| 3 | Speciation, sources, and risk assessment of heavy metals in suburban vegetable garden soil in Xianyang City, Northwest China显示文摘Intensive anthropogenic activities can lead tosoil heavy metal contamination resulting in potential risksto the environment and to human health. To reveal theconcentrations, speciation, sources, pollution level, andecological risk of heavy metals in vegetable garden soil, atotal of 136 soil samples were collected from threevegetable production fields in the suburbs of XianyangCity, Northwest China. These samples were analyzed byinductively coupled plasma- atomic emission spectrometryand atomic fluorescence spectrometry. The results showedthat the mean concentrations of Cd, Co, Cu, Mn, Pb, Zn,and Hg in vegetable garden soil were higher than thecorresponding soil element background values of ShaanxiProvince. The heavy metals studied in vegetable gardensoil were primarily found in the residual fraction,averaging from 31.26% (Pb)to 90.23% (Cr). Consideringthe non-residual fractions, the mobility or potential riskwas in the order of Pb (68.74%)〉 Co (60.54%)〉 Mn(59.28%)〉Cd (53.54%)〉〉Ni (23.36%)〉Zn(22.73%) 〉 Cu (14.93%) 〉 V (11.81%) 〉 Cr (9.78%). Cr,Mn, Ni, V, and As in the studied soil were related to soil-forming parent materials, while Cu, Hg, Zn, Cd, Co, andPb were associated with the application of plastic films,fertilizers, and pesticides, as well as traffic emissions andindustrial fumes. Cr, Ni, V, and As presented lowcontamination levels, whereas Co, Cu, Mn, Pb, and Znlevels were moderate, and Cd and Hg were high.Ecological risk was low for Co, Cr, Cu, Mn, Pb, Zn, andAs, with high risk observed for Cd and Hg. | 5 | ILF2 cooperates with E2F1 to maintain mitochondrial homeostasis and promote small cell lung cancer progression显示文摘Objective:Mitochondria play multifunctional roles in carcinogenesis.Deciphering uncertainties of molecular interactions within mitochondria will promote further understanding of cancer.Interleukin enhancer binding factor 2(ILF2)is upregulated in several malignancies,however,much remains unknown regarding ILF2 in small cell lung cancer(SCLC).In the current study,we explored ILF2's role in SCLC and demonstrated its importance in mitochondria quality control.Methods:Colony formation,cell proliferation,cell viability and xenograft studies were performed to examine ILF2's role on SCLC progression.Glucose uptake,lactate production,cellular oxygen consumption rate and extracellular acidification rate were measured to examine the effect of ILF2 on glucose metabolism.RNA-sequencing was utilized to explore genes regulated by ILF2.E2 F1 transcriptional activity was determined by dual luciferase reporter assay.Mitochondria quantification and mitochondrial membrane potential assays were performed to examine mitochondrial quality.Gene expression was determined by RT-qPCR,Western blot and IHC assay.Results:ILF2 promotes SCLC tumor growth in vitro and in vivo.ILF2 elevates oxidative phosphorylation expression and declines glucose intake and lactate production.Genome-wide analysis of ILF2 targets identified a cohort of genes regulated by E2 F1.In consistent with this,we found ILF2 interacts with E2 F1 in SCLC cells.Further studies demonstrated that suppression of E2 F1 expression could reverse ILF2-induced tumor growth and enhanced mitochondria function.Significantly,expression of ILF2 is progressively increased during SCLC progression and high ILF2 expression is correlated with higher histologic grades,which indicates ILF2's oncogenic role in SCLC.Conclusions:Our results demonstrate that ILF2 interacts with E2 F1 to maintain mitochondria quality and confers SCLC cells growth advantage in tumorigenesis. | Meng Zhao Yahui Liu Jiao Chang Jin Qi Ran Liu Yongwang Hou Yanhui Wang Xinwei Zhang Lu Qiao Li Ren | 2019 | Cancer Biology & Medicine2019,16,4: | 3 |
| 6 | Microelectrode array recordings of excitability of low Mg^(2+)-induced acute hippocampal slices显示文摘Neuronal connections can be detected by neuronal network discharges in hippocampal neurons cultured on multi-electrodes. However, the multi-electrode-array (MEA) has not been widely used in hippocampal slice culture studies focused on epilepsy. The present study induced spontaneous synchronous epileptiform activity using low Mg2+ artificial cerebrospinal fluid on acute hippocampal slices to record hippocampal discharges with MEA. Results showed that burst duration and average number of spikes in a burst were significantly greater in the CA3 compared with dentate gyrus and CA1 areas. In Schaffer cut-off group, CA1 area discharges disappeared, but synchronous discharges remained in the CA3 area. Moreover, synchronous discharge frequency in the Schaffer cut-off group was similar to control. However, burst duration and average number of spikes in a burst were significantly decreased compared with