1东亚地区春季二氧化硫的输送与转化过程研究 I.模式及其验证显示文摘结合新近评估的东亚地区污染源资料 ,作者利用一个耦合的区域化学输送模式系统以探讨东亚地区春季期间气象过程、气相与液相化学过程、非均相化学过程、气溶胶过程和干湿沉降过程对二氧化硫输送及转化过程的影响 ,并研究二氧化硫和硫酸盐气溶胶的空间分布及变化特征。模拟的二氧化硫和硫酸盐气溶胶的浓度值与 2 0 0 1年春季飞机和地面获取的观测值进行了比较。比较结果显示 ,模拟值与观测值具有很好的一致性 ,模式系统很好地反映了二氧化硫和硫酸盐气溶胶的分布特征和变化规律 ,再现了许多观测到的重要特征 。张美根 徐永福 Itsushi Uno Hajime Akimoto 2004大气科学2004,28,3:16
2A Numerical Study of Tropospheric Ozone in the Springtime in East Asia显示文摘The Models-3 Community Multi-scale Air Quality modeling system (CMAQ) coupled with the Regional Atmospheric Modeling System (RAMS) is applied to East Asia to study the transport and photochemical transformation of tropospheric ozone in March 1998. The calculated mixing ratios of ozone and carbon monoxide are compared with ground level observations at three remote sites in Japan and it is found that the model reproduces the observed features very well. Examination of several high episodes of ozone and carbon monoxide indicates that these elevated levels are found in association with continental outflow,demonstrating the critical role of the rapid transport of carbon monoxide and other ozone precursors from the continental boundary layer. In comparison with available ozonesonde data, it is found that the model-calculated ozone concentrations are generally in good agreement with the measurements, and the stratospheric contribution to surface ozone mixing ratios is quite limited.张美根 徐永福 Itsushi UNO Hajime AKIMOTO 2004Advances in Atmospheric Sciences2004,21,2:15
3Prevalence of linked angina and gastroesophageal reflux disease in general practice显示文摘AIM: To evaluate the association between gastroesophageal re? ux diseases (GERD) and coronary heart diseases. METHODS: One thousand nine hundred and seventy consecutive patients who attended our hospital were enrolled. All of the patients who first attend our hospital were asked to respond to the F-scale questionnaire regardless of their chief complaints. All patients had a careful history taken, and resting echocardiography (ECG) was performed by physicians if the diagnostic necessity arose. Patients with ECG signs of coronary artery ischemia were defined as STsegment depression based on the Minnesota code. RESULTS: Among 712 patients (36%) with GERD, ECG was performed in 171 (24%), and ischemic changes were detected in eight (5%). Four (50%) of these patients with abnormal findings upon ECG had no chest symptoms such as chest pain, chest oppression, or palpitations. These patients (0.6%; 4/712) were thought to have non-GERD heartburn, which may be related to ischemic heart disease. Of 281 patients who underwent ECG and did not have GERD symptoms, 20 (7%) had abnormal findings upon ECG. In patients with GERD symptoms and ECG signs of coronary artery ischemia, the prevalence of linked angina was considered to be 0.4% (8/1970 patients).CONCLUSION: The present study suggested that ischemic heart disease might be found although a patient was referred to the hospital with a complaint of GERD symptoms. 4HER2 heterogeneity is a poor prognosticator for HER2-positive gastric cancer显示文摘BACKGROUND The clinical significance of intratumoral human epidermal growth factor receptor 2(HER2)heterogeneity is unclear for HER2-positive gastric cancer,although it has been reported to be a significant prognosticator for HER2-positive breast cancer,which has received trastuzumab-based chemotherapy.AIM To clarify the clinical significance of intratumoral HER2 heterogeneity for HER2-positive gastric cancer,which has received trastuzumab-based chemotherapy.METHODS Patients with HER2-positive unresectable or metastatic gastric cancer who received trastuzumab-based chemotherapy as a first line treatment were included.The patients were classified into two groups according to their intratumoral HER2 heterogeneity status examined by immunohistochemistry(IHC)on endoscopic biopsy specimens before treatment,and their clinical response to chemotherapy and survival were compared.RESULTS