1Hepatocellular Carcinoma-Cause,Treatment and Metastasis显示文摘In the recent decades, the incidence of hepatocellular carcinoma (HCC) has been found to be increasing in males in some countries. In China, HCC ranked second of cancer mortality since 1990s. Hepatitis B and C viruses (HBV and HCV) and dietary aflatoxin intake remain the major causative factors of HCC. Surgery plays a major role in the treatment of HCC, particularly for small HCC. Downstaging unresectable huge HCC to smaller HCC and followed by resection will probably be a new approach for further study. Liver transplantation is indicated for small HCC, however, some issues remain to be solved.Different modes of 'regional cancer therapy for HCC' have been tried. Systemic chemotherapy has been disappointing in the past but the future can be promising.Biotherapy, such as cytokines, differentiation inducers,anti-angiogenic agents, gene therapy and tumor vaccine will probably play a role, particularly in the prevention of tumor recurrence. HCC invasiveness is currently the major target of study. 2Establishment of cell clones with different metastatic potential from the metastatic hepatocellular carcinoma cell line MHCC97显示文摘ALM To establish clone cells with different metastatic potential for the study of metastasis-related mechanisms. METHODS Cloning procedure was performed on parental hepatocellular carcinoma (HCC) cell line MHCC97. andbiological characteristics of the target clones selected by in vivo screening were studied.``RESULTS Two clones with high MHCC97-H and IowMHCC9--L1 metastatic potential were isolated from theparent cell line. Compared with MHCC97-L. MHCC97-H hadsmaller cell size average cell diameter 43 um vs 50 μmand faster in vitro and in vivo growth rate tumor celldoubling time was 34.2 h vs 60.0 h. The main ranges ofchromosomes were 5.5 58 in MHCC97-H and 57 62 inMHCC97-L. 3Role of mantle-derived magma in genesis of early Yanshanian granites in the Nanling Range, South China: in situ zircon Hf-O isotopic constraints显示文摘Although a number of petrographic observations and isotopic data suggest that magma mixing is common in genesis of many granite plutons, it is still controversial whether the mantle-derived magmas were involved in granites. We carried out in this study a systematic analysis of in situ zircon Hf-O isotopes for three early Yanshanian intrusions dated at ca. 160 Ma from the Nanling Range of Southeast China. The Qinghu monzonite has very homogeneous zircon Hf-O isotopic compositions, εHf(t) =11.6±0.3 and δ18O=5.4‰±0.3‰. In combination with whole-rock geochemical and Sr-Nd isotopic data, the parental magma of the Qinghu monzonite were likely derived from the partial melting of recently-metasomatized, phlogopite-bearing lithospheric mantle without appreciable crustal contamination. The Lisong and Fogang granites and the mafic microgranular enclaves (MME) within the Lisong granites have a wide range of zircon Hf-O isotopic compositions, with Hf and O isotopes being negatively correlated within each pluton. The Lisong MMEs were crystallized from a mantle-derived magma, similar to the parental magma of the Qinghu monzonite, with small amount of crustal assimilation. The Lisong and Fogang granites were formed by reworking of meta-sedimentary materials by mantle-derived magmas and mixing of the mantle-and sediment-derived melts to varying degrees. 10Modeling study of regional severe hazes over mid-eastern China in January 2013 and its implications on pollution prevention and control显示文摘The Nested Air Quality Prediction Model System(NAQPMS)was used to investigate the temporal and spatial variations of PM2.5over tropospheric central eastern China in January 2013.The impact of regional transport and its implications on pollution prevention and control were also examined.Comparison between simulated and observed PM2.5showed NAQPMS was able to reproduce the evolution of PM2.5during heavy haze episodes.The results indicated that regional transport of PM2.5played an important role in regional haze episodes in the city cluster including Hebei,Beijing and Tianjin(HBT).The cross-city clusters transport outside HBT and transport among cities inside HBT contributed 20%–35%and 26%–35%of PM2.5as compared with local emission,in HBT respectively.To meet the Air Quality Standards for Grade II,90%,90%and65%of emissions would have to be cut down in Hebei,Tianjin and Beijing,if non-control strategy was taken in the surrounding city clusters of HBT.This implicated that control of emissions in one city cluster is not sufficient to reduce regional haze events,and joint efforts among city clusters are essential.Besides regional transports,two-way feedback between boundary-layer evolution and PM2.5also significantly contributed to the formation of heavy hazes,which contributed 30%of monthly average PM2.5concentration in HBT.WANG ZiFa LI Jie WANG Zhe YANG WenYi TANG Xiao GE BaoZhu YAN PinZhong ZHU LiLi CHEN XueShun CHEN HuanSheng WAND Wei LI JianJun LIU Bing WANG XiaoYan WAND Wei ZHAO YiLin LU Ning SU DeBin 2014Science China Earth Sciences2014,57,1:106
