1Origin and phylogenetic analysis of Tibetan Mastiff based on the mitochondrial DNA sequence显示文摘At present,the Tibetan Mastiff is the oldest and most ferocious dog in the world. However,the origin of the Tibetan Mastiff and its phylogenetic relationship with other large breed dogs such as Saint Bernard are unclear. In this study,the primers were designed accord-ing to the mitochondrial genome sequence of the domestic dog,and the 2,525 bp mitochondrial sequence,containing the whole sequence of Cytochrome b,tRNA-Thr,tRNA-Pro,and control region of the Tibetan Mastiff,was obtained. Using grey wolves and coyotes as out-groups,the Tibetan Mastiff and 12 breeds of domestic dogs were analyzed in phylogenesis. Tibetan Mastiff,domestic dog breeds,and grey wolves were clustered into a group and coyotes were clustered in a group separately. This indicated that the Tibetan Mastiff and the other domestic dogs originated from the grey wolf,and the Tibetan Mastiff belonged to Carnivora,Canidae,Canis,Canis lupus,Canis lupus familiaris on the animal taxonomy. In domestic dogs,the middle and small breed dogs were clustered at first; German Sheepdog,Swedish Elkhound,and Black Russian Terrier were clustered into one group,and the Tibetan Mastiff,Old English Sheepdog,Leonberger,and Saint Bernard were clustered in another group. This confirmed the viewpoint that many of the famous large breed dogs worldwide such as Saint Bernard possibly had the blood lineage of the Tibetan Mastiff,based on the molecular data. 2Notch signaling pathway mediates the immunomodulatory mechanism of Yangfei Huoxue decoction alleviating bleomycin-induced pulmonary fibrosis in rats显示文摘OBJECTIVE: To investigate the immunomodulatory mechanism by which Yangfei Huoxue decoction(YHD) alleviates bleomycin(BLM)-induced pulmonary fibrosis(PF) in rats.METHODS: Rats were randomly divided into two time-point groups(day 14 and 28), and each timepoint group comprised the following six subgroups: control, BLM, dexamethasone(DXM), YHD high dose(YHD-H), YHD middle dose(YHD-M), and YHD low dose(YHD-L). Haematoxylin and eosin and Masson staining, flow cytometry, enzymelinked immunosorbent assay, Western blotting and UPLC-QT of analyses were examined.RESULTS: The results showed that YHD reduced the degree of alveolar inflammation and fibrosis;downregulated the expression of CD28, CD80,CD86, Delta-like 1, Notch2, and Notch3;and upregulated the proportions of Th1/Th2 and Tc1/Tc2. 3A New Species of the Genus Protobothrops(Squamata:Viperidae:Crotalinae) from the Dabie Mountains,Anhui,China显示文摘During a biological survey in July 2011, a pit viper was collected from the Yaoluoping Nature Reserve in the Dabie Mountains, Anhui, China. The pit viper's the total length measured 836 mm with a dorsal ground color of yellowbrown, decorated by 56 dark-brown transverse bands. The 4thsupralabial squama of the new species is separated from subocular by two squamae, and the tip of tail is orange; this combination of characters distinguish it from other species of the genus Protobothrops. 4Total body irradiation of donors can alter the course of tolerance and induce acute rejection in a spontaneous tolerance rat liver transplantation model显示文摘Liver transplantation is an established therapy for end-stage liver diseases. Graft rejection occurs unless the recipient receives immunosuppression after transplantation. This study aimed to explore the mechanism of acute rejection of liver allografts in rats pre-treated with total body irradiation to eliminate passenger lymphocytes and to define the role of CD4 + CD25 + regulatory T cells in the induction of immunotolerance in the recipient. Male Lewis rats were used as donors and male DA rats were recipients. Rats were randomly assigned to the following four groups: control group, homogeneity liver transplantation group, idio-immunotolerance group and acute rejection group. After transplantation, the survival time of each group, serum alanine aminotransferase, total bilirubin levels, number of Foxp3 + CD4 + CD25 + regulatory T cells, expression of glucocorticoid-induced tumor necrosis factor receptor on T cell subgroups, histopathology of the hepatic graft and spleen cytotoxic T lymphocyte lytic activity were