| 3 | A New Arg54Gly Transthyretin Gene Mutation Associated with Vitreous Amyloidosis in Chinese显示文摘Purpose:To analyse the hereditary features of a Chinese pedigree with familial vitreous amyloidosis in Liaoning Province,China,and to investigate the correlation between the clinical appearance of the disease and transthyretin(TTR)gene mutation,including the locus and type of TTR gene mutation.Methods:Five patients (10 eyes) from one Chinese family were diagnosed with vitreous amyloidosis between July 1996 and April 2009.Family members were followed up subsequently,and peripheral venous blood was obtained from 13 subjects (including 2 patients,and 11 controls without clinical signs of disease).DNA samples were extracted and 4 exons of the TTR gene were amplified by polymerase chain reaction (PCR).The gene fragments were subjected to sequencing analysis.The results were analyzed with DNAMAN Windows 5.2.2.0 and Chromas sequence chart analysis software,TTR gene exons were compared between affected patients and normal controls.Results:Family pedigree analysis revealed that patients were distributed in three generations.Male and female subjects had equal prevalence,and only one parent of affected patients had signs of disease.TTR gene exon sequencing showed that the sequence of patients was identical to that of normal individuals.No TTR gene mutations were noted in 10 unaffected family members.However,a TTR Gly-54 point mutation in the 2nd exon was detected in two patients and 1 unaffected family member (one of the patients' daughters).Vitreous samples in 4 cases (7 eyes) showed positive Congo red staining,suggesting that these family members suffered from familial vitreous amyloidosis.Conclusion:This pedigree affected with familial vitreous amyloidosis was characterized by autosomal dominant inheritance;.a TTR Gly-54 point mutation in the 2nd exon is presumed to be the cause.This Gly-54 point mutation of the TTR gene is a novel mutation in vitreous amyloidosis. | Yining Shi Jing Li Jia Hu Jun Hu Lijun Sun Huijin Li Rui Shi Le Yang Ye Sun Chan Li | 2011 | Eye Science2011,26,4: | 9 |

| 5 | The value of color Doppler ultrasonography in monitoring normal orthotopic liver transplantation and postoperative complications显示文摘OBJECTIVE: To assess the value of color Doppler ultrasonography in monitoring normal orthotopic livertransplantation and postoperative complications.METHODS: Forty-one patients after orthotopic liver transplantation were examined by using colorDoppler flow imaging to observe the hepatic blood flow and change of ultrasonography of the hepaticparenchyma and bile duct. The measured indexes included maximum blood flow velocity, time-averageblood flow velocity (TAV), resistance index (RI) and diameter of the bile duct.RESULTS: Among 41 patients, 17 (41.5%) suffered from liver transplant rejection. Of the 17patients, 13 (76.4%) showed decrease of TAV of the portal vein, 15 (88.25%) low-amplitudesingle-phase serrated wave or negative biphasic wave of the hepatic vein, 9 (52.9%) increased hepaticarterial RI, and 5 (29.4%) slightly dilated bile duct. Sonography showed disappearance of the hepaticartery blood flow around the portal vein in 5 (12.2%) of the 41 patients with hepatic artery thrombosisin the postoperative period. Slight dilatation of the intrahepatic bile duct was found in 3 (7.3%) of the41 patients in the early postoperational period and it normalized within 2 weeks. Ultrasonography of20 patients (48.8%) revealed a visible dilatation of the intrahepatic bile duct, which was worseninggradually. | 6 | Oxaliplatin and 5-fluorouracil hepatic infusion with lipiodolized chemoembolization in large hepatocellular carcinoma显示文摘AIM: To investigate transarterial chemoembolization(TACE) with hepatic infusion of oxaliplatin and 5-fluorouracil and Lipiodol chemoembolization in large hepatocellular carcinoma(HCC).METHODS: In this retrospective study, 132 patients with unresectable HCCs larger than 10 cm were treated with hepatic infusion of oxaliplatin and 5-fluorouracil followed by Lipiodol chemoembolization. The primary endpoint was overall survival(OS). Sixteen-week disease-control rate, time to progression(TTP), and major complications were also studied. Univariate and multivariate analyses were performed to identify prognostic factors affecting OS and TTP.RESULTS: A total of 319 procedures were performed in the 132 patients. Eleven(8.3%) patients received radical resection following TACE treatment(median time to initial TACE 4.3 ± 2.3 mo). The median OS and TTP were 10.3 and 3.0 mo respectively, with a 50.0% 16-wk disease-control rate. Major complications were encountered in 6.0%(8/132) of patients following TACE and included serious jaundice in 1.5%(2/132) patients, aleukia