1Effect of crocin carotenoid on BDNF and CREB gene expression in brain ventral tegmental area of morphine treated rats显示文摘Objective: To investigate the effect of crocin carotenoid on BNDF and CREB gene expression in the brain ventral tegmental area(VTA) and the serum level of BDNF in morphine-treated rats compared to control. Methods: In this study, 40 male Wistar rats(200-250 g) were used in 5 experimental groups: 1) non morphine treat rats(control); 2) non morphine-treated rats with 25 mg/kg crocin carotenoid(i.p., for 21 d); 3) morphine treated rats(10 mg/kg twice a day, s.c., 21 d); 4 and 5) morphine-treated rats with 12.5 and 25 mg/kg crocin carotenoid, respectively. By the end of research, BDNF and CREB expression was determined by real-time-PCR method. ELISA analysis was also applied for assessing the serum BDNF level. Results: The data indicated that morphine treatment could cause a significant decrease in BDNF and CREB gene expression(P<0.01 and P<0.001, respectively) in brain VTA as well as serum level of BDNF(P<0.01) in comparison to control group. Treatment with 25 mg/kg crocin carotenoid caused a significant enhancement in BDNF and CREF gene expression(P<0.01 and P<0.05, respectively) and serum level of BDNF(P<0.01) in morphine-treated rats in comparison to morphine-treated group. 2Protective effects of Balanites aegyptiaca extract, melatonin and ursodeoxycholic acid against hepatotoxicity induced by methotrexate in male rats显示文摘Objective: To compare the degree of ameliorative effects of Melatonin(MEL), Ursodeoxycholic acid(UDCA) and Balanites aegyptiaca(BA) against hepatotoxicity induced by MTX for one month. Methods: Eighty adult male rats(Sprague Dawely) weighing(190±10g), were randomly divided into eight equal groups: Control, MTX, MEL, BA, UDCA, MTX+MEL, MTX+BA, MTX+UDCA. Liver function biomarker enzymes, liver tissue oxidative stress parameters, together with total antioxidant capacity and tumor necrosis factor(TNF-α) were determined. Histopathological and immunohistochemistry examinations for TNF-α were also done. Results: MTX showed significant increase in alanine transaminase(ALT), aspartate transaminase(AST), alkaline phosphatase(ALP), gamma glutamyl transferase(GGT), total and direct bilirubin, as well as TNF-α levels, oxidized glutathione(GSSG), malodialdehyde(MDA) and nitric oxide(NO). whereas, total protein, albumin, total antioxidant capacity, reduced glutathione(GSH), glutathione peroxidase(GPx), glutathione reductase(GR), glutathione S-transferase(GST), superoxide dismutase(SOD) and catalase(CAT) levels were significantly decreased in MTX treated group. These alterations were improved by MEL and BA treatment, whereas no improvement was noticed in UDCA treatment. Conclusions: BA may be as promising as MEL in the hepatoprotection against MTX toxicity through their antioxidant and radical scavenging activities. 3Role of eIF4E on epithelial-mesenchymal transition,invasion,and chemoresistance of prostate cancer cells显示文摘Dear Editor,Prostate cancer(PCa)is the most common cancer and second leading cause of cancer death for men in the United States[1].PCa with similar Gleason score has been reported to show substantial interpatient heterogeneity and differential prostate cancer-specific mortality rate[2].Such heterogeneity in PCa often results in different therapeutic responses among patients,including therapy resistance,therapeutic failure,relapse,and metastasis[3].Numerous oncogenes,such as eukaryotic translation initiation factor 4E(eIF4E),have been reported to be involved in epithelial-mesenchymal transition(EMT)and/or drug resistance in PCa[4].We previously demonstrated that eIF4E overexpression was involved in chemoresistance of triple-negative breast cancer and silencing eIF4E significantly inhibited cancer cell proliferation and sensitized cancer cells to chemotherapy in a patient-derived xenograft mouse model[5].In addition,eIF4E phosphorylation is known to stimulate the translation of matrix metalloproteinase 3(MMP3)and Snail mRNAs to promote EMT in PCa[6].Furthermore,the complexity and dynamic nature of EMT contributes to the heterogeneity of aggressive cancer cells[7].The precise role of eIF4E in EMT,invasion,and chemoresistance in PCa is still to be established with consideration of different subpopulations in order to develop precision medicine for PCa.In this work,we aimed to explore the role of eIF4E in EMT,invasion,and chemoresistance in PCa for establishing a promising new therapeutic strategy by regulating eIF4E expression using(1-aminoethyl)iminobis[Noleicylcysteinyl-1-aminoethyl)propionamide](ECO)/small interfering RNA(siRNA)nanoparticles previously developed in our lab[8-10]for PCa therapy in the context of tumor heterogeneity.Xujie Liu Amita M Vaidya Da Sun Yan Zhang Nadia Ayat Andrew Schilb Zheng-Rong Lu 2020Cancer Communications2020,40,2:2
