2Co-Production of Lipids, Eicosapentaenoic Acid, Fucoxanthin, and Chrysolaminarin by Phaeodactylum tricornutum Cultured in a Flat-Plate Photobioreactor Under Varying Nitrogen Conditions显示文摘The marine diatom Phaeodactylum tricornutum is a polymorphological, ecologically significant, and well-studied model of unicellular microalga. This diatom can accumulate diverse important metabolites. Herein, we cultured P. tricornutum in an internally installed tie-piece flat-plate photobioreactor under 14.5 m mol L^(-1)(high nitrogen, HN) and 2.9 m mol L^(-1)(low nitrogen, LN) of KNO_3 and assessed its time-resolved changes in biochemical compositions. The results showed that HN was inductive to accumulate high biomass(4.1 g L^(-1)). However, the LN condition could accelerate lipid accumulation in P. tricornutum. The maximum total lipid(TL) content under LN was up to 42.5% of biomass on day 12. Finally, neutral lipids(NLs) were 63.8% and 75.7% of TLs under HN and LN, respectively. The content of EPA ranged from 2.3% to 1.5% of dry weight during the growth period under the two culture conditions. Peak volumetric lipid productivity of 128.4 mg L^(-1)d^(-1) was achieved in the HN group(on day 9). The highest volumetric productivity values of EPA, chrysolaminarin, and fucoxanthin were obtained in the exponential phase(on day 6) under HN, which were 9.6, 93.6, and 4.7 mg L^(-1)d^(-1), respectively. 3An App knock-in rat model for Alzheimer’s disease exhibiting Aβand tau pathologies,neuronal death and cognitive impairments显示文摘A major obstacle in Alzheimer’s disease(AD)research is the lack of predictive and translatable animal models that reflect disease progression and drug efficacy.Transgenic mice overexpressing amyloid precursor protein(App)gene manifest non-physiological and ectopic expression of APP and its fragments in the brain,which is not observed in AD patients.The App knock-in mice circumvented some of these problems,but they do not exhibit tau pathology and neuronal death.We have generated a rat model,with three familiar App mutations and humanized Aβsequence knocked into the rat App gene.Without altering the levels of full-length APP and other APP fragments,this model exhibits pathologies and disease progression resembling those in human patients:deposit of Aβplaques in relevant brain regions,microglia activation and gliosis,progressive synaptic degeneration and AD-relevant cognitive deficits.Interestingly,we have observed tau pathology,neuronal apoptosis and necroptosis and brain atrophy,phenotypes rarely seen in other APP models.This App knock-in rat model may serve as a useful tool for AD research,identifying new drug targets and biomarkers,and testing therapeutics.Keliang Pang Richeng Jiang Wei Zhang Zhengyi Yang Lin-Lin Li Makoto Shimozawa Simone Tambaro Johanna Mayer Baogui Zhang Man Li Jiesi Wang Hang Liu Ailing Yang Xi Chen Jiazheng Liu Bengt Winblad Hua Han Tianzi Jiang Weiwen Wang Per Nilsson Wei Guo Bai Lu 2022Cell Research2022,32,2:6

9SHANK2 is a frequently amplified oncogene with evolutionarily conserved roles in regulating Hippo signaling显示文摘Dysfunction of the Hippo pathway enables cells to evade contact inhibition and provides advantages for cancerous overgrowth.However,for a significant portion of human cancer,how Hippo signaling is perturbed remains unknown.To answer this question,we performed a genome-wide screening for genes that affect the Hippo pathway in Drosophila and cross-referenced the hit genes with human cancer genome.In our screen,Prosap was identified as a novel regulator of the Hippo pathway that potently affects tissue growth.Interestingly,a mammalian homolog of Prosap,SHANK2,is the most frequently amplified gene on 11 q13,a major tumor amplicon in human cancer.Gene amplification profile in this 11q13 amplicon clearly indicates selective pressure for SHANK2 amplification.More importantly,across the human cancer genome,SHANK2 is the most frequently amplified gene that is not located within the Myc amplicon.Further studies in multiple human cell lines confirmed that SHANK2 overexpression causes deregulation of Hippo signaling through competitive binding for a LATS1 activator,and as a potential oncogene,SHANK2 promotes cellular transformation and tumor formation in vivo.In cancer cell lines with deregulated Hippo pathway,depletion of SHANK2 restores Hippo signaling and ceases cellular proliferation.Taken together,these results suggest that SHANK2 is an evolutionarily conserved Hippo pathway regulator,commonly amplified in human cancer and potently promotes cancer.Our study for the first time illustrated oncogenic function of SHANK2,one of the most frequently amplified gene in human cancer.Furthermore,given that in normal adult tissues,SHANK2 s expression is largely restricted to the nervous system,SHANK2 may represent an interesting target for anticancer therapy.Liang Xu Peixue Li Xue Hao Yi Lu Mingxian Liu Wenqian Song Lin Shan Jiao Yu Hongyu Ding Shishuang Chen Ailing Yang Yi Arial Zeng Lei Zhang Hai Jiang 2021Protein & Cell2021,12,3:1