measured. In the acute rejection group, where donors were preconditioned with total body irradiation before liver transplantation, all recipients died between day 17 and day 21. On day 14, serum alanine aminotransferase increased signifi- cantly to (459.2±76.9) U L -1 , total bilirubin increased to (124.1±33.7) μmol L -1 (P<0.05) and the ratio of Foxp3 + CD4 + CD25 + regulatory T cells decreased significantly to 1.50%±0.50% (P<0.05) compared with the other groups. Analysis of the T cell subpopulations in the acute rejection group varied from the other groups. Histological analysis showed typical changes of acute rejection in the acute rejection group only. 5Energy Efficiency Based on Joint Data Packet Fragmentation and Cooperative Transmission显示文摘Energy efficiency(EE) is a key requirement for the design of short-range communication network.In order to alleviate energy consumption(EC) constraint,a novel layered heterogeneous mobile cloud architecture is proposed in this paper.Based on the proposed layered heterogeneous mobile cloud architecture,we establish an appropriate energy consumption model,and design an energy efficiency scheme based on joint data packet fragmentation and cooperative transmission and analyze the energy efficiency corresponding to different packet sizes and the cloud size.Simulation results show that,when all nodes of the cloud are accessing the same size of data packet fragmentation,the proposed layered heterogeneous mobile cloud architecture can provide significant energy savings.The results provide useful insights into the possible operation of the strategies and show that significant energy consumption reductions are possible.ZHANG Zufan YI Yinxue YANG Jing JING Xiaorong 2014China Communications2014,11,10:3
6Wildfire Susceptibility Assessment in Southern China:A Comparison of Multiple Methods显示文摘Wildfire is a primary forest disturbance.A better understanding of wildfire susceptibility and its dominant influencing factors is crucial for regional wildfire risk management.This study performed a wildfire susceptibility assessment using multiple methods,including logistic regression,probit regression,an artificial neural network,and a random forest(RF) algorithm.Yunnan Province,China was used as a case study area.We investigated the sample ratio of ignition and nonignition data to avoid misleading results due to the overwhelming number of nonignition samples in the models.To compare model performance and the importance of variables among the models,the area under the curve of the receiver operating characteristic plot was used as an indicator.The results show that a cost-sensitive RF had the highest accuracy(88.47%) for all samples,and 94.23% accuracy for ignition prediction.The identified main factors that influence Yunnan wildfire occurrence were forest coverage ratio,month,season,surface roughness,10 days minimum of the 6 h maximum humidity,and 10 days maxima of the 6 h average and maximum temperatures.These seven variables made the greatest contributions to regional wildfire susceptibility.Susceptibility maps developed from the models provide information regarding the spatial variation of ignition susceptibility,which can be used in regional wildfire risk management.Yinxue Cao Ming Wang Kai Liu 2017International Journal of Disaster Risk Science2017,8,2:3
7Low-frequency repetitive transcranial magnetic simulation prevents chronic epileptic seizure显示文摘Although low-frequency repetitive transcranial magnetic simulation can potentially treat epilepsy,its underlying mechanism remains unclear.This study investigated the influence of low-frequency repetitive transcranial magnetic simulation on changes in several nonlinear dynamic electroencephalographic parameters in rats with chronic epilepsy and explored the mechanism underlying repetitive transcranial magnetic simulation-induced antiepileptic effects.An epilepsy model was established using lithium-pilocarpine intraperitoneal injection into adult Sprague-Dawley rats,which were then treated with repetitive transcranial magnetic simulation for 7 consecutive days.Nonlinear electroencephalographic parameters were obtained from the rats at 7,14,and 28 days post-stimulation.Results showed significantly lower mean correlation-dimension and Kolmogorov-entropy values for stimulated rats than for non-stimulated rats.At 28 days,the complexity and point-wise correlation dimensional values were lower in stimulated rats.Low-frequency repetitive transcranial magnetic simulation has suppressive effects on electrical activity in epileptic rats,thus explaining its effectiveness in treating epilepsy.Yinxu Wang Xiaoming Wang Sha Ke Juan Tan Litian Hu Yaodan Zhang Wenjuan Cui 2013Neural Regeneration Research2013,8,27:2

