4A review on palaeogeographic implications and temporal variation in glaucony composition显示文摘This study presents a review on palaeogeographic implications and temporal variations of glaucony coveringboth modern and ancient records.Phanerozoic glaucony preferably forms in a shelf depositional setting.Deep marine conditions and low seawater temperature discourage formation of glaucony.Around 75%of glaucony is recorded from the Cretaceous to the Holocene sediments,which are related to the abundance of the most common substrates,faecal pellets and bioclasts.TFe_2O_3(total),Al_2O_3,K_2O and MgO contents of glaucony vary appreciably through geological time.While TFe_2O_3 content of most Mesozoic and Cenozoic glaucony exceeds 20%,it is always less than 20%in Precambrian varieties.High K_2O,Al_2O_3,MgO and low TFe_2O_3 distinguish the Precambrian glaucony from its Phanerozoic counterpart.Precambrian glaucony,preferably formed within a K-feldspar substrate,is always rich in potassium irrespective of its degree of evolution,while high K-content in Phanerozoic evolved glaucony indicates significant stratigraphic condensation.K_2O us.TFe_2O_3 relationship of glaucony exhibits three different evolutionary trends corresponding to three common modes of origin.Depositional conditions may influence the composition of glaucony as slightly reducing conditions favour Fe enrichment,whereas oxidising conditions cause Fe depletion in glaucony.Santanu Banerjee Udita Bansal Anup Vilas Thorat 2016Journal of Palaeogeography2016,5,1:10
5Graph Regularized L_p Smooth Non-negative Matrix Factorization for Data Representation显示文摘This paper proposes a Graph regularized Lpsmooth non-negative matrix factorization(GSNMF) method by incorporating graph regularization and L_p smoothing constraint, which considers the intrinsic geometric information of a data set and produces smooth and stable solutions. The main contributions are as follows: first, graph regularization is added into NMF to discover the hidden semantics and simultaneously respect the intrinsic geometric structure information of a data set. Second,the Lpsmoothing constraint is incorporated into NMF to combine the merits of isotropic(L_2-norm) and anisotropic(L_1-norm)diffusion smoothing, and produces a smooth and more accurate solution to the optimization problem. Finally, the update rules and proof of convergence of GSNMF are given. 6Mitochondrial dysfunction as a mechanism of drug-induced hepatotoxicity:current understanding and future perspectives显示文摘Mitochondria are critical cellular organelles for energy generation and are now also recognized as playing important roles in cellular signaling.Their central role in energy metabolism,as well as their high abundance in hepatocytes,make them important targets for drug-induced hepatotoxicity.This review summarizes the current mechanistic understanding of the role of mitochondria in drug-induced hepatotoxicity caused by acetaminophen,diclofenac,anti-tuberculosis drugs such as rifampin and isoniazid,anti-epileptic drugs such as valproic acid and constituents of herbal supplements such as pyrrolizidine alkaloids.The utilization of circulating mitochondrialspecific biomarkers in understanding mechanisms of toxicity in humans will also be examined.In summary,it is well-established that mitochondria are central to acetaminophen-induced cell death.However,the most promising areas for clinically useful therapeutic interventions after acetaminophen toxicity may involve the promotion of adaptive responses and repair processes including mitophagy and mitochondrial biogenesis,In contrast,the limited understanding of the role of mitochondria in various aspects of hepatotoxicity by most other drugs and herbs requires more detailed mechanistic investigations in both animals and humans.Development of clinically relevant animal models and more translational studies using mechanistic biomarkers are critical for progress in this area.Relevance for patients:This review focuses on the role of mitochondrial dysfunction in liver injury mechanisms of clinically important drugs like acetaminophen,diclofenac,rifampicin,isoniazid,amiodarone and others.A better understanding of the mechanisms in animal models and their translation to patients will be critical for the identification of new therapeutic targets.Anup Ramachandran Luqi Duan Jephte Y.Akakpo Hartmut Jaeschke 2018Journal of Clinical & Translational Research2018,4,1:8