A total of 88 patients were included in this study,and HER2 heterogeneity was observed in 23(26%)patients(Hetero group).The overall response rate was significantly better in patients without HER2 heterogeneity(Homo group)(Homo vs Hetero:79.5%vs 35.7%,P=0.002).Progression-free survival of trastuzumab-based chemotherapy was significantly better in the Homo group(median,7.9 vs 2.5 mo,HR:1.905,95%CI:1.109-3.268).Overall survival was also significantly better in the Homo group(median survival time,25.7 vs 12.5 mo,HR:2.430,95%CI:1.389-4.273).Multivariate analysis revealed IHC HER2 heterogeneity as one of the independent poor prognostic factors(HR:3.115,95%CI:1.610-6.024).CONCLUSION IHC of HER2 heterogeneity is the pivotal predictor for trastuzumab-based chemotherapy.Thus,HER2 heterogeneity should be considered during the assessment of HER2 expression.Akio Kaito Takeshi Kuwata Masanori Tokunaga Kohei Shitara Reo Sato Tetsuo Akimoto Takahiro Kinoshita 2019World Journal of Clinical Cases2019,7,15:5
5A novel synthesis method and up-conversion properties of hexagonal-phase NaYF_4:Er nano-crystals显示文摘A novel synthesis method for hexagonal(β)-phase NaYF4:Er nano-crystals(NCs)which showed up-conversion(UC)from infrared to visible spectral region was developed.The NaYF4:Er NCs were synthesized in oleic acid(OA)and 1-octadecene(ODE)with Y2(CO3)3· xH2O,Er2(CO3)3· xH2O,Na2CO3 and NH4F as precursors.This proposed method was simple and less toxic compared with generally used method so far.The XRD results showed that the molar ratio of OA/ODE and the temperature were key factors for phase control of NaYF4:Er NCs.The UC emission spectra were obtained with the emission wavelength at about 980 nm(4I11/2→4I15/2),800 nm(4I9/2→4I15/2),660 nm(4F9/2→4I15/2)and 540 nm(4S3/2→4I15/2)from Er3+ ions,by excitation wavelength of 1550 nm.The slope values,n,in the pump-power dependence,showed that the emission at 980 and 800 nm were generated by 2-step UC and at 660 nm and 540 nm were 3-step UC.The optical process for the UC excitation was discussed.罗显佳 Ryosuke Yuminami Takeaki Sakurai Katsuhiro Akimoto 2013Journal of Rare Earths2013,31,3:4
6Matrix metalloproteinase-2 as a superior biomarker for peritoneal deterioration in peritoneal dialysis显示文摘AIM: To investigate the efficacy of effluent biomarkers for peritoneal deterioration with functional decline in peritoneal dialysis(PD). METHODS: From January 2005 to March 2013, the subjects included 218 PD patients with end-stage renal disease at 18 centers. Matrix metalloproteinase-2(MMP-2), interleukin-6(IL-6), hyaluronan, and cancer antigen 125(CA125) in peritoneal effluent were quantified with enzyme-linked immunosorbent assay. Peritoneal solute transport rate was assessed by peritoneal equilibration test(PET) to estimate peritoneal deterioration. RESULTS: The ratio of the effluent level of creatinine(Cr) obtained 4 h after injection(D) to that of plasma was correlated with the effluent levels of MMP-2(ρ = 0.74, P < 0.001), IL-6(ρ = 0.46, P < 0.001), and hyaluronan(ρ = 0.27, P < 0.001), but not CA125(ρ = 0.13, P = 0.051). The area under receiver operating characteristic curve for the effluent levels of MMP-2, IL-6, and hyaluronan against high PET category were 0.90, 0.78, 0.62, and 0.51, respectively. No patient developed new-onset encapsulating peritoneal sclerosis for at least 1.5 years after peritoneal effluent sampling.CONCLUSION: The effluent MMP-2 level most closely reflected peritoneal solute transport rate. 8Successful biliary drainage using a metal stent through the gastric stoma显示文摘We report a case of biliary drainage for malignant stricture using a metal stent with an ultrathin endoscope through the gastric stoma. A 78-year-old female was referred to our hospital for jaundice and fever. She had undergone percutaneous endoscopic gastrostomy(PEG) for esophageal obstruction after radiation therapy for cancer of the pharynx. Abdominal contrast-enhanced computed tomography showed a 3-cm enhanced mass in the middle bile duct and dilatation of the intrahepatic bile duct. We initially performed endoscopic retrograde cholangiopancreatography(ERCP) with a trans-oral approach. However, neither the side-viewing endoscope nor the ultrathin endoscope passed through the esophageal orifice. Thus, we eventually performed ERCP via the PEG stoma using an ultrathin endoscope. We performed biliary drainage with a 6F introducer selfexpanding metal stent. The cytology findings obtained by brush cytology showed malignancy. 