2GeoScope:Full 3D Geospatial Information System Case Study显示文摘Aiming at the integrative management and comprehensive applications of large-scale 3D geospatial information covering the full 3D space of a city,this paper briefly introduces the design and implementation of a full 3D GIS platform:GeoScope,which provides a professional solution for the massive full three-dimensional geospatial data integration,management,analysis,visualization,and applications.GeoScope is characterized by:(1) extendible software architecture based on the hierarchical message bus,facilitates multimodal integrative applications of 2D GIS and 3D GIS;(2) unified 3D city models,support multiscale semantic representation of outdoor & indoor and aboveground & underground 3D objects;(3) high-efficient 3D geospatial database engine,supports integrated management of massive 3D geospatial data for real-time applications;and (4) high-performance visualization engine exploiting the massively parallel computation architecture of modern GPUs,supports real-time realistic rendering of large-scale complicated 3D geospatial environments.The successful pilot application of GeoScope is also illustrated with the 3D city models of 8494 km2 of the whole Wuhan City,the largest city in middle China.ZHANG Yeting ZHU Qing LIU Gang ZHENG Wenting LI Zhonghua DU Zhiqiang 2011Geo-Spatial Information Science2011,14,2:7
3B-box proteins:Pivotal players in light-mediated development in plants^FA显示文摘Light signals mediate a number of physio-logical and developmental processes in plants, such as flowering, photomorphogenesis, and pigment accu-mulation. Emerging evidence has revealed that a group of B-box proteins (BBXs) function as central players in these light-mediated developmental processes. B-box proteins are a class of zinc-coordinated transcription factors or regulators that not only directly mediate the transcription of target genes but also interact with various other factors to create a complex regulatory network involved in the precise control of plant growth and development. 4Efficacy of rigosertib, a small molecular RAS signaling disrupter for the treatment of KRAS-mutant colorectal cancer显示文摘Objective:Mutant KRAS,the principal isoform of RAS,plays a pivotal role in the oncogenesis of colorectal cancer by constitutively activating the RAF/MEK/ERK and PI3K/AKT pathways.Effective targeted therapies are urgently needed.We investigated whether rigosertib,a benzyl styryl sulfone RAS signaling disruptor,could selectively kill KRAS-mutant colorectal cancer cells.Methods:CCK-8 was used to determine the cell viability.Patient-derived tumor and cancer cell xenograft models were used to detect the inhibitory efficacy of rigosertib.Flow cytometry was used to evaluate the apoptosis and cell cycle progression.Apoptosis and cell cycle arrest markers were detected by Western blot.DCFH-DA was used to determine the reactive oxygen species.Immunohistochemistry staining and Western blot were performed to characterize RAS signaling markers in colorectal cancer tissues and cells.Results:Rigosertib(RGS)exhibited a cytotoxic effect against colorectal cancer cells,which was greater in KRAS-mutant cells.Furthermore,RGS induced mitotic arrest and oxidative stress-dependent apoptosis in KRAS-mutant DLD1 and HCT116 cells.Besides,RGS disrupted RAS signaling,and the inhibition of RAS/MEK/ERK was independent of cellular oxidative stress.Using patient-derived xenograft models,the response and tumor inhibition of RGS were significantly higher in the KRAS-mutant subgroup,while p-MEK,p-ERK,and p-AKT levels of RGS-treated tumors were significantly decreased.Finally,in a KRAS-mutant,chemotherapy-resistant patient-derived xenograft model,RGS showed a stronger therapeutic effect than the combination standard therapy involving fluoropyrimidine+oxaliplatin/irinotecan+bevacizumab.Conclusions:These data showed that targeting RAS signaling using RGS could be a therapeutic treatment for KRAS-mutant colorectal cancer patients.Xinyi Zhou Qian Xiao Dongliang Fu Haochen Zhang Yang Tang Jinjie He Yeting Hu Xiangxing Kong Fei Teng Xiangrui Liu Ying Yuan Kefeng Ding 2022Cancer Biology & Medicine2022,19,2:2
