| 1 | Identification of immunoreactive proteins of Brucella melitensis by immunoproteomics显示文摘Infection with Brucella causes brucellosis, a chronic disease in humans, which induces abortion and sterility in livestock. Among the different Brucella species, Brucella melitensis is considered the most virulent and is the predominant species associated with outbreaks in China. To date, no safe human vaccine is available against Brucella infection. The currently used live vaccines against Brucella in livestock induce antibodies that interfere with the diagnosis of field infection in vaccinated animals, which is harmful to eradication programs. However, there is as yet no complete profile of immunogenic proteins of B. melitensis. Towards the development of a safer, equally efficacious, and field infection-distinguishable vaccine, we used immunoproteomics to identify novel candidate immunogenic proteins from B. melitensis M5. Eighty-eight immunoreactive protein spots from B. melitensis M5 were identified by Western blotting and were assigned to sixty-one proteins by mass spectrometry, including many new immunoreactive proteins such as elongation factor G, F0F1 ATP synthase subunit beta, and OMP1. | 3 | Effects of urea plus nitrate pretreated rice straw and corn oil supplementation on fiber digestibility,nitrogen balance,rumen fermentation,microbiota and methane emissions in goats显示文摘Background: Urea pretreatment is an efficient strategy to improve fiber digestibility of low quality roughages for ruminants. Nitrate and oil are usually used to inhibit enteric methane(CH4) emissions from ruminants. The objective of this study was to examine the combined effects of urea plus nitrate pretreated rice straw and corn oil supplementation to the diet on nutrient digestibility, nitrogen(N) balance, CH4 emissions, ruminal fermentation characteristics and microbiota in goats. Nine female goats were used in a triple 3 × 3 Latin Square design(27 d periods). The treatments were: control(untreated rice straw, no added corn oil), rice straw pretreated with urea and nitrate(34 and 4.7 g/kg of rice straw on a dry matter [DM] basis, respectively, UN), and UN diet supplemented with corn oil(15 g/kg soybean and15 g/kg corn were replaced by 30 g/kg corn oil, DM basis, UNCO).Results: Compared with control, UN increased neutral detergent fiber(NDF) digestibility(P < 0.001) and copies of protozoa(P < 0.001) and R. albus(P < 0.05) in the rumen, but decreased N retention(-21.2%, P < 0.001), dissolved hydrogen concentration(-22.8%, P < 0.001), molar proportion of butyrate(-18.2%, P < 0.05),(acetate + butyrate) to propionate ratio(P < 0.05) and enteric CH4 emissions(-10.2%, P < 0.05). In comparison with UN, UNCO increased N retention(+34.9%, P < 0.001) and decreased copies of protozoa(P < 0.001) and methanogens(P < 0.001). Compared with control, UNCO increased NDF digestibility(+8.3%, P < 0.001), reduced ruminal dissolved CH4 concentration(-24.4%, P < 0.001) and enteric CH4 emissions(-12.6%, P < 0.05).Conclusions: A combination of rice straw pretreated with urea plus nitrate and corn oil supplementation of the diet improved fiber digestibility and lowered enteric CH4 emissions without negative effects on N retention. | 4 | Survival of people living with HIV after treatment with Traditional Chinese Medicine in Henan province of China: a retrospective cohortstudy显示文摘OBJECTIVE: To provide survival estimates of people living with human immunodeficiency virus(PLHIV) after treatment with Traditional Chinese Medicine(TCM) in rural China, to identify the prognostic factors at enrollment, and to explore the effectiveness of TCM intreating PLHIV.METHODS: PLHIV who enrolled in national TCM HIV treatment trial program in October 2004 were analyzed in this study and followed up to October 2010. Survival time was estimated by the Kaplan-Meier curve and hazard ratios, and identifying prognostic factors were computed through Cox proportional hazard models.RESULTS:A total of 1666 PLHIV were included with 102 591 person-months of follow-up. Overall, 312(18.7%) patients died. The total mortality rate over the study period was 3.6 per 100 person-years,which was lower than the worldwide rate. The cumulative survival rate was 95.9% at 1 year[95% confidence interval(CI)(94.8-96.8)] and 80.4% at 6years [95% CI(78.4-82.3)]. Elevated death risks emerged among males, older individuals, and thosewithlowerCD4+T-cellcounts.CONCLUSION: TCM could increase survival and lengthen the life span of PLHIV in Henan province of China, as shown by our retrospective cohort study. Factors such as sex, age, education, and CD4+ T-cell counts correlated to survival. | 9 | Applications and research trends of digital human models in the manufacturing industry显示文摘Virtual reality(VR)has been widely used in various manufacturing industries,and VR-based virtual manufacturing has received significant attention in the current intelligent manufacturing era.Digital human models(DHMs)are essential for virtual manufacturing applications.Additionally,researching new applications of DHMs has developed into an important academic research field.This paper aims to identify the applications and research trends of DHMs in the manufacturing industry and to provide a reference for the continued development of virtual manufacturing and DHMs.We selected a total of 49 related articles from a large number of articles published between 2014 and 2019.The applications of DHMs in the manufacturing