Dynamic modeling and optimal control of cystic echinococcosis显示文摘Background:Cystic echinococcosis is one of the most severe helminth zoonosis with a drastic impact on human health and livestock industry.Investigating optimal control strategy and assessing the crucial factors are essential for developing countermeasures to mitigate this disease. Methods:Two compartment models were formulated to study the dynamics of cystic echinococcosis transmission,to evaluate the effectiveness of various control measures, and to find the optimal control strategy.Sensitive analyses were conducted by obtaining PRCCs and contour plot was used to evaluate the effect of key parameters on the basic reproduction number.Based on forward-backward sweep method, numerical simulations were employed to investigate effects of key factors on the transmission of cystic echinococcosis and to obtain the optimal control strategy. Results:The food resources of stray dog and invalid sheep vaccination rate, which are always neglected, were significant to the transmission and control of cystic echinococcosis.Numerical simulations suggest that, the implementation of optimal control strategy can significantly reduce the infections.Improving the cost of health education and domestic dog deworming could not decrease human infections. Conclusions:Our study showed that only a long-term use of the optimal control measures can eliminate the disease.Meanwhile, during the intervention, sheep vaccination and stray dogs disposing should be emphasized ahead of domestic dogs deworming to minimize the control cost.Simultaneously reducing other wild intermediate hosts and strengthening the sheep vaccination as well as disposing the stray dogs would be most effective. | Xinmiao Rong Meng Fan Huaiping Zhu Yaohui Zheng | 2021 | Infectious Diseases of Poverty2021,10,2: | 4
Fusobacterium nucleatum enhances the efficacy of PD-L1 blockade in colorectal cancer显示文摘Given that only a subset of patients with colorectal cancer(CRC)benefit from immune checkpoint therapy, efforts are ongoing to identify markers that predict immunotherapeutic response.Increasing evidence suggests that microbes influence the efficacy of cancer therapies.Fusobacterium nucleatum induces different immune responses in CRC with different microsatellite-instability(MSI)statuses.Here,we investigated the effect of F.nucleatum on anti-PD-L1 therapy in CRC.We found that high F.nucleatum levels correlate with improved therapeutic responses to PD-1 blockade in patients with CRC.Additionally,F.nucleatum enhanced the antitumor effects of PD-L1 blockade on CRC in mice and prolonged survival.Combining F.nucleatum supplementation with immunotherapy rescued the therapeutic effects of PD-L1 blockade.Furthermore,F.nucleatum induced PD-L1 expression by activating STING signaling and increased the accumulation of interferon-gamma(IFN-γ)^(+)CD8^(+)tumor-infiltrating lymphocytes(TILs)during treatment with PD-L1 blockade, thereby augmenting tumor sensitivity to PD-L1 blockade.Finally,patient-derived organoid models demonstrated that increased F.nucleatum levels correlated with an improved therapeutic response to PD-L1 blockade. These findings suggest that F.nucleatum may modulate immune checkpoint therapy for CRC. | Yaohui Gao Dexi Bi Ruting Xie Man Li Jing Guo Hu Liu Xianling Guo Juemin Fang Tingting Ding Huiyuan Zhu Yuan Cao Meichun Xing Jiayi Zheng Qing Xu Qian Xu Qing Wei Huanlong Qin | 2021 | Signal Transduction and Targeted Therapy2021,6,12: | 3

Multi-channel multiplexing quantum teleportation based on the entangled sideband modes显示文摘Quantum teleportation is a key primitive across a number of quantum information tasks and represents a fundamental ingredient for many quantum technologies. Channel capacity, other than the fidelity, becomes another focus of quantum communication. Here, we present a 5-channel multiplexing continuous-variable quantum teleportation protocol in the optical frequency comb system, exploiting five-order entangled sideband modes.Because of the resonant electro-optical modulation(EOM) that is specifically designed, the fidelities of five channels are greater than 0.78, which are superior to the no-cloning limit of 2∕3. This work provides a feasible scheme for implementing efficient quantum information processing. | YIMIAO WU QINGWEI WANG LONG TIAN †XIAOLI ZHANG JIAWEI WANG SHAOPING SHI YAJUN WANG YAOHUI ZHENG | 2022 | Photonics Research2022,10,8: | 2
