1美国临床药学学科60年发展概况显示文摘目的介绍美国临床药学领域进展情况,为我国临床药学学科发展提供借鉴参考。方法通过Pubmed、中国知网等工具检索介绍美国临床药学教育、毕业后培训以及临床药学实践相关文献,结合美国专家学术报告内容,了解美国临床药学学科进展情况。结果经过60年发展,美国临床药学教育、毕业后培训及临床药学实践已形成较完善体系。Pharm.D已成为药师从业的唯一准入学位。美国药学教育越来越重视对学生能力的培养,注重药学实践的实际需求。完成PGY1项目培训已被作为从事临床药学实践的基本要求,完成PGY2项目培训并取得专业资格证书已逐渐成为在特殊专业领域工作的必需要求。药学实践不断实现专科化、标准化。临床药学研究主要针对临床药学服务及相关问题,而不是进行基础性药学科学研究。临床药学研究促进了临床药学学科发展和临床药学服务的有效实施。通过药学专业委员会(BPS)的专业资格认证保证了临床药师的专业胜任力。结论美国临床药学学科体系经过60年发展已趋于完善,相关经验值得我们学习和借鉴。余自成 Gary C YEE 朱珠 Wafa Y.DAHDAL 2019中国临床药学杂志2019,28,3:14

3Season of the year influences infection rates following total hip arthroplasty显示文摘AIM To research the influence of season of the year on periprosthetic joint infections.METHODS We conducted a retrospective review of the entire Medicare files from 2005 to 2014. Seasons were classified as spring, summer, fall or winter. Regional variations were accounted for by dividing patients into four geographic regions as per the United States Census Bureau(Northeast, Midwest, West and South). Acute postoperative infection and deep periprosthetic infections within 90 d after surgery were tracked. RESULTS In all regions, winter had the highest incidence of periprosthetic infections(mean 0.98%, SD 0.1%) and was significantly higher than other seasons in the Midwest, South and West(P < 0.05 for all) but not the Northeast(P = 0.358). Acute postoperative infection rates were more frequent in the summer and were significantly affected by season of the year in the West.CONCLUSION Season of the year is a risk factor for periprosthetic joint infection following total hip arthroplasty(THA). 5Novel role of STAT3 in microglia-dependent neuroinflammation after experimental subarachnoid haemorrhage显示文摘Background and purpose Signal transducer and activator of transcription 3(STAT3)may contribute to the proinflammation in the central nervous system diseases by modulating the microglial responses.Thus,this study was intended to investigate the effect of STAT3 on microglia-dependent neuroinflammation and functional outcome after experimental subarachnoid haemorrhage(SAH).Methods The SAH model was established by endovascular perforation in the mouse.Real-time PCR(RtPCR)and western blot were used to examine the dynamic STAT3 signalling pathway responses after SAH.To clarify the role of the STAT3 signalling pathway in the microglia-dependent neuroinflammation after SAH,the microglia-specific STAT3 knockout(KO)mice were generated by the Cre-LoxP system.The neurological functions were assessed by Catwalk and Morris water maze tests.Neuronal loss after SAH was determined by immunohistochemistry staining.Microglial polarisation status after STAT3 KO was then examined by RtPCR and immunofluorescence.Results The STAT3 and Janus kinase-signal transducer 2 activated immediately with the upregulation and phosphorylation after SAH.Downstream factors and related mediators altered dynamically and accordingly.Microglial STAT3 deletion ameliorated the neurological impairment and alleviated the early neuronal loss after SAH.To investigate the underlying mechanism,we examined the microglial reaction after STAT3 KO.STAT3 deletion reversed the increase of microglia after SAH.Loss of STAT3 triggered the early morphological changes of microglia and primed microglia from M1 to M2 polarisation.Functionally,microglial STAT3 deletion suppressed the SAH-induced proinflammation and promoted the anti-inflammation in the early phase.Conclusions STAT3 is closely related to the microglial polarisation transition and modulation of microglia-dependent neuroinflammation.Microglial STAT3 deletion improved neurological function and neuronal survival probably through promoting M2 polarisation and anti-inflammatory responses after SAH.STAT3 may serve as a promising therapeutic target to alleviate early brain injury after SAH.Zhiyuan Vera Zheng Junfan Chen Hao Lyu Sin Yu Erica Lam Gang Lu Wai Yee Chan George K C Wong 2022Stroke & Vascular Neurology2022,7,1:3
6机械负荷调节猪腰椎间盘细胞表达α5β1整合素的体外研究显示文摘目的研究α5β1整合素在体外培养的猪腰椎间盘细胞中的表达和机械负荷对其的影响。方法取10只年龄5~6周、体重25~30kg的猪,处死后12h内,无菌条件下切取完整的腰椎,剔除腰椎周围的韧带和软组织,尤其是椎间盘周围的韧带。从腹侧一次性切开椎间盘取出髓核(nucleuspulpous,NP),仔细分离纤维环(anulusfibrous,AF),将两者立即置于Hanks平衡盐溶液中,制成细胞悬液。分别对腰椎间盘的AF和NP细胞施加1MPa、1Hz,3h/d,共3d的周期性液压,通过对细胞的形态学观察、Western免疫印迹和免疫组织化学染色,检测α5β1整合素在正常腰椎间盘AF细胞和NP细胞中的表达及周期性压力对其的影响。结果经周期性液压后,NP细胞的存活率大于90%,AF细胞的存活率大于85%,加压后的AF细胞和NP细胞均可见体积缩小。α5β1整合素在正常腰椎间盘AF细胞和NP细胞中的表达呈强阳性。Western免疫印迹结果显示:加压后α5β1整合素在AF细胞中的表达均明显减少,P值分别为0.000、0.003,与对照组比较差异有统计学意义;α5β1整合素在NP细胞中的表达也明显减少,P值分别为0.001、0.015,与对照组比较差异有统计学意义。结论机械负荷可引起腰椎间盘细胞中α5β1整合素的变化,提示α5β1整合素在腰椎间盘细胞中可能发挥力学传感器的作用。于胜吉 邱贵兴 Yang B Roth S Whyne C Yee AJ 2006中华骨科杂志2006,26,6:2
