1Effect of phosphorylation of MAPK and Stat3 and expression of c-fos and c-jun proteins on hepatocarcinogenesis and their clinical significance显示文摘AIM To study the effect of phosphorylation ofMAPK and Stat3 and the expression of c-fos andc-jun proteins on hepatocellular carcinogenesisand their clinical significance.METHODS SP immunohistochemistry was usedto detect the expression of p42/44MAPK, p-Stat3,c-fos and c-jun proteins in 55 hepatocellularcarcinomas (HCC) and their surrounding livertissues.RESULTS The positive rates and expressionlevels of p42/44MAPK, p-Stat3, c-fos and c-junproteins in HCCs were significantly higher thanthose in pericarcinomatous liver tissues (PCLT).A positive correlation was observed between theexpression of p42/44MAPK and c-fos proteins, andbetween p-Stat3 and c-jun, but there was nosignificant correlation between p42/44MAPK and p-Stat3 in HCCs and their surrounding livertissues.CONCLUSION The abnormalities of Ras/Rat/MAPK and JAKs/ Stat3 cascade reaction maycontribute to malignant transformation ofhepatocytes. 3Lower Bifidobacteria counts in both duodenal mucosa-associated and fecal microbiota in irritable bowel syndrome patients显示文摘AIM: To determine the composition of both fecal and duodenal mucosa-associated microbiota in irritable bowel syndrome (IBS) patients and healthy subjects using molecular-based techniques. METHODS: Fecal and duodenal mucosa brush samples were obtained from 41 IBS patients and 26 healthy subjects. Fecal samples were analyzed for the composition of the total microbiota using fluorescent in situ hybridization (FISH) and both fecal and duodenal brush samples were analyzed for the composition of bif idobacteria using real-time polymerase chain reaction. RESULTS: The FISH analysis of fecal samples revealed a 2-fold decrease in the level of bifidobacteria (4.2 ± 1.3 vs 8.3 ± 1.9, P < 0.01) in IBS patients compared to healthy subjects, whereas no major differences in other bacterial groups were observed. At the species level, Bifidobacterium catenulatum levels were significantly lower (6 ± 0.6 vs 19 ± 2.5, P < 0.001) in the IBS patients in both fecal and duodenal brush samples than in healthy subjects.CONCLUSION: Decreased bifidobacteria levels in both fecal and duodenal brush samples of IBS patients compared to healthy subjects indicate a role for microbiotic composition in IBS pathophysiology.Angèle PM Kerckhoffs Melvin Samsom Michel E van der Rest Joris de Vogel Jan Knol Kaouther Ben-Amor Louis MA Akkermans 2009World Journal of Gastroenterology2009,15,23:33

5Darwin:a neuromorphic hardware co-processor based on Spiking Neural Networks显示文摘Broadly speaking, the goal of neuromorphic engineering is to build computer systems that mimic the brain. Spiking Neural Network(SNN) is a type of biologically-inspired neural networks that perform information processing based on discrete-time spikes, different from traditional Artificial Neural Network(ANN).Hardware implementation of SNNs is necessary for achieving high-performance and low-power. We present the Darwin Neural Processing Unit(NPU), a neuromorphic hardware co-processor based on SNN implemented with digitallogic, supporting a maximum of 2048 neurons, 20482= 4194304 synapses, and 15 possible synaptic delays.The Darwin NPU was fabricated by standard 180 nm CMOS technology with an area size of 5 × 5 mm2and70 MHz clock frequency at the worst case. It consumes 0.84 m W/MHz with 1.8 V power supply for typical applications. 6Treatment of allergic airway inflammation and hyperrespon-siveness by imiquimod modulating transcription factors T-bet and GATA-3显示文摘Background Imiquimod is an imidazoquinoline, which class of compounds are known to have antiviral and antitumoural properties. In recent studies, it was shown that imiquimod modulates the T helper cell type Th1/Th2 response by inducing the production of Th1 cytokines like IFN-(, and by inhibiting the Th2 cytokines like interleukin (IL)-4. Several investigators have shown that T-bet and GATA-3 are master Th1 and Th2 regulatory transcription factors. This study investigated whether imiquimod treatment inhibited airway inflammation by modulating transcription factors T-bet and GATA-3. Methods Thirty-six male SD rats were randomly divided into a control group, an asthmatic group, and an imiquimod group, which