2Maize ZmFDR3 localized in chloroplasts is involved in iron transport显示文摘Iron is an essential nutrient for plant metabolism such that Fe-limited plants display chlorosis and suffer from reduced photosynthetic efficiency. Differential display previously identified genes whose expression was elevated in Fe-deficient maize roots. Here,we describe the functional characterization of one of the genes identified in the screen,ZmFDR3 (Zea maize Fe-deficiency-related). Heterologous functional complementation assays using a yeast iron uptake mutant showed that ZmFDR3 functions in iron transport. ZmFDR3 contains a domain found in FliN-proteins of the type III secretion system and is predicted to localize to the thylakoid of plastids. Fluorescence immunocytochemistry showed that ZmFDR3 is localized in the plastids of roots,stems and leaves,with high expression found in guard cell chloroplasts. Transgenic tobacco expressing a 35S-ZmFDR3 construct contains elevated iron content,displays well arranged thylakoid membranes and has photosynthetic indices that are higher than those of the wild type. 3Xenobiotics and loss of tolerance in primary biliary cholangitis显示文摘Data from genome wide association studies and geoepidemiological studies established that a com-bination of genetic predisposition and environmental stimulation is required for the loss of tolerance in primary biliary cholangitis(PBC).The serologic hallmark of PBC are the presence of high titer anti-mitochondrial autoantibodies(AMA)that recognize the lipoyl domain of the mitochondrial pyruvate dehydrogenase E2(PDC--E2)subunit.Extensive efforts have been directed to investigate the molecular basis of AMA.Recently,experimental data has pointed to the thesis that the breaking of tolerance to PDC--E2 is a pivotal event in the initial etiology of PBC,including environmental xenobiotics including those commonly found in cos-metics and food additives,suggesting that chemical modification of the PDC--E2 epitope may render its vulnerable to become a neo-antigen and trigger an immune response in genetically susceptible hosts.Here,we will discuss the natural history,genetics and immunobiology of PBC and structural constraints of PDC--E2 in AMA recognition which makes it vulnerable to chemical modification.Jinjun Wang Guoxiang Yang Alana Mari Dubrovsky Jinjung Choi Patrick SC Leung 2016World Journal of Gastroenterology2016,22,1:4

5Proteomic analysis reveals distinctive protein profiles involved in CD8^(+) T cell-mediated murine autoimmune cholangitis显示文摘Autoimmune cholangitis arises from abnormal innate and adaptive immune responses in the liver,and T cells are critical drivers in this process.However,little is known about the regulation of their functional behavior during disease development.We previously reported that mice with T cell-restricted expression of a dominant negative form of transforming growth factor beta receptor type Ⅱ(dnTGFβ RII)spontaneously develop an autoimmune cholangitis that resembles human primary biliary cholangitis(PBC).Adoptive transfer of CD8^(+) but not CD4^(+) T cells into Rag1^(−/−)mice reproduced the disease,demonstrating a critical role for CD8^(+) T cells in PBC pathogenesis.Herein,we used SOMAscan technology to perform proteomic analysis of serum samples from dnTGFβRII and B6 control mice at different ages.In addition,we analyzed CD8 protein profiles after adoptive transfer of splenic CD8^(+) cells into Rag1^(−/−)recipients.The use of the unique SOMAscan aptamer technology revealed critical and distinct profiles of CD8 cells,which are key to biliary mediation.In total,254 proteins were significantly increased while 216 proteins were significantly decreased in recipient hepatic CD8^(+) cells compared to donor splenic CD8^(+) cells.In contrast to donor splenic CD8^(+) cells,recipient hepatic CD8^(+) cells expressed distinct profiles for proteins involved in chemokine signaling,focal adhesion,T cell receptor and natural killer cell-mediated cytotoxicity pathways.Weici Zhang Ren Zhang Jun Zhang Ying Sun Patrick SC Leung Guo-Xiang Yang Zongwen Shuai William M Ridgway M Eric Gershwin 2018Cellular & Molecular Immunology2018,15,8:3

7The effects of Spirulina on anemia and immune function in senior citizens显示文摘Anemia and immunological dysfunction(i.e.immunosenescence)are commonly found in older subjects and nutritional approaches are sought to counteract these phenomena.Spirulina is a filamentous and multicellular bule-green alga capable of reducing inflammation and also manifesting antioxidant effects.We hypothesized that Spirulina may ameliorate anemia and immunosenescence in senior citizens with a history of anemia.We enrolled 40 volunteers of both sexes with an age of 50 years or older who had no history of major chronic diseases.Participants took a Spirulina supplementation for 12 weeks and were administered comprehensive dietary questionnaires to determine their nutritional regimen during the study.Complete cell count(CCC)and indoleamine 2,3-dioxygenase(IDO)enzyme activity,as a sign of immune function,were determined at baseline and weeks 6 and 12 of supplementation.Thirty study participants completed the entire study and the data obtained were analyzed.Over the 12-week study period,there was a steady increase in average values of mean corpuscular hemoglobin in subjects of both sexes.In addition,mean corpuscular volume and mean corpuscular hemoglobin concentration also increased in male participants.Older women appeared to benefit more rapidly from Spirulina supplements.Similarly,the majority of subjects manifested increased IDO activity and white blood cell count at 6 and 12 weeks of Spirulina supplementation.Spirulina may ameliorate anemia and immunosenescence in older subjects.We encourage large human studies to determine whether this safe supplement could prove beneficial in randomized clinical trials.Carlo Selmi Patrick SC Leung Laura Fischer Bruce German Chen-Yen Yang Thomas P Kenny Gerry R Cysewski M Eric Gershwin 2011Cellular & Molecular Immunology2011,8,3:2
