| 1 | Laparoscopic Partial Nephrectomy With Segmental Renal Artery Clamping:Technique and Clinical Outcomes显示文摘Background:Warm ischemic injury is one of the most important factors affecting renal function in partial nephrectomy(PN).The technique of segmental renal artery clamping emerges as an alternative to conventional renal artery clamping for renal hilar control.Objective:To evaluate the feasibility and efficiency of laparoscopic PN(LPN) with segmental renal artery clamping in comparison with the conventional technique.Design,setting,and participants:A total of 75 patients underwent LPN from June 2007 to November 2009.All patients had T1a or T1b tumor in one kidney and a normal contralateral kidney.Thirty-seven patients underwent surgeries with main renal artery clamping,and 38 underwent surgeries with segmental artery clamping.Intervention:All procedures were performed by the same laparoscopic surgeon.Measurements:Blood loss,operation time,warm ischemia(WI) time,and complications affected renal function before and after operation were recorded.Results and limitations:All LPNs were completed without conversion to open surgery or nephrectomy.The novel technique slightly increased WI time(p < 0.001) and intraoperative blood loss(p = 0.006),while it provided better postoperative affected renal function(p < 0.001) compared with the conventional technique.The total complication rate was 12%.Among the 38 cases where segmental renal artery clamping was performed,7 had to convert to the conventional method.Tumor size and location influenced the number of clamped segmental arteries.Long-term postoperative renal function is still awaited.Conclusions:LPN with segmental artery clamping is safe and feasible in clinical practice.It minimizes the intraoperative WI injury and improves early postoperative affected renal function compared with main renal artery clamping.(C) 2010 European Association of Urology.Published by Elsevier B.V.All rights reserved. | Shao,P.F. | 4 | 阿尔茨海默病神经细胞凋亡与小胶质细胞活化的关系显示文摘目的 探讨阿尔茨海默病 (Alzheimer′sdisease ,AD)神经细胞凋亡与小胶质细胞活化的关系及活化的小胶质细胞在AD老年斑形成和进展中的作用。方法 收集 10例AD尸检材料及 4例非神经系统疾病死亡的老年尸检材料作为对照 ,进行白细胞介素 1α(IL 1α)免疫组织化学和原位DNA末端转移酶技术 (TUNEL)双重标记及淀粉样 β蛋白免疫组织化学和TUNEL双重标记。 结果 AD组静止的小胶质细胞数与对照组差异无显著性 (P >0 0 5 ) ,但活化的小胶质细胞数明显比对照组高 (P <0 0 1)。活化的小胶质细胞增大、变形 ,可进一步分为初级活化的小胶质细胞、增大的小胶质细胞及巨噬细胞 3种亚型。上述 3种活化的小胶质细胞数分别为 (5 4 0± 0 87)、(11 5 0± 1 2 5 )、(3 4 0± 0 32 )个 /mm2 ,占所有活化的小胶质细胞的比例分别为 2 6 6 0 % ,5 6 6 5 % ,16 75 %。AD组TUNEL阳性的凋亡的神经细胞数明显比对照组高 (P <0 0 5 ) ,且凋亡的神经细胞与活化的小胶质细胞之间具有相关性 (P <0 0 1)。活化的小胶质细胞在 4种类型老年斑中的分布比例不同 ,许多初级活化的小胶质细胞 (4 2 3% ) ,大多数增大的小胶质细胞和巨噬细胞 (分别是 5 6 2 %、70 6 % )分布在弥漫性神经突斑中。结论 小胶质细胞增生。 | 刘冬戈 何淑蓉 张伟 崔娣 Yue kui Li W. | 5 | TiZr-base Bulk Metallic Glass with over 50 mm in Diameter显示文摘Low-cost TiZr-base bulk metallic glasses(BMGs)(Ti36.1Zr33.2Ni5.8Be24.9)100-xCux(x=5,7 and 9) with a maximum size of over 50 mm in diameter were developed by optimizing the alloy composition.The idea is initiated by selecting a particular microstructure comprising primary β-Ti dendrite and amorphous phase.Afterwards,based on this composition of amorphous phase,a class of TiZr-base bulk metallic glasses was designed step by step to reach the optimum composition range.The glass transition temperature(Tg),initial crystallization temperature(Tx) and width of supercooled region(△T) of(Ti36.1Zr33.2Ni5.8Be24.9)91Cu9 BMG are 611,655 and 44 K,respectively.The(Ti36.1Zr33.2Ni5.8Be24.9)91Cu9 BMG exhibits low density of 5.541 g·cm-3 and high compressive fracture strength of 1800 MPa,which promises the potential application as structural materials. | M. | 9 | Insect Cell Culture and Biotechnology显示文摘The continued development of new cell culture technology is essential for the future growth and application of insect cell and baculovirus biotechnology. The use of cell lines for academic research and for commercial applications is currently dominated by two cell lines; the Spodoptera frugiperda