Deciphering the role of PGC-1α in neurological disorders: from mitochondrial dysfunction to synaptic failure显示文摘The onset and mechanisms underlying neurodegenerative diseases remain uncertain. The main features of neurodegenerative diseases have been related with cellular and molecular events like neuronal loss, mitochondrial dysfunction and aberrant accumulation of misfolded proteins or peptides in specific areas of the brain. The most prevalent neurodegenerative diseases belonging to age-related pathologies are Alzheimer's disease, Huntington's disease, Parkinson's disease and amyotrophic lateral sclerosis. Interestingly, mitochondrial dysfunction has been observed to occur during the early onset of several neuropathological events associated to neurodegenerative diseases. The master regulator of mitochondrial quality control and energetic metabolism is the transcriptional coactivator peroxisome proliferator-activated receptor gamma coactivator 1-alpha(PGC-1α). Additionally, it has been observed that PGC-1α appears to be a key factor in maintaining neuronal survival and synaptic transmission. In fact, PGC-1α downregulation in different brain areas(hippocampus, substantia nigra, cortex, striatum and spinal cord) that occurs in function of neurological damage including oxidative stress, neuronal loss, and motor disorders has been seen in several animal and cellular models of neurodegenerative diseases. Current evidence indicates that PGC-1α upregulation may serve as a potent therapeutic approach against development and progression of neuronal damage. Remarkably, increasing evidence shows that PGC-1α deficient mice have neurodegenerative diseases-like features, as well as neurological abnormalities. Finally, we discuss recent studies showing novel specific PGC-1α isoforms in the central nervous system that appear to exert a key role in the age of onset of neurodegenerative diseases and have a neuroprotective function in the central nervous system, thus opening a new molecular strategy for treatment of neurodegenerative diseases. The purpose of this review is to provide an up-to-date overview of the PGC-1α role in the physiopathology of neurodegenerative diseases, as well as establish the importance of PGC-1α function in synaptic transmission and neuronal survival.Jessica D.Panes Aline Wendt Oscar Ramirez-Molina Patricio A.Castro Jorge Fuentealba 2022Neural Regeneration Research2022,17,2:6
The inception of a Paleotethyan magmatic arc in Iberia显示文摘This paper presents a compilation of recent U-Pb(zircon) ages of late Carboniferous—early Permian(LCEP) calc-alkaline batholiths from Iberia, together with a petrogenetic interpretation of magma generation based on comparisons with Mesozoic and Tertiary Cordilleran batholiths and experimental melts.Zircon U-Pb ages distributed over the range ca.315—280 Ma,indicate a linkage between calc-alkaline magmatism,Iberian orocline generation and Paleotethys subduction. It is also shown that Iberian LC-EP calcalkaline batholiths present unequivocal subduction-related features comparable with typical Cordilleran batholiths of the Pacific Americas active margin, although geochemical features were partially obscured by local modifications of magmas at the level of emplacement by country rock assimilation. When and how LC-EP calc-alkaline batholiths formed in Iberia is then discussed, and a new and somewhat controversial interpretation for their sources and tectonic setting(plume-assisted relamination) is suggested. The batholiths are proposed to have formed during the subduction of the Paleotethys oceanic plate(Pangaea self-subduction) and, consequently, they are unrelated to Variscan collision. The origin of the Iberian batholiths is related to the Eurasian active margin and probably represents the inception of a Paleotethyan arc in the core of Pangaea.M.F.Pereira A.Castro C.Fernández 2015Geoscience Frontiers2015,6,2:3

SARS-CoV-2 productively infects primary human immune system cells in vitro and in CoviD-19 patients显示文摘The severe acute respiratory syndrome coronavirus 2(SARS-CovV-2)infection is associated with a hyperinflammatory state and lymphocytopenia,a hallmark that appears as both signature and prognosis of disease severity outcome.Although cytokine storm and a sustained inflammatory state are commonly associated with immune cell depletion,it is still unclear whether direct SARS-Cov-2 infection of immune cells could also play a role in this scenario by harboring viral replication. We found that monocytes,as well as both B and T lymphocytes, were susceptible to SARS-Cov-2 infection in vitro, accumulating double-stranded RNA consistent with viral RNA replication and ultimately leading to expressive T cell apoptosis. In addition, flow cytometry and immunofluorescence analysis revealed that SARS-Cov-2 was frequently detected in monocytes and B lymphocytes from