5应用BED-CEIA检测干血斑样本监测HIV-1发病率的可行性研究显示文摘目的研究干血斑样本应用于现场HIV-1BED—CEIA新近感染的检测以及监测HIV-1发病率的可行性。方法收集27个自愿咨询检测(VCT)哨点的10313名咨询者的血浆和干血斑样本,进行HIV抗体检测,对经免疫印迹方法确认的349例HIV感染者的血浆和干血斑样本同时进行BED—CEIA检测,检验干血斑样本BED—CEIA新近感染检测结果的稳定性和重复性,比较两种样本检测结果的差异及对HIV发病率估计的一致性。结果应用干血斑样本检测具有较好的稳定性和重复性,R^2值分别达到0.9551和0.95。349对样本的检测结果显示,294对样本被同时判定为长期感染,53对样本被同时判定为新近感染,两种样本对HIV-1是否为新近感染判断的一致性达到99.43%。而2对样本得到不同的结果,其An值均处于临界值附近。在人群总体水平上,两种样本计算得到的HIV-1发病率完全一致。结论在HIV-1BED—CEIA新近感染检测中,尽管个别样本检测结果存在差异,但在人群水平上十血斑样本的检测结果对HIV-1发病率的估计与血浆样本没有差异,因此在中国可以应用干血斑样本进行HIV-1BED-CEIA新近感染检测。沈圣 田飞 江华洲 孙国清 缪礼锋 郭晓黎 肖瑶 贾玉江 Sten H.Vermund 蒋岩 2009中华流行病学杂志2009,30,3:12

9Experimental and Numerical Investigation of Centrifugal Pumps with Asymmetric Inflow Conditions显示文摘Most of the times pumps operate off best point states.Reasons are changes of operating conditions,modifications,pollution and wearout or erosion.As consequences non-rotational symmetric flows,transient operational conditions,increased risk of cavitation,decrease of efficiency and unpredictable wearout can appear.Especially construction components of centrifugal pumps,in particular intake elbows,contribute to this matter.Intake elbows causes additional losses and secondary flows,hence non-rotational velocity distributions as intake profile to the centrifugal pump.As a result the impeller vanes experience permanent changes of the intake flow angle and with it transient flow conditions in the blade channels.This paper presents the first results of a project,experimentally and numerically investigating the consequences of non-rotational inflow to leading edge flow conditions of a centrifugal pump.Therefore two pumpintake-elbow systems are compared,by only altering the intake elbow geometry:a common single bended 90°elbow and a numerically optimized elbow(improved regarding rotational symmetric inflow conditions and friction coefficient).The experiments are carried out,using time resolved stereoscopic PIV on a full acrylic pump with refractions index matched(RIM)working fluid.This allows transient investigations of the flow field simultaneously for all blade leading edges.Additional CFD results are validated and used to further support the investigation i.e.for comparing an analog pump system with ideal inflow conditions.Sten Mittag Martin Gabi 2015Journal of Thermal Science2015,24,6:5

