| 1 | Characterization and mapping of QTLs on chromosome 2D for grain size and yield traits using a mutant line induced by EMS in wheat显示文摘Production of mutants with altered phenotypes is a powerful approach for determining the biological functions of genes in an organism. In this study, a high-grain-weight mutant line M8008 was identified from a library of mutants of the common wheat cultivar YN15 treated with ethylmethane sulfonate(EMS). F2 and F2:3generations produced from crosses of M8008 × YN15(MY) and M8008 × SJZ54(MS) were used for genetic analysis. There were significant differences between M8008 and YN15 in plant height(PH), spike length(SL),fertile spikelet number per spike(FSS), grain width(GW), grain length(GL), GL/GW ratio(GLW), and thousand-grain weight(TGW). Most simple correlation coefficients were significant for the investigated traits, suggesting that the correlative mutations occurred in M8008. Approximately 21% of simple sequence repeat(SSR) markers showed polymorphisms between M8008 and YN15, indicating that EMS can induce a large number of mutated loci. Twelve quantitative trait loci(QTLs) forming QTL clusters(one in MY and two in MS) were detected. The QTL clusters coinciding with(MY population) or near(MS population) the marker wmc41 were associated mainly with grain-size traits, among which the M8008 locus led to decreases in GW, factor form density(FFD), and TGW and to increases in GLW. | 8 | Effectiveness of herb-partitioned moxibustion combined with electroacupuncture on polycystic ovary syndrome in patients with symptom pattern of kidney deficiency and phlegm-dampness显示文摘OBJECTIVE:To observe the effect of herb-partitioned moxibustion combined with electroacupuncture on polycystic ovary syndrome(PCOS)in patients with symptom pattern of kidney deficiency and phlegm-dampness.METHODS:Totally 62 PCOS patients who met the inclusion criteria were randomly divided into intervention group and control group.In the intervention group,acupoints were subjected to(43±1)℃herb-partitioned moxibustion and electroacupuncture treatment.In the control group,acupoints were subjected to(37±1)℃herb-partitioned moxibustion and sham electroacupuncture treatment.Treatment was provided 3 times per week for 12 weeks,and the treatment was stopped during menstruation.After the treatment,the clinical efficacy,the Traditional Chinese Medicine(TCM)symptom pattern score,basal body temperature(BBT)levels,menstrual cycle,and related sex hormone levels of the two groups were evaluated.RESULTS:Sixty patients were eventually included in the study(30 in the intervention group and 30 in the control group).There were not obvious between-group differences in the baseline efficacy parameters(all P>0.05).(a)After treatment,the two groups have improved in terms of clinical efficacy,TCM symptom pattern score,menstrual cycle and,BBT(P<0.01,<0.05),and the intervention group was better than the control group(P<0.01,<0.05).(b)In the intervention group,the estradiol(E2)and P450 aromatase(P450 arom)levels were increased(all P<0.05),the testosterone(T)level was decreased(P<0.05),and the luteotropic hormone(LH)and anti-Müllerian hormone(AMH)levels were significantly decreased(P<0.01).In the control group,E2 level was increased(P<0.05),and LH level was decreased(P<0.05).After treatment,the T,LH,and AMH levels of the intervention group were decreased compared with those of the control group(all P<0.05),and the P450 arom level was increased(P<0.05).CONCLUSIONS:Herb-partitioned moxibustion combined with electroacupuncture can effectively improve the related clinical symptoms of PCOS patients with kidney deficiency and phlegm-dampness.Herb-partitioned moxibustion combined with electroacupuncture may inhibit the overexpression of AMH to increase the expression level of P450 arom in ovarian granulosa cells,thereby reconstructing the dependence of follicular development on FSH,and finally improving abnormal follicular development and hyperandrogenism in PCOS patients with kidney deficiency and phlegmdampness. | ZHAO Qingyi SUN Yi ZHOU Jing GAO Yanling MA Guizhi HU Zhihai WANG Yi SHI Yin | 2021 | Journal of Traditional Chinese Medicine2021,41,6: | 1 |
| 9 | Inhibitory effects of matrine on electrical signals and amino acid neurotransmitters in hippocampal brain slices显示文摘<正>BACKGROUND:Studies on electrical signals of hippocampal brain slices in vivo have shown that matrine inhibits benzylpenicillin sodium-induced activation of neuronal signal transduction. OBJECTIVE:To verify the inhibition effect of matrine on activation of electrical signals in rat brain slices and the role matrine plays in hippocampal amino acid transmitter release. DESIGN,TIME AND SEFFING:The in vitro,neurophysiological,controlled experiment was performed in the Zhejiang Province Key Laboratory of Cardio-cerebrovascular Disease and Nerve System Drugs Appraisement and Chinese Traditional Medicine Screening and Research between July 2003 and May 2004.The in vivo,neuronal,biochemical experiment was performed in the Zhejiang Province Key Laboratory of Chinese Traditional Medicine Quality Standardization from July 2005 to December 2006. MATERIALS:Forty healthy,Sprague Dawley rats,7-8 weeks old,and 120 healthy,ICR mice,5-6 weeks old,were included in this