1Therapeutic effect of acupuncture and massage for shoulder-hand syndrome in hemiplegia patients:a clinical two-center randomized controlled trial显示文摘OBJECTIVE:To evaluate the therapeutic effects of acupuncture and massage for shoulder-hand syndrome in hemiplegia patients.METHODS:One hundred and twenty hemiplegia patients with stage I shoulder-hand syndrome were randomly divided into a group treated with standardized electric acupuncture and massage,and a group treated with rehabilitation therapy for 6 weeks.The primary indices evaluated were pain on passive movement of the shoulder using the numeric pain rating scale(NPRS),and the number of patients with shoulder-hand syndrome at Steinbrocker stage II or III after treatment.The secondary indices were Fugl-Meyer evaluation of functional movement of the upper limb and hand using the modified rankin scale(MRS).RESULTS:At post-treatment evaluation and a 12-week follow-up visit,NPRS score,number of patients with stage II or III shoulder-hand syndrome,and MRS score were all improved in the acupuncture-massage group compared with the rehabilitation group(P<0.05).On Fugl-Meyer evaluation,functional movement of the upper limb was also improved in the acupuncture-massage group compared with the rehabilitation group(P<0.05).CONCLUSION:Standardized acupuncture-massage therapy may have curative effects on shoulder-hand syndrome in hemiplegia patients.Ning Li Fengwei Tian Chengwei Wang Pengming Yu Xi Zhou Qian Wen Xiulan Qiao Lu Huang 2012Journal of Traditional Chinese Medicine2012,32,3:25
2Clinical curative effect of electric acupuncture on acute cerebral infarction:a randomized controlled multicenter trial显示文摘OBJECTIVE: To examine whether electric acupuncture can improve the daily life of patients with ischemic cerebral apoplexy at acute stage.METHODS: A stratified-block randomized controlled multicenter trial was designed for this study.Totally 340 patients with acute ischemic cerebral apoplexy were randomly divided into an electric acupuncture group and a control group. In the electric acupuncture group, 170 patients were treated with electric acupuncture and routine therapy, and170 patients in the control group with routine therapy alone. Major indexes for judging curative effect were Barthel index at 3- and 6- months follow-up visits and number of re-hospitalized patients. Mi-nor indexes for judging curative effect were change in the score for nervous dysfunction at 4and 12 weeks follow-up visits and number of patients persisting in rehabilitation treatment with acupuncture during follow-up visit.RESULTS: Baseline data at the time of case selection between the two groups were similar. The odds ratio(OR) was 0.92, and the 95% confidence interval(CI) was 0.49-1.73 in disabled rate and 0.73 and 0.51-1.05 in the number of re-hospitalized patients in the electric acupuncture group at 6-month follow up visit compared with the control group.There was no difference in the score for nervous dysfunction at the end of 12-week follow-up visit between the two groups. The score for nervous dysfunction at the end of 4-week treatment in the electric acupuncture group was significantly higher than that in the control group(P<0.05). The number of patients discharged from hospital who persisted in rehabilitation treatment with acupuncture in the acupuncture group was significantly higher than that in the control group.CONCLUSION: Using electric acupuncture to treat patients with acute ischemic cerebral apoplexy can effectively improve the nervous dysfunction scores after 4-week treatment and their ability to deal with daily life after 6-month follow-up visit. 