16Saccharomyces cerevisiae CNCM I-3856 in irritable bowel syndrome: An individual subject meta-analysis显示文摘AIM To confirm previous conclusions on Saccharomyces cerevisiae(S. cerevisiae) CNCM I-3856 for irritable bowel syndrome(IBS) management.METHODS An individual patient data meta-analysis was performed on two randomized clinical trials studying the effect of S. cerevisiae CNCM I-3856 supplementation on gastrointestinal(GI) symptoms in IBS subjects. A total of 579 IBS subjects were included. Outcomes were the daily Likert scale scores of abdominal pain/discomfort and bloating [area under the curve(AUC) and weekly means], responder status, and bowel movements(stool frequency and consistency). Statistical analyses were conducted in Intent to Treat(ITT) population, IBS-C subjects and IBS-C subjects with an abdominal pain/discomfort score higher than or equal to 2 at baseline('IBS-C ≥ 2 subpopulation').RESULTS S. cerevisiae CNCM I-3856 significantly improved abdominal pain/discomfort and bloating during the second month of supplementation [AUC(W5-W8)]with improvement up to the minimal clinically relevant threshold of 10%: a 12.3% reduction of abdominal pain/discomfort in the ITT population compared to the Placebo group(P = 0.0134) has been observed. In the IBS-C ≥ 2 subpopulation, there were a 13.1% reduction of abdominal pain/discomfort and a 14.9% reduction of bloating compared to the Placebo group(P = 0.0194 and P = 0.0145, respectively). GI symptoms significantly decreased during supplementation but no statistical differences were reported between groups at the end of the supplementation period. Responder status was defined as a subject who experienced a decrease of 1 arbitrary unit(a.u.) or 50% of the abdominal discomfort score from baseline for at least 2 wk out of the last 4 wk of the study. A significant difference between groups was reported in the ITT population, when considering the first definition: subjects in the Active group had 1.510 higher odds to be a responder(reduction of 1 a.u. of abdominal pain/discomfort) compared with subjects in the Placebo group(P = 0.0240). At the end of supplementation period, stool consistency in the Active group of the ITT population was significantly improved and classified as 'normal' compared to Placebo(respectively 3.13 ± 1.197 a.u. vs 2.58 ± 1.020 a.u., P = 0.0003). 17Identifying the ‘unidentified’ fungi: a global-scale long-read third-generation sequencing approach显示文摘Molecular identification methods,in particular high-throughput sequencing tools,have greatly improved our knowledge about fungal diversity and biogeography,but many of the recovered taxa from natural environments cannot be identified to species or even higher taxonomic levels.This study addresses the phylogenetic placement of previously unrecognized fungal groups by using two complementary approaches:(i)third-generation amplicon sequencing analysis of DNA from global soil samples,screening out ITS reads of<90%similarity to other available Sanger sequences,and(ii)analysis of common fungal taxa that were previously indicated to be enigmatic in terms of taxonomic placement based on the ITS sequences alone(so-called top50 sequences).For the global soil samples,we chose to amplify the full rRNA gene operon using four partly overlapping amplicons and multiple newly developed primers or primer combinations that cover nearly all fungi and a vast majority of non-fungal eukaryotes.We extracted the rRNA 18S(SSU)and 28S(LSU)genes and performed phylogenetic analyses against carefully selected reference material.Both SSU and LSU analyses placed most soil sequences and top50 sequences to known orders and classes,but tens of monophyletic groups and single sequences remained outside described taxa.Furthermore,the LSU analyses recovered a few small groups of sequences that may potentially represent novel phyla.We conclude that rRNA genes-based phylogenetic analyses are efficient tools for determining phylogenetic relationships of fungal taxa that cannot be placed to any order or class using ITS sequences alone.However,in many instances,longer rRNA gene sequences and availability of both SSU and LSU reads are needed to improve taxonomic resolution.By leveraging third-generation sequencing from global soil samples,we successfully provided phylogenetic placement for many previously unidentified sequences and broadened our view on the fungal tree of life,with 10-20%new order-level taxa.In addition,the PacBio sequence data greatly extends fungal class-level information in reference databases.Leho Tedersoo Sten Anslan Mohammad Bahram Urmas Kõljalg Kessy Abarenkov 2020Fungal Diversity2020,,4:2

19Experimental Study of An Aquaculture Net Cage in Waves and Current显示文摘Model experiments of a floating fish cage subjected to waves and current have been performed. The objective was to study the dynamic behaviour of the fish cage model in waves and current. The fish cage model was composed of a model net, a flexible floating collar of the circular plastic type and a weight system. It was found that there are many wave periods in which cancellation of the wave-induced forces on the model occur. These cancellation wave periods are within the range of dimensioning wave periods commonly used for testing of fish farm structures and hence are important to be aware of. Large deformations of the net under realistic wave and current conditions were observed, where contact between the net and other parts of the structure were identified.