7Meta-analysis of Quantitative Diffusion-weighted MR Imaging in Differentiating Benign and Malignant Pancreatic Masses显示文摘There have been numerous studies done to explore the diagnostic performance of quantitative diffusion-weighted(DW) MR imaging to differentiate between benign and malignant pancreatic masses. However, the results have been inconsistent. We performed a meta-analysis to investigate whether DW-MR imaging can differentiate between these two diseases. Databases including MEDLINE, EMBASE and Cochrane Library were utilized to find relevant articles published between January 2001 and January 2014. A Stata version 12.0 and a Meta-Disc version 1.4 were used to describe primary results. Twelve studies with 594 patients, which fulfilled the inclusion criteria, were enrolled for the analysis. The pooled sensitivity and specificity of DW imaging was 0.91(95% CI: 0.84, 0.95) and 0.86(95% CI: 0.76, 0.93) respectively. The area under the curve of the summary receiver operating characteristic was 0.95(95% CI: 0.93, 0.96). 8Mechanisms of acetaminophen hepatotoxicity and their translation to the human pathophysiology显示文摘Acetaminophen(APAP)overdose is the most common cause of acute liver failure in the United States and mechanisms of liver injury induced by APAP overdose have been the focus of extensive investigation.Studies in the mouse model,which closely reproduces the human condition,have shown that hepatotoxicity is initiated by formation of a reactive metabolite N-acetyl-p-benzoquinone imine(NAPQI),which depletes cellular glutathione and forms protein adducts on mitochondrial proteins.This leads to mitochondrial oxidative and nitrosative stress,accompanied by activation of c-jun N-terminal kinase(JNK)and its translocation to the mitochondria.This then amplifies the mitochondrial oxidant stress,resulting in translocation of Bax and dynamin related protein 1(Drp1)to the mitochondria,which induces mitochondrial fission,and ultimately induction of the mitochondrial membrane permeability transition(MPT).The induction of MPT triggers release of intermembrane proteins such as apoptosis inducing factor(AIF)and endonuclease G into the cytosol and their translocation to the nucleus,causing nuclear DNA fragmentation and activation of regulated necrosis.Though these cascades of events were primarily identified in the mouse model,studies on human hepatocytes and analysis of circulating biomarkers from patients after APAP overdose,indicate that a number of mechanistic events are identical in mice and humans.Circulating biomarkers also seem to be useful in predicting the course of liver injury after APAP overdose in humans and hold promise for significant clinical use in the near future.Relevance for patients:This review focuses on the mechanisms behind APAP-induced hepatotoxicity and the relevance of these to the human pathophysiology.Current investigations on various biomarkers which may be useful in clinical management of APAP overdose patients are also discussed.Anup Ramachandran Hartmut Jaeschke 2017Journal of Clinical & Translational Research2017,3,1:6

10Nerve-sparing robot-assisted radical prostatectomy:Current perspectives显示文摘Robotic-assisted radical prostatectomy(RARP)is the current standard of care with long term cure in organ-confined disease.The introduction of nerve-sparing(NS)to standard RARP has shown positive results in terms of functional outcomes in addition to the oncological outcomes.This article reviews the current perspectives of NS-RARP in terms of applied anatomy of the prostatic fascial planes,the neurovascular bundle(NVB),various NS techniques and postoperative functional outcomes.A non-systematic review was done using PubMed,Embase and Medline databases to retrieve and analyse articles in English,with following keywords“prostate cancer”,“robotic radical prostatectomy”,“nerve-sparing”.The Delphi method was used with an expert panel of robotic surgeons in urology to analyse the potency outcomes of various published comparative and non-comparative studies.The literature has shown that NS-RARP involves various techniques and approaches while there is a lack of randomized studies to suggest the superiority of one over the other.Variables such as preoperative risk assessments,baseline potency,surgical anatomy of individual patients and surgeons’expertise play a major role in the outcomes.A tailored approach for each patient is required for applying the NS approach during RARP.Anup Kumar Vipul RPatel Sridhar Panaiyadiyan Kulthe Ramesh Seetharam Bhat Marcio Covas Moschovas Brusabhanu Nayak 2021Asian Journal of Urology2021,8,1:4