4Generation of super-resolved optical needle and multifocal array using graphene oxide metalenses显示文摘Ultrathin flat metalenses have emerged as promising alternatives to conventional diffractive lenses,offering new possibilities for myriads of miniaturization and interfacial applications.Graphene-based materials can achieve both phase and amplitude modulations simultaneously at a single position due to the modification of the complex refractive index and thickness by laser conversion from graphene oxide into graphene like materials.In this work,we develop graphene oxide metalenses to precisely control phase and amplitude modulations and to achieve a holistic and systematic lens design based on a graphene-based material system.We experimentally validate our strategies via demonstrations of two graphene oxide metalenses:one with an ultra-long(~16λ)optical needle,and the other with axial multifocal spots,at the wavelength of 632.8 nm with a 200 nm thin film.Our proposed graphene oxide metalenses unfold unprecedented opportunities for accurately designing graphene-based ultrathin integratable devices for broad applications.Hongtao Wang Chenglong Hao Han Lin Yongtian Wang Tian Lan Chengwei Qiu Baohua Jia 2021Opto-Electronic Advances2021,4,2:10
5Discriminated sgRNAs-Based SurroGate System Greatly Enhances the Screening Efficiency of Plant Base-Edited Cells显示文摘The development of CRISPR/Cas9-mediated base editing has made genomic modification more efficient. However, selection of genetically modified cells from millions of treated cells, especially plant cells, is still challenging. In this study, an efficient surrogate reporter system based on a defective hygromycin resistance gene was established in rice to enrich base-edited cells. After step-by-step optimization, the Discriminated sgRNAs-based SurroGate system (DisSUGs) was established by artificially differentiating the editing abilities of a wild-type single guide RNA (sgRNA) targeting the surrogate reporter gene and an enhanced sgRNA targeting endogenous sites. The DisSUGs enhanced the efficiency of screening base-edited cells by 3- to 5-fold for a PmCDA1-based cytosine-to-tyrosine base editor (PCBE), and 2.5- to 6.5-fold for an adenine base editor (ABE) at endogenous targets. These targets showed editing efficiencies of <25% in the conventional systems. The DisSUGs greatly enhanced the frequency of homozygous substitutions and expanded the activity window slightly for both a PCBE and an ABE. Analyses of the total number of single-nucleotide variants from whole-genome sequencing revealed that, compared with the no-enrichment PCBE strategy, the DisSUGs did not alter the frequency of genome-wide sgRNA-independent off-target mutations, but slightly increased the frequency of target-dependent off-target mutations. 6HY5-HDA9 Module Transcriptionally Regulates Plant Autophagy in Response to Light-to-Dark Conversion and Nitrogen Starvation显示文摘Light is arguably one of the most important environmental factors that determines virtually all aspects of plant growth and development,but the molecular link between light signaling and the autophagy pathway has not been elucidated in plants.In this study,we demonstrate that autophagy is activated during light-to-dark conversion though transcriptional upregulation of autophagy-related genes(ATGs).We showed that depletion of the ELONGATED HYPOCOTYL 5(HY5),a key component of light signaling,leads to enhanced autophagy activity and resistance to extended darkness and nitrogen starvation treatments,contributing to higher expression oiATGs.HY5 interacts with and recruits HISTONE DEACETYLASE 9(HDA9)to ATG5 and ATG8e loci to repress their expression by deacetylation of the Lys9 and Lys27 of histone 3.Furthermore,we found that both darkness and nitrogen depletion induce the degradation of HY5 via 26S proteasome and the concomitant disassociation of HDA9 from ATG5 and ATG8e loci,leading to their depression and thereby activated autophagy.Genetic analysis further confirmed that HY5 and HDA9 act synergistically and function upstream of the autophagy pathway.Collectively,our study unveils a previously unknown transcriptional and epigenetic network that regulates autophagy in response to light-to-dark conversion and nitrogen starvation in plants.Chao Yang Wenjin Shen Lianming Yang Yun Sun Xibao Li Minyi Lai Juan Wei Chaojun Wang Yingchao Xu Faqiang Li Shan Liang Chengwei Yang Shangwei Zhong Ming Luo Caiji Gao 2020Molecular Plant2020,13,3:8