coronavirus disease 2019(CoVID-19)patients.The rates of SARS-Cov-2-infected monocytes in peripheral blood mononuclear cells from CoVID-19 patients increased over time from symptom onset, with SARS-CoV-2-positive monocytes,B cells, and CD4+T lymphocytes also detected in postmortem lung tissue. These results indicated that SARS-CoV-2 infection of blood-circulating leukocytes in covID-19 patients might have important implications for disease pathogenesis and progression, immune dysfunction, and virus spread within the host.Marjorie C.Pontelli italo A.Castro Ronaldo B.Martins Leonardo La Serra Flavio P.Veras Daniele C.Nascimento Camila M.Silva Ricardo S.Cardoso Roberta Rosales Rogério Gomes Thais M.Lima Juliano P.Souza Brenda C.Vitti Diego B.Caetite Mikhael H.F.de Lima Spencer D.Stumpf Cassandra E.Thompson Louis-Marie Bloyet Juliana E Toller-Kawahisa Marcela C.Giannini Leticia P.Bonjorno Marial.F.Lopes Sabrina S.Batah Li Siyuan Rodrigo Luppino-Assad Sergio C.L.Almeida Fabiola R.Oliveira Maira N.Benatti Lorena L.F.Pontes Rodrigo C.Santana Fernando C.Vilar Maria Auxiliadora-Martins Pei-Yong Shi Thiago M.Cunha Rodrigo T.Calado JoséC.Alves-Filho Dario S.Zamboni Alexandre T.Fabro Paulo Louzada-Junior Rene D.R.Oliveira Sean P.J.Whelan Fernando Q.Cunha Eurico Arruda 2022Journal of Molecular Cell Biology2022,14,4:1
Alexandre F.FERREIRA Ivaldo L.FERREIRA Rangel P.ALMEIDA Jose A.CASTRO Roberto C.SALES Zilmar A.JUNIOR 2021Transactions of Nonferrous Metals Society of China2021,31,7:1

Perovskite solid solutions with multiferroic morphotropic phase boundaries and property enhancement显示文摘Recently, large phase-change magnetoelectric response has been anticipated by a first-principles investigation of phases in the BiFeO_(3)–BiCoO_(3)perovskite binary system, associated with the existence of a discontinuous morphotropic phase boundary(MPB)between multiferroic polymorphs of rhombohedral and tetragonal symmetries. This might be a general property of multiferroic phase instabilities, and a novel promising approach for room temperature magnetoelectricity. We review here our current inves-tigations on the identification and study of additional material systems, alternative to BiFeO_(3)–BiCoO_(3)that has only been obtained by high pressure synthesis.Three systems, whose phase diagrams were,in principle, liable to show multiferroic MPBs have been addressed:the BiMnO_(3)–PbTiO_(3)and BiFeO_(3)–PbTiO_(3)binary systems, and the BiFeO_(3)–BiMnO_(3)–PbTiO_(3)ternary one.A compre-hensive study of multiferroism across different solid solutions was carried out based on electrical and magnetic characterizations, complemented with mechanical and electromechanical measurements. An in-depth structural analysis was also accomplished when necessary.M.Algueró H.Amorín C.M.Fernandez-Posada O.Peña P.Ramos E.Vila A.Castro 2016Journal of Advanced Dielectrics2016,6,2:0
The adjustment of γ-aminobutyric acid_A tonic subunits in Huntington's disease:from transcription to translation to synaptic levels into the neostriatum显示文摘γ-Aminobutyric acid(GABA),plays a key role in all stages of life, also is considered the main inhibitory neurotransmitter.GABA activates two kind of membrane receptors known as GABAA and GABAB,the first one is responsible to render tonic inhibition by pentameric receptors containing α4-6,β3,δ,or ρ1-3 subunits, they are located at perisynaptic and/or in extrasynaptic regions. The biophysical properties of GABAA tonic inhibition have been related with cellular protection against excitotoxic injury and cell death in presence of excessive excitation. On this basis,GABAA tonic inhibition has been proposed as a potential target for therapeutic intervention of Huntington's disease.Huntington's disease is a neurodegenerative disorder caused by a genetic mutation of the huntingtin protein. For experimental studies of Huntington's disease mouse models have been developed, such as R6/1,R6/2,Hdh Q92,Hdh Q150,as well as YAC128.In all of them, some key experimental reports are focused on neostriatum. The neostriatum is considered as the most important connection between cerebral cortex and basal ganglia structures, its cytology display two pathways called direct and indirect constituted by medium sized spiny neurons expressing dopamine D1 and D2 receptors respectively, they display strong expression of many types of GABAA receptors, including tonic subunits. The studies about of GABAA tonic subunits and Huntington's disease into the neostriatum are rising in recent years, suggesting interesting changes in their expression and localization which can be used as a strategy to delay the cellular damage caused by the imbalance between excitation and inhibition,a hallmark of Huntington's disease.Abraham Rosas-Arellano Argel Estrada-Mondragón Carola A.Mantellero Carlos Tejeda-Guzmán Maite A.Castro 2018Neural Regeneration Research2018,13,4:0
