| 1 | Rethinking fish biology and biotechnologies in the challenge era for burgeoning genome resources and strengthening food security显示文摘Fish biology has been developed for more than 100 years,but some important breakthroughs have been made in the last decade.Early studies commonly concentrated on morphology,phylogenetics,development,growth,reproduction manipulation,and disease control.Recent studies have mostly focused on genetics,molecular biology,genomics,and genome biotechnologies,which have provided a solid foundation for enhancing aquaculture to ensure food security and improving aquatic environments to sustain ecosystem health.Here,we review research advances in five major areas:(1)biological innovations and genomic evolution of four significant fish lineages including non-teleost ray-finned fishes,northern hemisphere sticklebacks,East African cichlid fishes,and East Asian cyprinid fishes;(2)evolutionary fates and consequences of natural polyploid fishes;(3)biological consequences of fish domestication and selection;(4)development and innovation of fish breeding biotechnologies;and(5)applicable approaches and potential of fish genetic breeding biotechnologies.Moreover,five precision breeding biotechniques are examined and discussed in detail including gene editing for the introgression or removal of beneficial or detrimental alleles,use of sex-specific markers for the production of mono-sex populations,controllable primordial germ cell on-off strategy for producing sterile offspring,surrogate broodstock-based strategies to accelerate breeding,and genome incorporation and sexual reproduction regainbased approach to create synthetic polyploids.Based on these scientific and technological advances,we propose a blueprint for genetic improvement and new breed creation for aquaculture species and analyze the potential of these new breeding strategies for improving aquaculture seed industry and strengthening food security. | Jian-Fang Gui Li Zhou Xi-Yin Li | 2022 | Water Biology and Security2022,1,1: | 10 |
| 2 | Problems and solutions regarding generalized functional safety in cyberspace显示文摘The common endogenous security problems in cyberspace and related attack threats have posed subversive challenges to conventional theories and methods of functional safety.In the current design of the cyber physical system(CPS),functional safety and cyber security are increasingly intertwined and inseparable,which evolve into the generalized functional safety(S&S)problem.The conventional reliability and cybersecurity technologies are unable to provide security assurance with quanti able design and veri cation metrics in response to the cyberattacks in hardware and software with common endogenous security problems,and the functional safety of CPS facilities or device has become a frightening ghost.The dynamic heterogeneity redundancy(DHR)architecture and coding channel theory(CCT)proposed by the cyberspace endogenous security paradigm could handle random failures and uncertain network attacks in an integrated manner,and its generalized robust control mechanism can solve the universal problem of quantitative design for functional safety under probability or improbability perturbation.As a generalized functional safety enabling structure,DHR opens up a new direction to solve the common endogenous security problems in the cross-disciplinary elds of cyberspace. | JiangXing Wu | 2022 | Security and Safety2022,1,1: | 5 |

| 6 | Methodology for fish biodiversity monitoring with environmental DNA metabarcoding:The primers,databases and bioinformatic pipelines显示文摘Environmental DNA(eDNA)originates from cellular material shed by organisms into aquatic or terrestrial environments and can be sampled and monitored using metabarcoding technology,which is revolutionizing fish biodiversity monitoring.Several reviews concerning fish eDNA have focused on standard sampling methods and its applications,though a systematic review focused on marker genes,databases,and bioinformatic pipelines has not yet been published.Here,we present a comprehensive literature review of studies applying metabarcoding technology to fish eDNA for the purpose of fish biodiversity monitoring.We systematically provide the available universal primers used to amplify barcoding sequences from fish eDNA,and then discuss reference barcoding databases,relevant bioinformatic analyses,as well as developed pipelines.The performances of universal primers and their relevant reference databases are summarized.Combined use of multiple primer pairs targeted for more than one gene marker(e.g.,12S,16S,Cytb,COI),and use of both local and public databases are recommended as approaches to improve the sensitivity and reliability of fish eDNA analyses.We also compare the effectiveness of eDNA metabarcoding to traditional approaches for monitoring fish biodiversity and highlight challenges and future perspectives associated with this new tool.Ultimately,we advocate for greater incorporation of eDNA analysis into fish biodiversity assessments to assist environmental managers. | Fan Xiong Lu Shu Honghui Zeng Xiaoni Gan Shunping He Zuogang Peng | 2022 | Water Biology and Security2022,1,1: | 3 |

| 11 | Evolutionary rates of mitochondrial sequences and gene orders in Spirurina(Nematoda)are episodic but synchronised显示文摘In contrast to highly conserved mitogenomic architecture in most metazoan lineages,which indicates that rearrangement events are generally strongly selected against,a limited number of often unrelated lineages exhibit highly elevated architectural evolution rates.The underlying reasons for this discontinuity in the mitogenomic evolution remain unknown.Previously we sequenced the mitochondrial genome of the first Camallanoidea species,Camallanus cotti(Nematoda:Chromadorea:Spirurina:Camallanidae),and found that it exhibited a highly disrupted architecture.We hypothesised that disrupted architecture might be a synapomorphic feature of the sister-clades Camallanoidea and Dracunculoidea.In this study,we sequenced mitogenomes of three freshwater fish-parasitic nematodes:Camallanus lacustris(Camallanidae),and two Philometridae(Dracunculoidea)species,Clavinema parasiluri,and Philometra sp.In partial agreement with the working hypothesis,both Camallanoidea species had exceptionally large mitogenomes of 18–19 Kbp,albeit the underlying reasons differed:in C.lacustris it was the existence of a single enlarged noncoding region of5.5 Kbp.A segment of this region exhibited an inverted base composition skew,which is indicative of a sequence inversion or recombination event.Camallanidae is the second identified chromadorean(first for Spirurina)family that exhibits within-family protein-coding gene rearrangements,and the absence of trnL1 and trnF may be a synapomorphy for Camallanoidea.The underlying reason for the disrupted architecture of Camallanidae does not appear to be a particular event shared by their common ancestor,but rather an underlying mechanism that makes disruptive events more likely in this lineage.In disagreement with the working hypothesis,Spiruromorpha and Oxyuridomorpha exhibited even more highly rearranged gene orders and greater overall branch lengths than Camallanomorpha.However,withininfraorder architecture was highly conserved and leaf nodes very short.This indicates that common ancestors of Spiruromorpha and Oxyuridomorpha clades underwent a period of rapid mitochondrial evolution(both sequence and architecture),followed by a stabilisation after the taxonomic radiation.In contrast to this,Camallanomorpha,and particularly Camallanidae,appear to have entered a period of elevated evolutionary rates after the initial radiations of these two taxa.As a result of this evolutionary discontinuity,there was a strong correlation between the gene order rearrangement rate(GORR)and the overall branch length(0.81),but there was no correlation between the strength of purifying selection(ω?dN/dS)and the overall branch lengths(0.05)and GORR(0.04).These findings have important repercussions for future phylogenetic and other evolutionary studies of Spirurina. | Hong Zou Hong-Peng Lei Rong Chen Fang-Lin Chen Wen-Xiang Li Ming Li Dong Zhang Ivan Jakovlic Gui-Tang Wang | 2022 | Water Biology and Security2022,1,2: | 2 |