5Carbon emissions induced by farmland expansion in China during the past 300 years显示文摘Scientific assessment of the accounting over carbon in the terrestrial ecosystem in the process land use/land cover changes caused by human activities will help reduce the uncertainty in estimating carbon emissions from the terrestrial ecosystem. This study employs a bookkeeping model to estimate the carbon emissions from farmland reclamation in China during the past 300 years based on the annual rate of land use changes(derived from historical natural vegetation, farmland data), preset carbon density and coefficients of disturbance curves. We find out that:(1) there was a net increase of 79.30×10~4km^2 in national farmland; about 65% of reclaimed farmland had been forest land and 26% of that had been grass land previously;(2) the total amount of carbon emissions from farmland expansion in China had been between 2.94 and 5.61 Pg with the median 3.78 Pg during the past 300 years; specifically, carbon emissions of vegetation were 1.58 Pg while those of soil ranged from 1.35 Pg to4.03 Pg with the median 2.20 Pg;(3) carbon emissions vary greatly across various ecosystems: the emissions were most from forest land, and then grass land and swamps, and the least from shrubs; deserts functioned more likely to be carbon stock in the process of land reclamation;(4) along the time line, carbon emissions had decreased first and then increased while the peak emissions occurred in the first half of 20 th century; and spatially, carbon emissions were most released in Northeast and Southwest China; Northwest China was of the minimum carbon emissions.Xuhong YANG Xiaobin JIN Xiaomin XIANG Yeting FAN Jing LIU Wei SHAN Yinkang ZHOU 2019Science China Earth Sciences2019,62,2:2

12Proximity-enabled covalent binding of IL-2 to IL-2Rα selectively activates regulatory T cells and suppresses autoimmunity显示文摘Interleukin-2(IL-2)is a pleiotropic cytokine that orchestrates bidirectional immune responses via regulatory T cells(Tregs)and effector cells,leading to paradoxical consequences.Here,we report a strategy that exploited genetic code expansion-guided incorporation of the latent bioreactive artificial amino acid fluorosulfate-L-tyrosine(FSY)into IL-2 for proximity-enabled covalent binding to IL-2Rαto selectively promote Treg activation.We found that FSY-bearing IL-2 variants,such as L72-FSY,covalently bound to IL-2Rαvia sulfur-fluoride exchange when in proximity,resulting in persistent recycling of IL-2 and selectively promoting the expansion of Tregs but not effector cells.Further assessment of L72-FSY-expanded Tregs demonstrated that L72-FSY maintained Tregs in a central memory phenotype without driving terminal differentiation,as demonstrated by simultaneously attenuated expression of lymphocyte activation gene-3(LAG-3)and enhanced expression of programmed cell death protein-1(PD-1).Subcutaneous administration of L72-FSY in murine models of pristane-induced lupus and graft-versus-host disease(GvHD)resulted in enhanced and sustained therapeutic efficacy compared with wild-type IL-2 treatment.The efficacy of L72-FSY was further improved by N-terminal PEGylation,which increased its circulatory retention for preferential and sustained effects.This proximity-enabled covalent binding strategy may accelerate the development of pleiotropic cytokines as a new class of immunomodulatory therapies.Bo Zhang Jiaqi Sun Yeshuang Yuan Dezhong Ji Yeting Sun Yudong Liu Shengjie Li Xingxing Zhu Xunyao Wu Jin Hu Qiu Xie Ling Wu Lulu Liu Boyang Cheng Yuanjie Zhang Lingjuan Jiang Lidan Zhao Fei Yu Wei Song Min Wang Yue Xu Shiliang Ma Yunyun Fei Lihe Zhang Demin Zhou Xuan Zhang 2023Signal Transduction and Targeted Therapy2023,8,2:0
