| 1 | Simultaneous determination of telmisartan and amlodipine in human plasma by LC-MS/MS and its application in a human pharmacokinetic study显示文摘A rapid and sensitive liquid chromatography-tandem mass spectrometric(LC-MS/MS) assay method has been developed and fully validated for the simultaneous quantification of telmisartan and amlodipine in human plasma.Carbamazepine was used as an internal standard.Analytes and the internal standard were extracted from human plasma by solid-phase extraction technique using Waters Oasis HLB 1 cm 3(30 mg) extraction cartridge.The reconstituted samples were chromatographed on a Hypurity advance C 18 column(50 mm×4.6 mm,5 μm) using a mixture of acetonitrile-5 mM ammonium acetate buffer(pH-4.0)(50:50,v/v) as the mobile phase at a flow rate of 0.8 mL/min.The calibration curve obtained was linear(r≥0.99) over the concentration range of 2.01-400.06 ng/mL for telmisartan and 0.05-10.01 ng/mL for amlodipine.Method validation was performed as per FDA guidelines and the results met the acceptance criteria.A run time of 2.5 min for each sample made it possible to analyze more than 400 human plasma samples per day.The proposed method was found to be applicable to clinical studies. | Vasu Babu Ravi Jaswanth Kumar Inamadugu Nageswara Rao Pilli Vudagandla Sreenivasulu Venkateswarlu Ponneri | 2012 | Journal of Pharmaceutical Analysis2012，2，5： | 8 |
| 2 | Effect of fusion welding processes on tensile properties of armor grade, high thickness, non-heat treatable aluminium alloy joints显示文摘AA5059 is one of the high strength armor grade aluminium alloy that finds its applications in the military vehicles due to the higher resistance against the armor piercing (AP) threats. This study aimed at finding the best suitable process among the fusion welding processes such as gas tungsten arc welding (GTAW) and gas metal arc welding (GMAW) by evaluating the tensile properties of AA5059 aluminium alloy joints. The fracture path was identified by mapping the low hardness distribution profile (LHDP) across the weld cross section under tensile loading. Optical and scanning electron microscopies were used to characterize the microstructural features of the welded joints at various zones. It is evident from the results that GTAW joints showed superior tensile properties compared to GMAW joints and this is primarily owing to the presence of finer grains in the weld metal zone (WMZ) and narrow heat-affected zone (HAZ). The lower heat input associated with the GTAW process effectively reduced the size of the WMZ and HAZ compared to GMAW process. Lower heat input of GTAW process results in faster cooling rate which hinders the grain growth and reduces the evaporation of magnesium in weld metal compared to GMAW joints. The fracture surface of GTAW joint consists of more dimples than GMAW joints which is an indication that the GTAW joint possess improved ductility than GMAW joint. | K. Vasu H. | 3 | A rapid and sensitive liquid chromatography-tandem mass spectrometric assay for duloxetine in human plasma:Its pharmacokinetic application显示文摘This paper describes a simple, rapid and sensitive liquid chromatography-tandem mass spectrometry assay for the determination of duloxetine in human plasma. A duloxetine stable labeled isotope (duloxetine d 5 ) was used as an internal standard. Analyte and the internal standard were extracted from 100 mL of human plasma via solid phase extraction technique using Oasis HLB cartridges. The chromatographic separation was achieved on a C 18 column by using a mixture of acetonitrile-5 mM ammonium acetate buffer (83:17, v/v) as the mobile phase at a flow rate of 0.9 mL/min. The calibration curve obtained was linear (r 2 Z0.99) over the concentration range of 0.05-101 ng/mL. Multiple-reaction monitoring mode (MRM) was used for quantification of ion transitions at m/z 298.3/154.1 and 303.3/159.1 for the drug and the internal standard, respectively. Method validation was performed as per FDA guidelines and the results met the acceptance criteria. A run time of 2.5 min for each sample made it possible to analyze more than 300 plasma samples per day.
