| 1 | The role of cupric in maintaining the structure of CopC显示文摘The CopC protein from pseudomonas syringae pathovar tomato is expressed as one of four proteins encoded by the operon CopABCD that is responsible for copper resistance. And there are one trypto-phan (83), one tyrosine (79), and three phenylalanines (35, 43, 99) in apoCopC. The fluorescence peak of apoCopC is located near 320 nm, and the peak shifts toward 353 nm in the presence of 10 mmol·L^(-1) urea with excitation at 280 nm. Using urea as a chemical agent, the conformational stabilities of apoCopC and Cu_N^(2+) -CopC were monitored by fluorescence spectrum in 20 mmol·L^(-1) phosphate buffer and 100 mmol·L^(-1) sodium chloride at pH 6.0. The free energy of stabilization for apoCopC and Cu_N^(2+)-CopC is 16.29±0.65 kJ·mol^(-1) and 26.26±0.35 kJ·mol^(-1), respectively. The distance between the tryptophan residue and the Cu^(2+) in Cu_N^(2+) -CopC has been studied by observing Frster type nonradia-tive energy transfer. | 2 | Detailed resume of RNA m^(6)A demethylases显示文摘N6-Methyladenosine(m^(6)A) is the most abundant internal modification in eukaryotic mRNA,playing critical role in various bioprocesses. Like other epigenetic modifications, m^(6)A modification can be catalyzed by the methyltransferase complex and erased dynamically to maintain cells homeostasis. Up to now, only two m^(6)A demethylases have been reported, fat mass and obesity-associated protein(FTO)and alkylation protein AlkB homolog 5(ALKBH5), involving in a wide range of mRNA biological progress, including mRNA shearing, export, metabolism and stability. Furthermore, they participate in many significantly biological signaling pathway, and contribute to the progress and development of cancer along with other diseases. | 5 | DLX5 promotes Col10a1 expression and chondrocyte hypertrophy and is involved in osteoarthritis progression显示文摘Osteoarthritis(OA)has been considered non-reversible as articular cartilage wears down with limited repair capacity.Enhanced chondrocyte hypertrophy and increased type X collagen gene(COL10A1)expression have been associated with OA.Therefore,regulators controlling collagen X expression and chondrocyte hypertrophy may play a role in OA intervention.Here,we investigated how Distal-less homeobox 5(DLX5),the distal-less homeobox family member,controls murine Col10a1 gene expression and chondrocyte hypertrophy in chondrogenic cell models and its role in a murine OA model.Through qRT-PCR and Western blot analyses,we detected significantly increased levels of COL10A1 and DLX5 in hypertrophic MCT and ATDC5 cells compared to their proliferative stage.Forced expression of Dlx5 further increases,while knockdown of Dlx5 decreases COL10A1 expression in hypertrophic MCT cells.We have performed dual-luciferase reporter and ChiP assays and demonstrated that DLX5 promotes reporter activity through direct interaction with Col10a1 cis-enhancer.We established a murine OA model and detected markedly increased COL10A1 and DLX5 in the articular cartilage and subchondral bone of the OA mice compared with the controls.Notably,forced overexpression of DLX5 in hypertrophic MCT cells up-regulates RUNX2,and adjacent DLX5 and RUNX2 binding sites have previously been found within the Col10a1 cis-enhancer.Together,our data suggest that DLX5 may cooperate with RUNX2 to control cell-specific Col10a1 expression and chondrocyte hypertrophy and is involved in OA pathogenesis. | Jinnan Chen Fangzhou Chen Xuan Wu Huiqin Bian Chen Chen Xiaojing Zhang Ruoxuan Hei Xiaotong Yang Haochun Yuan Qian Wang Yaojuan Lu Longwei Qiao Qiping Zheng | 2023 | Genes & Diseases2023,10,5: | 1 |

| 7 | Field-assisted conductive substrate sparks the redox kinetics of Co_(9)S_(8) in Li-and Na-ion batteries显示文摘As a promising candidate electrode material in both Li-and Na-ion batteries(L/SIBs),the application of Co_(9)S_(8) is being hindered by its unsatisfactory electrochemical performance caused by the sluggish ion diffusion kinetics and drastic volume expansion.Herein,a hybrid material composed of Co_(9)S_(8-x),N-doped carbon foam that seeded with Co nanoparticles(Co_(9)S_(8-x)@Co-NC) is constructed.Particularly,theoretical and experimental results imply that a built-in electric field at the interface of Co and NC is observed due to the variation of Fermi levels,forming rich Mott-Schottky-like heterointerfaces,which can significantly enhance the charge transfer capability between the active materials of Co_(9)S_(8) and conductive NC skeleton.Moreover,the sulfur defects in Co_(9)S_(8-x)can not only effectively lower the energy barrier of the ion diffusion and charge transfer processes,but also endow the target sample with more storage/adsorption/active sites for Li^(+)/Na^(+) ions,thus improving the rate performance of the Co_(9)S_(8-x)@Co-NC composite.As a result,the Co_(9)S_(8-x)@Co-NC