Evaluation of inflammatory activity in Crohn's disease and ulcerative colitis显示文摘Crohn's disease and ulcerative colitis evolve with a relapsing and remitting course.Determination of inflammatory state is crucial for the assessment of disease activity and for tailoring therapy. However,no simple diagnostic test for monitoring intestinal inflammation is available.Noninvasive markers give only indirect assessments of disease activity.Histopathological or endoscopical examinations accurately assess inflammatory activity, but they are invasive, time consuming and expensive and therefore are unsuitable for routine use.Imaging procedures are not applicable for ulcerative colitis. The usefulness of ultrasound and Doppler imag-ing in assessing disease activity is still a matter of discussion for Crohn's disease, and an increased interest in computed tomography enterograph (CTE) has been seen, mainly because it can delineate the extent and severity of bowel wall inflammation, besides detecting extraluminal findings.Until now,the available data concerning the accuracy of magnetic resonance enterography in detecting disease activity is less than CTE.Due to this, clinical activity indices are still commonly used for both diseases. | Eduardo Garcia Vilela Henrique Osvaldo da Gama Torres Fabiana Paiva Martins Maria de Lourdes de Abreu Ferrari Marcella Menezes Andrade Aloísio Sales da Cunha | 2012 | World Journal of Gastroenterology2012,18,9: | 23
Optimization of high-speed railway pantographs for improving pantograph-catenary contact显示文摘A crucial system for the operation of high-speed trains is the pantograph catenary interface as it is the sole responsible to deliver electrical power to the train. Being the catenary a stationary system with a long lifespan it is also less likely to be redesigned and upgraded than the pantographs that fit the train vehicles. This letter proposes an optimization procedure for the improvement of the contact quality between the pantograph and the catenary solely based on the redesign of the pantograph head suspension characteristics. A pantograph model is defined and validated against experimental dynamic characteristics of existing pantographs. An optimization strategy based on the use of a global optimization method, to find the vicinity of the optimal solution, followed by the use of a deterministic optimization algorithm, to fine tune the optimal solution, is applied here. The spring stiffness, damping characteristics and bow mass are the design variables used for the pantograph optimization. The objective of the optimal problem is the minimization of the standard deviation of the contact force history, which is the most important quantity to define the contact quality. The pantograph head suspension characteristics are allowed to vary within technological realistic limits. It is found that current high-speed railway pantographs have a limited potential for mechanical improvements, not exceeding 10%-15% on the decrease of the standard deviation of the contact force. | Jorge Ambrósio Joo Pombo Manuel Pereira | 2013 | Theoretical & Applied Mechanics Letters2013,3,1: | 15
Focus on emerging drugs for the treatment of patients with non-alcoholic fatty liver disease显示文摘Non-alcoholic fatty liver disease(NAFLD)has become the most common liver disorder in Western countries and is increasingly being recognized in developing nations.Fatty liver disease encompasses a spectrum of hepatic pathology, ranging from simple steatosis to non-alcoholic steatohepatitis, cirrhosis, hepatocellular carcinoma and end-stage liver disease. Moreover,NAFLD is often associated with other metabolic conditions, such as diabetes mellitus type 2,dyslipidemia and visceral obesity. The most recent guidelines suggest the management and treatment of patients with NAFLD considering both the liver disease and the associated metabolic co-morbidities.Diet and physical exercise are considered the first line of treatment for patients with NAFLD,but their results on therapeutic efficacy are often contrasting.Behavior therapy is necessary most of the time to achieve a sufficient result.Pharmacological therapy includes a wide variety of classes of molecules with different therapeutic targets and, often, little evidence supporting the real efficacy.Despite the abundance of clinical trials,NAFLD therapy remains a challenge for the scientific community, and there are no licensed therapies for NAFLD.Urgently, new pharmacological approaches are needed. Here,we will focus on the challenges facing actual therapeutic strategies and the most recent investigated molecules. | Alessandro Federico Claudio Zulli Ilario de Sio Anna Del Prete Marcello Dallio Mario Masarone Carmela Loguercio | 2014 | World Journal of Gastroenterology2014,20,45: | 10

