1Observation and analysis of the diluted water and red tide in the sea off the Changjiang River mouth in middle and late June 2003显示文摘An interdisciplinary comprehensive survey was conducted in middle and late June 2003 with the Multi-Pa- rameter Environmental Monitoring System YSI6600 and water sample analysis in the sea off the Changjiang River mouth. The Changjiang diluted water (CDW) extended off- shore with a bimodal structure during the observation, one extending toward the southeast, the other toward the north- east. The main axis of the CDW extended toward the north- east. Asevere red tide with wide spatial extent and brown water color happened. Chlorophyll-a (Chl-a) distribution near the Changjiang River mouth also presented a bimodal structure, and its position and shape were roughly consistent with the extension of the CDW. Water sample analysis indi- cated that the serious eutrophication produced by the huge amount of nutrient load via the Changjiang River was the main cause of red tide bloom. The dominant algal specie at the most measurement stations was skeletonema costatum. There existed three centers of higher Chl-a concentration, locating at (122.45°E, 31.5°N), (122.4°E, 30.8°N) and (123.25°E, 30.0°N), respectively. The red tide at (122.45°E, 31.5°N) was located in the major modal of CDW and higher turbid seawater, its dominant algal specie was prorocentrum dentatum with density 2.23×106 ind/L. The red tide at (122.4°E, 30.8°N) was located in the second modal of CDW and lower turbid seawater, its dominant algal specie was skeletonema costatum with density 1.0×107 ind/L. 2The chimeric genes in the hybrid lineage of Carassius auratus cuvieri(♀)×Carassius auratus red var.(♂)显示文摘Hybridization can combine the genomes of different strains or species, which leads to changes of genotype and phenotype in the hybrids. In this study, we aimed to investigate the genetic variations of hybrids(WR-F1 and WR-F2) derived from the intraspecific hybridization of white crucian carp(Carassius auratus cuvieri, WCC, ♀) and red crucian carp(Carassius auratus red var., RCC, ♂). Here, we compared the orthologous genes in the liver transcriptomes of hybrids with those of WCC and RCC, and classified the orthologous genes into eight gene patterns within three categories(chimera, mutant, and biparental origin genes).The results revealed 19.04%, 4.17% chimeric genes and 6.90%, 5.05% mutations of orthologous genes in WR-F1 and WR-F2 respectively. Seventeen of twenty-three characterized genes(77%) were confirmed to be the chimeras at the genomic DNA level.The GO classification discovered that some chimeric and mutant genes were related to metabolic process, immune system and developmental process in WR-F1. Our results provide the new evidence that hybridization can combine the parental genomes,leading to changes in the genotype of the resultant hybrids. 4Targeted disruption of tyrosinase causes melanin reduction in Carassius auratus cuvieri and its hybrid progeny显示文摘The white crucian carp(Carassius auratus cuvieri,WCC) not only is one of the most economically important fish in Asia,characterized by strong reproductive ability and rapid growth rates,but also represents a good germplasm to produce hybrid progenies with heterosis.Gene knockout technique provides a safe and acceptant way for fish breeding.Achieving gene knockout in WCC and its hybrid progeny will be of great importance for both genetic studies and hybridization breeding.Tyrosinase(TYR) is a key enzyme in melanin synthesis.Depletion of tyr in zebrafish and mice results in mosaic pigmentation or total albinism.Here,we successfully used CRISPR-Cas9 to target tyr in WCC and its hybrid progeny(WR) derived from the cross of WCC(♀) and red crucian carp(Carassius auratus red var.,RCC,♂).The level of TYR protein was significantly reduced in mutant WCC.Both the mutant WCC and the mutant WR showed different degrees of melanin reduction compared with the wild-type sibling control fish,resulting from different mutation efficiency ranging from 60% to 90%.In addition,the transcriptional expression profiles of a series of pivotal pigment synthesis genes,i.e.tyrp1,mitfa,mitfb,dct and sox10,were down-regulated in tyr-CRISPR WCC,which ultimately caused a reduction in melanin synthesis.These results demonstrated that tyr plays a key role in melanin synthesis in WCC and WR,and CRISPR-Cas9 is an effective tool for modifying the genome of economical fish.Furthermore,the tyr-CRISPR models could be valuable in understanding fundamental mechanisms of pigment formation in non-model fish.Qingfeng Liu Yanhua Qi Qiuli Liang Jia Song Junmei Liu Wuhui Li Yuqin Shu Min Tao Chun Zhang Qinbo Qin Jing Wang Shaojun Liu 2019Science China(Life Sciences)2019,62,9:4
