1A holistic strategy for quality and safety control of traditional Chinese medicines by the “iVarious” standard system显示文摘An effective quality control system is the key to ensuring the quality, safety and efficacy of traditional Chinese medicines(TCMs). However, the current quality standard research lacks top-level design and systematic design,mostly based on specific technologies or evaluation methods. To resolve the challenges and questions of quality control of TCMs, a brand-new quality standard system, named 'iVarious', was proposed. The system comprises eight elements in a modular format. Meaning of every element was specifically illustrated via corresponding research instances. Furthermore, frankincense study was taken as an example for demonstrating standards and research process, based on the 'i Various' system. This system highlighted a holistic strategy for effectiveness,security, integrity and systematization of quality and safety control standards of TCMs. The establishment of'i Various' integrates multi-disciplinary technologies and progressive methods, basis elements and key points of standard construction. 2PARylation regulates stress granule dynamics, phase separation, and neurotoxicity of disease-related RNA-binding proteins显示文摘Mutations in RNA-binding proteins (RBPs) localized in ribonucleoprotein (RNP) granules, such as hnRNP A1 and TDP-43, promote aberrant protein aggregation, which is a pathological hallmark of various neurodegenerative diseases, such as amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). Protein posttranslational modifications (PTMs) are known to 佗gulate RNP granules. In this study, we investigate the function of poly(ADP-ribosyl)ation (PARylation), an important PTM involved in DNA damage repair and cell death, in RNP granule-related neurodegeneration. We reveal that PARylation levels are a major regulator of the assembly-disassembly dynamics of RNP granules containing disease-related RBPs, hnRNP A1 and TDP-43. We find that hnRNP A1 can both be PARylated and bind to PARylated proteins or poly(ADP-ribose)(PAR). We further uncover that PARylation of hnRNP A1 at K298 controls its nucleocytoplasmic transport, whereas PAR-binding via the PAR-binding motif (PBM) of hnRNP Al regulates its association with stress granules. Moreover, we reveal that PAR not only dramatically enhances the liquid-liquid phase separation of hnRNP A1, but also promotes the co-phase separation of hnRNP A1 and TDP-43 in vitro and their interaction in vivo. Finally, both genetic and pharmacological inhibition of PARP mitigates hnRNP Al- and TDP-43-mediated neurotoxicity in cell and Drosophila models of ALS. 3Comprehensive analysis of NAC transcription factors in diploid Gossypium: sequence conservation and expression analysis uncover their roles during fiber development显示文摘Determining how function evolves following gene duplication is necessary for understanding gene expansion.Transcription factors(TFs)are a class of proteins that regulate gene expression by binding to specific cis-acting elements in the promoters of target genes,subsequently activating or repressing their transcription.In the present study,we systematically examined the functional diversification of the NAC transcription factor(NAC-TFs)family by analyzing their chromosomal location,structure,phylogeny,and expression pattern in Gossypium raimondii(Gr)and G.arboreum(Ga).The 145 and 141 NAC genes identified in the Gr and Ga genomes,respectively,were annotated and divided into 18 subfamilies,which showed distinct divergence in gene structure and expression patterns during fiber development.In addition,when the functional parameters were examined,clear divergence was observed within tandem clusters,which suggested that subfunctionalization had occurred among duplicate genes.The expression patterns of homologous gene pairs also changed,suggestive of the diversification of gene function during the evolution of diploid cotton.These findings provide insights into the mechanisms underlying the functional differentiation of duplicated NAC-TFs genes in two diploid cotton species.Haihong Shang Zhongna Wang Changsong Zou Zhen Zhang Weijie Li Junwen Li Yuzhen Shi Wankui Gong Tingting Chen Aiying Liu Juwu Gong Qun Ge Youlu Yuan 2016Science China(Life Sciences)2016,59,2:12