in 1.5%(2/132), and hepatic failure in 3.0%(4/132). One patient died within one month due to serious hepatic failure and severe sepsis after receiving the second TACE. | 8 | Score model for predicting acute-on-chronic liver failure risk in chronic hepatitis B显示文摘AIM: To establish a clinical scoring model to predict risk of acute-on-chronic liver failure(ACLF) in chronic hepatitis B(CHB) patients.METHODS: This was a retrospective study of 1457 patients hospitalized for CHB between October 2008 and October 2013 at the Beijing Ditan Hospital, Capital Medical University, China. The patients were divided into two groups: severe acute exacerbation(SAE) group(n = 382) and non-SAE group(n = 1075). The SAE group was classified as the high-risk group based on the higher incidence of ACLF in this group than in the non-SAE group(13.6% vs 0.4%). Two-thirds of SAE patients were randomly assigned to risk-model derivation and the other one-third to model validation. Univariate risk factors associated with the outcome were entered into a multivariate logistic regression model for screening independent risk factors. Each variable was assigned an integer value based on the regression coefficients, and the final score was the sum of these values in the derivation set. Model discrimination and calibration were assessed using area under the receiver operating characteristic curve and the Hosmer-Lemeshow test. RESULTS: The risk prediction scoring model includedthe following four factors: age ≥ 40 years, total bilirubin ≥ 171 μmol/L, prothrombin activity 40%-60%, and hepatitis B virus DNA > 107 copies/m L. The sum risk score ranged from 0 to 7; 0-3 identified patients with lower risk of ACLF, whereas 4-7 identified patients with higher risk. The Kaplan-Meier analysis showed the cumulative risk for ACLF and ACLF-related death in the two risk groups(0-3 and 4-7 scores) of the primary cohort over 56 d, and log-rank test revealed a significant difference(2.0% vs 33.8% and 0.8% vs 9.4%, respectively; both P < 0.0001). | 9 | Upregulation of miR-489-3p and miR-630 inhibits oxaliplatin uptake in renal cell carcinoma by targeting OCT2显示文摘Renal cell carcinoma(RCC) is one of the most common malignant tumors affecting the urogenital system, accounting for 90% of renal malignancies. Traditional chemotherapy options are often the front-line choice of regimen in the treatment of patients with RCC, but responses may be modest or limited due to resistance of the tumor to anticarcinogen. Downregulated expression of organic cation transporter OCT2 is a possible mechanism underlying oxaliplatin resistance in RCC treatment. In this study, we observed that mi R-489-3 p and mi R-630 suppress OCT2 expression by directly binding to the OCT2 30-UTR. Meanwhile, via 786-O-OCT2-mi RNAs stable expression cell models, we found that mi RNAs could repress the classic substrate 1-methyl-4-phenylpyridinium(MPP+), fluorogenic substrate N,N-dimethyl-4-(2-pyridin-4-ylethenyl) aniline(ASP+), and oxaliplatin uptake by OCT2 both in vitro and in xenografts. In 33 clinical samples, mi R-489-3 p and mi R-630 were significantly upregulated in RCC, negatively correlating with the OCT2 expression level compared to that in adjacent normal tissues, using tissue microarray analysis and q PCR validation. The increased binding of c-Myc to the promoter of pri-mi R-630, responsible for the upregulation of mi R-630 in RCC, was further evidenced by chromatin immunoprecipitation and dual-luciferase reporter assay. | 11 | Regulation of microbiota–GLP1 axis by sennoside A in diet-induced obese mice显示文摘Sennoside A(SA) is a bioactive component of Chinese herbal medicines with an activity of irritant laxative, which is often used in the treatment of constipation and obesity. However, its activity remains unknown in the regulation of insulin sensitivity. In this study, the impact of SA on insulin sensitivity was tested in high fat diet(HFD)-induced obese mice through dietary supplementation. At a dosage of 30 mg/kg/day, SA improved insulin sensitivity in the mice after 8-week treatment as indicated by HOMA-IR(homeostatic model assessment for insulin resistance) and glucose tolerance test(GTT). SA restored plasma level of glucagon-like peptide 1(GLP1) by 90% and mRNA expression of Glp1 by 80% in the large intestine of HFD mice. In the mechanism, SA restored the gut microbiota profile, short chain fatty acids(SCFAs), and mucosal structure in the colon. A mitochondrial stress was observed in the enterocytes of HFD mice with ATP elevation, structural damage, and complex dysfunction. The mitochondrial response was induced in enterocytes by the dietary fat as the same responses were induced by palmitic acid in the cell culture. The mitochondrial