1美洲远志根提取物对苯并芘致肺癌小鼠的抗肿瘤作用(英文)显示文摘目的:研究美洲远志(Polygala senega)根的乙醇提取物对苯并芘致肺癌小鼠的抗肿瘤作用。方法:瑞士白化小鼠被分为5组,每组6只。组1为对照组,予口服橄榄油;组2予苯并芘(50mg/kg体质量,溶于橄榄油中),每周2次,连续4周;组3除予与组2相同的苯并芘外,再予48%乙醇;组4予与组2相同的苯并芘,并同时给予美洲远志的乙醇提取物,每日1次,连续16周;组5仅给予美洲远志的乙醇提取物,每日1次,连续16周。16周后,处死所有小鼠并检测和评价以下指标:2,2-二苯基-1-苦基肼(2,2-diphenyl-1-picrylhydrazyl,DPPH)法测定美洲远志乙醇提取物的抗氧化功效;分别称取各组小鼠的肺组织质量及体质量;彗星实验及酶联免疫吸附法检测DNA损伤情况;分别测定过氧化氢酶、超氧化物歧化酶、谷胱甘肽过氧化物酶、谷胱甘肽还原酶、脂质过氧化反应及总含硫化合物含量。结果:给予美洲远志乙醇提取物治疗的组4小鼠与组3模型组小鼠相比,体质量明显增加而肺组织质量明显降低(P<0.01)。彗星实验结果显示组4小鼠的DNA损伤较组3小鼠明显减轻(P<0.01)。酶联免疫吸附法测定p53蛋白水平,组4明显高于组3(P<0.01)。组2和组3的模型小鼠与组1比较,脂质过氧化反应水平明显升高,而抗氧化标志物的水平明显降低(P<0.05),组4小鼠的这些指标被明显逆转(P<0.01)。结论:美洲远志对于化学制剂致小鼠肺癌有明确的治疗效果。Saili Paul Soumya Sundar Bhattacharyya Asmita Samaddar Naoual Boujedaini Anisur Rahman Khuda-Bukhsh 2011中西医结合学报2011,9,3:4
2Diarylheptanoid-myricanone isolated from ethanolic extract of Myrica cerifera shows anticancer effects on HeLa and PC3 cell lines:signalling pathway and drug-DNA interaction显示文摘OBJECTIVE:To test if myricanone(C_(21)H_(24)O_5),a cyclic diarylheptanoid,has anticancer effects on two different cancer cell lines HeLa and PC3.The present study was conducted with a note on the drug-DNA interaction and apoptotic signalling pathway.METHODS:Several studies like cytotoxicity,nuclear damage,annexin-V-fluorescein isothiocyanate(FITC)/propidium iodide(Pl)-labelled apoptotic assay and cell cycle arrest,immunoblot and reverse transcriptase-polymerase chain reaction(RT-PCR)were used following standard protocols.Circular dichroism(CD)spectroscopy was also done to evaluate whether myricanone effectively interacted with DNA to bring about conformational changes that could strongly inhibit the cancer cell proliferation.RESULTS:Myricanone showed a greater cytotoxic effect on PC3 cells than on HeLa cells.Myricanone promoted G_0/G_1 arrest in HeLa cells and S phase arrest in PC3 cells.Nuclear condensation and annexin V-FITC/PI studies revealed that myricanone promoted apoptotic cell death.CD spectroscopic data indicated that myricanone had an interaction with calf thymus DNA that changed DNA structural conformation.RT-PCR and immunoblot studies revealed that myricanone activated the apoptotic signalling cascades through down-regulation of transcription factors like nuclear factor-κB(NF-κB)(p65),and signal transducers and activators of transcription 3(STAT3);cell cycle regulators like cyclin D1,and survivin and other signal proteins like Bcl-2 and up-regulation of Bax,caspase-9 and caspase-3.CONCLUSION:Myricanone induced apoptosis in both types of cancer cells by triggering caspase activation,and suppression of cell proliferation by down-regulation of NF-κB and STAT3signalling cascades,which makes it a suitable candidate for possible use in the formulation of therapeutic agent for combating cancer.Avijit Paul Sreemanti Das Jayeeta Das Asmita Samadder Kausik Bishayee Ratan Sadhukhan Anisur Rahman Khuda-Bukhsh 2013Journal of Integrative Medicine2013,11,6:4
3Electrochemical characterization of Ni-Co and Ni-Co-Fe for oxidation of methyl alcohol fuel with high energetic catalytic surface显示文摘Non Pt based metals and alloys as electrode materials for methyl alcohol fuel cells have been investigated w ith an aim of finding high electrocatalytic surface property for the faster electrode reactions.Electrodes w ere fabricated by electrodeposition on pure Al foil,from an electrolyte of Ni,Co,Fe salts.The optimum condition of electrodeposition w ere found out by a series of experiments,varying the chemistry of the electrolyte,pH valve,temperature,current and cell potential.Polarization study of the coated Ni-Co or Ni-CoFe alloy on pure Al w as found to exhibit high exchange current density,indicating an improved electro catalytic surface w ith faster charge-discharge reactions at anode