3A new loss-of-function allele 28y reveals a role of ARGONAUTE1 in limiting asymmetric division of stomatal lineage ground cell显示文摘In Arabidopsis thaliana L., stomata are produced through a series of divisions including asymmetric and symmetric divisions. Asymmetric entry division of meristemoid mother cell produces two daughter cells, the smaller meristemoid and the larger sister cell, a stomatal lineage ground cell(SLGC). Stomatal lineage ground cells can differentiate into epidermal pavement cells but have the potential to divide asymmetrically, spacing divisions, to create satellite meristemoids. Peptide ligands and TOO MANY MOUTHS(TMM) and ERECTA family receptors regulate the initiation of stomatal lineages, activity, and orientation of spacing divisions. Here, we reported that a natural mutant 28 y displayed an increased stomatal density and index. Using map-based cloning, we identified mutation in ARGONAUTE1(AGO1)as the cause of 28 y phenotypes. Time-lapse tracing of stomatal lineage cells reveals that stomatal overproduction in 28 y is caused by the excessive asymmetric spacing division of SLGCs.Further genetic results demonstrated that AGO1 acts downstream of TMM and negatively regulates the SPCH transcripts,but in a brassinosteroid-independent manner. 4Polymerase synthesis and potential interference of a small-interfering RNA targeting hPim-2显示文摘AIM: To synthesize three small-interference RNAs (siRNAs) by T7 RNA polymerase-catalyzed reaction, and to investigate their efficacy on modulating the expression of serine/threonine kinase Pim-2 in human colon cancer cell line.METHODS: siRNA Ⅰ,Ⅱ and Ⅲ were synthesized by T7 RNA polymerase-directed in vitro transcription, then transfected into human colon cancer cells SW-480. After incubation for 6 h at 37℃, 100 mL/L FBS in RPMI 1640 was substituted in each well. After the transfection was repeated twice to three times in each kind of siRNA, hPim-2 mRNA and protein expression were measured by RT-PCR and Western blotting, respectively.RESULTS: Compared to the control group, after transfected for 48 h with hPim-2 siRNA Ⅰ, Ⅱ and Ⅲ, the relative inhibition rates of hPim-2 mRNA expression in colon cancer cell swere 65.4% (P<0.05), 46.2% (P<0.05) and 56.1% (P<0.05),respectively. The protein level of hPim-2 was decreased at 72 h compared to the untransfected cells. The relative inhibition percentages of hPim-2 protein by siRNA Ⅰ, Ⅱ, Ⅲ were 61.6% (P<0.05), 45.8% (P<0.05) and 55.6% (P<0.05),respectively.CONCLUSION: The in vitro transcribed siRNAs can be useful for silencing oncogene hPim-2 expression specifically and efficiently. 6Concentrations of “elemental carbon” in samples from the Peking Man Site at Zhoukoudian and the possibility of their application in the development of evidence for the use of fire by humans显示文摘Elemental carbon?EC) is a C-rich, O-H-S-N-depleted substance that is necessarily produced in the process of combustion. Due to the logn-term use of fire by cave-inhabiting ape-men, considerable amounts of 揺lemen-tal carbon?would be necessarily left behind in the corre-sponding strata inside the cave. The sepaation of EC was effected by the chemical method and the contents of carbon were determined on an element analyser and a self-made measuring system. The concentration of Ec in the hearth,the exhibited ash samples collected from near the hearth un-earthed in the 1930s, and the three samples of accumulated material collected from the tenth bed are 43.74%, 1.77%,3.88%,1.87% and 1.11%,respectively; those of the equivalent samples from the tenth bed outside the cave and samples from the fourth and seventh beds are at least one order of magnitude lower than the former抯. The above re-sults show that the sampling location of the tenth bed may be close to the hearth. Sampling over a wider range may lead to the finding of a location where the concentrations of EC are corresponding to the maximum values of EC in the hearth. 20the National Natural Science Foundation of China(41877029,41961130383);Royal SocietyNewton Advanced Fellowship(NAF\R1\191017);Wuhan Science and Technology Bureau(2019020701011469).显示文摘Due to the tremendous diversity of microbial organisms in topsoil,the estimation of saturated richness in a belowground ecosystem is still challenging.Here,we intensively surveyed the 16S rRNA gene in four 1 m2 sampling quadrats in a typical grassland,with 141 biological or technical replicates generating over 11 million sequences per quadrat.Through these massive data sets and using both non-asymptotic extrapolation and non-parametric asymptotic approaches,results revealed that roughly 15919±193,27193±1076 and 56985±2347 prokaryotic species inhabited in 1 m2 topsoil,classifying by DADA2,UPARSE(97%cutoff)and Deblur,respectively,and suggested a huge difference among these clustering tools.Nearly 500000 sequences were required to catch 50%species in 1 m2,while any estimator based on 500000 sequences would still lose about a third of total richness.Insufficient sequencing depth will greatly underestimate both observed and estimated richness.At least~911000,~3461000,and~1878000 sequences were needed for DADA2,UPARSE,and Deblur,respectively,to catch 80%species in 1 m2 topsoil,and the numbers of sequences would be nearly twice to three times on this basis to cover 90%richness.In contrast,α-diversity indexes characterized by higher order of Hill numbers,including Shannon entropy and inverse Simpson index,reached saturation with fewer than 100000 sequences,suggesting sequencing depth could be varied greatly when focusing on exploring differentα-diversity characteristics of a microbial community.Our findings were fundamental for microbial studies that provided benchmarks for the extending surveys in large scales of terrestrial ecosystems.Shuzhen Li Xiongfeng Du Kai Feng Yueni Wu Qing He Zhujun Wang Yangying Liu Danrui Wang Xi Peng Zhaojing Zhang Arthur Escalas Yuanyuan Qu Ye Deng 2022Soil Ecology Letters2022,4,3:1
