1Antisense MMP-9 RNA inhibits malignant glioma cell growth in vitro and in vivo显示文摘The matrix-degrading metalloproteinases (MMPs), particularly MMP-9, play important roles in the pathogenesis and development of malignant gliomas. In the present study, the oncogenic role of MMP-9 in malignant glioma cells was investigated via antisense RNA blockade in vitro and in vivo. TJ905 malignant glioma cells were transfected with pcDNA3.0 vector expressing antisense MMP-9 RNA (pcDNA-AS-MMP9), which significantly decreased MMP-9 expression, and cell proliferation was assessed. For in vivo studies, U251 cells, a human malignant glioma cell line, were implanted subcutaneously into 4-to 6-week-old BALB/c nude mice. The mice bearing well-established U251 gliomas were treated with intratumoral pcDNA-AS-MMP9-Lipofectamine complex (AS-MMP-9-treated group), subcutaneous injection of endostatin (endostatin-treated group), or both (combined therapy group). Mice treated with pcDNA (empty vector)-Lipofectamine served as the control group. Four or eight weeks later, the volume and weight of tumor, MMP-9 expression, microvessel density and proliferative activity were assayed. We demonstrate that pcDNA-AS-MMP9 significantly decreased MMP-9 expression and inhibited glioma cell proliferation. Volume and weight of tumor, MMP-9 expression, microvessel density and proliferative activity in the antisense-MMP-9-treated and therapeutic alliance groups were significantly lower than those in the control group. The results suggest that MMP-9 not only promotes malignant glioma cell invasiveness, but also affects tumor cell proliferation. Blocking the expression of MMP-9 with antisense RNA substantially suppresses the malignant phenotype of glioma cells, and thus can be used as an effective therapeutic strategy for malignant gliomas.Cuiyun Sun Qian Wang Hongxu Zhou Shizhu Yu Alain R. 3A approach for generating high precision digitaltime-delay with low computational load显示文摘In this paper a novel method to generate high-precision and continuously adjustable digital time-delay by utilizing the linear phase of a FIR filter is presented. The generated time-delay can be continuously changed by adjusting the filter weights. The time-delay is pre cise and valid for wide-band, while the computational load is low. The proposed approach can find applications in wide-band digital beamforming, adaptive delay estimation, temporal analy sis of reflected waveform, etc.. In the paper, the basic principle, design steps and simulation re sults are given. 4Experimental researches on underwater target scattering properties in non-incident direction显示文摘In the bistatic case, theoretical analysis and experimental researches on underwater acoustic scattering properties of some submarine model are made in this paper. When sourcet target and receiver have complicated triangular configuration, the relationships among target strength, incidence angle and bistatic angle are obtained. The validity of this theory is verified by theoretical calculations and tank experiments. 5Study on the performance of data fusion system for sonar signal detection显示文摘The mathematical model and fusion algorithm for multisensor data fusion are presented, and applied to integrate the decisions obtained by multiple sonars in a distributed detection system. Assuming that all the sonars and the fusion system operate at the same false alarm probability, the expression for the detection probability of the fusion system is obtained. 