| 1 | Aortic heterogeneity across segments and under high fat/salt/glucose conditions at the single-cell level显示文摘The aorta,with ascending,arch,thoracic and abdominal segments,responds to the heartbeat,senses metabolites and distributes blood to all parts of the body.However,the heterogeneity across aortic segments and how metabolic pathologies change it are not known.Here,a total of 216612 individual cells from the ascending aorta,aortic arch,and thoracic and abdominal segments of mouse aortas under normal conditions or with high blood glucose levels,high dietary salt,or high fat intake were profiled using single-cell RNA sequencing.We generated a compendium of 10 distinct cell types,mainly endothelial(EC),smooth muscle(SMC),stromal and immune cells.The distributions of the different cells and their intercommunication were influenced by the hemodynamic microenvironment across anatomical segments,and the spatial heterogeneity of ECs and SMCs may contribute to differential vascular dilation and constriction that were measured by wire myography.Importantly,the composition of aortic cells,their gene expression profiles and their regulatory intercellular networks broadly changed in response to high fat/salt/glucose conditions.Notably,the abdominal aorta showed the most dramatic changes in cellular composition,particularly involving ECs,fibroblasts and myeloid cells with cardiovascular risk factor-related regulons and gene expression networks.Our study elucidates the nature and range of aortic cell diversity,with implications for the treatment of metabolic pathologies. | Dongxu He Aiqin Mao Chang-Bo Zheng Hao Kan Ka Zhang Zhiming Zhang Lei Feng Xin Ma | 2020 | National Science Review2020,7,5: | 2 |
| 2 | Gene analysis of an antiviral protein SP-2 from Chinese wild silkworm, Bombyx mandarina Moore and its bioactivity assay显示文摘The cDNA encoding an antiviral protein SP-2 against BmNPV was cloned from the midgut of Chinese wild silkworm, Bombyx mandarina Moore (GenBank access AY945210) based on the available informa- tion of the domesticated silkworm. Its cDNA was 855 bp encoding 284 amino acids with predicted mo- lecular weight of 29.6 kDa. Its full length in genomics was 1376 bp, including 5 exons and 4 introns. The expression analysis indicated that it was only expressed in midgut, and its expression level was higher during feeding stage of larval instars while very lower during the moltism and mature stages. The de- duced amino acid sequence of this protein showed eight-amino-acid variation compared with the counterpart of domesticated silkworm. Its antiviral activity was assayed through in vitro test. | 5 | 低剂量辐射对环磷酰胺化疗毒性作用的影响(英文)显示文摘Objective:The aim of the study was to investigate the effects as well as the possible mechanisms of low dose γ-ray pre-irradiation on hepatic damage,DNA damage of peripheral lymphocytes and genetic material damage caused by high dosage of cyclophosphamide(CTX).Methods:Kunming strain male mice were randomly divided into five groups:control group,sham-irradiated group,low dose irradiation group(LDR group),cyclophosphamide chemotherapy group(CTX group) and low dose irradiation combined with chemotherapy group(LDR + CTX group).Having being raised for one week,all the mice were implanted subcutaneously with S180 cells in the left inguen(control group excluded).On days 8 and 11,mice of LDR and LDR + CTX groups were given 75 mGy whole-body γ-irradiation,30 h later mice of CTX and LDR + CTX groups were injected i.p.3.0 mg cyclophosphamide.All the mice were sacrificed on day 13.DNA damage of the peripheral lymphocytes was analyzed using single cell gel electrophoresis(SCGE);ALT activity,total protein(TP) and albumin(ALB) of the plasma were analyzed using automatic biochemistry analyzer;MDA content,SOD and GSH-PX activity of the hepatic homogenate were analyzed using chromometry;genetic material damage was analyzed using micronucleus frequency(MNF) of polychromatoerythrocytes(PCE) in bone marrow.Results:1.Differences of MDA contents,SOD and GSH-PX activity of hepatic homogenate between 5 groups had notable statistical significance(P < 0.01);in control group MDA content was the lowest,SOD and GSH-PX activity