| 2 | Inhibition of SARS-CoV-2 (previously 2019-nCoV) infection by a highly potent pan-coronavirus fusion inhibitor targeting its spike protein that harbors a high capacity to mediate membrane fusion显示文摘The recent outbreak of coronavirus disease(COVID-19)caused by SARS-CoV-2 infection in Wuhan,China has posed a serious threat to global public health.To develop specific anti-coronavirus therapeutics and prophylactics,the molecular mechanism that underlies viral infection must first be defined.Therefore,we herein established a SARS-CoV-2 spike(S)protein-mediated cell-cell fusion assay and found that SARS-CoV-2 showed a superior plasma membrane fusion capacity compared to that of SARS-CoV.We solved the X-ray crystal structure of six-helical bundle(6-HB)core of the HR1 and HR2 domains in the SARS-CoV-2 S protein S2 subunit revealing that several mutated amino acid residues in the HR1 domain may be associated with enhanced interactions with the HR2 domain.We previously developed a pan-coronavirus fusion inhibitor,EK1,which targeted the HR!domain and could inhibit infection by divergent human coronaviruses tested,including SARS-CoV and MERS-CoV.Here we generated a series of lipopeptides derived from EK1 and found that EK1C4 was the most potent fusion inhibitor against SARS-CoV-2 S protein-mediated membrane fusion and pseudovirus infection with IC50s of 1.3 and 15.8 nM,about 241-and 149-fold more potent than the original EK1 peptide,respectively.EK1C4 was also highly effective against membrane fusion and infection of other human coronavirus pseudoviruses tested,including SARS-CoV and MERS-CoV,as well as SARSr-CoVs,and potently inhibited the replication of 5 live human coronaviruses examined,including SARS-CoV-2.Intranasal application of EK1C4 before or after challenge with HCoV-OC43 protected mice from infection,suggesting that EK1C4 could be used for prevention and treatment of infection by the currently circulating SARS-CoV-2 and other emerging SARSr-CoVs. | Shuai Xia Meiqin Liu Chao Wang Wei Xu Qiaoshuai Lan Siliang Feng Feifei Qi Linlin Bao Lanying Du Shuwen Liu Chuan Qin Fei Sun Zhengli Shi Yun Zhu Shibo Jiang Lu Lu | 2020 | Cell Research2020,30,4: | 81 |

| 8 | Rapid Detection of Filoviruses by Real-time TaqMan Polymerase Chain Reaction Assays显示文摘Ebola virus (EBOV) and Marburg virus (MARV) are causative agents of severe hemorrhagic fever with high mortality rates in humans and non-human primates and there is currently no licensed vaccine or therapeutics. To date, there is no specific laboratory diagnostic test in China, while there is a national need to provide differential diagnosis during outbreaks and for instituting acceptable quarantine procedures. In this study, the TaqMan RT-PCR assays targeting the nucleoprotein genes of the Zaire Ebolavirus (ZEBOV) and MARV were developed and their sensitivities and specificities were investigated. Our results indicated that the assays were able to make reliable diagnosis over a wide range of virus copies from 103 to 109, corresponding to the threshold of a standard RNA transcript. The results showed that there were about 1010 RNA copies per milliliter of virus culture supernatant, equivalent to 10,000 RNA molecules per infectious virion, suggesting the presence of many non-infectious particles. | 10 | Viral Metagenomics Analysis of Planktonic Viruses in East Lake,Wuhan,China显示文摘East Lake(Lake Donghu),located in Wuhan,China,is a typical city freshwater lake that has been experiencing eutrophic conditions and algal blooming during recent years.Marine and fresh water are considered to contain a large number of viruses.However,little is known about their genetic diversity because of the limited techniques for culturing viruses.In this study,we conducted a viral metagenomic analysis using a high-throughput sequencing technique with samples collected from East Lake in Spring,Summer,Autumn,and Winter.The libraries from four samples each generated 234,669,71,837,12,820,and 34,236 contigs(>90 bp each),respectively.The genetic structure of the viral community revealed a high genetic diversity covering 23 viral families,with the majority of contigs homologous to DNA viruses,including members of Myoviridae,Podoviridae,Siphoviridae,Phycodnaviridae,and Microviridae,which infect bacteria or algae,and members of Circoviridae,which infect invertebrates and vertebrates.The highest viral genetic diversity occurred in samples collected in August,then December and June,and the least diversity in March.Most contigs have low-sequence identities with known viruses.PCR detection targeting the conserved sequences of genes(g20,psbA,psbD,and DNApol)of cyanophages further confirmed that there are novel cyanophages in the East Lake.Our viral metagenomic data provide the first preliminary understanding of the virome in one freshwater lake in China and would be helpful for novel virus discovery and the control of algal blooming in the future. | Xingyi Ge Yongquan Wu Meiniang Wang Jun Wang Lijun Wu Xinglou Yang Yuji Zhang Zhengli Shi | 2013 | Virologica Sinica2013,28,5: | 8 |