15Bilateral Choroidal Occlusion in Antiphospholipid Syndrome Associated with Systemic Lupus Erythematosus显示文摘This article reports a rare case of bilateral choroidal occlusion that occurred in a 24-year-old woman with antiphospholipid syndrome (APS) associated with systemic lupus erythematosus (SLE). This young lady concurred with aorta ventralis thrombosis and bilateral iliac artery occlusion when presented, and experienced a rapid deterioration of vision. She also has a history of recurrent miscarriage. Corticosteroid,immunosuppression and anticoagulation therapy were administered. 16Extreme temperature increases the severity of intracerebral hemorrhage: An analysis based on the cold region of China显示文摘Objective:The purpose of this study was to find a suitable model to evaluate the relationship between temperature and intracerebral hemorrhage(ICH)and explore the effects of cold spells and heat waves on the clinicopathological parameters of ICH patients.Methods:We conducted a retrospective study based on the ICH admission in the First Affiliated Hospital of Harbin Medical University from 2015 to 2020(N=11124).The relationship between different seasons and the number of patients with ICH was explored.Poisson Akaike information criterion(AIC)was used to select the optimal model for temperature and ICH.Binary logistic regression analysis was used to investigate the association between extreme temperatures and clinicopathological features.Results:Hospital admissions for patients with ICH showed monthly changes.The optimal cold spell was defined as the daily average temperature<3rd percentile,lasting for five days,while the optimal heat wave was defined as the daily average temperature>97th percentile,lasting for three days.Based on the generalized extreme weather model,cold climate significantly increased the risk of hematoma volume expansion(OR 1.003;95%CI:1.000-1.005,P=0.047).In the optimal model,the occurrence of cold spells and heat waves increased the risk of midline shift in both conditions(OR 1.067;95%CI:1.021-1.115,P=0.004;OR 1.077;95%CI:1.030-1.127,P=0.001).Conclusion:Our study shows that seasonal cold spells and heat waves are essential factors affecting ICH severity,and targeted preventive measures should be taken to minimize the pathological impacts.Xun Xu Chunyang Liu Rui Liu Qiuyi Jiang Enzhou Lu Chao Yuan Yanchao Liang Huan Xiang Boxian Zhao Xin Chen Ailing Lian Qi Zhou Guang Yang 2022Frigid Zone Medicine2022,2,3:1

18Neuropeptide Y and nestin expression in the hippocampal CA3 region following restrained and inverted stress in rats显示文摘Our preliminary study demonstrated that neuropeptide Y(NPY)/nestin-positive cells exhibit a consistent spatial distribution in the hippocampus of normal adult rats.However,following severe acute and chronic stress-induced impaired learning and memory,synchronous decreased expression of nestin and NPY takes place in the hippocampus,and the underlying mechanisms remain unclear.In the present study,acute and chronic stress rat models were established using combined restrained and inverted stress.Results showed that learning and memory significantly decreased in acute and chronic stress rats.In addition,hippocampal cells were damaged,in particular in the acute stress rats,and nestin and NPY expression,as well as the number of NPY/nestin-positive cells in the CA3 region,significantly decreased.Furthermore,mature neurofilament 200-positive neurons were absent in the chronic stress rats.The NPY and cytoskeletal protein system equally contributed to stress-induced early learning and memory deficits,as well as sustained cerebral injury in the adult hippocampus.Sun, Guogang Chen, Bo Fan, Guangbi Xiao, Hongwen Chen, Yue Xu, Jie Nie, Ye Zhang, Bing Gong, Lin Li, Ailing 2011Neural Regeneration Research2011,6,3:1
19p-Type CaFe_2O_4 semiconductor nanorods controllably synthesized by molten salt method显示文摘Pure phase, regular shape and well crystallized nanorods of p-type semiconductor CaFe_2O_4 have been fabricated for the first time by a facile molten salt assisted method, as confirmed by XRD, TEM, SEM and HRTEM. UV-vis diffuse reflectance spectra and Mott–Schottky plots show that the band structure of the CaFe_2O_4 nanorods is narrower than that of the CaFe_2O_4 nanoparticles synthesized by conventional method. The enhancement of the visible-light absorption is due to narrowness of the band gap in CaFe_2O_4 nanorods. The appropriate ratio between the molten salt and the CaFe_2O_4 precursors plays an important role in inhibiting the growth of the crystals along the(201) plane to give the desired nanorod morphology. This work not only demonstrates that highly pure p-type CaFe_2O_4 semiconductor with tunable band structure and morphology could be obtained using the molten salt strategy, but also affirms that the bandgap of a semiconductor may be tunable by monitoring the growth of a particular crystal plane.Furthermore, the facile eutectic molten salt method developed in this work may be further extended to fabricate some other semiconductor nanomaterials with a diversity of morphologies.Xin Liu Junzhe Jiang Yushuai Jia Ailing Jin Xiangshu Chen Fei Zhang Hongxian Han 2016Journal of Energy Chemistry2016,25,3:1
