| 1 | DNA immunization with fusion genes encoding different regions of hepatitis C virus E2 fused to the gene for hepatitis B surface antigen elicits immune responses to both HCV and HBV显示文摘AIM: Both Hepatitis B virus (HBV) and Hepatitis C virus(HCV) are major causative agents of transfusion-associatedand community-acquired hepatitis worldwide. Developmentof a HCV vaccine as well as more effective HBV vaccines isan urgent task. DNA immunization provides a promisingapproach to elicit protective humoral and cellular immuneresponses against viral infection. The aim of this study is toachieve immune responses against both HCV and HBV by DNAimmunization with fusion constructs comprising various HCVE2 gene fragments fused to HBsAg gane of HBV.METHODS: C57BL/6 mice were immunized with plasmid DNAexpressing five fragments of HCV E2 fused to the gene forHBsAg respectively. After one primary and one boostingimmunizations, antibodies against HCV E2 and HBsAg weretested and subtyped in ELISA. Splenic cytokine expressionof IFN-γ and IL-10 was analyzed using an RT-PCR assay.Post-immune mouse antisera also were tested for theirability to capture HCV viruses in the serum of a hepatitis Cpatient in vitro.RESUTLTS: After immunization, antibodies against bothHBsAg and HCV E2 were detected in mouse sera, withIgG2a being the dominant immunoglobulin sub-class. High-level expression of INF-γ was deuetected in cultured splenic cells.Mouse antisera against three of the five fusion constructs wereable to capture HCV viruses in an in vitro assay.CONCLUSION: The results indicate that these fusionconstructs could efficiently elicit humoral and Th1 dominantcellular immune responses against both HBV S and HCV E2antigens in DNA-immunized mice. They thus could serve ascandidates for a bivalent vaccine against HBV and HCVinfection. Three-dimensional bio-printing technology also holds promise for future use in medical applications. At present 3D bio-printing is mainly used for simulating and reconstructing some hard tissues or for preparing drug-delivery systems in the medical area. The fabrication of 3D structures with living cells and bioactive moieties spatially distributed throughout will be realisable. Fabrication of complex tissues and organs is still at the exploratory stage. Kalman filter is employed to update object model by filtering object kernel-histogram using previous model and current candidate. A self-tuning method is used for adaptively adjust all the parameters of the filters under the analysis of the filtering residuals. In addition, hypothesis testing servers as the criterion for determining whether to accept filtering result. Therefore, the tracker has the ability to handle occlusion so as to avoid over-update. | 6 | Bolt insertion damage and mechanical behaviors investigation of CFRP/CFRP interference fit bolted joints显示文摘Interference fit has advantages in improving fatigue behaviors of composite bolted joints,however, interference fit bolt insertion tends to cause damages in laminates weakening joint mechanical properties. Therefore, an experimental study was conducted to investigate bolt insertion damages of Carbon Fiber Reinforced Polymer(CFRP)/CFRP interference fit bolted joints.Mechanical behaviors of joints were also evaluated experimentally under both quasi-static loads and cyclic loads. Scanning Electron Microscope(SEM) and high-resolution X-ray micro-CT scan were used to examine micro damages in laminates. Damage and failure behaviors of joints were characterized. The results demonstrated that the hole entrance in upper laminate and the laminate boundary near the hole wall were the most critical regions for damages during bolt insertions. However, the influence of those damages on quasi-static failure loads and fatigue failure modes of joints was minimal. Delamination and matrix cracking occurred first in laminates following fiber and matrix fracture in quasi-static tensile tests.