study,irrespective of gender.Matrine powder was provided by the National Institute for the Control of Pharmaceutical and Biological Products,China.Matrine injection was purchased from Zhuhai Biochemical Pharmaceutical Factory,China.Penicillin was bought from Shijiazhuang Pharmaceutical Group Co.,Ltd.,China. METHODS:(1) Rats were randomly assigned to four groups:control,penicillin model,and matrine high-dose and low-dose,with 10 rats in each group.The control group was perfused with artificial cerebrospinal fluid.In the remaining three groups,hippocampal brain slices were perfused with normal artificial cerebrospinal fluid containing 1×10~6 U/L penicillin for the first 10 minutes.The penicillin model group received artificial cerebrospinal fluid for an additional 30 minutes,while the matrine high-dose and low-dose groups received 0.1 g/L and 0.05 g/L matrine,respectively,for an additional 30 minutes.(2) Mice were randomly assigned to four groups(n=30).The matrine high-, medium-,and low-dose groups were separately injected with 58.5,39.0,and 19.5 mg/kg matrine via caudal vein,respectively.No intervention was administered to the normal group. MAIN OUTCOME MEASURES:The field potential value in the CA1 region of penicillin-induced rat hippocampal brain slices was analyzed using the evoked field potential technique;chromatography was utilized to determine y-aminobutyric acid and glutamic acid content in the mouse hippocampus. RESULTS:(1) Both 0.1 g/L and 0.05 g/L matrine reduced the number of evoked field potentials in the penicillin-induced rat hippocampal brain slices(P<0.05 or P<0.01).In addition,0.1 g/L matrine led to a reduction of evoked field potential amplitude(P<0.05).(2) Compared with normal mice, y-aminobutyric levels were dramatically increased at 20 minutes after high-dose matrine treatment (P<0.05).In addition,significantly increased y-aminobutyric acid levels were observed at 40 minutes after medium-and low-dose matrine treatments(P<0.05 or P<0.01).The glutamic acid/y-aminobutyric acid ratio was significantly less at 20 minutes after high-dose matrine treatment compared with the normal mice group(P<0.05). | 14 | Interictal Electrophysiological Source Imaging Based on Realistic Epilepsy Head Model in Presurgical Evaluation:A Prospective Study显示文摘Invasive techniques are becoming increasingly important in the presurgical evaluation of epilepsy.Adopting the electrophysiological source imaging(ESI)of interictal scalp electroencephalography(EEG)to localize the epileptogenic zone remains a challenge.The accuracy of the preoperative localization of the epileptogenic zone is key to curing epilepsy.The T1 MRI and the boundary element method were used to build the realistic head model.To solve the inverse problem,the distributed inverse solution and equivalent current dipole(ECD)methods were employed to locate the epileptogenic zone.Furthermore,a combination of inverse solution algorithms and Granger causality connectivity measures was evaluated.The ECD method exhibited excellent focalization in lateralization and localization,achieving a coincidence rate of 99.02%(p<0.05)with the stereo electroencephalogram.The combination of ECD and the directed transfer function led to excellent matching between the information flow obtained from intracranial and scalp EEG recordings.The ECD inverse solution method showed the highest performance and could extract the discharge information at the cortex level from noninvasive low-density EEG data.Thus,the accurate preoperative localization of the epileptogenic zone could reduce the number of intracranial electrode implantations required. | Ruowei Qu Zhaonan Wang Shifeng Wang Le Wang Alan Wang Guizhi Xu | 2023 | Chinese Journal of Electrical Engineering2023,9,1: | 1 |

| 18 | Assessing the Forecasting of Comprehensive Loss Incurred by Typhoons:A Combined PCA and BP Neural Network Model显示文摘This paper develops a joint model utilizing the principal component analysis(PCA)and the back propagation(BP)neural network model optimized by the Levenberg Marquardt(LM)algorithm,and as an application of the joint model to investigate the damages caused by typhoons for a coastal province,Fujian Province,China in 2005-2015(latest).First,the PCA is applied to analyze comprehensively the relationship between hazard factors,hazard bearing factors and disaster factors.Then five integrated indices,overall disaster level,typhoon intensity,damaged condition of houses,medical rescue and self-rescue capability,are extracted through the PCA;Finally,the BP neural network model,which takes the principal component scores as input and is optimized by the LM algorithm,is implemented to forecast the comprehensive loss of typhoons.It is estimated that an average annual loss of 138.514 billion RMB occurred for 2005-2015,with a maximum loss of 215.582 in 2006 and a decreasing trend since 2010 though the typhoon intensity increases.The model was validated using three typhoon events and it is found that the error is less than 1%.These results provide information for the government to increase medical institutions and medical workers and for the communities to promote residents’self-rescue capability. | Shuai Yuan Guizhi Wang Jibo Chen Wei Guo | 2019 | Journal on Artificial Intelligence2019,1,2: | 1 |
