1Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. 2Synergistic anti-inflammatory and osteogenic n-HA/resveratrol/chitosan composite microspheres for osteoporotic bone regeneration显示文摘The development of functional materials for osteoporosis is ultimately required for bone remodeling.However,grafts were accompanied by increasing pro-inflammatory cytokines that impaired bone formation.In this work,nano-hydroxyapatite(n-HA)/resveratrol(Res)/chitosan(CS)composite microspheres were designed to create a beneficial microenvironment and help improve the osteogenesis by local sustained release of Res.Study of in vitro release confirmed the feasibility of n-HA/Res/CS microspheres for controlled Res release.Notably,microspheres had anti-inflammatory activity evidenced by the decreased expression of pro-inflammatory cytokines TNF-α,IL-1βand iNOS in RAW264.7 cells in a dose dependent manner.Further,enhanced adhesion and proliferation of BMSCs seeded onto microspheres demonstrated that composite microspheres were conducive to cell growth.The ability to enhance osteo-differentiation was supported by up-regulation of Runx2,ALP,Col-1 and OCN,and substantial mineralization in osteogenic medium.When implanted into bone defects in the osteoporotic rat femoral condyles,enhanced entochondrostosis and bone regeneration suggested that the n-HA/Res/CS composite microspheres were more favorable for impaired fracture healing.The results indicated that optimized n-HA/Res/CS composite microspheres could serve as promising multifunctional fillers for osteoporotic bone defect/fracture treatment.Limei Li Mali Yu Yao Li Qing Li Hongcai Yang Meng Zheng Yi Han Di Lu Sheng Lu Li Gui 2021Bioactive Materials2021,6,5:6

7Sequential gastrodin release PU/n-HA composite scaffolds reprogram macrophages for improved osteogenesis and angiogenesis显示文摘Wound healing is a highly orchestrated process involving a variety of cells,including immune cells.Developing immunomodulatory biomaterials for regenerative engineering applications,such as bone regeneration,is an appealing strategy.Herein,inspired by the immunomodulatory effects of gastrodin(a bioactive component in traditional Chinese herbal medicine),a series of new immunomodulatory gastrodin-comprising biodegradable polyurethane(gastrodin-PU)and nano-hydroxyapatite(n-HA)(gastrodin-PU/n-HA)composites were developed.RAW 264.7 macrophages,rat bone marrow mesenchymal stem cells(rBMSCs),and human umbilical vein endothelial cells(HUVECs)were cultured with gastrodin-PU/n-HA containing different concentrations of gastrodin(0.5%,1%,and 2%)to decipher their immunomodulatory effects on osteogenesis and angiogenesis in vitro.Results demonstrated that,compared with PU/n-HA,gastrodin-PU/n-HA induced macrophage polarization toward the M2 phenotype,as evidenced by the higher expression level of pro-regenerative cytokines(CD206,Arg-1)and the lower expression of pro-inflammatory cytokines(iNOS).The expression levels of osteogenesis-related factors(BMP-2 and ALP)in the rBMSCs and angiogenesis-related factors(VEGF and BFGF)in the HUVECs were significantly up-regulated in gastrodin-PU/n-HA/macrophage-conditioned medium.The immunomodulatory effects of gastrodin-PU/n-HA to reprogram macrophages from a pro-inflammatory(M1)phenotype to an anti-inflammatory and pro-healing(M2)phenotype were validated in a rat subcutaneous implantation model.And the 2%gastrodin-PU/n-HA significantly decreased fibrous capsule formation and enhanced angiogenesis.Additionally,2%gastrodin-PU/n-HA scaffolds implanted in the rat femoral condyle defect model showed accelerated osteogenesis and angiogenesis.Thus,the novel gastrodin-PU/n-HA scaffold may represent a new and promising immunomodulatory biomaterial for bone repair and regeneration.Limei Li Qing Li Li Gui Yi Deng Lu Wang Jianlin Jiao Yingrui Hu Xiaoqian Lan Jianhong Hou Yao Li Di Lu 2023Bioactive Materials2023,,1:1