Oral pharmacological treatment for patent ductus arteriosus in premature neonates with hemodynamic repercussions显示文摘Objective: To evaluate the efficacy of oral indomethacin, ibuprofen, and paracetamol in oral dosage form on patent ductus arteriosus(PDA) in premature neonates with significant clinical and hemodynamic repercussions(CHRs) and to determine the effect of these respective treatments on renal function. Methods: A retrospective study of cases of PDA in premature neonates in the Neonatal Intensive Care Unit was conducted. The treatments consisted of indomethacin[0.2 mg/(kg$d), 3-day cycle], ibuprofen [10 mg/(kg$d) followed by 5 mg/(kg$d), 3-day cycle], and paracetamol(15 mg/kg every 6 h, 5-day cycle). The drugs were administered as an oral solution. The following variables were considered: gestational age, newborn weight at birth, Apgar score, diuresis, serum creatinine and urea levels, and serum electrolyte levels(sodium and potassium). Results: Treatment with indomethacin presented efficacy of 87.5% in closure of the ductus with a mean outcome period of 3.5 d. In premature neonates with CHRs and contraindications for indomethacin, the initial treatment with either ibuprofen or paracetamol failed to close the ductus. However, when this treatment was followed by indomethacin, closure occurred in 66.7% of the neonates, with an outcome period of9.66 d. The initial treatment with one cycle of ibuprofen followed by one or two cycles of paracetamol failed to close the ductus. Conclusions: Oral indomethacin was effective for closure of the PDA in premature neonates with severe CHRs. Oral paracetamol or ibuprofen for PDA closure in premature neonates with severe CHRs and contraindications for indomethacin was ineffective. However, results in clinical improvements of neonates allowed the subsequent use of indomethacin and successful closure of the ductus. A significant reduction of diuresis occurred in neonates who were treated with indomethacin, either as a first-line treatment or after the failure of ibuprofen or paracetamol. | Clarissa de Albuquerque Botura Bruno Ambrósio da Rocha Thiely Balensiefer Franciele Queiroz Ames Ciomar Aparecida Bersani-Amado Roberto Kenji Nakamura Cuman | 2017 | Asian Pacific Journal of Tropical Medicine2017,10,11: | 4
Coexpression of CD163 and CD141 identifies human circulating IL-10-producing dendritic cells (DC-10)显示文摘Tolerogenic dendritic cells(DCs)are key players in maintaining immunological homeostasis, dampening immune responses, and promoting tolerance.DC-10,a tolerogenic population of human IL-10-producing DCs characterized by the expression of HLA-G and ILT4,play a pivotal role in promoting tolerance via T regulatory type 1(Tr1)cells.Thus far,the absence of markers that uniquely identify DC-10 has limited in vivo studies.By in vitro gene expression profiling of differentiated human DCs,we identified CD141 and CD163 as surface markers for DC-10.The coexpression of CD141 and CD163 in combination with CD14 and CD16 enables the ex vivo isolation of DC-10 from the peripheral blood.CD14+CD16+CD141+CD163+cells isolated from the peripheral blood of healthy subjects(ex vivo DC-10)produced spontaneously and upon activation of IL-10 and limited levels of IL-12.Moreover,in vitro stimulation of allogeneic naive CD4+T cells with ex vivo DC-10 induced the differentiation of alloantigen-specific CD49b+LAG-3+Tr1 cells. Finally,ex vivo DC-10 and in vitro generated DC-10 exhibited a similar transcriptional profile, which are characterized by an anti-inflammatory and pro-tolerogenic signature. These results provide new insights into the phenotype and molecular signature of DC-10 and highlight the tolerogenic properties of circulating DC-10.These findings open the opportunity to track DC-10 in vivo and to define their role in physiological and pathological settings. | Michela Comi Daniele Avancini Francesca Santoni de Sio Matteo Villa Molly Javier Uyeda Matteo Floris Daniela Tomasoni Alessandro Bulfone Maria Grazia Roncarolo Silvia Gregori | 2020 | Cellular & Molecular Immunology2020,17,1: | 4