was exposed to an aerosol of 0.15% imiquimod. Twenty-four hours after the last ovalbumin (OVA) challenge, airway responsiveness was measured and changes in airway histology were observed. The concentrations of IL-4, IL-5 and IFN-( in bronchoalveolar lavage fluid (BALF) and serum were measured by enzyme linked immunosorbent assay (ELISA). The mRNA expressions of IL-4, IL-5, IFN-(, T-bet and GATA-3 in lung and in CD4+ T cells were determined by reverse transcription polymerase chain reaction (RT-PCR). The protein expressions of T-bet and GATA-3 were measured by Western blot. Results It was demonstrated that imiquimod 1) attenuated OVA induced airway inflammation; 2) diminished the degree of airway hyperresponsiveness (AHR); 3) decreased the Th2 type cytokines and increased Th1 type cytokines mRNA and protein levels; 4) modulated the Th1/Th2 reaction by inhibiting GATA-3 production and increasing T-bet production. 8Immunohistochemical study of hepatic oval cells in human chronic viral hepatitis显示文摘AIM To detect immunohistochemically the presence of oval cells in chronic viral hepatitis with antibody against c-kit.METHODS We detected oval cells in paraffin-embedded liver sections of 3 normal controls and 26 liver samples from patients with chronic viral hepatitis, using immunohistochemistry with antibodies against c-kit, π-class glutathione Stransferase ( Tr-GST ) and cytokeratins 19(CK19).RESULTS Oval cells were not observed in normal livers. In chronic viral hepatitis, hepatic oval cells were located predominantly in the periportal . region and fibrosis septa,characterized by an ovoid nucleus, small size,and scant cytoplasm. Antibody against stem cell factor receptor, c-kit, had higher sensitivity and specificity than π-GST and CK19. Many studies have identified a large number of upregulated oncogenic mi RNAs and downregulated tumour-suppressor mi RNAs in this type of cancer. In this review, we provide an overview of the role of mi RNAs, pointing to their potential to be useful as diagnostic and/or prognostic biomarkers in gastric cancer. 13Apatite-fission-track geochronology and its tectonic correlation in the Dabieshan orogen,central China显示文摘AFT data of granitoid and metamorphic samples from the Dabieshan orogen have an age range between 96.4 Ma and 41.9 Ma coupled with confined track lengths of 11.5—14.0 μm, reflecting the cooling time of rocks differentially through the 100℃± isotherm. The Juras- sic-Eogene sediments from the North Huaiyang and Hefei basin, however, yield their AFT ages of 128.8—62.0 Ma with mean track lengths of 8.6—11.9 μm, recording a complicated thermal de- velopment of the basin. These AFT data structurally allowing the coupling between the orogen and the basin are to a great extent considered as the result from the control of NNE-trending regional tectonics. It took place approximately at 95—90 Ma that the orogen developed with an abrupt shift from the doming extension to the differential transtension, which is assumed to be related with the changes of the convergence between the Western Pacific and Eurasia plates. The differentially sinistral strike-slipping from the transtension between Tanlu and Shang- cheng-Macheng faults brought the doming extension of the orogen shrunk sharply in Late Cre- taceous. In Eogene, the full-grown transtension that dominated in East Asia was caused mainly by a rapid decrease of the rates of the Western Pacific convergence, which can be therefore re- garded as a major mechanism creating the zonal anomalies of 60—40 Ma in the orogen. The thermal anomaly of 70—40 Ma zonally along the Tanlu fault cooled more slowly than the anomaly of 60—55 Ma zoning along the Shangcheng-Macheng fault, but the 60—40 Ma anomaly trending E-W in hinterlands of the orogen seems to be relicts of the doming extension as a result of inten- sive rebuilding by the transtension. In addition, the orogen was also reactivated by the far-field effect from India-Asia collision in Eogene, where major NWW-trending faults played an important role in facilitating such far-field tectonic transferring. In Oligocene to Miocene, the Dabieshan orogen remained a few thermal disturbances with their AFT ages particularly in coincidence with the time of opening and closing of the South China Sea.P. 16A pair of light signaling factors