13Trimetaphosphate-induced chiral selection between amino acid and nucleoside using ^(15)N-^(31)P coupling NMR显示文摘Life on Earth uses a common set of L-amino acids(L-aa)to construct proteins and D-nucleosides(D-Nu)to form nucleic acids,which serve as the carrier of genetic information.Herein,we reveal the in-trinsic mechanism of chiral selection of L-aa and D-Nu from the perspective of chemical origin of life.This work employed^(15)N-labeled L-aa and performed one-pot synthesis of nucleotide amidate of amino acid(N-aa-NMP)using equal amounts of L-^(15)N-aa and D-^(14)N-aa with D-/L-Nu in the aqueous solution of trimetaphosphate,generating L-^(15)N-aa-NMP and D-^(14)N-aa-NMP,respectively.The ^(31)P-NMR data indicated that L-aa was preferentially selected during the formation of N-aa-NMP in the presence of D-Nu.Surpris-ingly,D-aa was preferred over L-aa in the presence of L-Nu.Further analysis revealed that L-^(15)N-aa-D-NMP vs.D-^(14)N-aa-L-NMP and D-^(14)N-aa-D-NMP vs.L-^(15)N-aa-L-NMP were mirror isomers of each other,respec-tively.These data suggest that there could be a set of chiral systems opposite to that on Earth,which infers there might be a world of life that is a mirror image of the Earth.Jianxi Ying Ruiwen Ding Yumeng Zhang Bowen Han Yeting Guo Ning Wang Dandan Guo Yile Wu Junwei Huang Yan Liu Feng Ni Shaohua Huang Yufen Zhao 2022Chinese Chemical Letters2022,33,2:0
14Influence Mechanism of New-type Urbanization on Urban Land Use Efficiency in the Yangtze River Delta, China显示文摘Rapid urbanization in China has led to inefficient use of urban land and spatial structure disorder,attracting attention from academia and society.Taking the Yangtze River Delta,China as an example,this study constructed an index evaluation system that quantitatively analyses the impact of new-type urbanization(NTU)on urban land use efficiency(ULUE)from 2000 to 2020 using a panel data vector autoregressive model.The results show that NTU in the Yangtze River Delta promotes ULUE improvement.However,the promotion of NTU to ULUE is limited,and the level of urban economic development also plays a role in promoting the change of ULUE.Moreover,the study further analyzed the results of urbanization decomposition and found that population urbanization(PU),economic urbanization(EU),spatial urbanization(SU),and social urbanization(SCU)can explain changes in ULUE in the Yangtze River Delta to a certain extent.In terms of variance decomposition,PU contributed the most to ULUE,followed by EU,SU,and SCU.Some necessary measures should be taken to coordinate the development of different types of urbanization,improve the sustainable utilization level of land resources,and provide a reference for high-quality development in the Yangtze River Delta.YANG Qingke DING Linlin WANG Lei LIU Chao FAN Yeting LI Ying WANG Yazhu 2023Chinese Geographical Science2023,33,3:0

16Task-Based Resource Allocation Bid in Edge Computing Micro Datacenter显示文摘Edge computing attracts online service providers(SP)to offload services to edge computing micro datacenters that are close to end users.Such offloads reduce packet-loss rates,delays and delay jitter when responding to service requests.Simultaneously,edge computing resource providers(RP)are concerned with maximizing incomes by allocating limited resources to SPs.Most works on this topic make a simplified assumption that each SP has a fixed demand;however,in reality,SPs themselves may have multiple taskoffloading alternatives.Thus,their demands could be flexibly changed,which could support finer-grained allocations and further improve the incomes for RPs.Here,we propose a novel resource bidding mechanism for the RP in which each SP bids resources based on the demand of a single task(task-based)rather than the whole service(servicebased)and then the RP allocates resources to these tasks with following the resource constraints at edge servers and the sequential rule of task-offloading to guarantee the interest of SPs.We set the incomes of the RP as our optimization target and then formulate the resource allocation problem.Two typical greedy algorithms are adopted to solve this problem and analyze the performance differences using two different bidding methods.Comprehensive results show that our proposal optimizes resource utilization and improves the RP’s incomes when resources in the edge computing datacenter are limited.Yeting Guo Fang Liu Nong Xiao Zhengguo Chen 2019Computers, Materials & Continua2019,,8:0