and cathode and low overvoltage.Electrochemical impedance studies on coated and uncoated surface clearly show ed that the polarization resistance and impedance w ere decreased by Ni-Co or Ni-Co-Fe coating.X-ray diffraction(XRD),energy dispersive X-ray spectroscopy(EDX)and atomic absorption spectroscopy(AAS)studies confirmed the presence of alloying elements and constituents of the alloy.The morphology of the deposits from scanning electron microscope(SEM)images indicated that the electrode surface w as a three dimensional space w hich increased the effective surface area for the electrode reactions to take place.Subir Paul Sk Naimuddin Asmita Ghosh 2014燃料化学学报2014,42,1:4

5Apoptotic and autophagic death union by Thuja occidentalis homeopathic drug in cervical cancer cells with thujone as the bioactive principle显示文摘Objective‘Multi-targeting’drugs can prove fruitful to combat drug-resistance of multifactorial disease—cervical cancer.This study envisioned to reveal if Thuja homeopathic mother tincture(MT)and its bioactive component could combat human papillomavirus(HPV)-16-infected SiHa cervical cancer cells since it is globally acclaimed for HPV-mediated warts.Methods Thuja MT was studied for its antiproliferative and antimigratory properties in SiHa cells followed by microscopic determination of reactive oxygen species(ROS)generation by 2’,7’-dichlorodihydrofluorescein diacetate(DCFDA)staining and loss in mitochondrial membrane potential(MMP)by rhodamine(Rh)123 staining.Apoptosis and autophagy inductions were studied by acridine orange(AO)ethidium bromide(EB)staining and immunoblot analyses of marker proteins.The bioactive component of Thuja MT detected by gas chromatography-mass spectrometry was studied for antiproliferative and antimigratory properties along with in silico prediction of its cellular targets by molecular docking and oral drug forming competency.Results Thuja MT showed significant antiproliferative and antimigratory potential in SiHa cells at a 50%inhibitory concentration(IC50)dosage of 17.3μL/mL.An increase in DCFDA fluorescence and loss in Rh123 fluorescence prove that Thuja MT acted through the burst of ROS and loss in MMP respectively.AO/EB-stained cells under the microscope and immunoblot analyses supported Thuja-induced cellular demise via dual pathways—apoptosis and autophagy.Immunoblots showed cleavage of caspase-3 and poly adenosine diphosphate-ribose polymerase-1(PARP-1)along with upregulation of Beclin-1,microtubule-associated protein 1 light chain 3B(LC3B)-II,and p62 proteins.Hence,the apoptotic cascade followed a caspase-3-dependent pathway supported by PARP-1 cleavage,while autophagic death was Beclin-1-dependent and mediated by accumulation of LC3BII and p62 proteins.Thujone,detected as the bioactive principle of Thuja MT showed greater anti-proliferative and anti-migratory potential at an IC5077μg/mL along with excellent oral drug competency with the ability for gastrointestinal absorption and blood-brain-barrier permeation with nil toxicity.Molecular docking depicted thujone with the strongest affinity for mammalian target of rapamycin,phosphoinositide 3-kinase,and protein kinase B followed by B-cell lymphoma 2,murine double minute 2 and adenosine monophosphate-activated protein kinase,which might act as upstream triggers of apoptotic-autophagic crosstalk.Conclusion Robust‘multi-targeting’anticancer potential of Thuja drug and thujone for HPV-infected cervical cancer ascertain its therapeutic efficacy for HPV infections.Asmita Pal Sucharita Das Soumalee Basu Rita Kundu 2022Journal of Integrative Medicine2022,20,5:2
