| 1 | A high-quality genome sequence of alkaligrass provides insights into halophyte stress tolerance显示文摘Alkaligrass(Puccinellia tenuiflora) is a monocotyledonous halophytic forage grass widely distributed in Northern China. It belongs to the Gramineae family and shares a close phylogenetic relationship with the cereal crops, wheat and barley. Here, we present a high-quality chromosome-level genome sequence of alkaligrass assembled from Illumina, Pac Bio and 10× Genomics reads combined with genome-wide chromosome conformation capture(Hi-C) data. The ~1.50 Gb assembled alkaligrass genome encodes 38,387 protein-coding genes, and 54.9% of the assembly are transposable elements, with long terminal repeats being the most abundant. Comparative genomic analysis coupled with stress-treated transcriptome profiling uncovers a set of unique saline-and alkaline-responsive genes in alkaligrass. The high-quality genome assembly and the identified stress related genes in alkaligrass provide an important resource for evolutionary genomic studies in Gramineae and facilitate further understanding of molecular mechanisms underlying stress tolerance in monocotyledonous halophytes. | 2 | Evaluating Common Land Model Energy Fluxes Using FLUXNET Data显示文摘Given the crucial role of land surface processes in global and regional climates, there is a pressing need to test and verify the performance of land surface models via comparisons to observations. In this study, the eddy covariance measurements from 20 FLUXNET sites spanning more than 100 site-years were utilized to evaluate the performance of the Common Land Model(Co LM) over different vegetation types in various climate zones. A decomposition method was employed to separate both the observed and simulated energy fluxes, i.e., the sensible heat flux, latent heat flux, net radiation, and ground heat flux, at three timescales ranging from stepwise(30 min) to monthly. A comparison between the simulations and observations indicated that Co LM produced satisfactory simulations of all four energy fluxes, although the different indexes did not exhibit consistent results among the different fluxes. A strong agreement between the simulations and observations was found for the seasonal cycles at the 20 sites, whereas Co LM underestimated the latent heat flux at the sites with distinct dry and wet seasons, which might be associated with its weakness in simulating soil water during the dry season. Co LM cannot explicitly simulate the midday depression of leaf gas exchange, which may explain why Co LM also has a maximum diurnal bias at noon in the summer. | 3 | Profiles and Source Apportionment of Nonmethane Volatile Organic Compounds in Winter and Summer in Xi’an, China, based on the Hybrid Environmental Receptor Model显示文摘Summer and winter campaigns for the chemical compositions and sources of nonmethane hydrocarbons(NMHCs)and oxygenated volatile organic compounds(OVOCs)were conducted in Xi’an.Data from 57 photochemical assessment monitoring stations for NMHCs and 20 OVOC species were analyzed.Significant seasonal differences were noted for total VOC(TVOC,NMHCs and OVOCs)concentrations and compositions.The campaign-average TVOC concentrations in winter(85.3±60.6 ppbv)were almost twice those in summer(47.2±31.6 ppbv).Alkanes and OVOCs were the most abundant category in winter and summer,respectively.NMHCs,but not OVOCs,had significantly higher levels on weekends than on weekdays.Total ozone formation potential was higher in summer than in winter(by 50%)because of the high concentrations of alkenes(particularly isoprene),high temperature,and high solar radiation levels in summer.The Hybrid Environmental Receptor Model(HERM)was used to conduct source apportionment for atmospheric TVOCs in winter and summer,with excellent accuracy.HERM demonstrated its suitability in a situation where only partial source profile data were available.The HERM results indicated significantly different seasonal source contributions to TVOCs in Xi’an.In particular,coal and biomass burning had contributions greater than half in winter(53.4%),whereas traffic sources were prevalent in summer(53.1%).This study’s results highlight the need for targeted and adjustable VOC control measures that account for seasonal differences in Xi’an;such measures should target not only the severe problem with VOC pollution but also the problem of consequent secondary pollution(e.g.,from ozone and secondary organic aerosols). | Jian SUN Zhenxing SHEN Yue ZHANG Wenting DAI Kun HE Hongmei XU Zhou ZHANG Long CUI Xuxiang LI Yu HUANG Junji CAO | 2021 | Advances in Atmospheric Sciences2021,38,1: | 2 |
| 4 | Association between TSHR gene polymorphism and the risk of Graves' disease:a meta-analysis显示文摘Thyroid stimulating hormone receptor(TSHR) is thought to be a significant candidate for genetic susceptibility to Graves' disease(GD).However,the association between TSHR gene polymorphism and the risk of GD remains controversial.In this study,we investigated the relationship between the two conditions by meta-analysis.We searched all relevant case-control studies in PubMed,Web of Science,CNKI and Wanfang for literature available until May2015,and chose studies on two single nucleotide polymorphisms(SNPs):rs 179247 and rsl2101255,within TSHR intron-1.Bias of heterogeneity test among studies was determined by the fixed or random effect pooled measure,and publication bias was examined by modified Begg's and Egger's test.Eight eligible studies with 15 outcomes were involved in this meta-analysis,including 6,976 GD cases and 7,089 controls from China,Japan,Poland,UK and Brazil.Pooled odds ratios(ORs) for allelic comparisons showed that both TSHR rsl79247A/G and rsl2101255T/C polymorphism had significant association with GD(OR=1.422,95%CI=1.353—1.495,P<0.001,P_(heterogeneity)=0.448;OR= 1.502,95%CI:1.410-1.600,P<0.001,P_(heterogeneity)=0.642),and the associations were the same under dominant,recessive and co-dominant models.In subgroup analyses,the conclusions are also consistent with all those in Asian,European and South America subgroups(P<0.001).Our meta-analysis revealed a significant association between TSHR rsl79247A/G and rsl2101255T/C polymorphism with GD in five different populations from Asia,Europe and South America.Further studies are needed in other ethnic backgrounds to independently confirm our findings. | Wei Qian Kuanfeng Xu Wenting Jia Ling Lan Xuqin Zheng Xueyang Yang Dai Cui | 2016 | The Journal of Biomedical Research2016,30,6: | 2 |

The results showed that through the alignment analysis by DNAMAN software,three pairs of unknown proteins with higher comprehensive matching degrees were obtained,and these proteins were predicted to be coded by hrp gene cluster of type III secretion system;through Bioedit and Prot Param,the amino acid composition,size,molecular weight,isoelectric point and hydrophobicity of the each of the six unknown proteins were obtained; and corresponding secondary structures were obtained using DNAMAN. [Method] The occurrence status of gray mold in protected tomato in Shawan County, Manas County, the experimental field of Shihezi University and Shihezi Huayu base were investigated during the occurrence period of tomato gray mold, and the resistance of intro-duced variety against tomato gray mold was identified at seedling stage through indoor artificial inoculation method. [Result] The diseased leaf rate of tomato gray mold was 17.52%-100%, the diseased fruit rate was 3.1%-55.69%, and the disease index was 15.98-100. [Methods] BCr,BCs and F_2 were constructed from tomato CLN2037 and susceptible cultivar T2-03,and their resistance to late blight was identified and analyzed by inoculating the disease in the seedling stage of tomatoes. The genetic linkage analysis was made based on the genetic linkage map constructed by ICu GI using group segregation method and 90 pairs of tomato SSR primers. [Results]The resistance of CLN2037 to late blight was controlled by a single recessive gene. The polymorphic bands amplified by primer DM0231 had a linkage relationship with resistance genes.