were the highest,while in CTX group MDA content was the highest,SOD and GSH-PX activity were the lowest;compared with CTX group MDA content decreased significantly(P < 0.01) and SOD and GSH-PX activity increased significantly(P < 0.05) in LDR + CTX group.2.Differences of ALT activity of plasma between 5 groups had no statistical significance(F = 1.262,P > 0.05).Differences of TP and ALB of plasma between 5 groups had statistical significance(F = 12.879 and 6.336 respectively,P < 0.01);TP and ALB in control group were higher than those of other groups and compared with sham-irradiated group,TP and ALB in LDR group elevated significantly(P < 0.05).3.Differences of DNA damage of peripheral lymphocytes had notable statistical significance(F = 6.383,P < 0.01);DNA damage in control group was the lightest,while DNA damage in CTX group was the severest;compared with CTX group,DNA damage in LDR + CTX group was much lighter(P < 0.05).4.MNF of PCE between 5 groups had remarkable significance(F = 179.652,P < 0.01);compared with control group and sham-irradiated group,MNF in CTX group increased significantly(P < 0.01);compared with CTX group,MNF in LDR + CTX group had a tendency of decline,which had no statistical significance(P > 0.05).Conclusion:1.CTX can damage the hepatic tissue through oxidative stress;75 mGy γ-irradiation before CTX chemotherapy can induce activities of anti-oxidative enzymes,promote elimination of free radicals,so as to alleviate the damaging effects of oxidative stress to hepatic tissue caused by high-dose chemotherapy.2.A 75 mGy γ-irradiation before CTX chemotherapy has no obvious effect on ALT activity of plasma,but may have protective effect on the protein synthesis function of liver.3.High-dose CTX chemotherapy can cause DNA damage of peripheral lymphocytes;75 mGy γ-irradiation before chemotherapy may have certain protective effect on DNA damage.4.CTX has potent mutagenic effect,can cause significant increase of MNF of PCE;75 mGy γ-ray pre-irradiation did not show obvious protection against genetic toxicity of high-dose CTX chemotherapy. | Hongsheng Yu Li Yu Aiqin Song Zimin Liu Yeling Zhang Xinjia He | 2011 | The Chinese-German Journal of Clinical Oncology2011,10,2: | 1 |

| 7 | Knockdown Annexin A8 inhibits the proliferation and invasion of cervical cancer cells显示文摘This study aimed to explore the expression,function,and molecular mechanism of ANXA8,the gene for annexin 8,in cervical cancer.Methods:The gene expression of the ANX family members in cervical cancer tissues was classified via The Cancer Genome Atlas(TCGA)database.The expression of ANXA8 in paracancerous tissues,cervical cancer tissues,and cell lines was identified by fluorescence quantitative polymerase chain reaction(PCR)and immunohistochemistry.The effects of ANXA8 knockdown on the cellular growth and cell invasion of cervical cancer were examined by MTT,clone-formation assay,scratch test,and Transwell assay.The effect of ANXA8 knockdown on the proliferative potency of cervical cancer cells was assessed through an in vivo nude-mouse tumor-formation test.The gene expression levels of Ki-67 and ANXA8 in tumor-bearing tissues were measured through Immunohistochemical analysis.Results:TCGA data analysis and fluorescence quantitative PCR exhibited significantly increased expression of ANXA8 in cervical cancer tissues.Further examination exhibited that ANXA8 was expressed exceedingly in cervical cancer tissue,and it was associated with lymph node metastasis,FIGO stage,degree of differentiation,and infiltration depth of cancer patients.Cell-function assays revealed that knocking down ANXA8 may substantially suppress the propagation,colony formation,invasion,and migration of cervical cancer cells.In vivo experiments demonstrated that ANXA8 knockdown had a substantial inhibitory effect on the propagation of cervical cancer cells in nude mice and inhibited the expression of Ki-67.Conclusion:ANXA8 is specifically and significantly upregulated within cervical cancer tissues.The knockdown of this gene showed remarkable outcomes,as evidenced by the substantial inhibition of cervical cancer cell proliferation in in vitro and in vivo experimentations.Therefore,ANXA8 is a potential target or a marker that can serve as a therapeutic or diagnostic tool for cervical cancer. | WEILING ZHANG YONG LI CAN ZHANG QING HAN YU ZHANG AIQIN HE WEIPEI ZHU | 2023 | BIOCELL2023,47,12: | 0 |