exhibits fast surface-controlled redox kinetics and robust cycling stability.For instance,the Co_(9)S_(8-x)@Co-NC displays impressive Li-storage properties in both half and full cells with a high reversible capacity of 1007.4 mA h g^(-1)at 0.1 A g^(-1)after 100 cycles and superior rate capability up to 5 A g^(-1).Moreover,based on these comprehensive merits,the Co_(9)S_(8-x)@Co-NC composite shows decent electrochemical performance(472.2 and 311.1 mA h g^(-1)at 0.1 and 10 A g^(-1),respectively) as an anode for SIBs.This work presents an effective strategy for the construction of Mott-Schottky-like heterointerfaces in Co_(9)S_(8) based materials and provides specific inspiration for future works designing high-performance electrodes via interfacial engineering. | Huanhuan Li Yuxin Chen Huiqin Chen Yantao Wang Jinlong Li Yanping Zheng Linlin Zhang | 2023 | Journal of Energy Chemistry2023,,12: | 0 |
| 8 | Causal genetic regulation of DNA replication on immune microenvironment in colorectal tumorigenesis: Evidenced by an integrated approach of trans-omics and GWAS显示文摘The interplay between DNA replication stress and immune microenvironment alterations is known to play a crucial role in colorectal tumorigenesis,but a comprehensive understanding of their association with and relevant biomarkers involved in colorectal tumorigenesis is lacking.To address this gap,we conducted a study aiming to investigate this association and identify relevant biomarkers.We analyzed transcriptomic and proteomic profiles of 904 colorectal tumor tissues and 342 normal tissues to examine pathway enrichment,biological activity,and the immune microenvironment.Additionally,we evaluated genetic effects of single variants and genes on colorectal cancer susceptibility using data from genome-wide association studies(GWASs)involving both East Asian(7062 cases and 195745 controls)and European(24476 cases and 23073 controls)populations.We employed mediation analysis to infer the causal pathway,and applied multiplex immunofluorescence to visualize colocalized biomarkers in colorectal tumors and immune cells.Our findings revealed that both DNA replication activity and the flap structure-specific endonuclease 1(FEN1)gene were significantly enriched in colorectal tumor tissues,compared with normal tissues.Moreover,a genetic variant rs4246215 G>T in FEN1 was associated with a decreased risk of colorectal cancer(odds ratio=0.94,95%confidence interval:0.90–0.97,P_(meta)=4.70×10^(-9)).Importantly,we identified basophils and eosinophils that both exhibited a significantly decreased infiltration in colorectal tumors,and were regulated by rs4246215 through causal pathways involving both FEN1 and DNA replication.In conclusion,this trans-omics incorporating GWAS data provides insights into a plausible pathway connecting DNA replication and immunity,expanding biological knowledge of colorectal tumorigenesis and therapeutic targets. | Sumeng Wang Silu Chen Huiqin Li Shuai Ben Tingyu Zhao Rui Zheng Meilin Wang Dongying Gu Lingxiang Liu | 2024 | The Journal of Biomedical Research2024,38,1: | 0 |

| 10 | Oxygen-deficient MoO_(x)/Ni_(3)S_(2) heterostructure grown on nickel foam as efficient and durable self-supported electrocatalysts for hydrogen evolution reaction显示文摘High-performance and ultra-durable electrocatalysts are vital for hydrogen evolution reaction(HER)during water splitting.Herein,by one-pot solvothermal method,MoO_(x)/Ni_(3)S_(2) spheres comprising Ni_(3)S_(2) nanoparticles inside and oxygen-deficient amorphous MoO_(x) outside in situ grow on Ni foam(NF),to assembly the heterostructure composites of MoO_(x)/Ni_(3)S_(2)/NF.By adjusting volume ratio of the solvents of ethanol to water,the optimized MoO_(x)/Ni_(3)S_(2)/NF-11 exhibits the best HER performance,requiring an extremely low overpotential of 76 mV to achieve the current density of 10 mA∙cm‒2(η_(10)=76 mV)and an ultra-small Tafel slope of 46 mV∙dec^(-1) in 0.5 mol∙L^(-1) H_(2)SO_(4).More importantly,the catalyst shows prominent high catalytic stability for HER(>100 h).The acid-resistant MoO_(x) wraps the inside Ni_(3)S_(2)/NF to ensure the high stability of the catalyst under acidic conditions.Density functional theory calculations confirm that the existing oxygen vacancy and MoO_(x)/Ni_(3)S_(2) heterostructure are both beneficial to the reduced Gibbs free energy of hydrogen adsorption(|∆G_(H*)|)over Mo sites,which act as main active sites.The heterostructure effectively decreases the formation energy of O vacancy,leading to surface reconstruction of the catalyst,further improving HER performance.The MoO_(x)/Ni_(3)S_(2)/NF is promising to serve as a highly effective and durable electrocatalyst toward HER. | Zihuan Yu Haiqing Yan Chaonan Wang Zheng Wang Huiqin Yao Rong Liu Cheng Li Shulan Ma | 2023 | Frontiers of Chemical Science and Engineering2023,17,4: | 0 |