response was inhibited in HFD mice by SA treatment. | 12 | Heritability of growth traits in the Asian seabass (Lates calcarifer)显示文摘Growth is an economically important trait in aquaculture.To improve growth trait of the Asian seabass(Lates calcarifer)we have been carrying out,since 2004,a selective breeding program.This study focuses on growth traits in the F2 fish generation,comprised of offspring from 23 mass crosses from 383 F_(1) brooders.Using genotyping analysis for 10 microsatellites from both brood stock and progeny we have reconstructed the pedigree of each mass-cross.For F_(2) generation at 90 days post hatch(dph),we have recorded body weight(BW)for 12,117 individuals and total length(TL),standard length(SL)and condition factor(K_(tl) and K_(sl))for 3530 individuals;and all five traits for 2136 individuals at 270 dph.At 90 dph the average BW was 46.88±20.95 g.Combining pedigree information,recorded growth traits and Restricted Maximum Likelihood method,we have estimated that the narrow sense heritability(h^(2))in F_(1) fish for BW,TL,SL,K_(tl) and K_(sl) was at,90 dph,0.12±0.03,0.11±0.03,0.10±0.03,0.20±0.04 and 0.11±0.03,respectively and,at 270 dph,0.34±0.07,0.32±0.07,0.30±0.06,0.13±0.04 and 0.11±0.04,respectively.At 90 dph the realised heritability for BW was 0.13.Comparing with F_(1) generation,the growth performance of F_(2) fish was increased by 14.4%.Heritability of growth traits will be useful for future genetic improvement programmes of the Asian seabass. | Baoqing Ye Ziyi Wan Le Wang Hongyan Pang Yanfei Wen Huiming Liu Bing Liang Huan Sein Lim Junhui Jiang Genhua Yue | 2017 | Aquaculture and Fisheries2017,2,3: | 4 |

| 14 | Individualized minimally invasive treatment for adult testicular hydrocele: A pilot study显示文摘BACKGROUND Hydrocelectomy is the gold standard for the treatment of hydrocele,but it often causes complications after surgery,including hematoma,infection,persistent swelling,hydrocele recurrence,and chronic pain. In recent years,several methods for minimally invasive treatment of hydrocele have been introduced,but they all have limitations. Herein,we introduce a new method of individualized minimally invasive treatment for hydrocele.AIM To present a new method for the treatment of adult testicular hydrocele.METHODS Fifty-two adult patients with idiopathic testicular hydrocele were included. The key point of this procedure was that the scope of the resection of the sheath of the tunica vaginalis was determined according to the maximum diameter(d) of the effusion measured by ultrasound and the maximum diameter of the portion of the sheath pulled out of the scrotum was approximately πd/2. The surgical procedure consisted of a 2-cm incision in the anterior wall of the scrotum,drainage of the effusion,and dissection of part of the sheath of the tunica vaginalis. After the sheath was peeled away to the predetermined target extent,the pulled-out sheath was removed. The intraoperative findings and postoperative complications were analyzed.RESULTS All patients were successfully treated with a median operation time of 18 min.The median maximum diameter of the effusion on ultrasound was 3.5 cm,and the median maximum diameter of the resected sheath was 5.5 cm. Complications occurred in four(7.7%) patients: two(3.8%) cases of mild scrotal edema,one(1.9%) case of scrotal hematoma,and one(1.9%) case of wound infection. All of the complications were grade I-II. Recurrent hydrocele,chronic scrotal pain,and testicular atrophy were not observed during a median follow-up of 12 mo.CONCLUSION We report a new technique for individualized treatment of testicular hydrocele,which is quantitative and minimally invasive and yields good outcomes. | 16 | Estimating health damage cost from secondary sulfate particles—a case study of Hunan Province,China显示文摘China’s coal dominated energy pattern has resulted in large amount of SO 2 emissions. Estimate of the sulfur related health damage cost is necessary to help perform systematic cost benefit analysis and set national energy and emissions control priorities. Current researches were confined to gaseous SO 2 in urban areas; however, secondary sulfate (SO 2- 4) particles can exert serious impact in a wider region. Based on the concept of “intake fraction', CALPUFF long range dispersion model and 180 sample emission sources, multiple regression equation was obtained with good correlation( r =0 85), which illustrates that populations were key parameters to determine intake fraction but source characteristics were insignificant. Based on the formula and the population distribution data, county level intake fractions were mapped for Hunan Province(range: 1 1×10 -6 -3 2×10 -6 ) of China. A combination of county level