11Mechanism for the formation of the January 2013 heavy haze pollution episode over central and eastern China显示文摘In January 2013,a long-lasting episode of severe haze occurred in central and eastern China,and it attracted attention from all sectors of society.The process and evolution of haze pollution episodes were observed by the'Forming Mechanism and Control Strategies of Haze in China'group using an intensive aerosol and trace gases campaign that simultaneously obtained data at 11 ground-based observing sites in the CARE-China network.The characteristics and formation mechanism of haze pollution episodes were discussed.Five haze pollution episodes were identified in the Beijing-Tianjin-Hebei(Jing-Jin-Ji)area;the two most severe episodes occurred during 9–15 January and 25–31 January.During these two haze pollution episodes,the maximum hourly PM2.5mass concentrations in Beijing were 680 and 530μg m 3,respectively.The process and evolution of haze pollution episodes in other major cities in the Jing-Jin-Ji area,such as Shijiazhuang and Tianjin were almost the same as those observed in Beijing.The external cause of the severe haze episodes was the unusual atmospheric circulation,the depression of strong cold air activities and the very unfavorable dispersion due to geographical and meteorological conditions.However,the internal cause was the quick secondary transformation of primary gaseous pollutants to secondary aerosols,which contributed to the'explosive growth'and'sustained growth'of PM2.5.Particularly,the abnormally high amount of nitric oxide(NOx)in the haze episodes,produced by fossil fuel combustion and vehicle emissions,played a direct or indirect role in the quick secondary transformation of coal-burning sulphur dioxide(SO2)to sulphate aerosols.Furthermore,gaseous pollutants were transformed into secondary aerosols through heterogeneous reactions on the surface of fine particles,which can change the particle’s size and chemical composition.Consequently,the proportion of secondary inorganic ions,such as sulphate and nitrate,gradually increased,which enhances particle hygroscopicity and thereby accelerating formation of the haze pollution.WANG YueSi YAO Li WANG LiLi LIU ZiRui JI DongSheng TANG GuiQian ZHANG JunKe SUN Yang HU Bo XIN JinYuan 2014Science China Earth Sciences2014,57,1:189
12The prognostic molecular markers in hepatocellular carcinoma显示文摘The prognosis of hepatocellular carcinoma (HCC) stillremains dismal, although many advances in its clinicalstudy have been made. It is important for tumor control toidentity the factors that predispose patients to death. Withnew discoveries in cancer biology, the pathological andbiological prognostic factors of HCC have been studied quiteextensively. Analyzing molecular markers (biomarkers) withprognostic significance is a complementary method. A largenumber of molecular factors have been shown to associatewith the invasiveness of HCC, and have potential prognosticsignificance. One important aspect is the analysis ofmolecular markers for the cellular malignancy phenotypeThese include alterations in DNA ploidy, cellularproliferation markers (PCNA, Ki-67, Mcm2, MIB1, MIA, andCSE1L/CAS protein), nuclear morphology, the p53 geneand its related molecule MDM2, other cell cycle regulators(cyclin A, cyclin D, cyclin E, cdc2, p27, p73), oncogenesand their receptors (such as ras, c-myc, c-fms, HGF, c-met, and erb-B receptor family members ), apoptosisrelated factors (Fas and FasL), as well as telomeraseactivity. Another important aspect is the analysis ofmolecular markers involved in the process of cancerinvasion and metastasis. Adhesion molecules (E-cadherin,catenins, serum intercellular adhesion molecule-1, CD44variants), proteinases involved in the clegradation ofextracellular matrix (MMP-2, MMP-9, uPA, uPAR, PAl), aswell as other molecules have been regarded as biomarkersfor the malignant phenotype of HCC, and are related toprognosis and therapeutic outcomes. Tumor angiogenesisis critical to both the growth and metastasis of cancersincluding HCC, and has drawn much attention in recentyears. Many angiogenesis-related markers, such as vascularendothelial growth factor (VEGF), basic fibroblast growthfactor (bFGF), platelet-derived endothelial cell growth factor( PD-ECGF ), thrombospondin ( TSP ), angiogenin,pleiotrophin, and endostatin (ES) levels, as well asinratumor microvessel density (MVD) have been evaluatedand found to be of prognostic significance. Body fluid(particularly blood and urinary) testing for biomarkers iseasily accessible and useful in clinical patients. Theprognostic significance of circulating DNA in plasma orserum, and its genetic alterations in HCC are otherimportant trends. More attention should be paid to thesetwo areas in future. As the progress of the human genomeproject advances, so does a clearer understanding of tumorbiology, and more and more new prognostic markers withhigh sensitivity and specificity will be found and used inclinical assays. 