FHY3 and FAR1 regulates plant immunity by modulating chlorophyll biosynthesis显示文摘Light and chloroplast function is known to affect the plant immune response; however, the underlying mechanism remains elusive. We previously demonstrated that two light signaling factors, FAR-RED ELONGATED HYPOCOTYL 3(FHY3)and FAR-RED IMPAIRED RESPONSE 1(FAR1), regulate chlorophyll biosynthesis and seedling growth via controlling HEMB1 expression in Arabidopsis thaliana. In this study, we reveal that FHY3 and FAR1 are involved in modulating plant immunity. We showed that the fhy3 far1 double null mutant displayed high levels of reactive oxygen species and salicylic acid(SA) and increased resistance to Pseudomonas syringae pathogen infection. Microarray analysis revealed that a large proportion of pathogen-related genes, particularly genes encoding nucleotide-binding and leucine-rich repeat domain resistant proteins, are highly induced in fhy3 far1. Genetic studies indicated that the defects of fhy3 far1 can be largely rescued by reducing SA signaling or blocking SA accumulation, and by overexpression of HEMB1, which encodes a 5-aminolevulinic acid dehydratase in the chlorophyll biosynthetic pathway.Furthermore, we found that transgenic plants with reduced expression of HEMB1 exhibit a phenotype similar to fhy3 far1.Taken together, this study demonstrates an important role of FHY3 and FAR1 in regulating plant immunity, through integrating chlorophyll biosynthesis and the SA signaling pathway.Wanqing Wang Weijiang Tang Tingting Ma De Niu Jing Bo Jin Haiyang Wang Rongcheng Lin 2016Journal of Integrative Plant Biology2016,58,1:8
17Cytomegalovirus frequency in neonatal intrahepatic cholestasis determined by serology, histology, immunohistochemistry and PCR显示文摘AIM: To determine cytomegalovirus (CMV) frequency in neonatal intrahepatic cholestasis by serology, histological revision (searching for cytomegalic cells), immunohistochemistry, and polymerase chain reaction (PCR), and to verify the relationships among these methods.METHODS: The study comprised 101 non-consecutive infants submitted for hepatic biopsy between March 1982 and December 2005. Serological results were obtained from the patient's f iles and the other methods were performed on paraffin-embedded liver samples from hepatic biopsies. The following statistical measures were calculated: frequency, sensibility, specif ic positive predictive value, negative predictive value, and accuracy.RESULTS: The frequencies of positive results were as follows: serology, 7/64 (11%); histological revision, 0/84; immunohistochemistry, 1/44 (2%), and PCR, 6/77 (8%). Only one patient had positive immunohistochemical fi ndings and a positive PCR. The following statistical measures were calculated between PCR and serology: sensitivity, 33.3%; specificity, 88.89%; positive predictive value, 28.57%; negative predictive value, 90.91%; and accuracy, 82.35%. CONCLUSION: The frequency of positive CMV varied among the tests. Serology presented the highest positive frequency. 18Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. 19Insights into gastric neuroendocrine tumors burden显示文摘Type 1 gastric neuroendocrine tumors(g NETs)are usually small lesions,restricted to mucosal and sub-mucosal layers of corpus and fundus,with low aggressive behavior,for the majority of cases.Nevertheless,some cases present aggressive behavior.The increasing incidence of g NETs brings together a new relevant problem:how to identify potentially aggressive type 1 g NETs.The challenging problem seems to be finding out signs or features able to predict potentially aggressive cases,allowing a tailored approach,since the involved societies dedicated to provide guidelines for management of these neoplasms apparently failed in producing staging systems able to accurately predict prognosis of these tumors.Additionally,it is also important to try to find out explanations for increasing incidence,as well as to identify potential targets aiming to reach better control of this neoplasia.Here,we discuss potential pathways implicated in aggressive behavior,as well as new strategies to improve clinical management of these tumors.Taíssa Maíra Thomaz Araújo Williams Fernandes Barra André Salim Khayat Paulo Pimentel de Assumpcao 2017Chinese Journal of Cancer Research2017,29,2:7