6Blind equalization of underwater acoustic channel using a combined constant modulus algorithm显示文摘A kind of Combined Constant Modulus Algorithm (CCMA) is presented to compensate the defects of the Constant Modulus Algorithm (CMA) and the Sign Error CMA (SECMA). And CCMA is applied to the equalization of the underwater acoustic channel (UWAC). Based on the decision of the equalizer's output, its iteration process switches between CMA and SECMA automatically. It is more robust than SECMA, and more computationally efficient than CMA. Therefore, it is very suitable for the recovery of the underwater data transmission. 7机械力诱导MHC-Ⅰ构象变化增强TCR抗原识别及T细胞活化显示文摘CD8+T细胞主要通过T细胞表面受体(T cell recep tor,TCR)识别并与Ⅰ型主要组织性复合物提呈的抗原(pMHC-Ⅰ)相互作用[1]。TCR对刺激型抗原的识别在CD8+T细胞毒性和适应性免疫中发挥着关键作用。许多证据表明机械力可以延长TCR与刺激性pMHC作用的键合时间(bond lifetime)形成抗原特异性逆锁键(catch bond)[2],并且这种逆锁键对抗原识别非常重要。然而,机械力调控TCR抗原识别的具体结构机制仍不清楚。通过分子动力学模拟、单分子生物膜力学探针、磁镊、T细胞活化实验和动物模型对此问题展开了系统的研究[4]。发现作用在TCR-pMHC-Ⅰ复合体上的拉力可以作用在TCR-pMHC-Ⅰ复合体上的拉力可以打破MHC-I分子内部α1-α2和β2结构域间的相互作用,导致α1-α2结构域旋转并发生构象变化。力诱导的MHC-I构象变化可以进一步别构地调节TCR与刺激性抗原肽及α1-α2结构域的构象及相互作用,诱导产生新的氢键,增强TCR-pMHC-Ⅰ之间的键合时间,但并不能增强TCR与抑制性pMHC-Ⅰ之间的作用。当用点突变阻断这些新形成的氢键,或者α1-α2和β2结构域被二硫键锁住时,最佳力诱导的TCR-pMHC-Ⅰ作用的键合时间明显缩短并且T细胞的活化受到抑制。另外,在人TCR和HLA-A2相互作用中发现了类似机制,并且与肿瘤相关的HLA-A2点突变[3]可以通过限制HLA-A2α1--α2和β2结构域之间的构象打开减弱TCR对肿瘤抗原的识别及T细胞的功能。研究结果表明,机械力诱导的MHC-I构象变化对TCR抗原识别和T细胞活化非常重要,进一步地阐明了机械力调控TCR抗原识别机制,为临床肿瘤的免疫治疗和药物设计提供了新思路和新靶点。Peng Wu Tongtong Zhang Baoyu Liu Panyu Fei Lei Cui Rui Qin Huaying Zhu Danmei Yao Ryan Martinez Wei Hu Chenyi An Yong Zhang Junwei Liu Weiwei Yin Jie Sun Chun Zhou Xun Zeng Jianan Wang Brian Evavold Cheng Zhu Jizhong Lou Wei Chen 2019医用生物力学2019,34,A01:0

9Effects of Loading-Dose Statins Combined with PCSK9 Inhibitor Pre-Treatment before Primary Percutaneous Coronary Intervention on the Short-Term Prognosis in Patients with ST-Segment Elevation Myocardial Infarction显示文摘Objective:This study was aimed at investigating the effects of preoperative treatment with a loading dose of statins combined with a PCSK9 inhibitor on coronary blood perfusion and short-term cardiovascular adverse events in patients with ST-segment elevation myocardial infarction(STEMI).Method:Sixty-five patients with STEMI who had visited the Shanxi Cardiovascular Disease Hospital between May 2018 and May 2021 were enrolled in the study.The enrolled patients had no history of oral statins or antiplatelet therapy.The patients were divided into a combined treatment group(loading dose of statins combined with PCSK9 inhibitors,35 patients)and a routine treatment group(loading dose of statins only,30 patients).The primary endpoints were thrombolysis in myocardial infarction(TIMI)blood flow grading,corrected TIMI frame count(CTFC),and TIMI myocardial perfusion grading(TMPG),immediately after and 30 days after the operation.The secondary endpoint was a composite endpoint of cardiovascular death,nonfatal myocardial infarction,and target vessel revascularization 30 days after the operation.Results:The combined treatment group had significantly lower CTFC(14.09±8.42 vs 26±12.42,P=0.04)and better TMPG(2.74±0.61 vs 2.5±0.73,P=0.04)than the routine treatment group immediately after the operation.Similarly,the combined treatment group had a significantly lower CTFC(16.29±7.39 vs 26.23±11.53,P=0.04)and significantly better TMPG(2.94±0.24 vs 2.76±0.43,P=0.01)than the routine treatment group 1 month after the operation.Conclusion:Preoperative treatment with a loading dose of high-intensity statins combined with PCSK9 inhibitors increased coronary blood flow and myocardial perfusion after emergency thrombus aspiration in patients with STEMI.However,the treatment did not significantly decrease the incidence of cardiovascular death,nonfatal myocardial infarction,or target vessel revascularization.Zhe Wang Qingbo Bao Xiaojian Song Hengjie Song Shoudong Wei Junwei Lv Fei Wang Jian An 2022Cardiovascular Innovations and Applications2022,7,3:0