| 12 | Shuanghuanglian injection downregulates nuclear factor-kappa B expression in mice with viral encephalitis显示文摘A mouse model of viral encephalitis was induced by intracranial injection of a Coxsackie virus B3 suspension. Quantitative real-time reverse transcription-PCR and western blot assay were applied to detect mRNA and protein expression of intelectin-2 and nuclear factor-kappa B in the viral encephalitis and control groups. Nuclear factor-kappa B and intelectin-2 mRNA and protein expression were significantly increased in mice with viral encephalitis. After intraperitoneal injection of Shuanghuanglian at a dose of 1.5 mg/kg for 5 successive days, intelectin-2 and nuclear factor-kappa B protein and mRNA expression were significantly decreased. To elucidate the relationship between intelectin-2 and nuclear factor-kappa B, mice with viral encephalitis were administered an intracerebral injection of 107 pfu recombinant lentivirus expressing intelectin shRNA. Both protein and mRNA levels of intelectin and nuclear factor-kappa B in brain tissue of mice were significantly decreased. Simulation results show that particle interaction is responsible for axial segregation,the patterns of which are influenced by the end wall effect. The two influential factors vary with various rotational speeds and end wall friction levels. | 15 | Dynamic analysis of the Wenchuan Earthquake disaster and reconstruction with 3-year remote sensing data显示文摘Earth observation is an effective technique that plays an important role in earthquake damage reduction and reconstruction.This paper introduces the results of dynamic analysis on monitoring and assessing heavily impacted areas affected by the Wenchuan Earthquake using remote sensing data acquired in the past 3 years from 2008 to 2010.Immediately after the disaster on 12 May 2008,the Chinese Academy of Sciences launched a project entitled‘Wenchuan Earthquake Disasters Monitoring and Assessment Using Remote Sensing Technology.’More than 400 images from 17 satellites and 20.2TB airborne remote sensing data were acquired to facilitate quick monitoring and evaluation of severely damaged areas in 14 counties.Results of the image analyses were forwarded on a timely basis to assist with consultative service and decisionmaking support.In subsequent years,in order to monitor the process of environmental restoration and reconstruction,airborne optical remote sensing images covering most of the severely damaged areas were again acquired in May 2009 and April 2010.These images were analyzed and compared along with images from 2008.Results were useful in support of further work on environmental protection and reconstruction in earthquake-damaged areas.Three typical areas were selected for illustrative purposes including Tangjiashan Barrier Lake,Beichuan County,and counties of Yingxiu and the new Beichuan.These results well demonstrate the importance and effectiveness of the utility of earth observation for disaster mitigation and reconstruction. | Huadong Guo Liangyun Liu Liping Lei Yanhong Wu Liwei Li Bing Zhang Zhengli Zuo Zhen Li | 2010 | International Journal of Digital Earth2010,3,4: | 5 |
| 16 | Development and Validation of a Novel Reverse Phase HPLC for Determination of Aloperine in Aloperine Tablets显示文摘Aim To develop a reverse phase HPLC method for the determination of aloperine, an alkaloid that is newly extracted from Sophora alopecuraides and has shown wide pharmacological effects including antibacterial and antimflammatory actions. Methods The samples were analyzed on a ODS column with methanol-water-triethylamine (3:97:0.1 V/V) as a mobile phase, The flow rate was 1.0 mL·min^-1, and UV detection wavelength 205 nm. Results Linear regression equation was A = 1.6920C + 1.7455 (r^2 = 0.9999, n = 5) in concentratim ranging from 20 to 120μg·mL^-1. The recoveries were 101.2±1.46 % at 80 μg·mL^-1, 100.5±0.75% at 100μg·mL^-1, and 100.7±1.10% at 120 μg·mL^-1, respectively, and the precisions of aloperine within or between run were from 0.80% to 1.98% ( n = 5). | 19 | Cellular immunotherapy for hematological malignancy: recent progress and future perspectives显示文摘Advancements in the field of cellular immunotherapy have accelerated in recent years and have changed the treatment landscape for a variety of hematologic malignancies.Cellular immunotherapy strategies exploit the patient’s immune system to kill cancer cells.The successful use of CD19 chimeric antigen receptor(CAR)T-cells in treating B-cell malignancies is the paradigm of this revolution,and numerous ongoing studies are investigating and extending this approach to other malignancies.However,resistance to CAR-Tcell therapy and non-durable efficacy have prevented CAR-T-cells from becoming the ultimate therapy.Because natural killer(NK)cells play an essential role in antitumor immunity,adoptively transferred allogeneic NK and CAR-modified NK cell therapy has been attempted in certain disease subgroups.Allogenic hematopoietic stem cell transplantation(allo-HSCT)is the oldest form of cellular immunotherapy and the only curative option for hematologic malignancies.Historically,the breadth of application of allo-HSCT has been limited by a lack of identical sibling donors(ISDs).However,great strides have recently been made in the success of haploidentical allografts worldwide,which enable everyone to have a donor.Haploidentical donors can achieve comparable outcomes to those of ISDs and even better outcomes in certain circumstances because of a stronger graft vs.tumor effect.Currently,novel strategies such as CAR-T or NK-based immunotherapy can be applied as a complement to allo-HSCT for curative effects,particularly in refractory cases.Here,we introduce the developments in cellular immunotherapy in hematology. | Zhengli Xu Xiaojun Huang | 2021 | Cancer Biology & Medicine2021,18,4: | 3 |
| 20 | Complex duality quantum computers acting on pure and mixed states显示文摘The aim of this paper is to establish a mathematical fundamental of complex duality quantum computers(CDQCs) acting on vector-states(pure states) and operator-states(mixed states),respectively.A CDQC consists of a complex divider,a group of quantum gates represented by unitary operators,or quantum operations represented by completely positive and trace-preserving mappings,and a complex combiner.It is proved that the divider and the combiner of a CDQC are an isometry and a contraction,respectively.It is proved that the divider and the combiner of a CDQC acting on vector-states are dual,and in the finite dimensional case,it is proved that the divider and the combiner of a CDQC acting on operator-states(matrix-states) are also dual.Lastly,the loss of an input state passing through a CDQC is measured. | CAO HuaiXin CHEN ZhengLi GUO ZhiHua REN FangGuo | 2012 | Science China(Physics,Mechanics & Astronomy)2012,55,12: | 3 |