4Influence of Substrate Negative Bias on Structure and Properties of TiN Coatings Prepared by Hybrid HIPIMS Method显示文摘TiN coatings were deposited using a hybrid home-made high power impulse magnetron sputtering(HIPIMS)technique at room temperature.The effects of substrate negative bias voltage on the deposition rate,composition,crystal structure,surface morphology,microstructure and mechanical properties were investigated.The results revealed that with the increase in bias voltage from-50 to-400 V,TiN coatings exhibited a trend of densification and the crystal structure gradually evolved from(111) orientation to(200)orientation.The growth rate decreased from about 12.2 nm to 7.8 nm per minute with the coating densification.When the bias voltage was-300 V,the minimum surface roughness value of 10.1 nm was obtained,and the hardness and Young's modulus of TiN coatings reached the maximum value of 17.4 GPa and 263.8 GPa,respectively.Meanwhile,the highest adhesion of 59 N was obtained between coating and substrate.Zhenyu Wang Dong Zhang Peiling Ke Xincai Liu Aiying Wang 2015Journal of Materials Science & Technology2015,31,1:10
5Preparation of Ti_2AlC MAX Phase Coating by DC Magnetron Sputtering Deposition and Vacuum Heat Treatment显示文摘Due to the excellent corrosion resistance and high irradiation damage resistance,Ti_2AlC MAX phase is considered as a candidate for applications as corrosion resistant and irradiation resistant protective coating.MAX phase coatings can be fabricated through firstly depositing a coating containing the three elements M,A,and X close to stoichiometry of the MAX phases using physical vapor deposition,followed by heat treatment in vacuum.In this work,Ti-Al-C coating was prepared on austenitic stainless steels by reactive DC magnetron sputtering with a compound Ti_(50)Al_(50) target,and CH4 used as the reactive gas.It was found that the as-deposited coating is mainly composed of Ti_3AlC antiperovskite phase with supersaturated solid solution of Al.Additionally,the ratio of Ti/Al remained the same as that of the target composition.Nevertheless,a thicker thermally grown Ti_2AlC MAX phase coating was obtained after being annealed at 800℃ in vacuum for 1 h.Meanwhile,the ratio of Ti/Al became close to stoichiometry of Ti_2AlC MAX phases.It can be understood that owing to the higher activity of Al,it diffused quickly into the substrate during annealing,and then more stable Ti_2AlC MAX phases transformed from the Ti_3AlC antiperovskite phase.Zongjian Feng Peiling Ke Aiying Wang 2015Journal of Materials Science & Technology2015,31,12:10
6Implications from protein uptake kinetics onto dextran-grafted Sepharose FF coupled with ion exchange and affinity ligands显示文摘Our previous studies have reported the presence of 'chain delivery' effects of protein adsorption onto ion exchangers with polymer-grafted ion-exchange groups, such as dextran-grafted and poly(ethylenimine)-modified Sepharose gels. However, it is unclear if the 'chain delivery' occurs on affinity adsorption with specific interactions. This work is designed to address this issue. A dextran-grafted Sepharose gel was prepared, and then the matrix was modified using diethylaminoethyl, a typical ion-exchange group, or octapeptide(FYCHWQDE), an affinity ligand for human immunoglobulin G(h Ig G) to prepare ion-exchange or affinity adsorbents, respectively.Results of h Ig G adsorption showed that the uptake rate represented by the effective diffusivity of h Ig G onto the dextran-grafted ion exchangers was obviously enhanced by the dextran grafting, indicating the presence of'chain delivery' of the bound proteins on the charged groups on the dextran chains. By contrast, the effective diffusivity of h Ig G changed little as ligand density increased on the dextran-grafted FYCHWQDE adsorbents.Their adsorption capacities decreased and effective diffusivities were not accelerated by the dextran grafting.Thus, this work clarified that grafted dextran could not accelerate h Ig G uptake rate on the affinity resins, or in other words, chain delivery did not occur on the specific interaction-based affinity adsorption.Aiying Xue Linling Yu Yan Sun 2017Chinese Journal of Chemical Engineering2017,25,7:10
