1Curcumin suppresses gastric NF-κB activation and macromolecular leakage in Helicobacter pylori-infected rats显示文摘AIM:To investigate whether curcumin could attenuate nuclear factor(NF)-κB p65 expression and macromolecular leakage in the gastric mucosa of Helicobacter pylori(H.pylori)-infected rats.METHODS:Twenty-five male Sprague-Dawley rats were equally divided into five groups:control rats(Control),control rats supplemented with 600 mg/kg curcumin,H.pylori-infected rats(Hp),H.pylori-infected rats supplemented with 200 mg/kg curcumin(Hp + curIn H.pylori-infected groups,rats were inoculated with H.pylori suspension twice a day at an interval of 4 h for 3 d.Two weeks later,200 or 600 mg/kg curcumin was given once daily to curcuminsupplemented groups for 7 d.On the day of the experiment,macromolecular leakage in gastric mucosa was examined by intravital fluorescence microscopy.The stomach tissue was removed to examine NF-κB p65 expression in gastric epithelial cells by immunohistochemistry.RESULTS:The expression of NF-κB p65 in gastric epithelial cells and the macromolecular leakage from gastric mucosal microcirculation significantly increased in the Hp group compared with the Control group.The percentages of NF-κB p65 immunoreactive cells in Control and Hp groups were 10.72% ± 2.10% vs 16.02% ± 2.98%,P = 0.004,respectively.The percentages of macromolecular leakage in Control and Hp groups were 10.69% ± 1.43% vs 15.41% ± 2.83%,P = 0.001,respectively.Curcumin supplementation in Hp + cur-CONCLUSION:H.pylori-induced gastric inflammation in rats is associated with increased NF-κB activation and macromolecular leakage which can be reduced by curcumin supplementation.Kawiya Sintara Duangporn Thong-Ngam Suthiluk Patumraj Naruemon Klaikeaw Tanittha Chatsuwan 2010World Journal of Gastroenterology2010,16,32:28

3Anti-cancer and anti-angiogenic effects of curcumin and tetrahydrocurcumin on implanted hepatocellular carcinoma in nude mice显示文摘AIM: To determine the effect of tetrahydrocurcumin (THC) on tumor angiogenesis compared with curcumin (CUR) by using both in vitro and in vivo models of human hepatocellular carcinoma cell line (HepG2). METHODS: The 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyl-tetrazolium bromide (MTT) assay was used for testing the anti-proliferating activities of CUR and THC. In male BALB/c nude mice, 2 × 106 human HepG2 cells were inoculated onto a dorsal skin-fold chamber. One day after HepG2 inoculation, the experimental groups were fed oral daily with CUR or THC (300 mg/kg or 3000 mg/kg). On d 7, 14 and 21, the tumor microvasculature was observed using fluorescence videomicroscopy and capillary vascularity (CV) was measured. RESULTS: Pathological angiogenic features including microvascular dilatation, tortuosity, and hyper-permeability were observed. CUR and THC could attenuate these pathologic features. In HepG2-groups, the CV were significantly increased on d 7 (52.43%), 14 (69.17%), and 21 (74.08%), as compared to controls (33.04%,P < 0.001). Treatment with CUR and THC resulted in significant decrease in the CV (P < 0.005 and P < 0.001, respectively). In particular, the anti-angiogenic effects of CUR and THC were dose-dependent manner. However, the beneficial effect of THC treatment than CUR was observed, in particular, from the 21 d CV (44.96% and 52.86%, P < 0.05). CONCLUSION: THC expressed its anti-angiogenesis without any cytotoxic activities to HepG2 cells even at the highest doses. 