industry are analyzed from different perspectives and various relevant technical limitations are discussed.The results indicate that the applications of DHMs differ significantly between different types of fields.The automotive industry is the main application field for DHMs,and assembly/maintenance simulations and evaluations are the main application types.Additionally,there are still some limitations in the establishment of virtual environments,motion control,and DHM evaluation that should be addressed.Finally,research trends in the application of DHMs are illustrated and discussed,including the planning and assessment of human-robot collaboration systems,the combination of DHMs and augmented reality,and improved motion planning for DHMs.In summary,the application of DHMs can improve the realism and effectiveness of virtual manufacturing,and DHMs will be more widely and deeply studied and applied in various manufacturing industries in the near future. | Wenmin ZHU Xiumin FAN Yanxin ZHANG | 2019 | Virtual Reality & Intelligent Hardware2019，1，6： | 5 |
| 10 | Prevalence of liver injury among patients with acquired immunodeficiency syndrome treated with highly active antiretroviral therapy in China显示文摘OBJECTIVE: To estimate the prevalence of liver injury among patients with acquired immunodeficiency syndrome(AIDS) who received highly active antiretroviral therapy(HAART) in rural Henan Province in China, and to explore whether Traditional Chinese Medicine(TCM) treatment based on HAART would increase this risk.METHODS: This was a retrospective cross-sectional study. We collected medical information on patients with AIDS from two treatment databases in2014. Criteria established by the AIDS Clinical Trials Group in 1996 were used for grading liver injury,classified based on the limit of normal(ULN) for alanine transaminase and aspartate aminotransferase:grade 1(1.25-2.5 × ULN); grade 2(2.6-5 × ULN);grade 3(5.1-10 × ULN); and grade 4(> 10 × ULN).Factors associated with liver injury were evaluated using a logistic regression model.RESULTS: A total 6953 patients with AIDS(3324 male and 3629 female patients) were enrolled into this study. The prevalence of liver injury was 22.0%(18.0% grade 1, 3.1% grade 2, 0.9% grade 3). In multivariate analysis, patients aged 34-45 years were more likely to have liver injury than patients in other age groups [adjusted odds ratio(AOR), 1.39; 95%CI, 1.01-1.91)]. Other factors associated with liver injury included male sex(AOR, 1.64; 95% CI,1.46-1.85), HIV infection via blood(AOR, 1.47; 95%CI, 1.19-1.82), hepatitis B virus antibody positive(AOR, 1.07; 95% CI, 0.85-1.36), and hepatitis C virus(HCV) antibody positive(AOR, 2.76; 95% CI,2.28-3.34).CONCLUSION: The prevalence of liver injury was relatively high among HAART-experienced patients. Several factors associated with liver injury included male sex, age 35-45 years old, HIV infection through blood, and concurrent HCV infection. | 15 | Assessment of estrogen disrupting potency in animal foodstuffs of China by combined biological and chemical analyses显示文摘Food has been documented as one of major routes for human exposure to environmental estrogens(EEs), but information on the occurrence of EEs in animal foodstuffs is still scarce.This study analyzed estrogenic activity in 16 types of animal foodstuffs(n = 142) collected from four cities(Wuhan, Guangzhou, Wenzhou and Yantai) of China by combined yeast estrogen screen(YES) bioassay and chemical analysis. By bioassay, all samples' extracts were found to induce estrogenic activities and the bioassay-derived 17β-estradiol equivalent(EEQbio) ranged from 8.29 to 118.32 ng/g. In addition, the samples were analyzed by liquid chromatography coupled to tandem mass spectrometry for further chemical analysis. 17β-Estradiol was found in all samples in this survey at levels of 0.44 to 15.04 ng/g.All samples had 33.1% detection rate of 17α-ethinylestradiol(EE2), and the maximum concentration was 2.80 ng/g. Bisphenol A and 4-nonylphenols were detected in 83.8% and83.1% of samples, with concentrations up to 12.56 ng/g and 35.76 ng/g, respectively.However, the concentrations of estrone, diethylstilbestrol and 4-t-octylphenol were found to be below the limit of detection. A comparison of EEQbiomeasured from the YES assay and EEQchemcalculated from chemical analysis showed good correlation(R2= 0.84). AVP(4-8), different from its precursor AVP, plays memory enhancement roles in the CNS without peripheral side-effects. However, it is not clear whether AVP(4-8) can improve cognitive behaviors and synaptic plasticity in the APP/PS1 mouse model of AD. Here, we investigated for the first time the neuroprotective effects of AVP(4-8) on memory behaviors and in vivo long-term potentiation(LTP) in APP/PS1-AD mice.The results showed that:(1) APP/PS1-AD mice had lower spontaneous alternation in the Y-maze than wild-type(WT) mice, and this was significantly reversed by AVP(4-8);(2) the prolonged escape latency of APP/PS1-AD mice in the Morris water maze was significantly decreased by AVP(4-8), and the decreased swimming time in target quadrant recovered significantly after AVP(4-8) treatment;(3) in vivo hippocampal LTP induced by high-frequency stimulation had a significant deficit in the AD mice, and this was partly rescued by AVP(4-8);(4) AVP(4-8)significantly up-regulated the expression levels of postsynaptic density 95(PSD95) and nerve growth factor(NGF)in the hippocampus of AD mice.
