1Targeted ribonuclease can inhibit replication of hepatitis B virus显示文摘AIM: To study the effect of a novel targeted ribonuclease(TN), the fusion protein of HBVc and human eosinophil-derived neurotoxin (hEDN), on the HBV replication in vitro.METHODS: The gene encoding the targeted ribonucleasewas cloned into pcDNA3.1 (-) to form recombinant eukaryoticexpression vector p/TN. Control plasmids, including p/hEDN,p/HBVc, and p/TNmut in which a Lys113→Arg mutation wasintroduced by sequential PCR to eliminate the ribonucleaseactivity of hEDN, were also constructed. Liposome-mediatedtransfection of 2.2.15 cells by p/TN, p/TNmut, p/hEDN, p/HBVc, and pcDNA3.1 (-), or mock transfection wasperformed. After that, RT-PCR was used to verify thetransgene expression. Morphology of the transfected cellswas observed and MTT assay was performed to detect thecytotoxicity of transgene expression. Concentration of HBsAgin the supernatant of the transfected cells was measuredusing solid-phase radioimmunoassay.RESULTS: Transgenes were successfully expressed in 2.2.15cells. No obvious cytotoxic effect of transgene expressionon 2.2.15 cells was found. The HBsAg concentration in thep/TN transfected cells was reduced by 58 % compared withthat of mock transfected cells. 2Synthesis and catalytic performance of macroporous La1-xCexCoO3 perovskite oxide catalysts with high oxygen mobility for catalytic combustion of soot显示文摘The disordered macroporous-mesoporous La1-xCexCoO3 catalysts were prepared by complexcombustion method with ethylene glycol as complexing agent at relatively low calcination temperature.The samples were characterized by means of X-ray diffraction,N2 adsorption-ndash;desorption,Xray photoelectron spectroscopy,transmission electron microscopy,hydrogen temperature-programmed reduction and soot temperature-programmed reduction,and so on.The results show that the use of complexing agent and relatively low calcination temperature increase the specific surface area of the catalyst and have abundant pore structure.The Ce ions introduced into lattice of LaCoO3 mainly exist in the form of tetravalent.At the same time,Ce ions enhance the redox performance of the catalyst and the mobility of active oxygen species,which enhances the catalytic activity of the catalyst for soot combustion.The results of activity test show that La0.9Ce0.1CoO3 catalyst exhibits the highest activity in the absence of NO and NO2,and its T10,T50 and T90 are 371,444,and 497℃,respectively.At the same time,a possible reaction mechanism is proposed in this study based on the turnover frequency(TOF) calculated by isothermal anaerobic titrations,XPS and XRD results.Jiachen Sun Zhen Zhao Yazhao Li Xuehua Yu Linlin Zhao Jianmei Li Yuechang Wei Jian Liu 2020Journal of Rare Earths2020,38,6:8
3Combined effect of boundary layer formation and surface smoothing on friction and wear rate of lubricated point contacts during normal running-in processes显示文摘The combined effect of boundary layer formation and surface smoothing on friction and wear rate of metallic surfaces under lubricated point contact condition was investigated. The double trend of friction coefficient variations was revealed during running-in and sub-running-in processes. The evolution of surface topography was measured on-site using white-light interference profilometer and analyzed using bearing area curves. Comprehensive theoretical equations that explicitly express the contributions of boundary friction, adhesive friction and wear have been derived, and results obtained by these equations were compared with experimental observations. It is concluded that the theoretical models are quantitatively adequate to describe the combined effect of surface smoothing due to mechanical wear and formation of boundary films on the changes in friction and wear rate during normal running-in processes.Yazhao ZHANG Alexander KOVALEV Yonggang MENG 2018Friction2018,6,3:4
4Quantifying anti-HBV effect of targeted ribonuclease by real-time fluorescent PCR显示文摘AIM: To quantify the inhibition of HBV replication by targetedribonuclease by using real-time fluorescent PCR.METHODS: Targeted ribonudease was introduced into 2.2.15cells by liposome-mediated transfection or HIV-TAT mediatedprotein transduction. Forty-eight hours after the transfectionand 24 h after the transduction, supernatants of 2.2.15 cellswere collected and HBV DNA in the supernatants wasquantified by real-ljrne fluorescent PCR with a commercial kit.RESULTS: HBV DNA concentrations in the supernatants of2.2.15 cells transfected or transducted with targetedribonudease were 4.9~2.4x 108 copies/L and 8.3~4.0x 108copies/L, respectively. 5Effect of vector-expressed siRNA on HBV replication in hepatoblastoma cells显示文摘AIM: To study the effect of siRNA expressed from DNA vector on HBV replication.METHODS: Human U6 promoter was amplified from genomic DNA and cloned into plasmid pUC18 to construct a mammalian siRNA expression vector pUC18U6. Then oligonucleotides coding for a short hairpin RNA against HBV were cloned into pUC18U6 to form pUC18U6HBVsir which was introduced into 2.2.15 cells by using liposome-mediated transfection.2.2.15 cells transfected by pUC18U6 and pUC18U6GFPsir which expressed siRNA against green fluorescent protein and mock-transfected 2.2.15 cells were used as controls.Concentration of HBsAg in the supernatant of the transfected cells was measured by using solid-phase radioimmunoassay.RESULTS: A mammalian siRNA expression vector pUC18U6 was constructed successfully. 