73-year Treatment of Tenofovir Alafenamide vs.Tenofovir Disoproxil Fumarate for Chronic HBV Infection in China显示文摘Background and Aims:Tenofovir alafenamide(TAF)has similar efficacy to tenofovir disoproxil fumarate(TDF)but with improved renal and bone safety in chronic hepatitis B patients studied outside of China.We report 3-year results from two phase 3 studies with TAF in China(Clinicaltrials.gov:NCT02836249 and NCT02836236).Methods:Chinese hepatitis B e antigen(HBeAg)-positive and-negative chronic hepatitis B patients with viremia and elevated alanine aminotransferase were randomized 2:1 to TAF or TDF treatment groups and treated in a double-blind fashion for 144 weeks(3 years).Efficacy responses were assessed by individual study while safety was assessed by a pooled analysis.Results:Of the 334 patients(180 HBeAg-positive and 154 HBeAg-negative)randomized and treated,baseline characteristics were similar between groups.The overall mean age was 38 years and 73%were male.The mean HBV DNA was 6.4 log10 IU/mL.The median alanine aminotransferase was 88 U/L,and 37%had a history of antiviral use.At week 144,the proportion with HBV DNA<29 IU/mL was similar among the two groups,with TAF at 83%vs.TDF at 79%,and TAF at 93%vs.TDF at 92%for the HBeAg-positive and-negative patients,respectively.In each study,higher proportions of TAF than TDF patients showed normalized alanine aminotransferase(via the American Association for the Study of Liver Diseases and the China criteria)and showed loss of HBsAg;meanwhile,the HBeAg seroconversion rates were similar.Treatment was well-tolerated among the TAF patients,who showed a smaller median decline in creatinine clearance(−0.4 vs.−3.2 mL/min;p=0.014)and less percentage change in bone mineral density vs.TDF at hip(−0.95%vs.−1.93%)and spine(+0.35%vs.−1.40%).Conclusions:In chronic hepatitis B patients from China,TAF treatment provided efficacy similar to TDF but with better renal and bone safety at 3 years.Jinlin Hou Qin Ning Zhongping Duan You Chen Qing Xie Fu-Sheng Wang Lunli Zhang Shanming Wu Hong Tang Jun Li Feng Lin Yongfeng Yang Guozhong Gong John FFlaherty Anuj Gaggar Shuyuan Mo Cong Cheng Gregory Camus Chengwei Chen Yan Huang Jidong Jia Mingxiang Zhang GS-US-320-0110 and GS-US-320-0108 China Investigators 2021Journal of Clinical and Translational Hepatology2021,9,3:7
8Well-aligned carbon nanotube array membrane synthesized in porous alumina template by chemical vapor deposition显示文摘A new simple approach was developed for preparing well-aligned and monodispersed carbon nanotuoe (CNT) array membrane within the cylindrical pores of anodic aluminum oxide (AAO) template by chemical vapor deposition (CVD). Acetylene and hydrogen were used in the CVD process with Fe-catalyzer at 700℃ under 250 Pa. Scanning electron microscope (SEM) and transmission electron microscope (TEM) were employed to characterize the resulting highly-oriented uniform hollow tube array which had a diameter of about 250 nm, a tube density of 5.3×108cm2 and a length of about 60 μm. The length and diameter of the tubes depend on the thickness and pore diameter of the template. The growth properties of the CNT array film can be achieved by controlling the structure of the template, the particle size of Fe-catalyzer, the temperature in the reactor, the flow ratio and the deposition time. 9Function of Ca^(2+)-/calmodulin-dependent protein kinase Ⅳ in Ca^(2+)-stimulated neuronal signaling and behavior显示文摘The activity of Ca2+/calmodulin-dependent protein kinase IV(Ca MKIV) is sensitive to activity-dependent changes in the level of intracellular Ca2+.Following neuronal stimulation,the activation of Ca MKIV may trigger synaptic modifications and transcriptional responses,both of which are involved in regulating cognitive and emotional behavior.Here,we used Ca MKIV knockout(KO) neurons and mice to examine the function of Ca MKIV in Ca2+-stimulated intracellular signaling and animal behavior,respectively.Following NMDA receptor activation or membrane depolarization,the up-regulation of CREB(c AMP responsive element binding protein) and its target gene Bdnf(brain-derived neurotrophic factor) was intact in cortical neurons obtained from Ca MKIV KO mice.Ca MKIV KO mice displayed severe impairment in contextual