Novel SFTSV Phylogeny Reveals New Reassortment Events and Migration Routes显示文摘Severe fever with thrombocytopenia syndrome virus(SFTSV),the causative agent of a febrile human disease, was first identified from central and eastern provinces in China, and later in Japan and South Korea.Hubei Province is one of the major SFTS epidemic areas in the central part of China. This study reported the isolation of 11 new SFTSV strains from patients in Hubei Province collected in 2017.Extensive phylogenetic analyses were conducted based on the complete coding sequences of SFTSV segments including the new strains. It was suggested that five different SFTSV genotypes were circulating in Hubei, and 15 reassortment patterns and migration pathways correlated with each genotype were identified, which was more than previously recognized.Hubei Province was more involved in the evolutionary events of SFTSV than that previously thought in which the evolutionary events of SFTSV were reported to be independent from those in other epidemic regions.Further divergence of SFTSV strains was suggested by pairwise comparison of SFTSV sequences from each genotype and sequence identity normalized to representative strain in genotype C1.Subsequently,amino acid variations specific for genotype(s),strain(s),or cluster(s)were inspected, which may be related to differential biological activity of SFTSV strains/genotypes.In conclusion,we analyzed the current status of SFTSV phylogeny in Hubei Province and discussed the possible events correlated to SFTSV evolution. It provided an in-depth insight into SFTSV evolution, raising concerns for the use of proper SFTSV strains in future studies. | Xiaoli Wu Mingyue Li Yanfang Zhang Boyun Liang Junming Shi Yaohui Fang Zhengyuan Su Mengmeng Li Wenjing Zhang Ling Xu Jun Wang Qiaoli Wu Shuang Tang Hualin Wang Tao Zhang Cheng Peng Xin Zheng Fei Deng Shu Shen | 2021 | Virologica Sinica2021,36,2: | 2
Experimental Study on the Improvement of Yaw Stability by Coordination Control between the Caudal Fin and Anal Fin显示文摘Due to the unique locomotion, the head-shaking problem of biomimetic robotic fish inevitably occurs during rectilinear locomotion, which strongly hinders its practical applications. In this paper,we experimentally study this problem by proposing the method of coordination control between the caudal fin and anal fin.First,an untethered biomimetic robotic fish, equipped with an anal fin,a caudal fin and two pectoral fins,is developed as the experimental platform.Second,a Central Pattern Generator(CPG)-based controller is used to coordinate the motions of the anal fin and caudal fin.Third, extensive experiments are conducted to explore different combinations of the flapping frequencies, the flapping amplitudes, and the phase differences between the anal fin and caudal fin.Notably, through proper control of the anal fin,the amplitude of the yaw motion can be as small as 4.32°,which sees a 65%improvement compared to the scenario without anal fin,and a 57%improvement compared to that with a stationary anal fin. This paper provides a novel way to alleviate the head-shaking problem for biomimetic robotic fish, and first test this method on an untethered, freely swimming robotic platform, which can shed light on the development of underwater robotics. | Jiang Ding Changzhen Zheng Chaocheng Song Qiyang Zuo Yaohui Xu Bingbing Dong Jiaxu Cui Kai He Fengran Xie | 2022 | Journal of Bionic Engineering2022,19,5: | 1

Weakening of the anisotropy of surface roughness in ultra-precision turning of single-crystal silicon显示文摘Ultra-precision machining causes materials to undergo a greatly strained deformation process in a short period of time. The effect of shear strain rates on machining quality, in particular on surface anisotropy, is a topic deserving of research that has thus far been overlooked. This study analyzes the impact of the strain rate during the ultra-precision turning of single-crystal silicon on the anisotropy of surface roughness.Focusing on the establishment of cutting models considering the tool rake angle and the edge radius, this is the first research that takes into account the strain rate dislocation emission criteria in studying the effects of the edge radius, the cutting speed, and the cutting thickness on the plastic deformation of single-crystal silicon. The results of this study show that the uses of a smaller edge radius, faster cutting speeds, and a reduced cutting thickness can result in optimally uniform surface roughness, while the use of a very sharp cutting tool is essential when operating with smaller cutting thicknesses.A further finding is that insufficient plastic deformation is the major cause of increased surface roughness in the ultra-precision turning of brittle materials. On this basis, we propose that the capacity of single-crystal silicon to emit dislocations be improved as much as possible before brittle fracture occurs, thereby promoting plastic deformation and minimizing the anisotropy of surface roughness in the machined workpiece. | Wang Minghai Wang Ben Zheng Yaohui | 2015 | Chinese Journal of Aeronautics2015,28,4: | 1