14Relationship between different surgical methods,hemorrhage position,hemorrhage volume,surgical timing,and treatment outcome of hypertensive intracerebral hemorrhage显示文摘BACKGROUND:The present study aimed to explore the relationship between surgical methods,hemorrhage position,hemorrhage volume,surgical timing and treatment outcome of hypertensive intracerebral hemorrhage(HICH).METHODS:A total of 1 310 patients,who had been admitted to six hospitals from January 2004 to January 2008,were divided into six groups according to different surgical methods:craniotomy through bone fl ap(group A),craniotomy through a small bone window(group B),stereotactic drilling drainage(group C1 and group C2),neuron-endoscopy operation(group D) and external ventricular drainage(group E) in consideration of hemorrhage position,hemorrhage volume and clinical practice. A retrospective analysis was made of surgical timing and curative effect of the surgical methods.RESULTS:The effectiveness rate of the methods was 74.12% for 1 310 patients after onemonth follow-up. In this series,the disability rate was 44.82% 3–6 months after the operation. Among the 1 310 patients,241(18.40%) patients died after the operation. If hematoma volume was >80 mL and the operation was performed within 3 hours,the mortality rate of group A was signifi cantly lower than that of groups B,C,D,and E(P<0.05). If hematoma volume was 50–80 mL and the operation was performed within 6–12 hours,the mortality rate of groups B and D was lower than that of groups A,C and E(P<0.05). If hematoma volume was 20–50 mL and the operation was performed within 6–24 hours,the mortality rate of group C was lower than that of groups A,B and D(P<0.05).CONCLUSIONS:Craniotomy through a bone f lap is suitable for patients with a large hematoma and hernia of the brain. Stereotactic drilling drainage is suggested for patients with hematoma volume less than 80 mL. 16Ferroptosis:molecular mechanisms and health implications显示文摘Cell death can be executed through different subroutines.Since the description of ferroptosis as an iron-dependent form of nonapoptotic cell death in 2012,there has been mounting interest in the process and function of ferroptosis.Ferroptosis can occur through two major pathways,the extrinsic or transporter-dependent pathway and the intrinsic or enzyme-regulated pathway.Ferroptosis is caused by a redox imbalance between the production of oxidants and antioxidants,which is driven by the abnormal expression and activity of multiple redox-active enzymes that produce or detoxify free radicals and lipid oxidation products.Accordingly,ferroptosis is precisely regulated at multiple levels,including epigenetic,transcriptional,posttranscriptional and posttranslational layers.The transcription factor NFE2L2 plays a central role in upregulating anti-ferroptotic defense,whereas selective autophagy may promote ferroptotic death.Here,we review current knowledge on the integrated molecular machinery of ferroptosis and describe how dysregulated ferroptosis is involved in cancer,neurodegeneration,tissue injury,inflammation,and infection.Daolin Tang Xin Chen Rui Kang Guido Kroemer 2021Cell Research2021,31,2:182

19A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 20Multimodality treatment in hepatocellular carcinoma patients with tumor thrombi in portal vein显示文摘AIM To compare the therapeutic effect andsignificances of multimodality treatment forhepatocellular carcinoma (HCC) with tumorthrombi in portal vein (PVTT).METHODS HCC patients (n = 147) with tumortrombi in the main portal vein or the first branchof portal vein were divided into four groups bythe several therapeutic methods. There wereconservative treatment group in 18 out ofpatients (group A); and hepatic artery ligation(HAL) and/or hepatic artery infusion (HAl)group in 18 patients (group B), in whompostoberative chemoembolization was doneperiodically; group of removal of HCC with PVTTin 79 (group C) and group of transcatheterhepatic arterial chemoembolization (TACE) orHAl and/or portal vein infusion (PVI) afteroperation in 32 (group D).RESULTS The median survival period was 12months in our series and the 1-, 3-, and 5-yearsurvival rates were 44.3%, 24.5% and 15.2%,respectively. The median survival times were 2,5, 12 and 16 months in group A, B, C and D,respectively. The 1-, 3- and 5-year survival rateswere 5.6%, 0% and 0% in group A; 22.2%,5.6% and 0% in group B; 53.9%, 26.9% and16.6% in group C; 79.3%, 38.9% and 26.8% ingroup D, respectively.
