| 2 | The HDAC3 enzymatic activity regulates skeletal muscle fuel metabolism显示文摘Histone deacetylase 3 (HDAC3) is a major HDAC, whose enzymatic activity is targeted by small molecule inhibitors for treating a variety of conditions. However, its enzymatic activity is largely dispensable for its function in embryonic development and hepatic lipid metabolism.HDAC3 plays a pivotal role in regulating muscle fuel metabolism and contractile function. Here, we address whether these muscular functions of HDAC3 require its enzymatic activity. By mutating the NCoR/SMRT corepressors in a knock - in mouse model named NS-DADm, we ablated the enzymatic activity of HDAC3 without affecting its protein levels. Compared to the control mice, skeletal muscles from NS-DADm mice showed lower force generation, enhanced fatigue resistance, enhanced fatty acid oxidation, reduced glucose uptake during exercise, upregulated expression of metabolic genes involved in branchedchain amino acids catabolism, and reduced muscle mass during aging, without changes in the muscle fiber-type composition or mitochondrial protein content. These muscular phenotypes are similar to those observed in the HDAC3-depleted skeletal muscles, which demonstrates that, unlike that in the liver or embryonic development, the metabolic function of HDAC3 in skeletal muscles requires its enzymatic activity. | 5 | Dual roles of trifluoroborate in nickel-catalyzed ethylene polymerization:Electronic perturbation and anchoring for heterogenization显示文摘Brookhart-typeα-diimine nickel and palladium catalysts have been extensively studied over the past several decades;however,the heterogenization of these metal complexes has received much less attention.In this contribution,we installed a trifluoroborate potassium substituent on anα-diimine framework.The ionic nature of trifluoroborate potassium endowed theα-diimine nickel complex with a strong affinity for the SiO_(2)support,while its electron-donating nature enhanced the catalyst stability and polyethylene molecular weight.In the presence of only 100 equiv.of Et2AlCl cocatalyst,the SiO_(2)-supported catalyst demonstrated significantly better performance than its homogeneous analog during ethylene polymerization,with extremely high activity(1.42–6.53×10^(7)g mol^(−1)h^(−1))and high thermal stability.The heterogeneous system led to the formation of high-molecular-weight polyethylenes(Mn 142,500–732,800 g/mol),narrow polydispersities(2.18–3.00),tunable branching densities(21–64 per 1000 carbon atoms),and great mechanical properties.Moreover,the efficient copolymerization of ethylene with comonomers such as methyl 10-undecenoate,6-chloro-1-hexene or 5-hexenylacetate was achieved.These superior properties enabled by the trifluoroborate potassium moiety may inspire its applications in other polymerization catalyst systems. | Changwen Hong Zihao Wang Hui Jiang Guifu Si Maoping Song Changle Chen | 2023 | Chinese Chemical Letters2023,34,8: | 0 |
| 6 | Pincer iridium(Ⅲ)-catalyzed enantioselective C(sp3)-H functionalization via carbenoid C–H insertion of 3-diazooxindoles with 1,4-cyclohexadiene显示文摘The asymmetric carbenoid C–H insertion of 3-diazooxindoles into 1,4-cyclohexadiene has been accomplished in the presence of chiral bis(imidazoline) NCN pincer iridium(Ⅲ) complexes as the catalysts. With a catalyst loading of 0.5 mol%, the reactions proceeded smoothly at 0℃ to afford a variety of chiral 3-substituted oxindoles in good yields with moderate to excellent enantioselectivities(up to 99% ee). The protocol exhibits good functional group tolerance with respect to 3-diazooxindoles and is readily scaled up to 2 mmol scale without any loss in activity and enantioselectivity.