Plasma from healthy donors protects blood-brain barrier integrity via FGF21 and improves the recovery in a mouse model of cerebral ischaemia显示文摘Background Healthy plasma therapy reverses cognitive deficits and promotes neuroplasticity in ageing brain disease. However, whether healthy plasma therapy improve blood-brain barrier integrity after stroke remains unknown. Methods Here,we intravenously injected healthy female mouse plasma into adult female ischaemic stroke C57BL/6 mouse induced by 90 min transient middle cerebral artery occlusion for eight consecutive days.Infarct volume, brain atrophy and neurobehavioural tests were examined to assess the outcomes of plasma treatment.Cell apoptosis,blood-brain barrier integrity and fibroblast growth factor 21 knockout mice were used to explore the underlying mechanism. Results Plasma injection improved neurobehavioural recovery and decreased infarct volume, brain oedema and atrophy after stroke.Immunostaining showed that the number of transferase dUTP nick end labelling+/NeuN+cells decreased in the plasma-injected group.Meanwhile, plasma injection reduced ZO-1,occluding and claudin-5 tight junction gap formation and IgG extravasation at 3 days after ischaemic stroke.Western blot results showed that the FGF21 expression increased in the plasma-injected mice. However, using FGF21 knockout mouse plasma injecting to the ischaemic wild-type mice diminished the neuroprotective effects. Conclusions Our study demonstrated that healthy adult plasma treatment protected the structural and functional integrity of blood-brain barrier, reduced neuronal apoptosis and improved functional recovery via FGF21,opening a new avenue for ischaemic stroke therapy. | Muyassar Mamtilahun Lu Jiang Yaying Song Xiaojing Shi Chang Liu Yixu Jiang Lidong Deng Haoran Zheng Hui Shen Yongfang Li Zhijun Zhang Yongting Wang Yaohui Tang Guo-Yuan Yang | 2021 | Stroke & Vascular Neurology2021,6,4: | 1

Functional characterization of Bmcap in uric acid metabolism in the silkworm显示文摘After a millennium of domestication, numerous silkworm mutants have emerged that exhibit transparent epidermis, which is caused by abnormally low levels of uric acid.We identified the Bombyx mori gene Bmcap(BMSK0003832)as the homolog of cappuccino,a subunit of the biogenesis of lysosome-related organelles complex-1(BLOC-1)that has been extensively characterized in human, mouse, and insect species,by analyzing the amino acid sequences of putative purine metabolism genes.Using the clustered regularly interspaced palindromic repeats(CRISPR)/CRISPR-associated protein 9 system,we disrupted Bmcap, resulting in decreased uric acid levels in the silkworm epidermis and a translucent skin phenotype. In the Bmcap mutant, the purine metabolism, nitrogen metabolism, pyrimidine metabolism, and membrane system were altered compared to the wild type.Biogenesis of lysosome-related organelle complex genes play a role in the pigmentation and biogenesis of lysosome-related organelles(LROs)in platelets, melanocytes, and megakaryocytes.LROs exhibit unique morphologies and functions in various tissues and cells.Investigation of the Bmcap mutant will enhance our understanding of the uric acid metabolic pathway in silkworms, and this mutant offers a valuable silkworm model for LRO studies. | Linmeng Tang Dehong Yang Zhiwei Liu Yaohui Wang Xu Yang Yujia Liu Dongbin Chen Zheng Tang Yongping Huang | 2024 | Insect Science2024,31,1: | 0

Arabidopsis SYP121 acts as an ROP2 effector in the regulation of root hair tip growth显示文摘Tip growth is an extreme form of polarized cell expansion that occurs in all eukaryotic kingdoms to generate highly elongated tubular cells with specialized functions, including fungal hyphae, animal neurons, plant pollen tubes, and root hairs (RHs). RHs are tubular structures that protrude from the root epidermis to facilitate water and nutrient uptake, microbial interactions, and plant anchorage. RH tip growth requires polarized vesicle targeting and active exocytosis at apical growth sites. However, how apical exocytosis is spatially and temporally controlled during tip growth remains elusive. Here, we report that the Qa-Soluble N-ethylmaleimide-sensitive factor attachment protein receptor (SNARE) SYP121 acts as an effector of Rho of Plants 2 (ROP2), mediating the regulation of RH tip growth. We show that active ROP2 promotes SYP121 targeting to the apical plasma membrane. Moreover, ROP2 directly interacts with SYP121 and promotes the interaction between SYP121 and the R-SNARE VAMP722 to form a SNARE complex, probably by facilitating the release of the Sec1/Munc18 protein SEC11, which suppresses the function of SYP121. Thus, the ROP2-SYP121 pathway facilitates exocytic trafficking during RH tip growth. Our study uncovers a direct link between an ROP GTPase and vesicular trafficking and a new mechanism for the control of apical exocytosis, whereby ROP GTPase signaling spatially regulates SNARE complex assembly and the polar distribution of a Q-SNARE. | Xiankui Cui Shuwei Wang Yaohui Huang Xuening Ding Zirong Wang Lidan Zheng Yujing Bi Fanghui Ge Lei Zhu Ming Yuan Shaul Yalovsky Ying Fu | 2022 | Molecular Plant2022,15,6: | 0