13Recommendations for the use of the acetaminophen hepatotoxicity model for mechanistic studies and how to avoid common pitfalls显示文摘Acetaminophen(APAP)is a widely used analgesic and antipyretic drug,which is safe at therapeutic doses but can cause severe liver injury and even liver failure after overdoses.The mouse model of APAP hepatotoxicity recapitulates closely the human pathophysiology.As a result,this clinically relevant model is frequently used to study mechanisms of drug-induced liver injury and even more so to test potential therapeutic interventions.However,the complexity of the model requires a thorough understanding of the pathophysiology to obtain valid results and mechanistic information that is translatable to the clinic.However,many studies using this model are flawed,which jeopardizes the scientific and clinical relevance.The purpose of this review is to provide a framework of the model where mechanistically sound and clinically relevant data can be obtained.The discussion provides insight into the injury mechanisms and how to study it including the critical roles of drug metabolism,mitochondrial dysfunction,necrotic cell death,autophagy and the sterile inflammatory response.In addition,the most frequently made mistakes when using this model are discussed.Thus,considering these recommendations when studying APAP hepatotoxicity will facilitate the discovery of more clinically relevant interventions.Hartmut Jaeschke Olamide B.Adelusi Jephte Y.Akakpo Nga T.Nguyen Giselle Sanchez-Guerrero David S.Umbaugh Wen-Xing Ding Anup Ramachandran 2021Acta Pharmaceutica Sinica B2021,11,12:3

17Study of morphologic variability of incudostapedial angle and its relation with temporal bone pneumatization显示文摘Introduction:Optimal sound transmission across the ossicular chain is dependent on the appropriate alignment of the middle ear ossicles.Incudostapedial joint(ISJ)is conventionally considered to be at right angle.Objective:We intended to study the ISJ anatomy and the impact of temporal bone pneumatization on the same.Methods:In a cadaveric study comprising of 47 human temporal bones,canal wall down mastoidectomy was carried out under microscopic guidance keeping the ossicular chain intact.The morphology of ISJ was recorded and analysed with respect to the pneumatization status of the temporal bone.The data analysis was performed using statistical software Stata version 12.0.Results:The mean ISJ angle for the 47 bones was 90.5^0(SD-15^0;range:54^0-122^0).The mean angle in well pneumatized bones was 93.7^0(SD-16.5;Range:54°-122^0)and in sclerotic mastoids was 88.7^0(SD-14;Range:68°-118^0).The difference in the ISJ angle in these two conditions was not statistically significant(p=0.27).The mean angle was found to be significantly more obtuse in the cases with partially eroded ISJ(111.4^0{SD-8.8;range:100.3^0-221.9^0};p=0.0001)and in the cases with an‘adherent/tilted morphology’of the stapes suprastructure with the promontory(mean-95.8^0(SD-13.8;range:70.7^0-120.4^0);p-<0.00001).Conclusion:The ISJ angle shows considerable variations.This variability needs to be taken into account when undertaking middle ear reconstructive procedures,specifically the ones involving the stapes footplate.The mastoid pneumatization does not appear to have an impact on the ISJ angle.Anup Singh David Victor Kumar Irugu Rajeev Kumar Hitesh Verma Anandita Gupta Abha Kumari 2020Journal of Otology2020,15,2:3
18无铬Co-Cu/SBA-15催化剂催化生物质衍生α-,β-不饱和醛加氢制醇(英文)显示文摘Cr-free bi-metallic SBA-15-supported Co–Cu catalysts were examined in the conversion of biomass-derived α-, β-unsaturated aldehyde(furfural) to value-added chemical furfuryl alcohol(FOL).Co–Cu/SBA-15 catalysts with a fixed Cu loading of 10 wt% and varying Co loadings(2.5, 5, and 10 wt%) were prepared by the impregnation method. The catalysts were characterized by X-ray diffraction, N2 sorption, H2 temperature-programmed reduction, scanning electron microscopy, energy-dispersive X-ray spectroscopy, high-resolution transmission electron microscopy, CO chemisorption, and inductively coupled plasma mass spectrometry. The influence of different reaction parameters such as temperature, pressure, catalyst dosage, and furfural concentration on the catalyst performance was evaluated. Relative to catalysts supported on amorphous silica, the current SBA-15-supported Co–Cu catalysts displayed higher performance, attaining a furfural conversion of 99% and furfuryl alcohol selectivity of 80%.