| 8 | Knockdown of circular RNA (CircRNA)_001896 inhibits cervical cancer proliferation and stemness in vivo and in vitro显示文摘Objective:Previous studies indicated that aberrant circular RNA(circRNA)expression affects gene expression regulatory networks,leading to the aberrant activation of tumor pathways and promoting tumor cell growth.However,the expression,clinical significance,and effects on cell propagation,invasion,and dissemination of circRNA_001896 in cervical cancer(CC)tissues remain unclear.Methods:The Gene Expression Omnibus(GEO)datasets(GSE113696 and GSE102686)were used to examine differential circRNA expression in CC and adjacent tissues.The expression of circRNA_001896 was detected in 72 CC patients usingfluorescence quantitative PCR.Correlation analysis with clinical pathological features was performed through COX multivariate and univariate analysis.The effect of circRNA_001896 downregulation on CC cell propagation was examined using the cell counting kit-8(CCK-8)test,clonogenic,3D sphere formation,and in vivo tumorigenesis assays.Results:Intersection of the GSE113696 and GSE102686 datasets revealed an increased expression of four circRNAs,including circRNA_001896,in CC tissues.Fluorescence quantitative PCR confirmed circRNA_001896 as a circular RNA.High expression of circRNA_001896 was considerably associated with lymph node metastasis,International Federation of Gynecologists and Obstetricians(FIGO)stage,tumor diameter,and survival period in CC patients.Proportional hazards model(COX)univariate and multivariate analyses revealed that circRNA_001896 expressions are a distinct risk factor affecting CC patients’prognosis.Cellular functional experiments showed that downregulating circRNA_001896 substantially suppressed CC cell growth,colony formation,and 3D sphere-forming ability.In vivo,tumorigenesis analysis in nude mice demonstrated that downregulating circRNA_001896 remarkably reduced the in vivo proliferation capacity of CC cells.Conclusion:CircRNA_001896 is highly expressed in CC tissues and is substantially related to lymph node metastasis,FIGO stage,tumor size,and survival period in patients.Moreover,downregulating circRNA_001896 significantly inhibits both in vivo and in vitro propagation of CC cells.Therefore,circRNA_001896 might be used as a biomarker for targeted therapy in cervical cancer. | JIA SHAO CAN ZHANG YAONAN TANG AIQIN HE WEIPEI ZHU | 2024 | BIOCELL2024,48,4: | 0 |
| 9 | Carboxymethyl chitosan/poly(ethylene oxide)water soluble binder:Challenging application for 5V LiNi0.5Mn1.5O4 cathode显示文摘Carboxymethyl chitosan/poly(ethylene oxide)(CCTS/PEO) composite is firstly reported as a water soluble binder for the application of 5V LiNi_(0.5)Mn_(1.5)O_4 cathode in Li-ion batteries. Both CCTS and PEO show a high electrochemical oxidation potential of above 5.0 V(vs. Li/Li+). The electrochemical performances of LiNi_(0.5)Mn_(1.5)O_4(LNMO) cathodes with CCTS/PEO composite binders of different mass rates are investigated, it is found that LiNi_(0.5)Mn_(1.5)O_4 cathode with an optimized CCTS/PEO(85/15, w/w) composite exhibits a slightly better cycling performance than that of polyvinylidene fluoride(PVDF), retaining 81.4% capacity as compared with 79.8% for PVDF at 0.5C rate after 200 cycles. LNMO with PEO/CCTS(85/15,w/w) exhibited the better rate capability than that of PVDF. | 10 | Physiological levels of fluid shear stress modulate vascular function through TRPV4 sparklets显示文摘Endothelial calcium(Ca^(2+))signaling plays a major role in regulating vasodilation in response to fluid shear stress(FsS)generated by blood flow.Local Ca^(2+)influx through single transient receptor potential channel subfamily V member4(TRPV4)(termed'sparklets')activated bylowconcentrations of chemical andbiological stimuli has been revealed to modulate vascular function.However,the range in which FsS can initiate TRPV4 sparklets to induce vasodilation is unknown.Here,we assess theactivity of TPRV4 sparklets inducedbyvarious physiological levels of FSS and investigate the mechanisms involving these Ca?