7Correlation between BRAF^(V600E) mutation and clinicopathological features in pediatric papillary thyroid carcinoma显示文摘In adults, the presence of the BRAF^(V600E) mutation in papillary thyroid cancer(PTC) has been demonstrated to be strongly associated with aggressive cancer-cell characteristics and poor patient prognosis. In contrast, the frequency of this mutation in pediatric PTC has undergone limited study, and the few available estimates range from 0 to 63%. Furthermore, the role of the BRAF^(V600E) mutation in pediatric PTC is controversial; thus, the present study aimed to investigate the prevalence and role of the BRAF^(V600E) mutation in48 pediatric patients with PTC, aged 3–13 years. Of these patients, 41 were diagnosed with classic PTC, five were found to have a follicular variant of PTC, and two to exhibit a diffuse sclerosing PTC variant. The BRAF^(V600E) mutation was identified to be present in 35.4% of the 48 analyzed patients, and in 41.5% of the patients diagnosed with classical PTC. Furthermore, the presence of the BRAF^(V600E) mutation was found to be associated with a patient age at diagnosis of less than ten years(P=0.011), the performance of a thyroidectomy(P=0.03), exhibited tumor multifocality(P=0.02) and/or extra-thyroidal invasion(P=0.003), and both a low MACIS(Metastases, Age, Completeness of resection, Invasion, Size)(P=0.036) and AMES(Age, Metastasis, Extent of tumor,Size)(P=0.001)score. 8Manipulation of SlMXl for enhanced carotenoids accumulation and drought resistance in tomato显示文摘Plants are the ultimate source of nutrients in the human diet. To ensure adequate availability of high quality food for an increasing world population, traits including improved tolerance of stresses and nutrient levels need to be selected in crops, both individually and in combination. Here we report the identification of Sl MX1 encoding a MIXTA-like MYB transcription factor in tomato that simultaneously modulates drought resistance and metabolic processes through regulating key structural and regulatory genes of the corresponding pathways. Over-expression of Sl MX1 results in substantially increased drought tolerance and improved fruit quality, while knocking down Sl MX1 resulted in the opposite phenotypes. 9Erosion behavior and failure mechanism of Ti/TiAlN multilayer coatings eroded by silica sand and glass beads显示文摘Severe erosion by hard particles is a crucial problem to engine blades when aircraft take off and land in harsh environments, especially for the developed lightweight titanium alloy components. Here, we deposited the Ti/TiAlN multilayer coatings with various cycles on Ti–6 Al–4 V substrates by a home-made hybrid multisource cathodic arc system. The effects of the silica sand and glass beads on erosion behavior of the coatings were focused. Results showed that the Ti/TiAlN multilayer coatings eroded by the silica sand exhibited the predominant 'layer by layer' failure mechanism. In particular, increasing the number of cycles led to the dramatic increase in erosion rate for Ti/TiAlN multilayer coatings, due to the deterioration of their mechanical properties. Different from the silica sand case, however, the erosion rate of the coatings treated by glass beads indicated faint dependence upon the number of cycles, where the coating failure was dominated by the 'piece by piece' failure mechanism. Noted that the Ti layers along with the formed interfaces enhanced the erosion resistance of the coatings, although the failure mechanisms were differently eroded by silica sand and glass beads. 10Constructing a high-density linkage map for Gossypium hirsutum × Gossypium barbadense and identifying QTLs for lint percentage显示文摘To introgress the good fiber quality and yield from Gossypium barbadense into a commercial Upland cotton variety, a high-density simple sequence repeat(SSR) genetic linkage map was developed from a BC1F1 population of Gossypium hirsutum×Gossypium barbadense. The map comprised 2,292 loci and covered 5115.16 centi Morgan(c M) of the cotton AD genome, with an average marker interval of 2.23 c M. Of the marker order for 1,577 common loci on this new map, 90.36% agrees well with the marker order on the D genome sequence genetic map. Compared with five published high-density SSR genetic maps, 53.14% of marker loci were newly discovered in this map. Twenty-six quantitative trait loci(QTLs) for lint percentage(LP) were identified on nine chromosomes. Nine stable or common QTLs could be used for marker-assisted selection. Fifty percent of the QTLs were from G. barbadense and increased LP by 1.07%–2.41%. 