| 1 | Effect of Gubenfangxiao decoction on respiratory syncytial virus-induced asthma and expression of asthma susceptibility gene orosomucoid 1-like protein 3 in mice显示文摘OBJECTIVE: To investigate the effect of Gubenfangxiao decoction(GBFXD) on respiratory-syncytial-virus(RSV)-induced asthma and the expression of asthma susceptibility gene, orosomucoid 1-like protein 3(ORMDL3) in mice.METHODS: Seventy-two female BALB/c mice were randomly assigned to normal, model, GBFXD high dose, GBFXD moderate dose, GBFXD low dose and montelukast groups. An asthma model was induced via intraperitoneal injection and aerosol inhalation of ovalbumin(OVA) and repeated intranasal instillation of RSV in all mice, except those in the normal group. All treatments were administered at the first onset of asthma(within 8 weeks of model establishment) and the mice were euthanized after 28 days of treatment. The levels of transforming growth factor-β(TGF-β) and interleukin-6(IL-6) in bronchoalveolar lavage fluid(BALF) of the mice were measured and the expression of asthma susceptibility gene ORMDL3 in lung tissue was determined using real-time polymerase chain reaction(RT-PCR) and western blotting.RESULTS: Expression of ORMDL3 and levels of TGF-β and IL-6 were significantly higher in the model group(P < 0.05, P < 0.01) compared with the normal mice. Levels of ORMDL3, TGF-β and IL-6 were significantly lower in all three GBFXD treated groups(P < 0.05) compared with the model group.However, the levels in the GBFXD treatment groups did not differ significantly from the montelukast group.CONCLUSION: GBFXD had a therapeutic effect in this experimental model. | 2 | TRDMT1 exhibited protective effects against LPS-induced inflammation in rats through TLR4-NF-κB/MAPK-TNF-αpathway显示文摘Background:Inflammation is a complex physiological and pathological process.Although many types of inflammation are well characterized,their physiological func-tions are largely unknown.tRNA aspartic acid methyltransferase 1(TRDMT1)has been implicated as a stress-related protein,but its intrinsic biological role is unclear.Methods:We constructed a Trdmt1 knockout rat and adopted the LPS-induced sepsis model.Survival curve,histopathological examination,expression of inflammatory fac-tors,and protein level of TLR4 pathway were analyzed.Results:Trdmt1 deletion had no obvious impact on development and growth.Trdmt1 de-letion slightly increased the mortality during aging.Our data showed that Trdmt1 strongly responded in LPS-treated rats,and Trdmt1 knockout rats were vulnerable to LPS treat-ment with declined survival rate.We also observed more aggravated tissue damage and more cumulative functional cell degeneration in LPS-treated knockout rats compared with control rats.Further studies showed upregulated TNF-αlevel in liver,spleen,lung,and serum tissues,which may be explained by enhanced p65 and p38 phosphorylation.Conclusions:Our data demonstrated that Trdmt1 plays a protective role in inflamma-tion by regulating the TLR4-NF-κB/MAPK-TNF-αpathway.This work provides useful information to understand the TRDMT1 function in inflammation. | Zhengguang Li Xiaolong Qi Xu Zhang Lei Yu Lijuan Gao Weining Kong Wei Chen Wei Dong Lijun Luo Dan Lu Lianfeng Zhang Yuanwu Ma | 2022 | Animal Models and Experimental Medicine2022,5,2: | 5 |

| 11 | Optimization methods of production layer combination for coalbed methane development in multi-coal seams显示文摘Based on the productivity equation of coalbed methane(CBM) wells, three indexes, main production layer optimization index, main production layer expansion index and capacity contribution index are proposed, with which the three-step optimization method of production-layer combination is established. In selecting main production layer, the coal seam thickness, CBM content, coal seam permeability, coal seam reservoir pressure and coal structure are considered comprehensively to evaluate the potential of the production layer. In selecting expansion of the main production layer combination, on the premise of ensuring full and slow desorption of the main production layer and non-exposure of the main production layer out of liquid surface, the degree of mutual interference between the main and non-main production layers is comprehensively evaluated by coupling the critical desorption pressure, layer spacing and reservoir pressure gradient difference. In optimizing production layer combination, the main concern is the economic efficiency of the combined layers. Only when the contribution coefficient of the main production layer is greater than 30% and the contribution index of the other production layers is more than 10%, the economic benefit of a CBM well after being put into production can be ensured. | 19 | Development unit division and favorable area evaluation for joint mining coalbed methane显示文摘Based on the productivity equation of coalbed methane (CBM) well, considering the impact of coal reservoir reformability on gas well productivity, the main production layer optimization index in the “three-step method” of optimal combination of production layers is corrected, and then the CBM production layer potential index is introduced to evaluate favorable areas for commingled multi-coal seam production. Through analysis of the key parameters of coal reservoirs affecting the CBM productivity index, a development unit division method for areas with multi-coal seams is established, and a quantitative grading index system is proposed. On this basis, the evaluation process of CBM development favorable area is developed: the mature 3-D modeling technology is used to characterize the reservoir physical properties of multi-coal seams in full-scale;the production layer potential index of each grid is calculated, and the production layer potential index contour under single-layer or commingled multi-layer production are plotted;according to the distribution of the contour of production layer potential index, the quantitative index of CBM development unit is adopted to outline the grade I, II, III coal reservoir distribution areas, and thus to pick out the favorable development areas. | 20 | Monitoring model for predicting maize grain moisture at the filling stage using NIRS and a small sample size显示文摘The change in the maize moisture content during different growth stages is an important indicator to evaluate the growth status of maize.In particular,the moisture content during the grain-filling stage reflects the grain quality and maturity and it can also be used as an important indicator for breeding and seed selection.At present,the drying method is usually used to calculate the moisture content and the dehydration rate at the grain-filling stage,however,it requires large sample size and long test time.In order to monitor the change in the moisture content at the maize grain-filling stage using small sample set,the Bootstrap re-sampling strategy-sample set partitioning based on joint x-y distances-partial least squares(Bootstrap-SPXY-PLS)moisture content monitoring model and near-infrared spectroscopy for small sample sizes of 10,20,and 50 were used.To improve the prediction accuracy of the model,the optimal number of factors of the model was determined and the comprehensive evaluation thresholds RVP(coefficient of determination(R^(2)),the root mean square error of cross-validation(RMSECV)and the root mean square error of prediction(RMSEP))was proposed for sub-model screening.The model exhibited a good performance for predicting the moisture content of the maize grain at the filling stage for small sample set.For the sample sizes of 20 and 50,the R^(2) values were greater than 0.99.The average deviations of the predicted and reference values of the model were 0.1078%,0.057%,and 0.0918%,respectively.Therefore,the model was effective for monitoring the moisture content at the grain-filling stage for a small sample size.The method is also suitable for the quantitative analysis of different concentrations using near-infrared spectroscopy and small sample size. | Xue Wang Tiemin Ma Tao Yang Ping Song Zhengguang Chen Huan Xie | 2019 | International Journal of Agricultural and Biological Engineering2019,12,2: | 0 |