6Anti-HBV activity of TRL mediated by recombinant adenovirus显示文摘AIM: To investigate the inhibitive effect of hepatitis B virus (HBV)-TRL on HBV replication. METHODS: Based on previously constructed pcDNA3.1 (-)/TRL, TR, TRmut, HBV core protein (HBVc) and hEDN, interest gene sequences TRL, TR, HBVc and hEDN were inserted into adenovirus shuttle plasmid pDC316 respectively and co-transfected HEK293 cells with rescue plasmid pBHGlox(delta)E1,3Cre to acquire RAd/TRL, TR, HBVcand hEDN. And then RAds were identified, amplified and the titers in HEK293 cells were determined. RAd/TRL and TR were named as the experimental groups, and others were control ones. After HepG2.2.15 cells were infected, RAd/TRL expression was identified by indirect immunofluorescence staining. Supernatant HBV-DNA content was determined by fluorescent quantification PCR. Meanwhile, metabolism of HepG2.2.15 cells was evaluated by MTT colorimetry.RESULTS: RAd vectors with distinct interest gene sequence were successfully constructed. Effective expression of RAd/TRL in HepG2.2.15 cells resulted in a significant decrease of supernatant HBV-DNA content compared to RAd/TR (0.63±0.14 vs 1.60±0.47, P = 0.0266, <0.05) andother control groups (0.63±0.14 vs 8.50±2.78, 8.25±2.26,8.25±2.29, 8.50±1.51, 8.57±1.63, P<0.01). MTT assaysuggested that there were no significant differences in cell metabolic activity between groups (P>0.05).CONCLUSION: The construction and expression of RAd/TRL has been achieved and it could inhibit HBV replication successfully, which has laid the foundation for further research on anti-HBV activity in vivo.Wei-DongGong YaZhao JunYi JinDing JunLiu Cai-FangXue 2005World Journal of Gastroenterology2005,11,17:2

9Liquid-solid mass transfer in a rotating packed bed reactor with structured foam packing显示文摘A rotating packed bed(RPB) reactor has substantially potential for the process intensification of heterogeneous catalytic reactions. However, the scarce knowledge of the liquid–solid mass transfer in the RPB reactor is a barrier for its design and scale-up. In this work, the liquid–solid mass transfer in a RPB reactor installed with structured foam packing was experimentally studied using copper dissolution by potassium dichromate. Effects of rotational speed, liquid and gas volumetric flow rate on the liquid–solid mass transfer coefficient(kLS) have been investigated. The correlation for predicting kLSwas proposed, and the deviation between the experimental and predicted values was within±12%. The liquid–solid volumetric mass transfer coefficient(kLSaLS) ranged from 0.04–0.14 1^-1, which was approximately 5 times larger than that in the packed bed reactor. 12Numerical Simulation of Contamination Accumulation Characteristics of Composite Insulators in Salt Fog Environment显示文摘To investigate the fouling characteristics of the composite insulator surface under the salt fog environment,the FXBW-110/120-2 composite insulator was taken as the research object.Based on the field-induced charge mechanism,the multi-physical field coupling software COMSOL was used to numerically simulate the fouling characteristics,explored the calculation method of ESDD,and demonstrated its rationality.Based on this method,the pollution characteristics of the composite insulator under the pollution fog environment were studied,and the influence of wind speed,droplet size,and voltage type on the pollution characteristics of the composite insulator was analyzed.The results showed that:with the increase in wind speed,the amount of accumulated pollution of insulator increases in the range of droplet size,and the relationship between wind speed and accumulated pollution is approximately linear;at the same wind speed,the amount of accumulated pollution increases with the increase of droplet size under the action of DC voltage;when there is no voltage,the amount of dirt on the upper surface of the insulator is more than that on the lower surface,while it is the opposite under DC voltage.Yukun Lv Zeze Chen Quanzhi Ge Qian Wang Yazhao Zhang 2023Energy Engineering2023,120,2:0
13White light luminescence in Dy/Tm co-doped yttria stabilized zirconia single crystals显示文摘A series of high quality Dy_(2)O_(3) and Tm_(2)O_(3)co-doped yttria stabilized zirconia(YSZ) single crystals was synthesized.These crystals have a single cubic structure,and exhibit the characteristic of Dy^(3+)(blue and yellow emissions) and Tm^(3+)(blue emission) under excitation with NUV light.Impo rtantly,the hue can be varied between blue and white by modifying the Tm^(3+)/Dy^(3+)ratio.The photoluminescence and decay curves are consistent with the existence of resonance-type energy transfer from Tm^(3+)to Dy^(3+)ions via an electric multipole interaction mechanism.Maximum luminescence intensity is observed in YSZ doped with 0.50 mol% Tm_(2)O_(3) and 0.75 mol% Dy_(2)O_(3),and this sample has CIE coordinates(x = 0.33,y = 0.33) in accordance with those for white chromaticity.Thus,the outstanding luminescence properties of these single crystals suggest potential applications in white light emission devices.Xing Hong Lei Zhang Shoulei Xu Zeyu Cheng Yazhao Wang Bernard A.Goodman Dingkang Xiong Wen Deng 2023Journal of Rare Earths2023,41,12:0