Single-event-effect propagation investigation on nanoscale system on chip by applying heavy-ion microbeam and event tree analysis显示文摘The propagation of single-event effects(SEEs)on a Xilinx Zynq-7000 system on chip(SoC)was inves-tigated using heavy-ion microbeam radiation. The irradia-tion results reveal several functional blocks’sensitivity locations and cross sections, for instance, the arithmetic logic unit,register,D-cache,and peripheral, while irradi-ating the on-chip memory(OCM)region. Moreover, event tree analysis was executed based on the obtained microbeam irradiation results. This study quantitatively assesses the probabilities of SEE propagation from the OCM to other blocks in the SoC. | Wei-Tao Yang Xue-Cheng Du Yong-Hong Li Chao-Hui He Gang Guo Shu-Ting Shi Li Cai Sarah Azimi Corrado De Sio Luca Sterpone | 2021 | Nuclear Science and Techniques2021,32,10: | 3
Automatic Detection of COVID-19 Infection Using Chest X-Ray Images Through Transfer Learning显示文摘The new coronavirus(COVID-19),declared by the World Health Organization as a pandemic, has infected more than 1 million people and killed more than 50 thousand. An infection caused by COVID-19 can develop into pneumonia, which can be detected by a chest X-ray exam and should be treated appropriately. In this work,we propose an automatic detection method for COVID-19 infection based on chest X-ray images. The datasets constructed for this study are composed of194 X-ray images of patients diagnosed with coronavirus and 194 X-ray images of healthy patients.Since few images of patients with COVID-19 are publicly available,we apply the concept of transfer learning for this task.We use different architectures of convolutional neural networks(CNNs)trained on Image Net,and adapt them to behave as feature extractors for the X-ray images. Then, the CNNs are combined with consolidated machine learning methods, such as k-Nearest Neighbor,Bayes,Random Forest, multilayer perceptron(MLP),and support vector machine(SVM).The results show that, for one of the datasets, the extractor-classifier pair with the best performance is the Mobile Net architecture with the SVM classifier using a linear kernel, which achieves an accuracy and an F1-score of 98.5%.For the other dataset, the best pair is Dense Net201 with MLP,achieving an accuracy and an F1-score of 95.6%.Thus,the proposed approach demonstrates efficiency in detecting COVID-19 in X-ray images. | Elene Firmeza Ohata Gabriel Maia Bezerra João Victor Souza das Chagas Aloísio Vieira Lira Neto Adriano Bessa Albuquerque Victor Hugo Cde Albuquerque Pedro Pedrosa Rebouças Filho | 2021 | IEEE/CAA Journal of Automatica Sinica2021,8,1: | 3
Proton nuclear magnetic resonance-based metabonomic models for non-invasive diagnosis of liver fibrosis in chronic hepatitis C:Optimizing the classification of intermediate fibrosis显示文摘AIM To develop metabonomic models(MMs), using 1 H nuclear magnetic resonance(NMR) spectra of serum, to predict significant liver fibrosis(SF: Metavir ≥ F2), advanced liver fibrosis(AF: METAVIR ≥ F3) and cirrhosis(C: METAVIR = F4 or clinical cirrhosis) in chronic hepatitis C(CHC) patients. Additionally, to compare the accuracy of the MMs with the aspartate aminotransferase to platelet ratio index(APRI) and fibrosis index based on four factors(FIB-4). METHODS Sixty-nine patients who had undergone biopsy in the previous 12 mo or had clinical cirrhosis were included. The presence of any other liver disease was a criterion for exclusion. The MMs, constructed using partial least squares discriminant analysis and linear discriminant analysis formalisms, were tested by cross-validation, considering SF, AF and C. RESULTS Results showed that forty-two patients(61%) presented SF, 28(40%) AF and 18(26%) C. The MMs showed sensitivity and specificity of 97.6% and 92.6% to predict SF; 96.4% and 95.1% to predict AF; and 100% and 98.0% to predict C. Besides that, the MMs correctly classified all 27(39.7%) and 25(38.8%) patients with intermediate values of APRI and FIB-4, respectively. CONCLUSION The metabonomic strategy performed excellently in predicting significant and advanced liver fibrosis in CHC patients, including those in the gray zone of APRI and FIB-4, which may contribute to reducing the need for these patients to undergo liver biopsy. | Andrea Dória Batista Carlos Jonnatan Pimentel Barros Tássia Brena Barroso Carneiro Costa Michele Maria Goncalves de Godoy Ronaldo Dionísio Silva Joelma Carvalho Santos Mariana Montenegro de Melo Lira Norma ThoméJucá Edmundo Pessoa de Almeida Lopes Ricardo Oliveira Silva | 2018 | World Journal of Hepatology2018,10,1: | 2
