1Single-cell transcriptomics reveals regulators underlying immune cell diversity and immune subtypes associated with prognosis in nasopharyngeal carcinoma显示文摘Nasopharyngeal carcinoma(NPC)is an aggressive malignancy with extremely skewed ethnic and geographic distributions.Increasing evidence indicates that targeting the tumor microenvironment(TME)represents a promising therapeutic approach in NPC,highlighting an urgent need to deepen the understanding of the complex NPC TME.Here,we generated single-cell transcriptome profiles for 7581 malignant cells and 40,285 immune cells from fifteen primary NPC tumors and one normal sample.We revealed malignant signatures capturing intratumoral transcriptional heterogeneity and predicting aggressiveness of malignant cells.Diverse immune cell subtypes were identified,including novel subtypes such as CLEC9A^+dendritic cells(DCs).We further revealed transcriptional regulators underlying immune cell diversity,and cell–cell interaction analyses highlighted promising immunotherapeutic targets in NPC.Moreover,we established the immune subtype-specific signatures,and demonstrated that the signatures of macrophages,plasmacytoid dendritic cells(pDCs),CLEC9A^+DCs,natural killer(NK)cells,and plasma cells were significantly associated with improved survival outcomes in NPC.Taken together,our findings represent a unique resource providing in-depth insights into the cellular heterogeneity of NPC TME and highlight potential biomarkers for anticancer treatment and risk stratification,laying a new foundation for precision therapies in NPC.Yu-Pei Chen Jian-Hua Yin Wen-Fei Li Han-Jie Li Dong-Ping Chen Cui-Juan Zhang Jia-Wei Lv Ya-Qin Wang Xiao-Min Li Jun-Yan Li Pan-Pan Zhang Ying-Qin Li Qing-Mei He Xiao-Jing Yang Yuan Lei Ling-Long Tang Guan-Qun Zhou Yan-Ping Mao Chen Wei Ke-Xu Xiong Hong-Bo Zhang Shi-Da Zhu Yong Hou Ying Sun Michael Dean Ido Amit Kui Wu Dong-Ming Kuang Gui-Bo Li Na Liu Jun Ma 2020Cell Research2020,30,11:19
2Status and development of high-power laser facilities at the NLHPLP显示文摘In this paper, we review the status of the multifunctional experimental platform at the National Laboratory of High Power Laser and Physics(NLHPLP). The platform, including the SG-II laser facility, SG-II 9th beam, SG-II upgrade(SG-II UP) facility, and SG-II 5 PW facility, is operational and available for interested scientists studying inertial confinement fusion(ICF) and a broad range of high-energy-density physics. These facilities can provide important experimental capabilities by combining different pulse widths of nanosecond, picosecond, and femtosecond scales. In addition, the SG-II UP facility, consisting of a single petawatt system and an eight-beam nanosecond system, is introduced including several laser technologies that have been developed to ensure the performance of the facility. 3Evolution of low-frequency noise passing through a spatial filter in a high power laser system显示文摘The theoretical model of spatial noise passing through a spatial filter is established in high power laser system under the small signal approximation. The transmission characteristic for a noise signal passing through spatial filters with different magnifications is analyzed by numerical simulation, according to the actual structure of the high power laser system. The results show that the spatial modulation period of low-frequency noise getting through the pinhole will be proportional to the magnification of the spatial filter. When the magnification is less than 1, the safe low-frequency noise will be extruded into the high-frequency region, which is the fast increasing part, and finally develops into the most dangerous part which can damage the optical devices. 9Interlayer-confined two-dimensional manganese oxide-carbon nanotube catalytic ozonation membrane for efficient water purification显示文摘Catalytic ozonation technology has attracted copious attention in water purification owing to its favorable oxidative degradation of pollutants and mitigation of membrane fouling capacity.However,its extensive industrial application has been restricted by the low ozone utilization and limited mass transfer of the short-lived radical species.Interlayer space-confined catalysis has been theoretically proven to be a viable strategy for achieving high catalytic efficiency.Here,a two-dimensional MnO_(2)-incorporated ceramic membrane with tunable interspacing,which was obtained via the intercalation of a carbon nanotube,was designed as a catalytic ozonation membrane reactor for degrading methylene blue.Benefiting from the abundant catalytic active sites on the surface of two-dimensional MnO_(2) as well as the ultralow mass transfer resistance of fluids due to the nanolayer confinement,an excellent mineralization effect,i.e.,1.2 mg O_(3)(aq)mg^(-1) TOC removal(a total organic carbon removal rate of 71.5%),was achieved within a hydraulic retention time of 0.045 s of pollutant degradation.Further,the effects of hydraulic retention time and interlayer spacing on methylene blue removal were investigated.Moreover,the mechanism of the catalytic ozonation employing catalytic ozonation membrane was proposed based on the contribution of the Mn(III/IV)redox pair to electron transfer to generate the reactive oxygen species.This innovative twodimensional confinement catalytic ozonation membrane could act as a nanoreactor and separator to efficiently oxidize organic pollutants and enhance the control of membrane fouling during water purification.Dean Xu Tong Ding Yuqing Sun Shilong Li Wenheng Jing 2022Frontiers of Chemical Science and Engineering2022,16,5:1
