced surplus CaP crystal formation and caused stagnation of PT[Ca^(2+)]i,invoking PT cell injury,hence mixed stone formation. | Samuel Shin Cliff-Lawrence Ibeh Eugenia Awuah Boadi Bok-Eum Choi Sanjit K.Roy Bidhan C.Bandyopadhyay | 2022 | Genes & Diseases2022,9,2: | 2 |