fear memory but normal locomotor activity and anxiety level in the contextual training chamber.Although Ca MKIV KO mice showed normal memory in the standard passive avoidance task,they were defective in learning the temporal dissociative passive avoidance task.As indicated by the light/dark test and marble-burying test data,Ca MKIV KO mice showed less anxiety and normal perseveration.In the voluntary wheel-running test,Ca MKIV KO mice showed normal running time and distance but higher maximal running speed.Our results demonstrate the function of Ca MKIV in regulating different forms of fear memory,anxiety,and certain aspect of motor function.SONG ZiXiang CHEN Qi DING Qi ZHENG Fei LI ChengWei XU LePing WANG HongBing 2015Science China(Life Sciences)2015,58,1:6

11Modification of wettability of stainless steel by picosecond laser surface microstructuring显示文摘We report on the modification of the wettability of stainless steel by picosecond laser surface microstructuring in this paper. Compared with traditional methods, picosecond laser-induced surface modification provides a fast and facile method for surface modification without chemical damage and environmental pollution. As a result of treatment by 100 ps laser pulses, microstructures are fabricated on the stainless steel sample surface, contributing to the increase of the contact angle from 88° to 105°, which realizes a transformation from hydrophilicity to hydrophobicity. 13Outline of Metallogenic Regularity of Bauxite Deposits in China显示文摘Among the abundant aluminum ore resources in China, bauxite is dominated, which is mainly distributed in 19 provinces and regions, including Shanxi, Henan, Guizhou and Guangxi. The major deposit type of bauxite is paleo-weathering crust sedimentary type, and the other one is the accumulation type. The main metallogenic period is the late Paleozoic Era followed by the Cenozoic Era. The metallogenic tectonic background is characterized by a cratonic environment. This paper summarizes the bauxite metallogenic regularity based on the characteristics of bauxite resources, bauxite deposit type, bauxite metallogenic belt and metallogenic series in China, and 15 bauxite metallogenic belts, 8 bauxite metallogenic series and 7 bauxite ore concentrated areas were identified in the study. 14高分辨磁共振成像对颈动脉粥样硬化斑块表面钙化与斑块稳定性关系研究显示文摘目的通过高分辨磁共振成像(MRI)多序列扫描,分析颈动脉粥样硬化斑块的表面钙化的形状、位置与斑块稳定性的关系。方法运用高分辨MRI多序列(3D-TOF、DIR T1WI、FSE T2WI、PDWI)对34例颈动脉斑块患者进行扫描,分析钙化类型和位置与斑块表面溃疡的关系。结果 87层表面钙化斑块,59层(67.8%)见斑块表面溃疡。按钙化类型和部位划分,64层为点状、弧形的不规则钙化,23层为大片状钙化;58层为边缘钙化,29层为中央钙化。不规则钙化组伴斑块表面溃疡的概率显著高于大片状钙化组;边缘钙化组较中央钙化组更易伴斑块表面溃疡。结论表面钙化是斑块不稳定的重要因素之一,容易导致斑块表面溃疡形成;表面钙化的类型、部位对斑块的稳定性有重要影响。彭少华 王成伟 尹喜 PENG Shaohua WANG Chengwei YIN Xi 2012临床荟萃2012,27,6:4
15Expanding base editing scope to near-PAMless with engineered CRISPR/Cas9 variants in plants显示文摘Dear Editor,Base editors(BEs),including cytosine base editor(CBE)and adenine base editor(ABE),have been widely used to generate irreversible nucleotide substitution in plants and animals.However,their wide applications are largely hindered by the strict NG protospacer adjacent motif(PAM)sequences recognized by Streptococcus pyogenes Cas9(SpCas9)and its engineered variants,such as SpCas9-NG and xCas9(Hua et al.,2019;Ren et al.,2019;Wu et al.,2019;Zhong et al.,2019;Zhang et al.,2020).Most recently,it was reported that three new SpCas9 variants,SpCas9-NRRH,SpCas9-NRTH,and SpCas9-NRCH,could recognize non-G PAMs(NRNH,where R is A or G and H is A,C,or T)in human cells(Miller et al.,2020).Meanwhile,SPRY,another new SpCas9 variant,was developed to greatly expand the editing scope of BEs to nearly PAMless(Walton et al.,2020).In this study,we generated a series of efficient BE toolkits and almost achieved C-to-T mutation without PAM restriction except for NTG PAM,and largely expanded A-to-G mutation scope in stable transformed rice,providing a reference for application in other plants.Chengwei Zhang Yao Wang Feipeng Wang Si Zhao Jinling Song Feng Feng Jiuran Zhao Jinxiao Yang 2021Molecular Plant2021,14,2:4