11Prepartum body conditions affect insulin signaling pathways in postpartum adipose tissues in transition dairy cows显示文摘Background: Overconditioned dairy cows are susceptible to excessive lipolysis and increased insulin resistance during the transition period.The associations among body fat reserve,insulin resistance,and lipolysis in adipose tissues(AT) remain to be elucidated.Therefore,this study aimed to investigate whether excessive fat reserves influence the insulin signaling pathway in AT postpartum.Results: Twenty multiparous dairy cows were selected and assigned to one of two groups,according to prepartum body condition score(BCS): Control group(BCS = 3.0–3.5;n = 10) and Overconditioned group(BCS ≥ 4.0;n = 10).Blood samples were collected on days-14,-7,-4,-2,-1,0,1,2,4,7,and 14 relative to parturition.Subcutaneous AT were collected on day 2 following parturition for quantitative real-time polymerase chain reaction and western blot analyses.No differences were observed between the two groups in serum glucose,non-esterified fatty acids,β-hydroxybutyric acid,tumor necrosis factor(TNF)α,insulin,or leptin concentrations during the experimental period.Compared with the control cows,the overconditioned cows had lower serum triglyceride levels and higher adiponectin concentrations.In the AT postpartum,insulin receptor mRNA and protein levels were lower in the overconditioned cows than in the control cows,and no differences were found in glucose transporter 4 mRNA.Compared with the control cows,the overconditioned cows had lower mRNA levels of TNFα and higher mRNA levels of peroxisome proliferator-activated receptor gamma(PPARγ) in AT postpartum.The phosphorylated protein kinase B(AKT) content and phosphorylation rate of AKT were increased in the overconditioned cows compared with the control cows,which suggested that the downstream insulin signaling in AT was affected.Conclusions: In the present study,transition dairy cows with higher BCS did not show more fat mobilization.The changes of insulin signaling pathway in AT postpartum of overconditioned cows may be partly related to the expression of PPARγ and TNFα,and the secretion of adiponectin.Fanjian Zhang Dan Li Qiong Wu Jian Sun Wenyi Guan Yinxu Hou Yaohong Zhu Jiufeng Wang 2019Journal of Animal Science and Biotechnology2019,10,3:2
12Effect of bile salts and bile acids on human gastric mucosal epithelial cells显示文摘Objective:To explore the effect of bile salt and bile acid on cultured eternalized human gastric mucosa epithelium GES-1 cells. Methods:Cultured eternalized human gastric mucosa epithelium GES-1 cells were treated with media containing 6 different kinds of bile salts and 3 different kinds of bile acids and their mixture with different concentrations: GCDC(glycochenodeoxycholate),GDC(glycodeoxycholate),GC(glycocholate),TCDC(taurochenodeoxycholate),TDC(taurodeoxycholate),TC (taurocholate),LCA(lithocholicacid),CA(cholic acid),DCA(deoxycholic acid)(50μmol/L,250μmol/L,500μmol/L,1000μmol/L),DY(mixture of bile salts) and DS(mixture of bile acids)(250μmol/L,500μmol/L,1000μmol/L,1500μmol/L,2000μmol/L),in comparison with the control group(in normal media without bile salts and bile acids). Cell proliferation was assessed by MTT(3-[4,5-Dimethylthiaolyl]-2,5-diphenyl-tetrazolium bromide) assay for 72 hours with different concentrations and the apoptotic cells were assayed by flow cytometry (FCM) with Annex V-FITC conjugated with propidium iodide(PI) staining for 24 hours with different concentrations(1500,2000μmol/L). Results:There was no significant difference in morphology and cell proliferation in GC group after 24~72 h. Low concentration(50μmol/L) of GCDC,GDC,TCDC,TDC and TC accelerated gastric epithelial cell growth in a