control (P < 0.05). | 7 | Low phosphorus promotes NSP1–NSP2 heterodimerization to enhance strigolactone biosynthesis and regulate shoot and root architecture in rice显示文摘Phosphorus is an essential macronutrient for plant development and metabolism,and plants have evolved ingenious mechanisms to overcome phosphate(Pi)starvation.However,the molecular mechanisms underlying the regulation of shoot and root architecture by low phosphorus conditions and the coordinated utilization of Pi and nitrogen remain largely unclear.Here,we show that Nodulation Signaling Pathway 1(NSP1)and NSP2 regulate rice tiller number by promoting the biosynthesis of strigolactones(SLs),a class of phytohormones with fundamental effects on plant architecture and environmental responses.We found that NSP1 and NSP2 are induced by Oryza sativa PHOSPHATE STARVATION RESPONSE2(OsPHR2)in response to low-Pi stress and form a complex to directly bind the promoters of SL biosynthesis genes,thus markedly increasing SL biosynthesis in rice.Interestingly,the NSP1/2–SL signaling module represses the expression of CROWN ROOTLESS 1(CRL1),a newly identified early SL-responsive gene in roots,to restrain lateral root density under Pi deficiency.We also demonstrated that GR24^(4DO) treatment under normal conditions inhibits the expression of OsNRTs and OsAMTs to suppress nitrogen absorption but enhances the expression of OsPTs to promote Pi absorption,thus facilitating the balance between nitrogen and phosphorus uptake in rice.Importantly,we found that NSP1p:NSP1 and NSP2p:NSP2 transgenic plants show improved agronomic traits and grain yield under low-and medium-phosphorus conditions.Taken together,these results revealed a novel regulatory mechanism of SL biosynthesis and signaling in response to Pi starvation,providing genetic resources for improving plant architecture and nutrient-use efficiency in low-Pi environments. | Kun Yuan Hao Zhang Chaoji Yu Nan Luo Jijun Yan Shuang Zheng Qingliang Hu Dahan Zhang Liquan Kou Xiangbing Meng Yanhui Jing Mingjiang Chen Xinwei Ban Zongyun Yan Zefu Lu Jian Wu Yu Zhao Yan Liang Yonghong Wang Guosheng Xiong Jinfang Chu Ertao Wang Jiayang Li Bing Wang | 2023 | Molecular Plant2023,16,11: | 2 |

| 9 | Toxic metal enrichment characteristics and sources of arid urban surface soil in Yinchuan City, China显示文摘To investigate the environmental quality of the urban surface soil in Yinchuan City, the capital of Ningxia Hui Autonomous Region(Ningxia), China, we sampled surface soil and measured the concentrations of 8 toxic metals(Pb, Cr, Cu, Zn, Co, Bi, Ni and V) using X-ray fluorescence spectrometry. The enrichment characteristics and sources of these toxic metals in the soil were analyzed by the enrichment factor(EF) and multivariate statistical analysis. The results showed that the mean concentrations of these toxic metals in the soil samples were 25.0, 109.1, 16.8, 26.0, 37.2, 2.7, 25.3 and 59.9 mg/kg for Pb, Cr, Cu, Zn, Co, Bi, Ni and V, respectively, which were 1.2, 1.8, 0.8, 0.4, 3.2, 8.7, 0.7 and 0.8 times of the corresponding background values of Ningxia soil, respectively. The variations of Pb, Zn, Co, Bi and Ni concentrations in the surface soil of Yinchuan were larger than those of the other metals. Our results also showed that the toxic metals investigated in the soil had different enrichment levels. Both Co and Bi were significantly enriched, whereas Cr was only moderately enriched in the soil. There was a deficiency or minimal enrichment of the other toxic metals in the soil. Source analysis results based on the concentration, enrichment characteristics and multivariate statistical analysis indicated that Cr, V and Ni originated from a combination of fossil fuel combustion, traffic pollution and natural occurrence. | 10 | miR-495 andmiR-5688 are down-regulated in non-small cell lung cancer under hypoxia to maintain interleukin-11 expression显示文摘Background:Hypoxia is a hallmark of cancer and is associated with poor prognosis.However,the molecular mechanism by which hypoxia promotes tumor progression remains unclear.MicroRNAs dysregulation has been shown to play a critical role in the tumor and tumor microenvironment.Here,we investigated the roles ofmiR-495 and miR-5688 in human non-small cell lung cancer(NSCLC)and their underlying mechanism.Methods:The expression levels of miR-495 and miR-5688 in human NSCLC tissue specimens were measured by quantitative real-time polymerase chain reaction(qRT-PCR).Deferoxamine(DFO)was used to determine whether the regulation of miR-495 and miR-5688 under hypoxia was dependent on hypoxia-inducible factor 1-alpha(HIF-1α).Furthermore,the functions of miR-495 and miR-5688 in tumor progression were evaluated using colony formation,3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2Htetrazolium(MTS),wound healing,transwell assays,and xenograft model.Two algorithms,PicTAR