4Delta-like ligand 4 in hepatocellular carcinoma intrinsically promotes tumour growth and suppresses hepatitis B virus replication显示文摘AIM To investigate the role of Delta-like ligand 4(DLL4) on tumour growth in hepatitis B virus(HBV)-associated hepatocellular carcinoma(HCC) in vivo.METHODS We suppressed DLL4 expression in an HBV expressing HCC cell line, HepG2.2.15 and analysed the growth ability of cells as subcutaneous tumours in nude mice. The expression of tumour angiogenesis regulators, VEGF-A and VEGF-R2 in tumour xenografts were examined by western blotting. The tumour proliferation and neovasculature were examined by immunohistochemistry. The viral replication and viral protein expression were measured by quantitative PCR and western blotting, respectively.RESULTS Eighteen days after implantation, tumour volume in mice implanted with sh DLL4 HepG2.2.15 was significantly smaller than in mice implanted with control HepG2.2.15(P < 0.0001). The levels of angiogenesis regulators, VEGF-A and VEGF-R2 were significantly decreased in implanted tumours with suppressed DLL4 compared with the control group(P < 0.001 and P < 0.05, respectively). Furthermore, the suppression of DLL4 expression in tumour cells reduced cell proliferation and the formation of new blood vessels in tumours. 5Curcumin prevents indomethacin-induced gastropathy in rats显示文摘AIM:To investigate the effects of curcumin on gastric microcirculation and inflammation in rats with indomethacin-induced gastric damage.METHODS:Male Sprague-Dawley rats were randomly divided into three groups.Group 1(control group,n =5)was fed with olive oil and 5%NaHCO3-(vehicle).Group 2[indomethacin(IMN)group,n=5]was fed with olive oil 30 min prior to indomethacin 150 mg/kg body weight(BW)dissolved in 5%NaHCO3-at time 0th and 4th h.Group 3(IMN+Cur group,n=4)was fed with curcumin 200 mg/kg BW dissolved in olive oil 0.5 mL,30 min prior to indomethacin at 0th and 4th h.Leukocyte-endothelium interactions at postcapillary venules were recorded after acridine orange injection.Blood samples were determined for intercellular adhesion molecule(ICAM)-1 and tumor necrosis factor(TNF)-αlevels using enzyme linked immunosorbent assay method.Finally,the stomach was removed for histopathological examination for gastric lesions and grading for neutrophil infiltration.RESULTS:In group 2,the leukocyte adherence in postcapillary venules was significantly increased compared to the control group(6.40±2.30 cells/frame vs 1.20±0.83 cells/frame,P=0.001).Pretreatment with curcumin caused leukocyte adherence to postcapillary venule to decline(3.00±0.81 cells/frame vs 6.40 ±2.30 cells/frame,P=0.027).The levels of ICAM-1 and TNF-αincreased significantly in the indomethacintreated group compared with the control group(1106.50 ±504.22 pg/mL vs 336.93±224.82 pg/mL,P=0.011 and 230.92±114.47 pg/mL vs 47.13±65.59 pg/mL,P =0.009 respectively).Pretreatment with curcumin significantly decreased the elevation of ICAM-1 and TNF-α levels compared to treatment with indomethacin alone(413.66±147.74 pg/mL vs 1106.50±504.22 pg/mL,P =0.019 and 58.27±67.74 pg/mL vs 230.92±114.47 pg/mL,P=0.013 respectively).The histological appearance of the stomach in the control group was normal.In the indomethacin-treated group,the stomachs showed a mild to moderate neutrophil infiltration score.Gastric lesions were erosive and ulcerative.In rats treated with indomethacin and curcumin,stomach histopathology improved and showed only a mild neutrophil infiltration score and fewer erosive lesions in the gastric mucosa.CONCLUSION:The results indicate that curcumin prevents indomethacin-induced gastropathy through the improvement of gastric microcirculation by attenuating the level of ICAM-1 and TNF-α.Duangporn Thong-Ngam Sakonwan Choochuai Suthiluk Patumraj Maneerat Chayanupatkul Naruemon Klaikeaw 2012World Journal of Gastroenterology2012,18,13:3