11Enhanced hepatic differentiation in the subpopulation of human amniotic stem cells under 3D multicellular microenvironment显示文摘BACKGROUND To solve the problem of liver transplantation donor insufficiency,an alternative cell transplantation therapy was investigated.We focused on amniotic epithelial cells(AECs)as a cell source because,unlike induced pluripotent stem cells,they are cost-effective and non-tumorigenic.The utilization of AECs in regenerative medicine,however,is in its infancy.A general profile for AECs has not been comprehensively analyzed.Moreover,no hepatic differentiation protocol for AECs has yet been established.To this end,we independently compiled human AEC libraries,purified amniotic stem cells(ASCs),and co-cultured them with mesenchymal stem cells(MSCs)and human umbilical vein endothelial cell(HUVECs)in a 3D system which induces functional hepatic organoids.AIM To characterize AECs and generate functional hepatic organoids from ASCs and other somatic stem cells METHODS AECs,MSCs,and HUVECs were isolated from the placentae and umbilical cords of cesarean section patients.Amnion and primary AEC stemness characteristics and heterogeneity were analyzed by immunocytochemistry,Alkaline phosphatase(AP)staining,and flow cytometry.An adherent AEC subpopulation was selected and evaluated for ASC purification quality by a colony formation assay.AEC transcriptomes were compared with those for other hepatocytes cell sources by bioinformatics.The 2D and 3D culture were compared by relative gene expression using several differentiation protocols.ASCs,MSCs,and HUVECs were combined in a 3D co-culture system to generate hepatic organoids whose structure was compared with a 3D AEC sphere and whose function was elucidated by immunofluorescence imaging,periodic acid Schiff,and an indocyanine green(ICG)test.RESULTS AECs have certain stemness markers such as EPCAM,SSEA4,and E-cadherin.One AEC subpopulation was also either positive for AP staining or expressed the TRA-1-60 and TRA-1-81 stemness markers.Moreover,it could form colonies and its frequency was enhanced ten-fold in the adherent subpopulation after selective primary passage.Bioinformatics analysis of ribose nucleic acid sequencing revealed that the total AEC gene expression was distant from those of pluripotent stem cells and hepatocytes but some gene expression overlapped among these cells.TJP1,associated with epidermal growth factor receptor,and MET,associated with hepatocyte growth factor receptor,were upregulated and may be important for hepatic differentiation.In conventional flat culture,the cells turned unviable and did not readily differentiate into hepatocytes.In 3D culture,however,hepatic gene expression of the AEC sphere was elevated even under a two-step differentiation protocol.Furthermore,the organoids derived from the MSC and HUVEC co-culture showed 3D structure with polarity,hepatic-like glycogen storage,and ICG absorption/elimination.CONCLUSION Human amniotic epithelial cells are heterogeneous and certain subpopulations have high stemness.Under a 3D co-culture system,functional hepatic organoids were generated in a multicellular microenvironment.Kinji Furuya Yun-Wen Zheng Daisuke Sako Kenichi Iwasaki Dong-Xu Zheng Jian-Yun Ge Li-Ping Liu Tomoaki Furuta Kazunori Akimoto Hiroya Yagi Hiromi Hamada Hiroko Isoda Tatsuya Oda Nobuhiro Ohkohchi 2019World Journal of Stem Cells2019,11,9:2
