| 1 | Daily genetic profiling indicates JAK/STAT signaling promotes early hepatic stellate cell transdifferentiation显示文摘AIM: To identify signaling pathways and genes that initiate and commit hepatic stellate cells (HSCs) to transdifferentiation. METHODS: Primary HSCs were isolated from male Sprague-Dawley rats and cultured on plastic for 0-10 d. Gene expression was assessed daily (quiescent to day 10 culture-activation) by real time polymerase chain reaction and data clustered using AMADA software. The significance of JAK/STAT signaling to HSC transdifferentiation was determined by treating cells with a JAK2 inhibitor. RESULTS: Genetic cluster analyses, based on expression of these 21 genes, showed similar expression profiles on days 1-3, days 5 and 6, and days 7-10, while freshly isolated cells (day Q) and day 4 cells were genotypically distinct from any of the other days. Additionally, gene expression clustering revealed strong upregulation of interleukin-6, JAK2 and STAT3 mRNA in the early stages of activation. Inhibition of the JAK/STAT signaling pathway impeded the morphological transdifferentiation of HSCs which correlated with decreased mRNA expression of several profibrotic genes including collagens, α-SMA, PDGFR and TGFβR. | 4 | Apparent redox potential discontinuity(aRPD) depth as a relative measure of sediment oxygen content and habitat quality显示文摘As hypoxic conditions spread in our oceans,indices that quickly and efficiently assess oxygen content in sediment pore water,and habitat quality are increasingly becoming desirable.Depth to the apparent redox potential discontinuity(aRPD) in sediment is one such index;however,the relationship between aRPD depth and oxygen content is still unclear.We quantified oxygen content in sediment pore water with a dissolved oxygen(DO) probe,utilizing luminescent technology,to determine how DO concentration varies with depth in the sediment,and with position relative to the aRPD(above versus below) on an intertidal mudflat in the Bay of Fundy,Canada.Based on generalized linear regression models,we found that depth in the sediment was a poor predictor of pore water DO concentration,while position in the sediment relative to the aRPD was a good predictor.Further,sediment above the aRPD had higher DO content than sediment below.We maintain that aRPD depth is a good measure of relative oxygen content in sediment pore water,as well as habitat quality.However,we recommend caution as the factors that influence aRPD depth arc numerous and variable;therefore,the relationship between aRPD depth and DO content should be experimentally confirmed in any system of interest before aRPD depth is used as an indicator. | Travis G.GERWING Alyssa M.ALLEN GERWING Diana J.HAMILTON Myriam A.BARBEAU | 2015 | International Journal of Sediment Research2015，30，1： | 3 |

| 6 | Bioactive extracellular matrix scaffolds engineered with proangiogenic proteoglycan mimetics and loaded with endothelial progenitor cells promote neovascularization and diabetic wound healing显示文摘Diabetic ischemic wound treatment remains a critical clinical challenge.Neovascularization plays a significant role in wound healing during all stages of the tissue repair process.Strategies that enhance angiogenesis and neovascularization and improve ischemic pathology may promote the healing of poor wounds,particularly diabetic wounds in highly ischemic conditions.We previously identified a cyclic peptide LXW7 that specifically binds to integrinαvβ3 on endothelial progenitor cells(EPCs)and endothelial cells(ECs),activates vascular endothelial growth factor(VEGF)receptors,and promotes EC growth and maturation.In this study,we designed and synthesized a multi-functional pro-angiogenic molecule by grafting LXW7 and collagen-binding peptides(SILY)to a dermatan sulfate(DS)glycosaminoglycan backbone,named LXW7-DS-SILY,and further employed this multi-functional molecule to functionalize collagen-based extracellular matrix(ECM)scaffolds.We confirmed that LXW7-DS-SILY modification significantly promoted EPC attachment and growth on the ECM scaffolds in vitro and supported EPC survival in vivo in the ischemic environment.When applied in an established Zucker Diabetic Fatty(ZDF)rat ischemic skin flap model,LXW7-DS-SILY-functionalized ECM scaffolds loaded with EPCs significantly improved wound healing,enhanced neovascularization and modulated collagen fibrillogenesis in the ischemic environment.Altogether,this study provides a promising novel treatment to accelerate diabetic ischemic wound healing,thereby reducing limb amputation and mortality of diabetic patients. | Siqi He Tanaya Walimbe Hongyuan Chen Kewa Gao Priyadarsini Kumar Yifan Wei Dake Hao Ruiwu Liu Diana L.Farmer Kit S.Lam Jianda Zhou Alyssa Panitch Aijun Wang | 2022 | Bioactive Materials2022，7，4： | 2 |

| 11 | Reproductive behavior in horseshoe crabs:Does density matter?显示文摘While the four species of horseshoe crabs share many common reproductive traits with respect to their reproductivesystems,they do differ with respect to their mating behavior (monogamy vs.polygynandry).Past research has attributed thesedifferences to a number of factors including:spawning densities,operational sex ratios (OSR's),male condition (or age),environmentaland/or genetic factors,or a combination thereof.Mating behaviors in the three Asian horseshoe crab species (Tachypleusgigas,T.tridentatus,and Carcinoscorpius rotundicauda) with low spawning densities and 1:1 operational sex ratios aretypically monogamous.In Limulus polyphemus,mating behavior is more variable ranging from monogamy to polygynandry.Herewe provide evidence,through a long term behavioral study,that variation in mating behavior is influenced by population densityin L.polyphemus.Our study population on two beaches in Connecticut (Long Island Sound) have a spawning density 400 timesless than that found in Delaware Bay (0.002 females/m^2 vs.0.8 females/m^2) but similar operational sex ratios.Between 90%-95%of all spawning females in CT were paired with only one male,thus exhibiting monogamous behavior.In contrast,between 30and 60% of spawning females in Delaware Bay have more than one mate and produce clutches of eggs with multiple paternities.Male condition played no role in mating behavior in CT populations.We also observed that on average 18% of the females on thespawning beaches are single.These results suggest that population density is an important condition that determines mating behavior.Also,low population density may lead to decreased mate finding ability and lost opportunities for | Jennifer H.MATTEI Mark A.BEEKEY Adam RUDMAN Alyssa WORONIK | 2010 | Current Zoology2010，56，5： | 2 |
