1Development and analysis of a novel AF11-2 aptamer capable of enhancing the fluorescence of aflatoxin B1显示文摘Aflatoxin B1(AFB1)is one of the most common mycotoxins that threatens human health.As singlestranded oligonucleotides with high affinity and specificity,aptamers have incomparable effect on the targeted detection of AFB1.Herein,after 11 rounds of selection and analysis using a modified affinity chromatography-based SELEX strategy,the truncated 37 nt aptamer AF11-2 was successfully obtained.The aptamer shows good detection performance for AFB1,and can sensitively detect AFB1 in the range of 100-1000 nmol/L,with a detection limit of 42 nmol/L.In the detection of pretreated edible peanut oil samples,AF11-2 aptamer also showed a high recovery rate and good stability for AFB1,and achieved satisfactory results.In addition,AF11-2 aptamer can significantly enhance the fluorescence ability of AFB1,which is not available in traditional Afla17-2-3 aptamer.After molecular docking analysis,it was found that AF11-2 and Afla17-2-3 had different nucleotide binding sites for AFB1.Afla17-2-3 binds to the carbonyl O of AFB1,while AF11-2 binds to the pyrrolic O of AFB1,which may be the main reason that AF11-2 can enhance the fluorescence of AFB1.Wenjing Li Yian Pei Jine Wang 2022Chinese Chemical Letters2022,33,8:2

3Anti-neuroinflammatory responses in lipopolysaccharide-stimulated BV2 microglial cells upon treatment with methanol extract of Panax ginseng root显示文摘OBJECTIVE:To investigate the anti-neuroinflammatory properties of Panax ginseng(P.ginseng)root by measuring the levels of nitric oxide(NO),tumour necrosis factor-alpha(TNF-α),interleukin-6(IL-6),and interleukin-10(IL-10)in lipopolysaccharide(LPS)-stimulated BV2 microglia cells.METHODS:Maximal non-toxic dose(MNTD)of methanol extract of P.ginseng root culture on BV2 microglia cells was first determined via 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay,followed by treatment and LPS stimulation of cells,and the measurement of NO using Griess assay and TNF-α,IL-6,and IL-10 using ELISA assay.RESULTS:The MNTD of P.ginseng root extract was determined to be(587±57)µg/mL.Following that,NO and IL-6 levels were found to be insignificantly reduced by 6.88%and 0.14%respectively in stimulated cells upon treatment with MNTD.Treatment with½MNTD yielded similar insignificant result,with only a reduction of 3.58%and 0.08%in NO and IL-6 levels respectively.However,TNF-αand IL-10 levels were significantly downregulated by 15.64%and 34.96%respectively upon treatment with P.ginseng root extract at MNTD.CONCLUSION:Methanol extract of P.ginseng root culture did not show any significant anti-inflammatory effects on NO and IL-6 levels,but might potentially possess both anti-neuroinflammatory and pro-neuroinflammatory properties through the downregulation of TNF-αand IL-10 respectively.Chien Yi Koay Anna Pick Kiong Ling Ying Pei Wong Rhun Yian Koh Sobri Hussein 2021Journal of Traditional Chinese Medicine2021,41,2:0
4Experimental Evaluation of the Mechanical Properties of Cement Sheath under High-Temperature Conditions显示文摘The sealing integrity of cement sheath in offshore wells is seriously threatened under high-temperature conditions,resulting in gas channeling and other problems.Given the lack of experimental results,in this study relevant samples of a cement slurry sealing section of a typical offshore high-temperature well have been prepared and analyzed.In particular,the mechanical properties have been assessed with a triaxial pressure servo instrument and a high-temperature curing kettle.The density and the Poisson’s ratio of the samples have also been tested.The stress-strain curve has been drawn to obtain the elastic modulus and the compressive strength.The rock brittleness index has been calculated according to the measured elastic modulus and the Poisson’s ratio,together with the brittleness and the compressibility of the cement samples.The test results show that the mechanical properties and bonding strength of the cement samples are optimal at 130°C,medium at 150°C,and poor at 180°C.Pei Zou Zhiqiang Huang Yian Tong Leichuan Tan Rui Li Kai Wei Deng Qiao 2022Fluid Dynamics & Materials Processing2022,18,3:0
