1In vivo gene expression profiling of the entomopathogenic fungus Beauveria bassiana elucidates its infection stratagems in Anopheles mosquito显示文摘The use of entomopathogenic fungi to control mosquitoes is a promising tool for reducing vector-borne disease transmission.To better understand infection stratagems of insect pathogenic fungi,we analyzed the global gene expression profiling of Beauveria bassiana at 36,60,84 and 108 h after topical infection of Anopheles stephensi adult mosquitoes using RNA sequencing(RNA-Seq).A total of 5,354 differentially expressed genes(DEGs) are identified over the course of fungal infection.When the fungus grows on the mosquito cuticle,up-regulated DEGs include adhesion-related genes involved in cuticle attachment,Pthll-like GPCRs hypothesized to be involved in host recognition,and extracellular enzymes involved in the degradation and penetration of the mosquito cuticle.Once in the mosquito hemocoel,the fungus evades mosquito immune system probably through up-regulating expression of |3-l,3-glucan degrading enzymes and chitin synthesis enzymes for remodeling of cell walls.Moreover,six previous unknown SSCP(small secreted cysteine-rich proteins) are significantly up-regulated,which may serve as 'effectors' to suppress host defense responses.B.bassiana also induces large amounts of antioxidant genes to mitigate host-generated exogenous oxidative stress.At late stage of infection,B.bassiana activates a broad spectrum of genes including nutrient degrading enzymes,some transporters and metabolism pathway components,to exploit mosquito tissues and hemolymph as a nutrient source for hyphal growth.These findings establish an important framework of knowledge for further comprehensive elucidation of fungal pathogenesis and molecular mechanism of Beauveria-mosqaito interactions.Yiling Lai Huan Chen Ge Wei Guandong Wang Fang Li Sibao Wang 2017Science China(Life Sciences)2017,60,8:5

4Approaching the audience:engagement markers in Longinus'On the Sublime显示文摘Metadiscourse is a significant method in revealing audience awareness,but related studies have been confined to linguistic markers and current writing analyses.This essay aims to enrich metadiscourse engagement in particular with certain rhetorical figures and to investigate audience awareness in a highly acclaimed Roman-era Greek classic:Longinus'On the Sublime.Drawing from a refined framework of engagement,we find explicit evidence of audience aware-ness as manifested in the author's use of engagement markers.We have sorted these markers into four types,including reader mentions(apostrophe and pro-nouns),directives(modal verbs and imperatives),questions(erotema and rogatio),and appeals to shared knowledge(emphasizers and comment clauses).We sug-gest that the frequent use of these engagement markers,in particular reader mentions,could be taken as evidence of the author's strong audience awareness and the cueing effect in facilitating perception.This integration of rhetorical fig-ures into the metadiscourse engagement markers contributes to the audience awareness analysis in a more explicit and comprehensive way.Yiling Fang Ganlin Zhuang 2022Language and Semiotic Studies2022,8,3:0
5A novel cartridge for nucleic acid extraction,amplification and detection of infectious disease pathogens with the help of magnetic nanoparticles显示文摘Nucleic acid detection(NAD)based on real-time polymerase chain reaction(real-time PCR)is gold standard for infectious disease detection.Magnetic nanoparticles(MNPs)are widely used for nucleic acid extraction(NAE)because of their excellent properties.Microfluidic technology makes automated NAD possible.However,most of the NAD microfluidic chips are too complex to be applied to point-of-care(POC)testing.In this paper,a simple-structure cartridge was developed for POC detection of infectious diseases.This self-contained cartridge can be divided into a magnetic-controlled NAE part,a valve-piston combined fluidic control part and a PCR chip,which is able to extract nucleic acid from up to 500μL of liquid samples by MNPs and finish the detection process from“sample in”to“answer out”automatically.Performance tests of the cartridges show that it met the demands of automated NAD.Results of on-cartridge detection of hepatitis B virus(HBV)demonstrated that this system has good uniformity and no cross-contamination between different cartridges,and the limit of detection(LOD)of this system for HBV in serum is 50 IU/mL.Multiplex detections of severe acute respiratory syndrome coronaviruses 2(SARS-CoV-2)with a concentration of 500 copies/mL were carried out on the system and 100%positive detection rate was achieved.Yile Fang Yue Wang Liangxi Zhu Haoran Liu Xiangyi Su Yuan Liu Zhu Chen Hui Chen Nongyue He 2023Chinese Chemical Letters2023,34,8:0