5Comparison of diploid and triploid Carassius auratus provides insights into adaptation to environmental change显示文摘Focusing on adaptation of aquatic organisms, especially fish, can help elucidate complex dynamics in freshwater ecology. The differences in genetic and epigenetic regulation between diploid and triploid Carassius auratus affect survival under eutrophication. To identify the underlying mechanisms that lead to better adaption of triploids than diploids, we compared mRNA and microRNA(miRNA) expressions in liver tissue of diploid and triploid individuals obtained from the Dongting lake water system in central China. Differential expression analysis revealed that 566 transcripts were significantly up-regulated, whereas758 were down-regulated in triploids; of these differentially expressed transcripts, 33 transcripts including cacna1 d, nfkb2, hspa1 and fgfr4 were involved in the MAPK signaling pathway, and eight transcripts were determined to be regulated by seven miRNAs. Additionally, four of 25 differential expressed(DE) transcripts(mhc1, irf7, nfkb2 and pik3 c) involving the viral carcinogenesis pathway were regulated by four miRNAs. Furthermore, genetic polymorphisms analysis showed that more heterozygous mutations were detected in triploids than diploids. The d N/d S results revealed that 21 genes were under positive selection(d N/d S>1) in C. auratus complex. 6Heterogeneous electrolyte membranes enabling double-side stable interfaces for solid lithium batteries显示文摘The solid polymer electrolyte(SPE) is one of the most promising candidates for building solid lithium batteries with high energy density and safety due to its advantages of flexibility and light-weight.However,the conventional monolayered electrolytes usually exhibit unstable contacts with either high-voltage cathodes or Li-metal anodes during cell operation.Herein,heterogeneous dual-layered electrolyte membranes(HDEMs) consisting of the specific functional polymer matrixes united with the designed solid ceramic fillers are constructed to address the crucial issues of interfacial instability.The electrolyte layers composed of the high-conductivity and oxidation-resistance polyacrylonitrile(PAN) combined with Li_(0.33)La_(0.557)TiO_(3) nanofibers are in contact with the high-voltage cathodes,achieving the compatible interface between the cathodes and the electrolytes.Meanwhile,the electrolyte layers composed of the highstability and dendrite-resistance polyethylene oxide(PEO) with Li_(6.4)La_(3) Zr_(1.4)Ta_(0.6)O_(12) nanoparticles are in contact with the Li-metal anodes,aiming to suppress the dendrite growth,as well as avoid the passivation between the PAN and the Li-metal.Consequently,the solid LiNi_(0.6)Co_(0.2)Mn_(0.2)O2‖Li full cells based on the designed HDEMs show the good rate and cycling performance,i.e.the discharge capacity of 170.1 mAh g^(-1) with a capacity retention of 78.2% after 100 cycles at 0.1 C and 30℃.The results provide an effective strategy to construct the heterogeneous electrolyte membranes with double-side stable electrode/-electrolyte interfaces for the high-voltage and dendrite-free solid lithium batteries.Shuang Mu Weilin Huang Wuhui Sun Ning Zhao Mengyang Jia Zhijie Bi Xiangxin Guo 2021Journal of Energy Chemistry2021,30,9:2
7Evidence for the paternal mitochondrial DNA in the crucian carp-like fish lineage with hybrid origin显示文摘In terms of taxonomic status,common carp(Cyprinus carpio,Cyprininae)and crucian carp(Carassius auratus,Cyprininae)are different species;however,in this study,a newborn homodiploid crucian carp-like fish(2n=100)(2nNCRC)lineage(F1–F3)was established from the interspecific hybridization of female common carp(2n=100)×male blunt snout bream(Megalobrama amblycephala,Cultrinae,2n=48).The phenotypes and genotypes of 2 n NCRC differed from those of its parents but were closely related to those of the existing diploid crucian carp.We further sequenced the whole mitochondrial(mt)genomes of the 2n NCRC lineage from F1 to F3.The paternal mt DNA fragments were stably embedded in the mt-genomes of F1–F3 generations of 2n NCRC to form chimeric DNA fragments.Along with this chimeric process,numerous base sites of F1–F3 generations of 2 n NCRC underwent mutations.Most of these mutation sites were consistent with the existing diploid crucian carp.Moreover,the mt DNA organization and nucleotide composition of 2n NCRC were more similar to those of the existing diploid crucian carp than those of the parents.The inheritable chimeric DNA fragments and mutant loci in the mt-genomes of different generations of 2nNCRC provided important evidence of the mt DNA change process in the newborn lineage derived from hybridization of different species.Our findings demonstrated for the first time that the paternal mt DNA were transmitted into the mt-genomes of homodiploid lineage,which provided new insights into the existence of paternal mt DNA in the mt DNA inheritance.Shi Wang Ni Jiao Lu Zhao Meiwen Zhang Pei Zhou Xuexue Huang Fangzhou Hu Conghui Yang Yuqin Shu Wuhui Li Chun Zhang Min Tao Bo Chen Ming Ma Shaojun Liu 2020Science China(Life Sciences)2020,63,1:2