12Cluster observations of large-scale southward movement and dawnward-duskward flapping of Earth's magnetotail current sheet显示文摘In August 2001,Cluster satellites observed that the mid-tail current sheet(CS) moved southward continuously for almost seven hours.Meanwhile,Cluster crossed back and forth the CS repeatedly.This means that the large-scale southward movement of the CS was accompanied by a small-scale CS flapping during this period.Using the minimum-variation-analysis(MVA) method and the multi-spacecraft data,we calculated the normal vector,current density and the magnetic curvature of the CS,the results showed that the CS alternated between flattened CS and tilted CS for several times.Strong dawn-dusk oscillations were found for the tilted CS,which caused the repeated crossings of the center of CS by the satellites.This feature is obviously different from the previous observations of the vertical flapping of the CS induced by the kink instability.Two types of flapping were observed:One of them is accompanied with bursty bulk flows(BBFs) and the other is not.This suggests that in this event there was no direct relationship between the CS flapping and BBFs.DUAN AiYing CAO JinBin MA YuDuan WEI XinHua 2013Science China(Technological Sciences)2013,56,1:6

153D MHD simulation of the double-gradient instability of the magnetotail current sheet显示文摘Flapping motion of the current sheet(CS) is an important physical process in the Earth's magnetotail. The magnetic doublegradient model, which includes both the instability and wave modes, offers a reasonable explanation for the exciting and propagation of the flapping wave. In this paper, we apply an advanced numerical magnetohydrodynamic(MHD) scheme(conservation element and solution element(CESE)-MHD) to simulate the magnetic double-gradient instability in an idealized current sheet that mimics the magnetotail configuration. We initialize the simulations with a numerically relaxed magnetotail equilibrium, in which the normal component of the magnetic field has a tailward gradient. It is confirmed in our simulation that the instability develops in the current layer. The growth rate of the instability yielded from the simulation is very close to the prediction of theory, with a relative deviation of only ten percent. 16QTL mapping for fiber quality and yieldrelated traits across multiple generations in segregating population of CCRI 70显示文摘Background:Cotton is a significant economic crop that plays an indispensable role in many domains.Gossypium hirsutum L.is the most important fiber crop worldwide and contributes to more than 95%of global cotto n production.Identifying stable quantitative trait locus(QTLs)controlling fiber quality and yield related traits are necessary prerequisites for marker-assisted selection(MAS).Results:A genetic linkage map was constructed with 312 simple sequence repeat(SSR)loci and 35 linkage groups using JoinMap 4.0;the map spanned 1 929.9 cM,with an average interval between two markers of 6.19 cM,and covered approximately 43.37%of the cotton genome.A total of 74 QTLs controlling fiber quality and 41 QTLs controlling yield-related traits were identified in 4 segregating generations.These QTLs were distributed across 20 chromosomes and collectively explained 1.01%?27.80%of the observed phenotypic variations.In particular,35 stable QTLs could be identified in multiple generations,25 common QTLs were con sistent with those in previous studies,and 15 QTL clusters were found in 11 chromosome segments.Conclusion:These studies provide a theoretical basis for improving cotton yield and fiber quality for molecular marker-assisted selection.DENG Xiaoying GONG Juwu LIU Aiying SHI Yuzhen GONG Wankui GE Qun LI Junwen SHANG Haihong WU Yuxiang YUAN Youlu 2019Journal of Cotton Research2019,2,2:3