7Inhibition of glycogen synthase kinase 3βimproves cognitive function in aged mice by upregulating claudin presences in cerebral endothelial cells显示文摘Glycogen synthase kinase-3β(GSK-3β),a serine/threonine protein kinase,is widely distributed in mammalian brains.Since GSK-3βplays a vital role in the development of neurodegenerative disorders,the present study was designed to investigate the role of GSK-3βin the blood-brain barrier(BBB)permeability in aged mice.Morris water maze test was used to examine mouse cognitive function.BBB permeability was examined by the leakage of fluorescence signals of low-molecular weight dextran.GSK-3βinhibitor,4-benzyl-2-methyl-1,2,4-thiadiazolidine-3,5-dione(TDZD-8),was administrated in aged mice and in cultured mouse brain microvascular endothelial cells(bEnd.3).Compared with young mice,aged mice had increased leftover signals of dextran in the hippocampus and a lower score in the maze test,suggesting that aged mice have abnormal leakage of BBB and cognitive dysfunction.The protein expression of Toll-like receptor 4(TLR4)was increased,whereas the protein expressions of junction proteins(claudinl and claudin5)were reduced in endothelial cells of BBB in aged mice.Phosphorylated level of serine 9,an inhibitory residue in GSK-3βprotein,was decreased.TDZD-8 treatment downregulated TLR4 protein expression,upregulated claudinl and claudin5 protein expressions,and significantly improved cognitive function in aged mice.In bEnd.3 cells,TDZD-8 treatment reduced TLR4 expression and increased claudin5 expression in cells stimulated with lipopolysaccharides.In conclusion,the inhibition of GSK-3βactivity downregulates aging-induced TLR4 expression and restores the BBB integrity,resulting in the improvement of cognitive function in aged mice.Tao Liang Huihui Ju Yile Zhou Yajie Yang Yi Shi Hao Fang 2020Acta Biochimica et Biophysica Sinica2020,52,4:0
8Automated screening of primary cell-based aptamers for targeting and therapy of pancreatic cancer显示文摘Although it has been developed for many years, nucleic acid aptamer screening technology still fails to be widely used, a considerable part of it is due to the variability of tumor cell morphology, which leads to the use of immortalized cell lines in the laboratory to screen nucleic acid aptamers for recognition ability of tumor cells in the diseased body.To address this, primary cells that can be stably passaged were isolated and extracted from spontaneous tumors of genetically engineered pancreatic ductal adenocarcinoma model mice in this study.Next, an automated screening instrument for nucleic acid aptamers developed autonomously by our group was used to perform efficient aptamer screening using a limited number of cells, and the obtained nucleic acid aptamers were affinity verified at the cellular level.Finally, to answer the question of the cell growth environment difference on the recognition ability of nucleic acid aptamers, we verified its targeting ability to tumors in vivo on a nude mice xenograft tumor model, and further used a common antitumor drug doxorubicin combined with nucleic acid aptamers to verify the drug loading ability of this aptamer combined with the targeting therapeutic ability.Zhukang Guo Baijiang Jin Yile Fang Lian Jin Song Li Yan Deng Zhu Chen Hui Chen Yuanying Zhang Rabia Usman Nongyue He 2024Chinese Chemical Letters2024,35,2:0

10Bacterial community diversity of meltwater runoff and soil in Midre Lovénbreen glacier in Ny-Alesund,Arctic显示文摘Glacial meltwater runoff is a dynamic ecosystem.On the one hand,nutrient concentration changes as it flows from upstream to downstream,and on the other hand,bacterial community structure changes due to its contact with nearby soil during the flow process.We studied meltwater and soil in the Midre Lovénbreen glacier region,to explore changes in bacterial diversity as meltwater flows,and the relationship between meltwater and soil bacterial diversity.As glacial meltwater flows from upstream to downstream,the relative abundance of dominant bacterial groups changes.In addition,we found that during the flowing process,nutrient exchange and bacterial contact had occurred between the meltwater runoff and the soil.As a result,the distribution patterns of some bacteria in the meltwater are very similar to those in the soil.Finally,we combined distance-based redundancy analysis and weighted correlation network analysis to show that NO_(3)^(-)-N and NO_(2)^(-)-N are the most two significant factors affecting glacial meltwater and soil,respectively.Our results suggest that in such a close-knit ecosystem,the interaction of glacial meltwater with soil,as well as environmental factors,together determine bacterial community composition.LIN Lidong WANG Nengfei HAN Wenbing ZHANG Botao ZANGJiaye LI Qinxin QIN Yiling WANG Long ZHANG Fang LIU Jie 2022Advances in Polar Science2022,33,2:0

12Telocytes reduce oxidative stress by downregulating DUOX2 expression in inflamed lungs of mice显示文摘Telocytes(TCs),a novel type of interstitial cells,have been found to participate in tissue protection and repair.In this study,we investigated the antioxidative effects of TCs in inflamed lungs of mice.Acute respiratory distress syndrome(ARDS)mice were used as models of inflamed lungs of mice.Gene sequencing was used to screen the differentially expressed miRNAs in TCs after lipopolysaccharide(LPS)stimulation.AntagomiR-146a-5p-pretreated TCs were first injected into mice,and antioxidant activity of TCs was estimated.TCs,RAW264.7 cells,and MLE-12 cells were collected for the detection of expressions of NOX1–4,DUOX1–2,SOD1–3,GPX1–2,CAT,Nrf2,miR-146a-5p,and miR-21a-3p after LPS stimulation.Silencing miRNAs were delivered to examine the involved signaling pathways.Oxidative stress was examined by measuring malondialdehyde(MDA)levels.We found that microRNA-146a-5p and microRNA-21a-3p were upregulated in TCs after LPS stimulation.ARDS mice that were preinfused with TCs had lower lung tissue injury scores,lung wet-dry ratios,white blood cell counts in alveolar lavage fluid and lower MDA concentrations in lung tissue.However,in antagomiR-146a-5p-pretreated ARDS mice,the infusion of TCs caused no corresponding changes.After LPS stimulation,DUOX2 and MDA concentrations were downregulated in TCs,while DUOX2 was restored by antagomiR-146a-5p in TCs.Dual-luciferase reporter assay confirmed that CREB1 was downregulated by miR-146a-5p,while DUOX2 was downregulated by CREB1,which was confirmed by treating TCs with a specific CREB1 inhibitor.This study demonstrates that LPS stimulation upregulates miR-146a-5p in TCs,which downregulates the CREB1/DUOX2 pathway,resulting in a decrease in oxidative stress in cultured TCs.TCs reduce LPS-induced oxidative stress by decreasing DUOX2 in inflamed lungs of mice.Haihong Tang Tao Liang Yile Zhou Huihui Ju Dongli Song Hao Fang 2022Acta Biochimica et Biophysica Sinica2022,54,4:0