SO 2 emissions with the intake fractions yields a total 1 98 tons of sulfate(SO 2- 4) inhalation, and resulting total health damage cost to be 0 76(willingness to pay approach, WTP) or 0 16(human capital approach, HC) billion USD in 1997, about 2.1% or 0 45% of GDP in Hunan in 1997. Average health damage cost per ton of SO 2 emission is 930(WTP) or 200 USD(HC). | 17 | Validation of the DNATyper^(TM)15 PCR Genotyping System for Forensic Application显示文摘We describe the optimization and validation of the DNATyper^(TM)15 multiplex polymerase chain reaction(PCR)genotyping system for autosomal short tandem repeat(STR)amplification at 14 autosomal loci(D6S1043,D21S11,D7S820,CSF1PO,D2S1338,D3S1358,D13S317,D8S1179,D16S539,Penta E,D5S818,vWA,D18S51,and FGA)and amelogenin,a sex‑determining locus.Several DNATyper^(TM)15 assay variables were optimized,including hot start Taq polymerase concentration,Taq polymerase activation time,magnesium concentration,primer concentration,annealing temperature,reaction volume,and cycle number.The performance of the assay was validated with respect to species specificity,sensitivity to template concentration,stability,accuracy,influence of the DNA extraction methods,and the ability to genotype the mixture samples.The performance of the DNATyper^(TM)15 system on casework samples was compared with that of two widely used STR amplification kits,Identifiler^(TM)(Applied Biosystems,Carlsbad,CA,USA)and PowerPlex 16®(Promega,Madison,WI,USA).The conditions for PCR‑based DNATyper^(TM)15 genotyping were optimized.Contamination from forensically relevant nonhuman DNA was not found to impact genotyping results,and full profiles were generated for all the reactions containing≥0.125 ng of DNA template.No significant difference in performance was observed even after the DNATyper^(TM)15 assay components were subjected to 20 freeze‑thaw cycles.The performances of DNATyper^(TM)15,Identifiler^(TM),and PowerPlex 16®were comparable in terms of sensitivity and the ability to genotype the mixed samples and case‑type samples,with the assays giving the same genotyping results for all the shared loci.The DNA extraction methods did not affect the performance of any of the systems.Our results demonstrate that the DNATyper^(TM)15 system is suitable for genotyping in both forensic DNA database work and case‑type samples. | Jian Ye Chengtao Jiang Xingchun Zhao Le Wang Caixia Li Anquan Ji Li Yuan Jing Sun Shuaifeng Chen | 2015 | Journal of Forensic Science and Medicine2015,1,1: | 3 |

| 19 | Dihydro-stilbene gigantol relieves CCl4-induced hepatic oxidative stress and inflammation in mice via inhibiting C5b-9 formation in the liver显示文摘In general,anti-inflammatory treatment is considered for multiple liver diseases despite the etiology.But current drugs for alleviating liver inflammation have defects,making it necessary to develop more potent and safer drugs for liver injury.in this study,we screened a series of(dihydro-)stillene or(dihydro-)phenanthrene derivatives extracted from Pholidota chinensis for theirpotential biological activities.Among 31 compounds,the dihydro-stlbene gigantol exerted most potent protective effects onhuman hepatocytes against lithocholic acid toxicity,and exhibited solid antioxidative and anti-inflammatory effect in vitro.In micewith CCl-induced acute liver injury,pre-administration of gigantol(10,20,40 mg kg^-1·d^-1,po,for 7 days)dose-dependentydecreased serum transaminase levels and improved pathological changes in liver tissues.The elevated lipid peroxidation andinflammatory responses in the livers were also signifhcantly alleviated by gigantol.The pharmacokinetic studies showed thatgigantol was highly concentrated in the mouse lvers,which consisted with its efficacy in preventing liver injury.Using a label-freequantitative proteomic analysis we revealed that gigantol mainly regulated the immune system process in liver tissues of CCl4-treated mice,and the complement and coagulation cascades was the predominant pathway gigantol markedly inhibited theexpression of complement component C9,which was a key component for the formation of terminal complerment complex(TCC)C5b-9.These results were validated by immunohistochemistry(IHC) or real time-PCR.Confocal microscopy analysis showed thatgigantol significantly inhibited the vascular deposition of TCC in the liver.In conclusion,we demonstrate for the first time that oral administration of gigantol potently relieves liver oxidative stress and inflammation,possibly via a novel mechanism of inhibiting the C5b-9 formation in the liver. | Ya-ru Xue Sheng Yao Qian Liu Zhao-liang Peng Qiang-qiang Deng—Bo Liu Zheng-hua Ma Le Wangf Hu Zhou Yang Ye Guo-yu Pan | 2020 | Acta Pharmacologica Sinica2020,41,11: | 3 |