6海南蒲桃提取物对L6细胞砷中毒缓解作用的体外实验(英文)显示文摘目的:研究海南蒲桃提取物对砷酸盐引起的L6骨骼肌细胞葡萄糖内稳态破坏及线粒体功能异常的缓解作用。方法:通过测量多个指标如丙酮酸激酶活性、葡糖激酶、线粒体膜电位等衡量细胞内葡萄糖水平及线粒体功能,并测量相关标志物的蛋白质及mRNA表达情况,如葡萄糖转运体4、胰岛素受体基质1、胰岛素受体基质2、葡糖激酶等以分析可能有关的信号通路。结果:海南蒲桃提取物能够通过葡萄糖转运体4通路改善砷中毒的L6细胞内诸多标志物的表达,使其正常化,与模型组相比差异有统计学意义。结论:海南蒲桃提取物对细胞砷中毒有明显的缓解作用,未来可考虑将其用于治疗砷中毒相关疾病如高血糖症等。Asmita Samadder Jayeeta Das Sreemanti Das Raktim Biswas Anisur Rahman Khuda-Bukhsh 2012中西医结合学报2012,10,11:2

13Psorinum 6× triggers apoptosis signals in human lung cancer cells显示文摘OBJECTIVE: To provide in vitro evidence of Psorinum treatment against cancer cells in a controlled study. METHODS: Effects of homeopathic Psorinum 6× on cell viability were initially determined in several cancer cell lines, including A549, Hep G2 and MCF-7, using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay, and an ethanol 6× control. The cell line that exhibited highest inhibition was selected and used in the following experiments. A range of Psorinum 6× doses was used to explore treatment effects on cell cycle arrest, cell death(apoptosis), generation of reactive oxygen species(ROS) and change in mitochondrial membrane potential(MMP) using fl ow cytometry and fl uorescence microscopy, respectively. Expression of several signal proteins related to apoptosis and cell survival were quantified with Western blotting and confocal microscopy. Further, circular dichroism(CD) spectroscopy was used to determine possible drug-DNA interactions, as well as the induction of conformational changes. RESULTS: Treatment of cancer cell lines with Psorinum showed greater anticancer effects in A549 cells than in others. In A549 cells Psorinum treatment inhibited cell proliferation at 24 h after treatment, and arrested cell cycle at sub-G1 stage. It also induced ROS generation, MMP depolarization, morphological changes and DNA damage, as well as externalization of phosphatidyl serine. Further, increases in p53 expression, Bax expression, cytochrome c release, along with reduction of Bcl-2 level and caspase-3 activation were observed after Psorinum 6× treatment, which eventually drove A549 cells towards the mitochondria-mediated caspase-3-dependent pathway. CD spectroscopy revealed direct interaction of Psorinum with DNA, using calf thymus-DNA as target.CONCLUSION: Psorinum 6× triggered apoptosis in A549 cells via both up- and down-regulations of relevant signal proteins, including p53, caspase-3, Bax and Bcl-2.Jesmin Mondal Asmita Samadder Anisur Rahman K huda-Bukhsh 2016Journal of Integrative Medicine2016,14,2:1

18新的聚苯乙烯负载锌化合物:高效苯酚氧化催化剂(英文)显示文摘The novel recyclable free –ONNO– tetradentate Schiff base ligand N,N′‐bis(2‐hydroxy‐3‐ methox‐ybenzaldehyde)4‐methylbenzene‐1,2‐diamine (3‐MOBdMBn) was synthesized. Complexation of this ligand with zinc(3‐MOBdMBn‐Zn) was performed, and the catalytic activity of the complex was evaluated. The polymer‐supported analog of this complex(P‐3‐MOBdMBn‐Zn) was synthesized, and its catalytic activity was studied. These free and polymer‐anchored zinc complexes were prepared by the reactions of metal solutions with one molar equivalent of unsupported 3‐MOBdMBn or P‐3‐MOBdMBn in methanol under nitrogen. The catalytic activity of 3‐MOBdMBn‐Zn and P‐3‐MOBdMBn‐Zn was evaluated in phenol oxidation. The activity of P‐3‐MOBdMBn‐Zn was signif‐icantly affected by the polymer support, and the rate of phenol conversion was around 50% for polystyrene‐supported 3‐MOBdMBn. 20变黑白头翁提取物降低亚砷酸盐对小鼠精细胞的毒性(英文)显示文摘目的:研究变黑白头翁提取物降低亚砷酸盐对小鼠精细胞毒性的作用。方法:给予实验小鼠亚砷酸钠(20mg/kg每日),并分别在第30、60、90天对小鼠进行检测。亚砷酸盐中毒小鼠被分为2组,其中一组给予变黑白头翁提取物(35mg/kg),另一组给予85%乙醇。通过检测小鼠精细胞凋亡标志蛋白CYP1A1、p53及caspase3的活性,确定细胞及DNA的损伤情况,并测定睾丸毒性标志物的水平。通过圆二色光谱仪及熔化温度数据检测变黑白头翁提取物与DNA的相互作用情况。结果:亚砷酸盐中毒小鼠的所有细胞凋亡标志蛋白及睾丸毒性标志物的水平均有所上升,而经变黑白头翁提取物治疗的小鼠上述各项指标均降低或恢复至正常水平。变黑白头翁提取物与DNA相互作用,引起了DNA结构和构象的变化。结论:变黑白头翁提取物可作为砷中毒引起的生殖功能损害的治疗药物给予研究和开发。Asmita Samadder Jayeeta Das Sreemanti Das Durba Das Arnab De Kakali Bhadra Anisur Rahman Khuda-Bukhsh 2012中西医结合学报2012,10,12:1