line, SF21 (and its clonal isolate, SF9), and the Trichoplusia ni line, BTI 5B1-4, commercially known as High Five cells. The long perceived prediction that the immense potential application of the baculovirus-insect cell system, as a tool in cell and molecular biology, agriculture, and animal health, has been achieved. The versatility and recent applications of this popular expression system has been demonstrated by both academia and industry and it is clear that this cell-based system has been widely accepted for biotechnological applications. Numerous small to midsize startup biotechnology companies in North America and the Europe are currently using the baculovirus-insect cell technology to produce custom recombinant proteins for research and commercial applications. The recent breakthroughs using the baculovirus-insect cell-based system for the development of several commercial products that will impact animal and human health will further enhance interest in this technology by pharma. | 10 | Quantitative study of tectonic geomorphology along Haiyuan fault based on airborne LiDAR显示文摘High-precision and high-resolution topography are the fundamental data for active fault research.Light detection and ranging(LiDAR)presents a new approach to build detailed digital elevation models effectively.We take the Haiyuan fault in Gansu Province as an example of how LiDAR data may be used to improve the study of active faults and the risk assessment of related hazards.In the eastern segment of the Haiyuan fault,the Shaomayin site has been comprehensively investigated in previous research because of its exemplary tectonic topographic features.Based on unprecedented LiDAR data,the horizontal and vertical coseismic offsets at the Shaomayin site are described.The measured horizontal value is about 8.6 m,and the vertical value is about 0.8 m.Using prior dating ages sampled from the same location,we estimate the horizontal slip rate as4.0±1.0 mm/a with high confidence and define that the lower bound of the vertical slip rate is 0.4±0.1 mm/a since the Holocene.LiDAR data can repeat the measurements of field work on quantifying offsets of tectonic landform features quite well.The offset landforms are visualized on an office computer workstation easily,and specialized software may be used to obtain displacement quantitatively.By combining precious chronological results,the fundamentallink between fault activity and large earthquakes is better recognized,as well as the potential risk for future earthquake hazards. | Tao Chen Pei Zhen Zhang Jing Liu Chuan You Li Zhi Kun Ren Kenneth W. | 18 | BubR1 phosphorylates CENP-E as a switch enabling the transition from lateral association to end-on capture of spindle microtubules显示文摘Error-free mitosis depends on accurate chromosome attachment to spindle microtubules, powered congression of those chromosomes, their segregation in anaphase, and assembly of a spindle midzone at mitotic exit. The centromere-associated kinesin motor CENP-E, whose binding partner is BubR1, has been implicated in congression of misaligned chromosomes and the transition from lateral kinetochore-microtubule association to end-on capture. Although previously proposed to be a pseudokinase, here we report the structure of the kinase domain of Drosophila melanogaster BubR1, revealing its folding into a conformation predicted to be catalytically active. BubR1 is shown to be a bona fide kinase whose phosphorylation of CENP-E switches it from a laterally attached microtubule motor to a plus-end microtubule tip tracker. Computational modeling is used to identify bubristatin as a selective BubR1 kinase antagonist that targets the αN1 helix of N-terminal extension and αC helix of the BubR1 kinase domain. Inhibition of CENP-E phosphorylation is shown to prevent proper microtubule capture at kinetochores and, surprisingly, proper assembly of the central spindle at mitotic exit.