+signals in FsS-induced vasodilation.Intact small mesenteric arteries are used for Ca^(2+)imaging with a GCaMP2(TRPV4-KO)mouse model and high-speed confocal systems.Markedly increased local Ca^(2+)signals are observed in the endothelium under 4-8 dyne/cm^(2)FsS,whereas FSS>8 dyne/cm^(2)causes global Ca^(2+)influx.Further analysis shows that TRPV4 channels form a four-channel group to mediate Ca^(2+)sparklets under certain levels of Fss.The large Ca^(2+)influx hyperpolarizes endothelial cells by stimulating intermediate(IK)-and small(SK)-conductance Ca^(2+)-sensitive potassium channels,leading to hyperpolarization of the surrounding smooth muscle cells and ultimately causing endothelium-dependent vasodilation.In conclusion,Ca^(2+)influx transits through a small number of endothelial TRPV4 channels opened by certain levels of FsS,which activates the Ca^(2+)-sensitive IK and SK channels to cause vasodilation. | Li Geng Chi Zhang Chao He Ka Zhang Hao Kan Aiqin Mao Xin Ma | 2022 | Acta Biochimica et Biophysica Sinica2022,54,9: | 0 |
| 11 | Spatial patterns of phytoplankton communities in an International Seabed Authority licensed area(COMRA,Clarion-Clipperton Zone)in relation to upper ocean biogeochemistry显示文摘The Clarion-Clipperton Zone(CCZ)hosts one of the largest known oceanic nodule fields worldwide and is regulated by the International Seabed Authority.A baseline assessment of diversity and distribution patterns is essential for reliable predictions of disturbed ecosystem response scenarios for sustained commercial activities in the future.In the present study,the spatial patterns and diversity of phytoplankton communities were analyzed along with upper ocean biogeochemistry,in the licensed China Ocean Mineral Resources R&D Association(COMRA)contract area and the surrounding western CCZ between August 21 and October 8,2017.Results indicated this was a typical low-nutrient low-chlorophyll a(Chl a)environment,characterized by low levels of phytoplankton abundance and diversity.In total 112 species belonging to 4 phyla were recorded(>10μm),with species counts including 82 diatoms,27 dinoflagellates,1 cyanobacteria and 2 chrysophyte.Dominant taxa in successive order of descending abundance and occurrence included Nizschia marina,Cyclotella stylorum,Dactyliosolen mediterraneus,Rhizosolenia setigera,Pseudo-nitzschia delicatissima,Thalassiothrix frauenfeldii,Synedra sp.,Chaetoceros simplex and Pseudo-nitzschia circumpora.The depth-averaged abundance and Chl a concentrations were(265±233)cells/L and(0.27±0.30)μg/L,respectively.Diatoms accounted for 90.94%of the community with(241±223)cells/L,while dinoflagellates accounted for 5.67%and(15±13)cells/L.The distribution pattern exhibited the same trend as abundance,Chl a and species richness,showing subsurface maximum levels at around 100 m,with stations near 10°N having higher levels than in the north.Cluster analysis was performed in two assemblages,relating to geographic locations to the south and north of 12°N.The subsurface maximum of abundance,Chl a,species richness,dissolved oxygen and nitrite were generally corresponding to the presence of high salinity North Pacific Central Water at depths of 50-120 m.Higher availability of nitrate,phosphate and silicic acid in the subsurface may account for the shift in phytoplankton distribution,as shown by redundancy correspondence and spearman correlation analysis.Diel variation in an anchor station demonstrated prominent species succession without significant differences in oceanographic variables,among which diatoms succession resulted from the light limitation,while dinoflagellate diel variation mainly related to lateral transport of water masses.The observed patchiness in spatial phytoplankton distributional patterns was attributed to upper ocean environmental gradients in the CCZ.The baseline generated in this study could be analyzed using current conservation strategy programs associated with deep-sea mining. | Yu Wang Aiqin Han Xuebao He Fangfang Kuang Feng Zhao Peng Xiang Kuidong Xu | 2022 | Acta Oceanologica Sinica2022,41,11: | 0 |
