| 3 | Role of autophagy in differential sensitivity of hepatocarcinoma cells to sorafenib显示文摘AIM: To investigate the role of sorafenib(SFN) in autophagy of hepatocellular carcinoma(HCC). We evaluated how SFN affects autophagy signaling pathway in human HCC cell lines. METHODS: Two different human HCC cell lines, Hep3 B and Huh7, were subjected to different concentrations of SFN. Cell viability and onset of apoptosis were determined with colorimetric assay and immunoblotting analysis, respectively. The changes in autophagy-related proteins, including LC3, ULK1, AMPK, and LKB, were determined with immunoblotting analysis in the presence or absence of SFN. To assess autophagic dynamics, autophagic flux was measured with chloroquine, a lysosomal inhibitor. The autophagic responsiveness between different HCC cell lines was compared under the autophagy enhancing conditions.RESULTS: Hep3 B cells were significantly more resistant to SFN than Huh7 cells. Immunoblotting analysis revealed a marked increase in SFN-mediated autophagy flux in Huh7 cells, which was, however, absent in Hep3 B cells. While both starvation and rapamycin enhanced autophagy in Huh7 cells, only rapamycin increased autophagy in Hep3 B cells. Immunoblotting analysis of autophagy initiation proteins showed that SFN substantially increased phosphorylation of AMPK and consequently autophagy in Huh7, but not in Hep3 B cells.CONCLUSION: The autophagic responsiveness to SFN is distinct between Hep3 B and Huh7 cells. | 5 | 用传统和超声提取的松果菊(紫锥菊属)的抗氧化和抗菌活性(英文)显示文摘Antioxidant and antimicrobial activities of Echinacea purpurea L. (Asteraceae) extracts obtained by classical and ultrasound solvent extraction were compared. The dry aerial part of plant was extracted by 70% etha-nol at a solid-to-liquid ratio of 1︰10 (m/v) and 25°C. The extract obtained by classical solvent extraction contained 29% larger amounts of phenolic compounds and 20% higher content of flavonoids. 2,2-diphenyl-1-picril hydrazyl radical (DPPH) scavenging reached 93.6% and the values of EC50 were (34.16±0.65) μg·ml-1 and (65.48±1.12) μg·ml-1 for the extracts obtained by the classical and ultrasound extractions, respectively. The extracts, independent of the extraction technique applied, showed a considerable growth inhibition on Candida albicans and Saccharo-myces cerevisiae, while no growth inhibition zones were observed for Aspergillus niger. | 6 | A new kinetic model for the common juniper essenstial oil extraction by microwave hydrodistillation显示文摘The main objective of the present study was to model the kinetics of essential oil extraction from swelled ground juniper berries by classic hydrodistillation(HD) and microwave-assisted hydrodistillation(MAHD). A new phenomenological kinetic model was developed on the basis of the juniper essential oil extraction mechanism that assumed three mass transfer processes occurring simultaneously: washing, unhindered diffusion and hindered diffusion. The new model was compared to the existing kinetic models. Among the tested models,the new model had the smallest mean relative percentage deviation and the highest corrected Akaike information criterion value. In addition, that, the new model was verified for HD and MAHD of essential oils from some other plant materials.
