| 1 | E2FBP1 antagonizes the p16^(INK4A)-Rb tumor suppressor machinery for growth suppression and cellular senescence by regulating promyelocytic leukemia protein stability显示文摘Cellular senescence is an irreversible cell cycle arrest triggered by the activation of oncogenes or mitogenic signaling as well as the enforced expression of tumor suppressors such as p53, p16 INK4A and promyelocytic leukemia protein (PML) in normal cells. E2F-binding protein 1 (E2FBP1), a transcription regulator for E2F, induces PML reduction and suppresses the formation of PML-nuclear bodies, whereas the down-regulation of E2FBP1 provokes the PML-dependent premature senescence in human normal fibroblasts. Here we report that the depletion of E2FBP1 induces the accumulation of PML through the Ras-dependent activation of MAP kinase signaling. The cellular levels of p16 INK4A and p53 are elevated during premature senescence induced by depletion of E2FBP1, and the depletion of p16 INK4A , but not p53 rescued senescent cells from growth arrest. Therefore, the premature senescence induced by E2FBP1 depletion is achieved through the p16 INK4A -Rb pathway. Similar to human normal fibroblasts, the growth inhibition induced by E2FBP1 depletion is also observed in human tumor cells with intact p16 INK4A and Rb. | 2 | Ever-increasing diversity of drug-induced pancreatitis显示文摘With over 100000 hospital admissions per annum,acute pancreatitis remains the leading gastrointestinal cause of hospitalization in the United States and has farreaching impact well beyond.It has become increasingly recognized that druginduced pancreatitis(DIP),despite accounting for less than 3%of all cases,represents an important and growing though often inconspicuous cause of acute pancreatitis.Nevertheless,knowledge of DIP is often curtailed by the limited availability of evidence needed to implicate given agents,especially for nonprescription medications.Indeed,the majority of available data is derived from case reports,case series,or case control studies.Furthermore,the mechanism of injury and causality for many of these drugs remain elusive as a definitive correlation is generally not established(<10%of cases).Several classification systems have been proposed,but no single system has been widely adopted,and periodic updates are required in light of ongoing pharmacologic expansion.Moreover,infrequently prescribed medications or those available over-thecounter(including herbal and other alternative remedies)are often overlooked as a potential culprit of acute pancreatitis.Herein,we review the ever-increasing diversity of DIP and the potential mechanisms of injury with the goal of raising awareness regarding the nature and magnitude of this entity.We believe this manuscript will aid in increasing both primary and secondary prevention of DIP,thus ultimately facilitating more expedient diagnosis and a decrease in DIPrelated morbidity. | Simcha Weissman Muhammad Aziz Ryan B Perumpail Tej I Mehta Rutwik Patel James H Tabibian | 2020 | World Journal of Gastroenterology2020,26,22: | 8 |
| 3 | Expression profile of cholecystokinin type-A receptor in gallbladder cancer and gallstone disease显示文摘BACKGROUND:Regulatory peptide receptors have attracted the interest of oncologists as a new promising approach for cancer pathology,imaging and therapy.Although cholecystokinin (CCK) is a potent modulator of gallbladder contractility and plays a potential role in pancreatic carcinogenesis through CCK type-A receptor (CCKAR),its role in gallbladder cancer (GBC) is still unknown and immunohistochemical detection of CCKAR in the gallbladder has not yet been reported.This novel case-control study aimed to investigate the expression profile of CCKAR in GBC and gallstone disease (GSD).METHODS:This study included 162 samples of gallbladder:94 from GBC and 68 from GSD.Expression of CCKAR was analyzed by immunohistochemistry and immunoblotting.The results were statistically correlated with disease history including age,sex,presence of gallstone,stage and differentiation.RESULTS:CCKAR was positive in 30/68 (44.1%) of GSD and 72/94 (76.6%) of GBC samples.Fifty-one of the 72 (70.8%) CCKAR-positive GBC samples showed over-expression.Interestingly,consistent results also appeared in the immunoblotting study.CONCLUSIONS:CCKAR expression was significantly increased in GBC compared to GSD.Moreover,CCKAR expression was associated with the degree of tumor differentiation,i.e.,less expression in poorly-differentiated tumors.Thus,it has future prognostic and therapeutic implications in the management of GBC. | Rajani Rai Mallika Tewari Mohan Kumar Tej Bali Singh Hari S Shukla | 2011 | Hepatobiliary & Pancreatic Diseases International2011,10,4: | 7 |
| 4 | New spinal robotic technologies显示文摘Robotic systems in surgery have developed rapidly. Installations of the da Vinci Surgical System® (Intuitive Surgical, Sunnyvale, CA,, USA), widely used in urological and gynecological procedures, have nearly doubled in the United States from 2010 to 2017. Robotics systems in spine surgery have been adopted more slowly;however, users are enthusiastic about their applications in this subspecialty. Spinal surgery often requires fine manipulation of vital structures that must be accessed via limited surgical corridors and can require repetitive tasks over lengthy periods of time — issues for which robotic assistance is well-positioned to complement human ability. To date, the United States Food and Drug Administration (FDA) has approved 7 robotic systems across 4 companies for use in spinal surgery. The available clinical data evaluating their efficacy have generally demonstrated these systems to be accurate and safe. A critical next step in the broader adoption of surgical robotics in spine surgery is the design and implementation of rigorous comparative studies to interrogate the utility of robotic assistance. | 6 | Effect of disturbance on biomass,production and carbon dynamics in moist tropical forest of eastern Nepal显示文摘Background: Forest biomass is helpful to assess its productivity and carbon(C) sequestration capacity. Several disturbance activities in tropical forests have reduced the biomass and net primary production(NPP) leading to climate change. Therefore, an accurate estimation of forest biomass and C cycling in context of disturbances is required for implementing REDD(Reducing Emissions from Deforestation and Forest Degradation) policy.Methods: Biomass and NPP of trees and shrubs were estimated by using allometric equations while herbaceous biomass was estimated by harvest method. Fine root biomass was determined from soil monolith. The C stock in vegetation was calculated by multiplying C concentration to dry weight.Results: Total stand biomass(Mg?ha^(-1)) in undisturbed forest stand(US) was 960.4 while in disturbed forest stand(DS) it was 449.1. The biomass(Mg?ha^(-1)) of trees, shrubs and herbs in US was 948.0, 4.4 and 1.4,respectively, while in DS they were 438.4, 6.1 and 1.2, respectively. Total NPP(Mg?ha^(-1)?yr^(-1)) was 26.58(equivalent to 12.26 Mg C?ha^(-1)?yr^(-1)) in US and 14.91(6.88 Mg C?ha^(-1)?yr^(-1)) in DS. Total C input into soil through litter plus root turnover was 6.78 and 3.35 Mg?ha^(-1)?yr^(-1)in US and DS, respectively.Conclusions: Several disturbance activities resulted in the significant loss in stand biomass(53 %), NPP(44 %), and C sequestration capacity of tropical forest in eastern Nepal. The net uptake of carbon by the vegetation is far greater than that returned to the soil by the turnover of fine root and litter. Therefore, both stands of present forest act as carbon accumulating systems.