and Targetscan,were used to predict the target gene of these two miRNAs,and dual-luciferase reporter assay was conducted to confirm the target.The unpaired two-tailed t test,Pearson correlation analysis,and Fisher’s exact probability test were performed for statistical analyses.Results:Two miRNAs,miR-495 and miR-5688,were found to participate in NSCLC progression under hypoxia.They were down-regulated in NSCLC tissues compared with normal tissues.We determined that hypoxia led to the down-regulation of miR-495 and miR-5688 in NSCLC cells,which was independent of HIF-1αand cellular metabolic energy.In addition,miR-495 and miR-5688 suppressed cell proliferation,migration,and invasion in vitro.The NSCLC xenograft model showed that miR-495 and miR-5688 inhibited tumor formation in vivo.Interestingly,we found that miR-495 and miR-5688 had the same target,interleukin-11(IL-11).Recombinant human IL-11 counteracted the effects of miR-495 and miR-5688 on NSCLC cells,suggesting that miR-495 and miR-5688 executed their tumor suppressive role by repressing IL-11 expression.Conclusion:We found that hypoxia down-regulated the expression levels of miR-495 and miR-5688 in NSCLCto enhance IL-11 expression and tumor progression,indicating that the miR-495/miR-5688/IL-11 axismay serve as a therapeutic target and potential biomarker for NSCLC. | Meng Zhao Jiao Chang Ran Liu Yahui Liu Jin Qi Yanhui Wang Xinwei Zhang Lu Qiao Yu Jin Haohua An Li Ren | 2020 | Cancer Communications2020,40,9: | 2 |

| 18 | Identification of hub genes and their novel diagnostic and prognostic significance in pancreatic adenocarcinoma显示文摘Objective:The main reasons for the poor prognoses of pancreatic adenocarcinoma(PA)patients are rapid early-stage progression,advanced stage metastasis,and chemotherapy resistance.Identification of novel diagnostic and prognostic biomarkers of PA is therefore urgently needed.Methods:Three mRNA microarray datasets were obtained from the Gene Expression Omnibus database to select differentially expressed genes(DEGs).Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analyses for hub genes were performed using DAVID.Correlations between expression levels of hub genes and cancer-infiltrating immune cells were investigated by TIMER.Cox proportional hazard regression analyses were also performed.Serum hub genes were screened using the HPA platform and verified for diagnostic value using ELISAs.Results:We identified 59 hub genes among 752 DEGs.GO analysis indicated that these 59 hub genes were mainly involved in the defense response to viruses and the type I interferon signaling pathway.We also discovered that RSAD2 and SMC4 were associated with immune cell infiltration in the PA microenvironment.Additionally,DLGAP5 mRNA might be used as an independent risk factor for the prognoses of PA patients.Furthermore,the protein encoded by ISG15,which exists in peripheral blood,was validated as a potential diagnostic biomarker that distinguished PA patients from healthy controls(area under the curve:0.902,95%confidence interval:0.819–0.961).Conclusions:Our study suggested that RSAD2 and SMC4 were associated with immune cell infiltration in the PA microenvironment,while DLGAP5 mRNA expression might be an independent risk factor for the survival prognoses of PA patients.Moreover,ELISAs indicated that serum ISG15 could be a potential novel diagnostic biomarker for PA. | Duo Zuo Yongzi Chen Xinwei Zhang Zhuozhi Wang Wenna Jiang Fan Tang Runfen Cheng Yi Sun Lu Sun Li Ren Rui Liu | 2022 | Cancer Biology & Medicine2022,19,7: | 1 |

| 20 | Long-term agricultural activity affects anthropogenic soil on the Chinese Loess Plateau显示文摘Anthropogenic activities largely influence the soil quality of agricultural fields and the composition of soil. Samples of typical anthropogenic Loutu soil in the Guanzhong area of the Loess Plateau, Shaanxi Province, China were collected and measured for soil compaction, bulk density, total organic carbon(TOC), active organic carbon(AOC), and soil enzyme activities to investigate spatial variations in soil quality. The results indicate that soil compaction and bulk density increased with increasing distance from the farm village, whereas soil TOC, AOC, and soil enzyme activities firstly increased and subsequently decreased with increasing distance from the farm village. All of the tested parameters presented clear concentric distribution. Vertically, soil compaction and bulk density in the topsoil were lower than those in the subsoil, but all other tested parameters in the topsoil were significantly higher than those in the subsoil. In addition, there was a significant positive correlation between organic carbon content and enzyme activities, confirming that the spatial distribution of Loutu soil characteristics has been affected by long-term anthropogenic activities to some extent.