12Determination of Periplocymarin in Human Blood and Urine by High-Performance Liquid Chromatography-Mass Spectrometry显示文摘A simple,rapid and sensitive liquid chromatography with tandem mass spectrometry method for the determination of periplocymarin in human blood and urine was developed.The digoxin‑d3 was used as an internal standard.Periplocymarin and digoxin‑d3(IS)were processed with ethyl acetate by liquid–liquid extraction.The chromatographic separation was performed on a Shim‑pack XR‑ODSIII C18 column with a 7 min gradient elution using methanol‑ammonium formate(5 mmol/L)as mobile phase at a flow rate of 0.3 mL/min(65:35,v/v).The detection was performed on a triple quadrupole tandem mass spectrometer using positive‑ion mode electrospray ionization in selected reaction monitoring mode.The periplocymarin was well separated from the internal standard.Two calibration curves were linear within the concentration range 0.01–1µg/mL.The limit of detection and quantification of blood and urine samples were both estimated at 0.005 and 0.01µg/mL.The interday and intraday precisions,accuracy,and recovery were assessed to verify this method.The results showed that the method was suitable for the determination of periplocymarin in forensic toxicological analysis and clinical diagnosis.Chen Jian Xia Wei Qing Tao Chen Fan Wu Hao Fu Ying Qing Wu Zhi Gui Dai Xin Hua Ye Yi Yan You Yi Liao Lin Chuan 2017Journal of Forensic Science and Medicine2017,3,4:0
133-to 24-month Follow-up on COVID-19 with Pulmonary Tuberculosis Survivors after Discharge:Results from a Prospective,Multicenter Study显示文摘Objective Coronavirus disease 2019(COVID-19)and tuberculosis(TB)are major public health and social issues worldwide.The long-term follow-up of COVID-19 with pulmonary TB(PTB)survivors after discharge is unclear.This study aimed to comprehensively describe clinical outcomes,including sequela and recurrence at 3,12,and 24 months after discharge,among COVID-19 with PTB survivors.Methods From January 22,2020 to May 6,2022,with a follow-up by August 26,2022,a prospective,multicenter follow-up study was conducted on COVID-19 with PTB survivors after discharge in 13 hospitals from four provinces in China.Clinical outcomes,including sequela,recurrence of COVID-19,and PTB survivors,were collected via telephone and face-to-face interviews at 3,12,and 24 months after discharge.Results Thirty-two COVID-19 with PTB survivors were included.The median age was 52(45,59)years,and 23(71.9%)were men.Among them,nearly two-thirds(62.5%)of the survivors were moderate,three(9.4%)were severe,and more than half(59.4%)had at least one comorbidity(PTB excluded).The proportion of COVID-19 survivors with at least one sequela symptom decreased from 40.6%at 3 months to 15.8%at 24 months,with anxiety having a higher proportion over a follow-up.Cough and amnesia recovered at the 12-month follow-up,while anxiety,fatigue,and trouble sleeping remained after 24months.Additionally,one(3.1%)case presented two recurrences of PTB and no re-positive COVID-19during the follow-up period.Conclusion The proportion of long symptoms in COVID-19 with PTB survivors decreased over time,while nearly one in six still experience persistent symptoms with a higher proportion of anxiety.The recurrence of PTB and the psychological support of COVID-19 with PTB after discharge require more attention.WANG Jing Ya ZONG Xing Yu WU Gui Hui QI Yuan Lin LI Hui Zhen JI Xin Yu TONG Lin ZHANG Lei YANG Ming Bo YANG Pu Ye LI Ji Ke XIAO Fu Rong ZHANG Lin Song HU Yun Hong LIU Hong De XU Shou Fang SUN Sheng WU Wei MAO Ya LI Min Qing HOU Hao Hua GONG Zhao Yuan GUO Yang JIAO Li Wen QIN Jin WANG Ding Yi WANG Fang GUAN Li LIN Gang MA Yan WANG Yan Ping SHI Nan Nan 2022Biomedical and Environmental Sciences2022,35,12:0