Dietary approach in the treatment of nonalcoholic fatty liver disease显示文摘Nonalcoholic fatty liver disease(NAFLD) has been identified as one of the most prevalent chronic liver disease in adults and children populations. NAFLD is usually associated with the metabolic syndrome(MS), which is chiefly related to insulin resistance and its consequences. Insulin resistance has a crucial role in the pathogenesis of hepatic steatosis and potentially nonalcoholic steatohepatitis(NASH). Because of the contemporary epidemics of MS and obesity, the burden of NAFLD is also expected to rise. Unhealthy diets, such as the so-called western diet, are enriched in fructose, trans-fatty acids and saturated fat and seem to be associated with the development of NAFLD. In human studies, certain dietary sugars, particularly fructose, are used as a substrate for lipogenesis leading to hepatic fatty infiltration, inflammation, and possibly fibrosis. Other investigations have shown that fat consumption especially cholesterol and trans/saturated fatty acids are also steatogenic and seem to increase visceral adiposity. The identification of specific dietary components that favor the development of NASH could be important for the management of this disorder. This review focuses on the effects of different dietary approaches to prevent and treat NAFLD emphasizing the macronutrients and energy composition. | Silvia Marinho Ferolla Luciana Costa Silva Maria de Lourdes Abreu Ferrari Aloísio Sales da Cunha Flaviano dos Santos Martins Cláudia Alves Couto Teresa Cristina Abreu Ferrari | 2015 | World Journal of Hepatology2015,7,24: | 2

Autophagy, apoptosis and organelle features during cell exposure to cadmiumč显示文摘Cadmium(Cd)induces several effects in different tissues, but our knowledge of the toxic effects on organelles is insuffi cient. To observe the progression of Cd effects on organelle structure and function,HuH-7 cells(human hepatic carcinoma cell line)were exposed to CdCl2 in increasing concentrations(1μM–20μM)and exposure times(2 h–24 h).During Cd treatment, the cells exhibited a progressive decrease in viability that was both time-and dose-dependent.Cd treated cells displayed progressive morphological changes that included cytoplasm retraction and nuclear condensation preceding a total loss of cell adhesion.Treatment with 10μM for 12 h led to irreversible damages. Before these drastic and irreparable damages, treated cells(5μM for 12 h)presented a progressive loss of mitochondrial function and cytoplasm acidifi cation as well as dysfunction and disorganization of microfi laments and endoplasmic reticulum. These damages led to the induction of apoptotic events and an increase in autophagic bodies in the cytoplasm. These results revealed that Cd affects multiple intra-cellular targets that induce alterations in the mitochondria, cytoskeleton, endoplasmic reticulum and acidic compartments, ultimately culminating in cell death via apoptotic and autophagic pathways. | CRISTIANE DOS SANTOS VERGILIO EDÉSIO JOSÉTENÓRIO DE MELO | 2013 | BIOCELL2013,37,2: | 2
Globalization and long-run co-movements in the stock market for the G7: An application of VECM under structural breaks显示文摘This paper analyzes the process of long-run co-movements and stock market globalization on the basis of cointegration tests and vector error correction (VEC) models. The cointegration tests used here allow for structural breaks to be explicitly modeled and breakpoints to be computed on a relative-time basis. The data used in our empirical analysis were drawn from Datastream and comprise the natural logarithms of relative stock market indexes since 1973 for the G7 countries. The main results point to the conclusion that significant causal cointegration effects occur in this context and that there is a long-run relationship that governs the worldwide process of market integration. Globalization, however, is a complex adjustment process and in many cases there is only evidence of weak market integration which means that non-proportional price transmission occurs in the market along with proportional changes. The worldwide markets, as expected, appear to be driven in general by the US stock market. | MENEZES Rui DIONíSIO Andreia | 2011 | Chinese Science Bulletin2011,56,34: | 2