8Quantification of groundwater recharge and evapotranspiration along a semi-arid wetland transect using diurnal water table fluctuations显示文摘Groundwater is a vital water resource in arid and semi-arid areas.Diurnal groundwater table fluctuations are widely used to quantify rainfall recharge and groundwater evapotranspiration(ET_(g)).To assess groundwater resources for sustainable use,we estimated groundwater recharge and ET_(g) using the diurnal water table fluctuations at three sites along a section with different depths to water table(DWT)within a wetland of the Mukai Lake in the Ordos Plateau,Northwest China.The water table level was monitored at an hourly resolution using a Keller DCX-22 A data logger that measured both the total pressure and barometric pressure,so that the effect of barometric pressure could be removed.At this study site,a rapid water table response to rainfall was observed in two shallow wells(i.e.,Obs1 and Obs2),at which diurnal water table fluctuations were also observed over the study period during rainless days,indicating that the main factors influencing water table variation are rainfall and ET_(g).However,at the deep-water table site(Obs3),the groundwater level only reacted to the heaviest rainfalls and showed no diurnal variations.Groundwater recharge and ET_(g) were quantified for the entire hydrological year(June 2017–June 2018)using the water table fluctuation method and the Loheide method,respectively,with depth-dependent specific yields.The results show that the total annual groundwater recharge was approximately 207 mm,accounting for 52%of rainfall at Obs1,while groundwater recharge was approximately 250 and 21 mm at Obs2 and Obs3,accounting for 63%and 5%of rainfall,respectively.In addition,the rates of groundwater recharge were mainly determined by rainfall intensity and DWT.The daily mean ET_(g) at Obs1 and Obs2 over the study period was 4.3 and 2.5 mm,respectively,and the main determining factors were DWT and net radiation.JIA Wuhui YIN Lihe ZHANG Maosheng ZHANG Xinxin ZHANG Jun TANG Xiaoping DONG Jiaqiu 2021Journal of Arid Land2021,13,5:1

20Centennial-scale monsoon collapses during Marine Isotope Stage(MIS)7e detected in a northern Chinese stalagmite record显示文摘Occurrence or absence of centennial-scale climatic variability during Marine Isotope Stage(MIS)7e,the first peak interval of the Penultimate Interglacial,remains ambiguous due to the limited available high-resolved records.Here,we present a decadal-resolved stalagmiteδ^(18)O record from northern China spanning from 242.3 to 236.8 ka BP,covering MIS7e.The composite KLSδ^(18)O record,integrating this study with the previous record in the same cave,shows a descending trend from TerminationⅢto MIS7e,which follows the increasing Northern Hemisphere summer insolation(NHSI).This observation further emphasizes the important influence of NHSI on the glacial/interglacial transition.There are more large-amplitude,millennial-to centennial-scale variability occurring during TerminationⅢcompared with MIS7e,implying that ice-sheet decay potentially plays an important role in climatic excursions.Four centennial-scale summer monsoon collapses,peaking at~242.1,~240.8,~239.3 and~238.2 ka BP,are detected in our new record,indicating the pervasiveness of the intra-interglacial climatic instability.Given the lack of solar activity and freshwater outbursts data during MIS7e,it is not possible to conclude about the drivers of the identified abrupt climatic anomalous.Following the dynamic mechanisms of sub-millennial climatic anomalous during early Holocene,we tentatively propose that slowdown of the meridional overturning circulation caused by freshwater fluxes into North Atlantic and/or attenuated solar irradiance is the potential forcing for the abrupt climate events within MIS7e.Additionally,attenuated solar irradiance could also result in climatic anomalies through low-latitude processes.Wuhui DUAN Ming TAN Lisheng WANG Xuefeng WANG Zhibang MA Linlin CUI Zhengyi XIAO 2023Science China Earth Sciences2023,66,12:0
