| 1 | Cholangiocarcinoma:principles and current trends显示文摘BACKGROUND:Cholangiocarcinoma(CCA)is a lethal cancer of the biliary epithelium,originating from the liver(intrahepatic),at the confluence of the right and left hepatic ducts(hilar)or in the extrahepatic bile ducts.It is a rare malignancy associated with poor prognosis.DATA SOURCES:We searched the PubMed/MEDLINE database for relevant articles published from 1989 to 2008.The search terms used were related to 'cholangiocarcinoma' and its ' treatment'.Although no language restrictions were imposed initially,for the full-text review and final analysis,our resources only permitted the review of articles published in English.This review deals with the treatment of cholangiocarcinoma,the principles and the current trends.RESULTS:The risks and prognostic factors,symptoms and differential diagnosis are thoroughly discussed.In addition,the tools of preoperative diagnosis such as endoscopic retrograde cholangiopancreatography,digital image analysis,fluorescence in situ hybridization and magnetic resonance cholangiopancreatography are reviewed.Moreover,the treatment of CCA is discussed.CONCLUSIONS:The only curative treatment available is surgical management.Unfortunately,many patients present with unresectable tumors,the majority of whom die within a year of diagnosis.Surgical treatment involves major resections of the liver,pancreas and bile duct,with considerable mortality and morbidity.However,in selected cases and where indicated,appropriate management with aggressive surgery may achieve a good outcome with a prolonged survival expectancy. | George N Zografos Athanasios Farfaras Flora Zagouri Dimosthenis Chrysikos Kostas Karaliotas | 2011 | Hepatobiliary & Pancreatic Diseases International2011，10，1： | 14 |

| 3 | 22-gauge core vs 22-gauge aspiration needle for endoscopic ultrasound-guided sampling of abdominal masses显示文摘AIM To compare the aspiration needle(AN) and core biopsy needle(PC) in endoscopic ultrasound-guided fine needle aspiration(EUS-FNA) of abdominal masses.METHODS Consecutive patients referred for EUS-FNA were included in this prospective single-center trial. Each patient underwent a puncture of the lesion with both standard 22-gauge(G) AN(Echo Tip Ultra; Cook Medical, Bloomington, Indiana, United States) and the novel 22 G PC(Echo Tip Pro Core; Cook Medical, Bloomington, Indiana, United States) in a randomized fashion; histology was attempted in the PC group only. The main study endpoint was the overall diagnostic accuracy, including the contribution of histology to the final diagnosis. Secondary outcome measures included material adequacy, number of needle passes, and complications.RESULTS Fifty six consecutive patients(29 men; mean age 68 years) with pancreatic lesions(n = 38), lymphadenopathy(n = 13), submucosal tumors(n = 4), or others lesions(n = 1) underwent EUS-FNA using both of the needles in a randomized order. AN and PC reached similar overall results for diagnostic accuracy(AN: 88.9 vs PC: 96.1, P = 0.25), specimen adequacy(AN: 96.4% vs PC: 91.1%, P = 0.38), mean number of passes(AN: 1.5 vs PC: 1.7, P = 0.14), mean cellularity score(AN: 1.7 vs PC: 1.1, P = 0.058), and complications(none). A diagnosis on the basis of histology was achieved in the PC group in 36(64.3%) patients, and in 2 of those as the sole modality. | 4 | Biomechanical characteristics of bone in streptozotocin-induced diabetic rats:An in-vivo randomized controlled experimental study显示文摘AIM: To investigate the in vivo effects of type Ⅰdiabetes on the mechanical strength of tibial bone in a rodent model.METHODS: The biomechanical effect of diabetes on the structural integrity of the tibia in streptozotocin induced diabetic Wistar rats was analysed. Induction of diabetes was achieved by an intra-peritoneal injection and confirmed by measuring serial blood glucose levels(> 150 mg/d L). After 8 wk the tibiae were harvested and compared to a control group. Biomechanical analysis of harvested tibiae was performed using a threepoint bending technique on a servo hydraulic MTS 858 MiniB ionix frame. Maximum force applied to failure(N), stiffness(N × mm) and energy absorbed(N/mm) were recorded and plotted on load displacement curves. A displacement control loading mode of 1 mm/min was selected to simulate quasi-static loading conditions. Measurements from load-displacement curves were directly compared between groups.RESULTS: Fourteen streptozotocin induced diabetic Wistar rats were compared against nineteen non-diabetic controls. An average increase of 155.2 g in body weight was observed in the control group compared with only 5 g in the diabetic group during the experimental study period. Levels of blood glucose increased to 440.25 mg/d L in the diabetic group compared to 116.62 mg/d L in the control group.The biomechanical results demonstrate a highly significant reduction in the maximum load to failure from 69.5 N to 58 N in diabetic group compared to control(P = 0.011). Energy absorption to fracture was reduced from 28.2 N in the control group to 23.5 N in the diabetic group(P = 0.082). | 15 | Whole-genome resequencing of Cucurbita pepo morphotypes to discover genomic variants associated with morphology and horticulturally valuable traits显示文摘Cucurbita pepo contains two cultivated subspecies,each of which encompasses four fruit-shape morphotypes(cultivar groups).The Pumpkin,Vegetable Marrow,Cocozelle,and Zucchini Groups are of subsp.pepo and the Acorn,Crookneck,Scallop,and Straightneck Groups are of subsp.ovifera.Recently,a de novo assembly of the C.pepo subsp.pepo Zucchini genome was published,providing insights into its evolution.To expand our knowledge of evolutionary processes within C.pepo and to identify variants associated with particular morphotypes,we performed wholegenome resequencing of seven of these eight C.pepo morphotypes.We report for the first time whole-genome resequencing of the four subsp.pepo(Pumpkin,Vegetable Marrow,Cocozelle,green Zucchini,and yellow Zucchini)morphotypes and three of the subsp.ovifera(Acorn,Crookneck,and Scallop)morphotypes.A high-depth resequencing approach was followed,using the BGISEQ-500 platform that enables the identification of rare variants,with an average of 33.5X.Approximately 94.5%of the clean reads were mapped against the reference Zucchini genome.In total,3,823,977 high confidence single-nucleotide polymorphisms(SNPs)were identified.Within each accession,SNPs varied from 636,918 in green Zucchini to 2,656,513 in Crookneck,and were distributed homogeneously along the chromosomes.Clear differences between subspecies pepo and ovifera in genetic variation and linkage disequilibrium are highlighted.In fact,comparison between subspecies pepo and ovifera indicated 5710 genes(22.5%)with Fst>0.80 and 1059 genes(4.1%)with Fst=1.00 as potential candidate genes that were fixed during the independent evolution and domestication of the two subspecies.Linkage disequilibrium was greater in subsp.ovifera than in subsp.pepo,perhaps reflective of the earlier differentiation of morphotypes within subsp.ovifera.Some morphotype-specific genes have been localized.Our results offer new clues that may provide an improved understanding of the underlying genomic regions involved in the independent evolution and domestication of the two subspecies.Comparisons among SNPs unique to particular subspecies or morphotypes may provide candidate genes responsible for traits of high economic importance. | Aliki Xanthopoulou Javier Montero-Pau Ifigeneia Mellidou Christos Kissoudis JoséBlanca Belén Picó Aphrodite Tsaballa Eleni Tsaliki Athanasios Dalakouras Harry S.Paris Maria Ganopoulou Theodoros Moysiadis Maslin Osathanunkul Athanasios Tsaftaris Panagiotis Madesis Apostolos Kalivas Ioannis Ganopoulos | 2019 | Horticulture Research2019，6，1： | 1 |