16Developing high-efficiency base editors by combining optimized synergistic core components with new types of nuclear localization signal peptide显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)–CRISPR-associated protein(Cas) system has been widely used for genome editing. In this system, the cytosine base editor(CBE) and adenine base editor(ABE) allow generating precise and irreversible base mutations in a programmable manner and have been used in many different types of cells and organisms. However, their applications are limited by low editing efficiency at certain genomic target sites or at specific target cytosine(C) or adenine(A) residues. Using a strategy of combining optimized synergistic core components, we developed a new multiplex super-assembled ABE(sABE) in rice that showed higher base-editing efficiency than previously developed ABEs. We also designed a new type of nuclear localization signal(NLS) comprising a FLAG epitope tag with four copies of a codon-optimized NLS(F4NLS^(r2)) to generate another ABE named F4NLS-sABE. This new NLS increased editing efficiency or edited additional A at several target sites. A new multiplex super-assembled CBE(sCBE) and F4NLS^(r2) involved F4NLS-sCBE were also created using the same strategy. F4NLS-sCBE was proven to be much more efficient than sCBE in rice. 18A Methodology of Epidemiologic Study in the General Population Focusing on Food Allergy—China, 2020显示文摘Introduction:Adverse reactions to food(ARF)are a major worldwide public health and food safety problem.Among the various causes of ARF,food allergies(FA)are particularly serious as the immune response that is triggered can be fatal even at very low doses.However,the prevalence of ARF and FA in the general population in China is presently unclear.This study aims to determine the epidemiological characteristics and risk factors for ARF and FA,which can be a basis for estimating thresholds for major food allergens.Methods:This is a multicenter,cross-sectional,epidemiologic survey with a case-control study nested among a selected population in China.Random individuals were recruited using stratified cluster random sampling;ARF and FA were comprehensively assessed using modified EuroPrevall FA Project questionnaires as well as structured interviews,sensitization testing,and double-blind placebocontrolled food challenges(DBPCFC).Results:This method of epidemiologic study on ARF and FA was a pilot application in Jiangxi Province from January 2020;among the total 21,273 children and adults that completed the questionnaire,5.8%reported ARF and 4.3%reported FA.ARF were determined to be associated with age,gender,and region.Animal-derived foods were the dominant offending foods,especially shrimp,and skin symptoms were the most commonly reported ARF.Discussion:This is the first multi-center,largescale,epidemiologic study on ARF and FA using standardized methods,including DBPCFC,in the Chinese general population.This study presents an important approach to assessing ARF and FA,provides significant insights about the prevalence of ARF and FA,and facilitates support for updating the list of allergenic food labels:which will be essential for improving ARF prevention and management in China.Hua Feng Yan Chen Hongbing Chen Chengwei Liu Wei Zhou Lianglu Wang Yongning Wu 2022China CDC weekly2022,4,34:2

20Synthesis of ordered Si nanowire arrays in porous anodic aluminum oxide templates显示文摘Highly ordered poly crystalline Si nanowire arrays were synthesized in porous anodic aluminum oxide (AAO) templates by the chemical vapor deposition (CVD) method. The morphological structure, the crystal character of Si nanowire arrays and the individual nanowire were analyzed by the transmission electron microscopy (TEM), scanning electron microscopy (SEM), atom force microscopy (AFM) and the X-ray diffraction spectrum (XRD), respectively. It is shown that most fabricated silicon nanowires (SiNWs) tend to be assembled parallelly in bundles and constructed with highly orientated arrays. This method provides a simple and low cost fabricating craftwork and the diameters and lengths of SiNWs can be controlled, the large area Si nanowire arrays can be achieved easily under such a way.