dosage-time dependent manner. At middle concentration(250-500μmol/L),it showed positive effect after 24~48 h,while negative effect after 72 h. At high concentration(1000μmol/L),it accelerated gastric epithelial cell growth after 24h and show consistent inhibition even leading to necrosis after 48~72 h. LCA and CA showed a positive effect on the concentration of 50μmol/L after 24~72 h,while 250-1000μmol/L showed a trend towards apoptosis after 24~72 h. At 50-500μmol/L,DCA showed proliferation after 24 h and apoptosis after 48~72 h,but showed necrosis after 24~72 h at 1000 μmol/L. DY and DS could facilitate normal gastric mucosa epithelial cell growth at low concentration (250-500μmol/L),however at 1000-2000μmol/L the trend shifted from apoptosis to necrosis. FCM with Annexin-V conjugated with PI staining revealed that GCDC,GDC,GC,TCDC,TDC,TC,LCA,CA,DCA,DY and DS induced apoptosis of human gastric mucosal epithelial cells. They were all significantly higher than that of the control(P < 0.05),but there was no significant difference in GC group (P > 0.05). The bile salts induced apoptosis in a time-dose-dependent manner. 13Utilizing phase-shifted long-period fiber grating to suppress spectral broadening of a high-power fiber MOPA laser system显示文摘Suppressing nonlinear effects in high-power fiber lasers based on fiber gratings has become a hotspot.At present,research is mainly focused on suppressing stimulated Raman scattering in a high-power fiber laser.However,the suppression of spectral broadening,caused by self-phase modulation or four-wave mixing,is still a challenging attribute to the close distance between the broadened laser and signal laser.If using a traditional fiber grating with only one stopband to suppress the spectral broadening,the signal power will be stripped simultaneously.Confronting this challenge,we propose a novel method based on phase-shifted long-period fiber grating(PS-LPFG)to suppress spectral broadening in a high-power fiber master oscillator power amplifier(MOPA)laser system in this paper.A PS-LPFG is designed and fabricated on 10/130 passive fiber utilizing a point-by-point scanning technique.The resonant wavelength of the fabricated PS-LPFG is 1080 nm,the full width at half maximum of the passband is 5.48 nm,and stopband extinction exceeds 90%.To evaluate the performance of the PS-LPFG,the grating is inserted into the seed of a kilowattlevel continuous-wave MOPA system.Experiment results show that the 30 dB linewidth of the output spectrum is narrowed by approximately 37.97%,providing an effective and flexible way for optimizing the output linewidth of highpower fiber MOPA laser systems.Yinxu Bian Kerong Jiao Xuecheng Wu Hua Shen Feiyan Yang Rihong Zhu 2021High Power Laser Science and Engineering2021,9,3:2
14Dental stem cell-derived extracellular vesicles as promising therapeutic agents in the treatment of diseases显示文摘Dental stem cells(DSCs), an important source of mesenchymal stem cells(MSCs), can be easily obtained by minimally invasive procedures and have been used for the treatment of various diseases. Classic paradigm attributed the mechanism of their therapeutic action to direct cell differentiation after targeted migration, while contemporary insights into indirect paracrine effect opened new avenues for the mystery of their actual low engraftment and differentiation ability in vivo. As critical paracrine effectors, DSC-derived extracellular vesicles(DSC-EVs) are being increasingly linked to the positive effects of DSCs by an evolving body of in vivo studies. Carrying bioactive contents and presenting therapeutic potential in certain diseases, DSC-EVs have been introduced as promising treatments. Here, we systematically review the latest in vivo evidence that supports the therapeutic effects of DSC-EVs with mechanistic studies. In addition, current challenges and future directions for the clinical translation of DSC-EVs are also highlighted to call for more attentions to the(Ⅰ) distinguishing features of DSC-EVs compared with other types of MSC-EVs,(Ⅱ)heterogeneity among different subtypes of DSC-derived EVs,(Ⅲ) action modes of DSC-EVs,(Ⅳ) standardization for eligible DSC-EVs and(Ⅴ) safety guarantee for the clinical application of DSC-EVs.