Imaging response predictors following drug eluting beads chemoembolization in the neoadjuvant liver transplant treatment of hepatocellular carcinoma显示文摘BACKGROUND Drug-eluting bead transarterial chemoembolization(DEB-TACE)is an endovascular treatment to release chemotherapeutic agents within a target lesion, minimizing systemic exposure and adverse effects to chemotherapeutics. Therefore, identifying which patient characteristics may predict imaging response to DEB-TACE can improve treatment results while selecting the best candidates.Predictors of the response after DEB-TACE still have not been fully elucidated. This is the first prospective study performed with standardized DEBTACE technique that aim to identify predictors of radiological response, assessing patients clinical and laboratory characteristics, diagnostic imaging and intraprocedure data of the hepatocellular carcinoma treated in the neoadjuvant context for liver transplantation.AIM To identify pre-and intraoperative clinical and imaging predictors of the radiological response of drug-eluting bead transarterial chemoembolization(DEB-TACE)for the neoadjuvant treatment of hepatocellular carcinoma(HCC).METHODS This is prospective, cohort study, performed in a single transplant center, from 2011 to 2014.Consecutive patients with HCC considered for liver transplant who underwent DEB-TACE in the first session for downstaging or bridging purposes were recruited.Pre and post-chemoembolization imaging studies were performed by computed tomography or magnetic resonance. The radiological response of each individual HCC was evaluated by objective response using mRECIST and the percentage of necrosis.RESULTS Two hundred patients with 380 HCCs were examined.Analysis of the objective response(nodule-based analysis)demonstrated that HCC with pseudocapsules had a 2.01 times greater chance of being responders than those without pseudocapsules(P=0.01),and the addition of every 1mg of chemoembolic agent increased the chance of therapeutic response in 4%(P<0.001).Analysis of the percentage of necrosis through multiple linear regression revealed that the addition of each 1mg of the chemoembolic agent caused an average increase of 0.65%(P<0.001)in necrosis in the treated lesion, whereas the hepatocellular carcinoma with pseudocapsules presented 18.27%(P<0.001)increased necrosis compared to those without pseudocapsules.CONCLUSION The presence of a pseudocapsule and the addition of the amount of chemoembolic agent increases the chance of an objective response in hepatocellular carcinoma and increases the percentage of tumor necrosis following drug-eluting bead chemoembolization in the neoadjuvant treatment, prior to liver transplantation. | Francisco Leonardo Galastri Felipe Nasser Breno Boueri Affonso Leonardo Guedes Moreira Valle Bruno Calazans Odísio Joaquim Mauricio Motta-Leal Filho Paolo Rogério Salvalaggio Rodrigo Gobbo Garcia Márcio Dias de Almeida Ronaldo Hueb Baroni Nelson Wolosker | 2020 | World Journal of Hepatology2020,12,1: | 2
Al_2O_3负载Cu,Fe或Ni掺杂氧化钼催化乙苯氧化脱氢(英文)显示文摘Molybdenum-based catalysts supported on Al_2O_3 doped with Ni, Cu, or Fe oxide were synthesized and used in ethylbenzene dehydrogenation to produce styrene. The molybdenum oxide was supported using an unconventional route that combined the polymeric precursor method(Pechini) and wet impregnation on commercial alumina. The samples were characterized by X-ray diffraction(XRD), N_2 adsorption-desorption isotherms, temperature-programmed reduction of H_2(H_2-TPR),and thermogravimetric(TG) analysis. XRD results showed that the added metals were well dispersed on the alumina support. The addition of the metal oxide(Ni, Cu, or Fe) of 2 wt% by wet impregnation did not affect the texture of the support. TPR results indicated a synergistic effect between the dopant and molybdenum oxide. The catalytic tests showed ethylbenzene conversion of 28%–53% and styrene selectivity of 94%–97%, indicating that the addition of the dopant improved the catalytic performance, which was related to the redox mechanism. Molybdenum oxides play a fundamental role in the oxidative dehydrogenation of ethylbenzene to styrene by its redox and acid–base properties. The sample containing Cu showed an atypical result with increasing conversion during the reaction, which was due to metal reduction. The Ni-containing solid exhibited the highest amount of carbon deposited, shown by TG analysis after the catalytic test, which explained its lower catalytic stability and selectivity. | Tiago Pinheiro Braga Ant?nio Narcísio Pinheiro Edson R.Leite Regina Cláudia R.dos Santos Antoninho Valentini | 2015 | 催化学报2015,36,5: | 2
