1不同基因型戊型肝炎病毒重组蛋白的抗原性分析显示文摘目的 :探讨不同基因型和亚型戊型肝炎病毒 (HEV)重组蛋白氨基酸序列的变化对HEV抗原性的影响 ,筛选出最佳的HEV抗原用于戊型肝炎的免疫诊断。方法 :用已知序列的HEV基因工程表达蛋白作为包被抗原 ,对已知血清标本进行酶联免疫吸附试验测定。结果 :6种不同抗原对 3 0份正常献血者血清无抗原性 ,对 17份急性戊型肝炎病人血清和 5份实验感染动物血清均呈阳性反应 ,但血清抗体滴度的高低与所用抗原的基因型有关 ,尤其是受到该抗原第 76位氨基酸变化的影响。结论孟继鸿 戴星 窦晓光 YE KHUDYAKOV H A FIELDS 2002东南大学学报(医学版)2002,21,1:30

3Transcriptomic landscape regulated by the 14 types of bone morphogenetic proteins(BMPs)in lineage commitment and differentiation of mesenchymal stem cells(MSCs)显示文摘Mesenchymal stem cells(MSCs)are ubiquitously-existing multipotent progenitors that can self-renew and differentiate into multiple lineages including osteocytes,chondrocytes,adipocytes,tenocytes and myocytes.MSCs represent one of the most commonly-used adult progenitors and serve as excellent progenitor cell models for investigating lineagespecific differentiation regulated by various cellular signaling pathways,such as bone morphogenetic proteins(BMPs).As members of TGFb superfamily,BMPs play diverse and important roles in development and adult tissues.At least 14 BMPs have been identified in mammals.Different BMPs exert distinct but overlapping biological functions.Through a comprehensive analysis of 14 BMPs in MSCs,we demonstrated that BMP9 is one of the most potent BMPs in inducing osteogenic differentiation of MSCs.Nonetheless,a global mechanistic view of BMP signaling in regulating the proliferation and differentiation of MSCs remains to be fully elucidated.Here,we conducted a comprehensive transcriptomic profiling in the MSCs stimulated by 14 types of BMPs.Hierarchical clustering analysis classifies 14 BMPs into three subclusters:an osteo/chondrogenic/adipogenic cluster,a tenogenic cluster,and BMP3 cluster.We also demonstrate that six BMPs(e.g.,BMP2,BMP3,BMP4,BMP7,BMP8,and BMP9)can induce ISmads effectively,while BMP2,BMP3,BMP4,BMP7,and BMP11 up-regulate Smad-independent MAP kinase pathway.Furthermore,we show that many BMPs can upregulate the expression of the signal mediators of Wnt,Notch and PI3K/AKT/mTOR pathways.While the reported transcriptomic changes need to be further validated,our expression profiling represents the first-of-its-kind to interrogate a comprehensive transcriptomic landscape regulated by the 14 types of BMPs in MSCs.Linghuan Zhang Qing Luo Yi Shu Zongyue Zeng Bo Huang Yixiao Feng Bo Zhang Xi Wang Yan Lei Zhenyu Ye Ling Zhao Daigui Cao Lijuan Yang Xian Chen Bin Liu William Wagstaff Russell R*Reid Hue H*Luu Rex C*Haydon Michael J*Lee Jennifer Moriatis Wolf Zhou Fu Tong-Chuan He Quan Kang 2019Genes & Diseases2019,6,3:11

7Consolidation and dynamics of 3D unsaturated porous seabed under rigid caisson breakwater loaded by hydrostatic pressure and wave显示文摘In this study,based on the dynamic Biot's theory 'u-p' approximation,a 3D finite element method(FEM) numerical soil model is developed,in which the Generalized Newmark-β method is adopted to determine the time integration.The developed 3D FEM soil model is a part of the coupled model PORO-WSSI 3D for 3D wave-seabed-marine structures interaction problem,and is validated by the analytical solution proposed by Wang(2000) for a laterally infinite seabed loaded by a uniform force.By adopting the developed 3D soil model,the consolidation of seabed under a caisson breakwater and hydrostatic pressure is investigated.The numerical results show that the caisson breakwater built on seabed has very significant effect on the stresses/displacements fields in the seabed foundation after the transient deformation and primary consolidation are completed.The parametric study indicates that the Young's modulus E of seabed is the most important parameter to affect the settlement of breakwater,and the displacement fields in seabed foundation.Taking the consolidation status as the initial condition,the interaction between ocean wave,caisson breakwater and seabed foundation is briefly investigated.The 3D ocean wave is determined by solving the Navier-Stokes equations with finite volume method(FVM).The numerical results indicate that there is intensive interaction betweenYE JianHong JENG DongSheng CHAN A H C 2012Science China(Technological Sciences)2012,55,8:4
8Ticagrelor inhibits the NLRP3 inflammasome to protect against inflammatory disease independent of the P2Y_(12 ) signaling pathway显示文摘Ticagrelor is the first reversibly binding oralP2Y_(12) receptor antagonist to inhibit platelet activation and has been approved by the Food and Drug Administration for the treatment of coronary artery disease. At present, the other pharmacological functions of ticagrelor remain poorly understood. The NOD-, LRR-, and pyrin domain-containing protein 3 (NLRP3) inflammasome plays a critical role in the innate immune system, but its excessive activation also contributes to the pathogenesis of complex diseases. In this study, we systematically examined the effects of ticagrelor on the NLRP3 inflammasome and found that ticagrelor inhibits NLRP3 inflammasome activation in macrophages independent of its classic inhibitory effect on theP2Y_(12) signaling pathway. Further mechanistic studies demonstrate that ticagrelor attenuates the oligomerization of apoptosis-associated speck-like protein containing a CARD (ASC) by blocking chloride efflux, an effect achieved through the degradation of chloride intracellular channel proteins (CLICs) and blockade of the translocation of CLICs to the plasma membrane. Moreover, experiments on lipopolysaccharide-induced sepsis and alum-induced peritonitis in mice confirmed that ticagrelor mitigates the severity of systemic inflammation independent ofP2Y_(12) receptor antagonism. Importantly, oral administration of ticagrelor rapidly and strongly inhibited NLRP3 inflammasome activation in peripheral blood mononuclear cells from patients with acute coronary syndrome.
